1PARylation regulates stress granule dynamics, phase separation, and neurotoxicity of disease-related RNA-binding proteins显示文摘Mutations in RNA-binding proteins (RBPs) localized in ribonucleoprotein (RNP) granules, such as hnRNP A1 and TDP-43, promote aberrant protein aggregation, which is a pathological hallmark of various neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Protein posttranslational modifications (PTMs) are known to 佗gulate RNP granules. In this study, we investigate the function of poly(ADP-ribosyl)ation (PARylation), an important PTM involved in DNA damage repair and cell death, in RNP granule-related neurodegeneration. We reveal that PARylation levels are a major regulator of the assembly-disassembly dynamics of RNP granules containing disease-related RBPs, hnRNP A1 and TDP-43. We find that hnRNP A1 can both be PARylated and bind to PARylated proteins or poly(ADP-ribose)(PAR). We further uncover that PARylation of hnRNP A1 at K298 controls its nucleocytoplasmic transport, whereas PAR-binding via the PAR-binding motif (PBM) of hnRNP Al regulates its association with stress granules. Moreover, we reveal that PAR not only dramatically enhances the liquid-liquid phase separation of hnRNP A1, but also promotes the co-phase separation of hnRNP A1 and TDP-43 in vitro and their interaction in vivo. Finally, both genetic and pharmacological inhibition of PARP mitigates hnRNP Al- and TDP-43-mediated neurotoxicity in cell and Drosophila models of ALS. 3Long-term adenosine A1 receptor activation-induced sortilin expression promotes α-synuclein upregulation in dopaminergic neurons显示文摘Prolonged activation of adenosine A1 receptor likely leads to damage of dopaminergic neurons and subsequent development of neurodegenerative diseases.However,the pathogenesis underlying long-term adenosine A1 receptor activation-induced neurodegeneration remains unclear.In this study,rats were intraperitoneally injected with 5 mg/kg of the adenosine A1 receptor agonist N6-cyclopentyladenosine(CPA)for five weeks.The mobility of rats was evaluated by forced swimming test,while their cognitive capabilities were evaluated by Y-maze test.Expression of sortilin,α-synuclein,p-JUN,and c-JUN proteins in the substantia nigra were detected by western blot analysis.In addition,immunofluorescence staining of sortilin andα-synuclein was performed to detect expression in the substantia nigra.The results showed that,compared with adenosine A1 receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine(5 mg/kg)+CPA co-treated rats,motor and memory abilities were reduced,surface expression of sortin andα-synuclein in dopaminergic neurons was reduced,and total sortilin and totalα-synuclein were increased in CPA-treated rats.MN9D cells were incubated with 500 nM CPA alone or in combination with 10μM SP600125(JNK inhibitor)for 48 hours.Quantitative real-time polymerase chain reaction analysis of sortilin andα-synuclein mRNA levels in MN9D cells revealed upregulated sortilin expression in MN9D cells cultured with CPA alone,but the combination of CPA and SP600125 could inhibit this expression.Predictions made using Jasper,PROMO,and Alibaba online databases identified a highly conserved sequence in the sortilin promoter that was predicted to bind JUN in both humans and rodents.A luciferase reporter assay of sortilin promoter plasmid-transfected HEK293T cells confirmed this prediction.After sortilin expression was inhibited by sh-SORT1,expression of p-JUN and c-JUN was detected by western blot analysis.Long-term adenosine A1 receptor activation levels upregulatedα-synuclein expression at the post-transcriptional level by affecting sortilin expression.The online tool Raptor-X-Binding and Discovery Studio 4.5 prediction software predicted that sortilin can bind toα-synuclein.Co-immunoprecipitation revealed an interaction between sortilin andα-synuclein in MN9D cells.Our findings indicate that suppression of prolonged adenosine A1 receptor activation potently inhibited sortilin expression andα-synuclein accumulation,and dramatically improved host cognition and kineticism.This study was approved by the University Committee of Animal Care and Supply at the University of Saskatchewan(approval No.AUP#20070090)in March 2007 and the Animals Ethics Committee of University of South China(approval No.LL0387-USC)in June 2017.Yun-Cheng Lv An-Bo Gao Jing Yang Li-Yuan Zhong Bo Jia Shu-Hui Ouyang Le Gui Tian-Hong Peng Sha Sun Francisco S.Cayabyab 2020Neural Regeneration Research2020,15,4:4
