1Knockdown of survivin gene expression by RNAi induces apoptosis in human hepatocellular carcinoma cell line SMMC-7721显示文摘AIM: To investigate the survivin gene expression in human hepatocellular carcinoma cell line SMMC-7721 and the effects of survivin gene RNA interference (RNAi) on cell apoptosis and biological behaviors of SMMC-7721 cells.METHODS: Eukaryotic expression vector of survivin gene RNAi and recombinant plasmid pSuppressorNeo-survivin (pSuNeo-SW), were constructed by ligating into the vector,pSupperssorNeo (pSuNeo) digested with restriction enzymes Xba I and Sa/I and the designed double-chain RNAi primers. A cell model of SMMC-7721 after treatment with RNAi was prepared by transfecting SMMC-7721 cells with the lipofectin transfection method. Strept-avidinbiotin-complex (SABC) immunohistochemical staining and RT-PCR were used to detect survivin gene expressions in SMMC-7721 cells. Flow cytometry was used for the cell cycle analysis. Transmission electron microscopy was performed to determine whether RNAi induced cell apoptosis, and the method of measuring the cell growth curve was utilized to study the growth of SMMC-7721 cells before and after treatment with RNAi.RESULTS: The eukaryotic expression vector of survivin gene RNAi and pSuNeo-SW, were constructed successfully. The expression level of survivin gene in SMMC-7721 cells was observed. After the treatment of RNAi, the expression of survivin gene in SMMC-7721 cells was almost absent,apoptosis index was increased by 15.6%, and the number of cells was decreased in G2/M phase and the cell growth was inhibited.CONCLUSION: RNAi can exert a knockdown of survivin gene expression in SMMC-7721 cells, and induce apoptosis and inhibit the growth of carcinoma cells.Sheng-QuanCheng Wen-LiangWang WeiYan Qing-LongLi LiWang Wen-YongWang 2005World Journal of Gastroenterology2005,11,5:42
2Pancreatic cancer mortality in China(1991-2000)显示文摘AIM: To describe the mortality rate of pancreatic cancer and its distribution in China during the period of 1991-2000.METHODS: Based on the data of demography and death collected through China′s Disease Surveillance Point System (DSPS) over the period of 1991-2000, the distribution of death rate of pancreatic cancer was described in terms of age group, gender, calendar year, rural/urban residence and administrative district.RESULTS: A total of 1 619 death cases attributed to pancreatic cancer (975 men and 644 women) were reported by DSPS during 1991-2000. The reported, adjusted and agestandardized mortality rates increased from 1.46, 1.75, and 2.18 per 100 000 populations in 1991 to 2.38, 3.06, and 3.26per 100 000 populations in 2000. The majority (69.62 %) of the deaths of pancreatic cancer were seen in the age group of 60 years and older. The mortality rate was higher in men than in women, but the male to female death rate ratios decreased during the 10 years. Our data also showed that the death rate of pancreatic cancer in urban areas was about 2-4 fold higher than that in rural areas, and in Northeast and East China, the death rates were higher than those in the other 5 administrative districts.CONCLUSION: The death rate due to pancreatic cancer was rising during the period of 1991-2000 and the peak mortality of pancreatic cancer might arrive in China.LiWang Gong-HuanYang Xing-HuaLu Zheng-JingHuang HuiLi 2003World Journal of Gastroenterology2003,9,8:38
3Ginkgo biloba extract reverses CCI4-induced liver fibrosis in rats显示文摘AIM: To study the reversing effect of Ginkgo biloba extract (GbE) on established liver fibrosis in rats. METHODS: Following confirmation of CCI4-induced liver fibrosis, GbE or saline was administrated to the rats for 4 weeks. The remaining rats received neither CCI 4 norGbE as normal control. The four groups were compared in terms of serum enzymes, tissue damage, expression of αSMA and tissue inhibitor-1 of metalloproteinase (TIMP-1) and metalloproteinase-1 (MMP-1). RESULTS: Compared with saline-treated group, liver fibrosis rats treated with GbE had decreased serum total bilirubin (P<0.01) and aminotransferase levels (P<0.01) and increased levels of serum albumin (P<0.01). Microscopic studies revealed that the livers of rats receiving GbE showed allieviation in fibrosis (P<0.05) as well as expression of αSMA (P<0.01). The liver collagen and reticulum contents were lower in rats treated with GbE than saline-treated group (P<0.01). RT-PCR revealed that the level of TIMP-1 decreased while the level of MMP-1 increased in GbE group. CONCLUSION: Administration of GbE improved CCI4-induced liver fibrosis. 