4In situ synthesis and hardness of TiC/Ti_5Si_3 composites on Ti-5Al-2.5Sn substrates by gas tungsten arc welding显示文摘TiC/Ti5Si3 composites were fabricated on Ti-5Al-2.5Sn substrates by gas tungsten arc welding (GTAW). Identification of the phases was performed using X-ray diffraction (XRD). The microstructures were analyzed using scanning electron microscopy (SEM) combined with energy-dispersive X-ray spectrometry (EDS) and optical microscopy (OM). The Vickers hardness was measured with a micro-hardness tester. 8Study on instability fracture and simulation of surrounding rock induced by fault activation under mining influence显示文摘The fault is potentially vulnerability's geological structure in the working face and its vicinity,and it is also a crucial geological factor affecting coal mine safety exploitation.To investigate the unstable failure of surrounding rock induced by fault activation under the influence of adoption,which was studied utilizing field case and numerical analysis for the deformation and failure process of surrounding rock near the fault-affected zone.Combined with field cases,this paper analyzes disturbance stress and roof abscission layer monitoring in effecting zones of fault activation.Using the discrete element 3DEC numerical analysis method,the model of surrounding rock unstable fracture induced by fault activation under adoption is established.The unstable fracture and stress variation characteristics of surrounding rock induced by fault activation during the excavation of the upper side wall and lower side wall of the faults are simulated and analyzed.Field analysis shows that as the coal working face continues to advance,the mining stress gradually increases.There is a zigzag wave on the relationship curve between coal mining and roof displacement near the fault,which reveals that the surrounding rock of the fault activation affected zone is in the superposition state of static load and dynamic load.Furthermore,the simulation results show that the stress and displacement of surrounding rock near the fault increase with the advance of coal mining face.The closer to the fault plane,the displacement gradually returns to zero,and the stress is also in a lower state.Jie Chen Kaiwen Shi Yuanyuan Pu Derek B.Apel Chuanjiu Zhang Yujun Zuo Jiongkun Chen Le Xu Zhen Gui Longfei Song 2023Rock Mechanics Bulletin2023,2,2:1

11Study on the green extraction of corncob xylan by deep eutectic solvent显示文摘Corn as one of the world's major food crops,its by-product corn cob is also rich in resources.However,the unreasonable utilization of corn cob often causes the environmental pollution,waste of resources and other problems.As one of the most abundant polymers in nature,xylan is widely used in food,medicine,materials and other fields.Corn cob is rich in xylan,which is an ideal raw material for extracting xylan.However,the intractable lignin is covalently linked to xylan,which increases the difficulty of xylan extraction.It has been reported that the deep eutectic solvent(DES)could preferentially dissolve lignin in biomass,thereby dissolving the xylan.Then,the xylan in the extract was separated by ethanol precipitation method.The xylan precipitate was obtained after centrifugation,while the supernatant was retained.The components of the supernatant after ethanol precipitation were separated by the rotary evaporator.The ethanol,water and DES were collected for the subsequent extraction of corn cob xylan.In this study,a novel way was provided for the green production of corn cob xylan.The DES was used to extract xylan from corn cob which was used as the raw material.The effects of solid-liquid ratio,reaction time,reaction temperature and water content of DES on the extraction rate of corn cob xylan were investigated by the single factor test.Furthermore,the orthogonal test was designed to optimize the xylan extraction process.The structure of corn cob xylan was analyzed and verified.The results showed that the optimum extraction conditions of corn cob xylan were as follows:the ratio of corn cob to DES was 1:15(g:mL),the extraction time was 3 h,the extraction temperature was 60℃,and the water content of DES was 70%.Under these conditions,the extraction rate of xylan was 16.46%.The extracted corn cob xylan was distinctive triple helix of polysaccharide,which was similar to the structure of commercially available xylan.Xylan was effectively and workably extracted from corn cob by the DES method.This study provided a new approach for high value conversion of corn cob and the clean production of xylan.Bingyu Jiao Le Wang Haitao Gui Zifu Ni Rong Du Yuansen Hu 2024Grain & Oil Science and Technology2024,7,1:0