5Genetic regulation by non-coding RNAs显示文摘Large scale cDNA sequencing and genome tiling array studies have shown that around 50% of genomic DNA in humans is transcribed, of which 2% is translated into proteins and the remaining 98% is non-coding RNAs (ncRNAs). There is mounting evidence that these ncRNAs play critical roles in regulating DNA structure, RNA expression, protein translation and protein functions through multiple genetic mechanisms, and thus affect normal development of organisms at all levels. Today, we know very little about the regulatory mechanisms and functions of these ncRNAs, which is clearly essential knowledge for understanding the secret of life. 8Experiment research on grain drying process in the heat pump assisted fluidized beds显示文摘A heat pump assisted fluidized bed grain drying experimental system was developed. Based on this system, a serial of ex- periments was performed under four kinds of air cycle conditions. According to the experimental analysis, an appropriate drying medium-air cycle for the heat pump assisted fluidized bed drying equipment was decided, which is different from the commonly used heat pump assisted drying system. The experimental results concerning the drying operation performance of the new system show that the averaged coefficient of performance (COP) can reach more than 2.5. 10Population pharmacokinetic model of valproate and prediction of valproate serum concentrations in children with epilepsy显示文摘AIM: Using sparse data of valproate (VPA) serum concentrations to build a population pharmacokinetic (PPK) model of VPA in Chinese children with epilepsy and to predict serum concentrations for new patients using a Bayesian approach. METHODS: Two hundred epileptic children, whose VPA serum concentrations were collected, were divided randomly into two groups (A and B, n=100 each). The PPK parameter values of group A were calculated to establish a PPK Model by using the NPEM Program of USC*PACK software. Based on it, VPA serum concentrations of group B were predicted with the Bayesian Fitting Program of the USC*PACK software. To assess the accuracy and precision of prediction, a paired-comparisons t-test was run between predicted and ob- served concentrations, and then the mean prediction error (MPE), mean square prediction error (MSPE), root mean square prediction error (RMSPE), and coincidence rates for different percentages of prediction error were all calculated. RESULTS: Optimum PPK parameters were: Ka, 2.522±2.743 h-1; Vs, 0.329±0.496 L/kg; and Kel, 0.0438±0.0384 h-1. For group B, there was no significant difference between predicted and observed concentrations. MPE was -0.43 mg/L, MSPE was 115.40 (mg/L)2, and RMSPE was 5.47 mg/L. The coincidence rates for percent- ages of prediction error, which were less than 5 %, 10 %, 15 %, 20 %, 25 %, and 30 %, were 62 %, 74 %, 82 %, 85 %, 89 %, and 93 %, respectively. CONCLUSION: A PPK model of VPA in epileptic children was successfully established. 11Effects of extracellular iron concentration on calcium absorption and relationship between Ca^(2+) and cell apoptosis in Caco-2 cells显示文摘AIM: To determine the method of growing small intestinal epithelial cells in short-term primary culture and to investigate the effect of extracellular iron concentration ([Fe3+]) on calcium absorption and the relationship between the rising intracellular calcium concentration ([Ca2+]i) and cell apoptosis in human intestinal epithelial Caco-2 cells. METHODS: Primary culture was used for growing small intestinal epithelial cells. [Ca2+]i was detected by a confocal laser scanning microscope. The changes in [Ca2+]i were represented by fluorescence intensity (FI). The apoptosis was evaluated by flow cytometry.RESULTS: Isolation of epithelial cells and preservation of its three-dimensional integrity were achieved using the digestion technique of a mixture of collagenase Ⅺ and dispase Ⅰ. Purification of the epithelial cells was facilitated by using a simple differential sedimentation method. The results showed that proliferation of normal gut epithelium in vitro was initially dependent upon the maintenance of structural integrity of the tissue. If 0.25% trypsin was used for digestion, the cells were severely damaged and very difficult to stick to the Petri dish for growing. The Fe3+ chelating agent desferrioxamine (100, 200 and 300 μmol/L) increased the FI of Caco-2 cells from 27.50±13.18 (control,n = 150) to 35.71±13.99 (n = 150, P<0.01), 72.19±35.40 (n = 150, P<0.01) and 211.34±29.03 (n = 150, P<0.01) in a concentration-dependent manner. There was a significant decrease in the FI of Caco-2 cells treated by ferric ammonium citrate (FAC, a Fe3+ donor; 10, 50 and 100 μmol/L). The FIvalue of Caco-2 cells treated by FAC was 185.85±33.77 (n = 150, P<0.01), 122.73±58.47 (n = 150, P<0.01), and 53.29±19.82 (n = 150, P<0.01), respectively, suggesting that calcium absorption was influenced by [Fe3+]. Calcium ionophore A23187 (0.1, 1.0 and 10 μmol/L) increased the FI of Caco-2 cells from 40.45±13.95 (control, n = 150) to 45.19±21.95 (n = 150, P<0.01), 89.87±43.29 (n = 150,P<0.01) and 104.64±51.07 (n = 150, P<0.01) in a concentration-dependent manner. The positive apoptotic cell number of the Caco-2 cells after being treated with indicating that the increase in the positive apoptotic cell~ increased from 0.32% to 0.69%, 0.90% and 1.10%, number was positively correlated with [Ca2+]i.CONCLUSION: Ca2+ absorbability is increased with the decrease of extracellular iron concentration Fe3+ and hindered with the increase of Fe3+ consistence out of them. 13A genome-wide association study uncovers a critical role of the RsPAP2 gene in red-skinned Raphanus sativus L.显示文摘Radish(Raphanus sativus L.)taproot contains high concentrations of flavonoids,including anthocyanins(ATCs),in redskinned genotypes.However,little information on the genetic regulation of ATC biosynthesis in radish is available.A genome-wide association study of radish red skin color was conducted using whole-genome sequencing data derived from 179 radish genotypes.The R2R3-MYB transcription factor production of anthocyanin pigment 2(PAP2)gene was found in the region associated with a leading SNP located on chromosome 2.The amino acid sequence encoded by the RsPAP2 gene was different from those of the other published RsMYB genes responsible for the red skin color of radish.The overexpression of the RsPAP2 gene resulted in ATC accumulation in Arabidopsis and radish,which was accompanied by the upregulation of several ATC-related structural genes.RsPAP2 was found to bind the RsUFGT and RsTT8 promoters,as shown by a dual-luciferase reporter system and a yeast one-hybrid assay.The promoter activities of the RsANS,RsCHI,RsPAL,and RsUFGT genes could be strongly activated by coinfiltration with RsPAP2 and RsTT8.These findings showed the effectiveness of GWAS in identifying candidate genes in radish and demonstrated that RsPAP2 could(either directly or together with its cofactor RsTT8)regulate the transcript levels of ATC-related genes to promote ATC biosynthesis,facilitating the genetic enhancement of ATC contents and other related traits in radish.Lianxue Fan YanWang Liang Xu Mingjia Tang Xiaoli Zhang Jiali Ying Cui Li Junhui Dong Liwang Liu 2020Horticulture Research2020,7,1:2

15Effect of ginkgo biloba extract on livers in aged rats显示文摘AIM: To investigate the protective effect of ginkgo biloba extract (GBE) on livers of aged rats and the associated mechanisms.METHODS: Two-mo- and 20-mo-old rats were treated with GBE/saline for 3 mo. Liver tissue samples from 5-moold rats treated with saline (group Y) and 23-mo-old rats treated with GBE (group E) or saline (group N) were used for histopathological examinations (hematoxylin-eosin and Masson staining, Lipofuscin staining-Schmorl staining) and determination of expression of tissue inhibitor-1 of metalloproteinase (TIMP-1) and the level of malondialdehyde (MDA), glutathione peroxidase (GPx) and superoxide dismutase (SOD). Blood samples were collected for determination of alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL) and albumin.RESULTS: Microscopic studies with Masson staining revealed mild liver fibrosis in aged rats (group N), while the livers of aged rats receiving GBE (group E) showed amelioration in fibrosis (2.2±0.1 vs 2.8±0.1, P<0.01) and deposition of lipofuscin (33.7±5.3 vs 62.8±5.7, P<0.01).The expression of TIMP-1 and the level of liver MDA (1.0±0.1 vs 1.2±0.2, P<0.05) also decreased but the activity of GPx (97.1±15.3 vs 61.8±14.5, P<0.01) increased in group E. Compared with group Y, the level of liver MDA (0.8±0.1 vs 1.2±0.2, P<0.01), lipofuscin (32.4±6.0 vs 62.8±5.7, P<0.01) and TIMP-1 expression were increased,while the activity of GPx (103.2±17.6 vs61.8±14.5, P<0.01)and SOD (16.7±4.4 vs 11.8±3.9, P<0.05) was decreased in group N. 16A state-of-the-art review of mechanical characteristics and cracking processes of pre-cracked rocks under quasi-static compression显示文摘The mechanical characteristics and failure behavior of rocks containing flaws or discontinuities have received wide attention in the field of rock mechanics.When external loads are applied to rock materials,stress-induced cracks would initiate and propagate from the flaws,ultimately leading to the irreversible failure of rocks.To investigate the cracking behavior and the effect of flaw geometries on the mechanical properties of rock materials,a series of samples containing one,two and multiple flaws have been widely investigated in the laboratory.In this paper,the experimental results for pre-cracked rocks under quasistatic compression were systematically reviewed.The progressive failure process of intact rocks is briefly described to reveal the background for experiments on samples with flaws.Then,the nondestructive measurement techniques utilized in experiments,such as acoustic emission(AE),X-ray computed tomography(CT),and digital image correlation(DIC),are summarized.The mechanical characteristics of rocks with different flaw geometries and under different loading conditions,including the geometry of pre-existing flaws,flaw filling condition and confining pressure,are discussed.Furthermore,the cracking process is evaluated from the perspective of crack initiation,coalescence,and failure patterns.Liwang Liu Haibo Li Xiaofeng Li 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,6:2

18Modal Analysis of Thread off Failure of Oil Round Thread Casing Connection显示文摘The dynamic modal model analysis of the oil casing connection is done by finite element method (FEM), and the first nature frequency, the second nature frequency, the vibration mode shape and the time domain of the node displacement and the element stress variation in the thread connection are gotten. The Von.Mises stresses of the connection in the static and dynamic state are also studied.