12Recombinant human thrombopoietin prior to mobilization chemotherapy facilitates platelet recovery in autologous transplantation in patients with lymphoma:Results of a prospective randomized study显示文摘Background:Chemotherapy plus granulocyte colony-stimulating factor (GCSF) regimen is one of the available approaches to mobilize peripheral blood progenitor cells (PBPCs).It causes thrombocytopenia and delays leukapheresis.This study aimed to evaluate the role of recombinant human thrombopoietin (rhTPO) before mobilization chemotherapy in facilitating leukapheresis in patients with lymphoma.Methods:In this randomized open-label phase 2 trial, patients were randomly assigned in a 1:2 ratio to receive mobilization with rhTPO plus GCSF in combination with chemotherapy (the rhTPO plus GCSF arm) or GCSF alone in combination with chemotherapy (the GCSF alone arm).The recovery of neutrophils and platelets and the amount of platelet transfusion were monitored.Results:Thirty patients were enrolled in this study between March 2016 and August 2018. Patients in the rhTPO plus GCSF arm (n = 10) had similar platelet nadir after mobilization chemotherapy (P=0.878) and similar amount of platelet transfusion (median 0 vs.1 unit,P=0.735) when compared with the GCSF alone arm (n = 20). On the day of leukapheresis, the median platelet count was 86 ×10^(9)/L (range 18-219) among patients who received rhTPO and 73 ×10^(9)/L (range 42-197) among those who received GCSF alone (P=0.982). 13Sulfation modification of dopamine in brain regulates aggregative behavior of animals显示文摘Behavioral plasticity and the underlying neuronal plasticity represent a fundamental capacity of animals to cope with environmental stimuli.Behavioral plasticity is controlled by complex molecular networks that act under different layers of regulation.While various molecules have been found to be involved in the regulation of plastic behaviors across species,less is known about how organisms orchestrate the activity of these molecules as part of a coherent behavioral response to varying environments.Here we discover a me chanism for the regulation of animal behavioral plasticity involving molecular sulfation in the brain,a modification of substrate molecules by sulfo transferase(ST)-catalyzed addition of a sulfonate group(SO_(3))from an obligate donor,3'-phosphoadenosine 5'-phosphosulfate(PAPS)to the substrates.We investigated aggregation behaviors of migratory locusts,which are well-known for extreme phase change plasticity triggered by population density.The processes of PAPS biosynthesis acted efficiently on induction of locust behavioral transition:Inhibition of PAPS synthesis solicited a behavioral shift from gregarious to solitarious states;external PAPS dosage,by contrast,promoted aggregation in solitarious locusts.Genetic or pharmacological intervention in the sulfation catalyzation resulted into pronounced solitarizing effects.Analysis of substrate-specific STs suggests a widespread involvement of sulfated neurotransmitters in the behavioral response.D opamine in the brain was finally identified to be actively sulfate conjugate d,and the sulfate conjugation enhanced the free DA-mediated behavioral aggregation.Similar results in Caenorhabditis elegans and mice indicate that sulfation may be involved more broadly in the modulation of animal aggregation.These findings reveal a general me chanism that effectively regulates animal social-like behavioral plasticity,possibly through sulfation-mediated modification of neural networks.Bing Chen Xiwen Tong Xia Zhang Wanying Gui Guoming Ai Lihua Huang Ding Ding Jiangxu Zhang Le Kang 2022National Science Review2022,9,4:0

15In situ growth of large-area and self-aligned graphene nanoribbon arrays on liquid metal显示文摘Intrinsic graphene features semi-metallic characteristics that limit its applications in electronic devices,whereas graphene nanoribbons(GNRs) are promising semiconductors because of their bandgap-opening feature. However, the controllable mass-fabrication of high-quality GNR arrays remains a major challenge.In particular, the in situ growth of GNR arrays through template-free chemical vapor deposition(CVD) has not been realized. Herein, we report a template-free CVD strategy to grow large-area, high-quality and self-aligned GNR arrays on liquid copper surface. The width of as-grown GNR could be optimized to sub-10 nm with aspect ratio up to 387, which is higher than those of reported CVD-GNRs. The study of the growth mechanism indicates that a unique comb-like etching-regulated growth process caused by a trace hydrogen flow guides the formation of the mass-produced self-aligned GNR arrays. 18Standardized quantitative measurements of wrist cartilage in healthy humans using 3T magnetic resonance imaging显示文摘AIM: To quantify the wrist cartilage cross-sectional area in humans from a 3D magnetic resonance imaging(MRI) dataset and to assess the corresponding reproducibility. METHODS: The study was conducted in 14 healthy volunteers(6 females and 8 males) between 30 and 58 years old and devoid of articular pain. Subjectswere asked to lie down in the supine position with the right hand positioned above the pelvic region on top of a home-built rigid platform attached to the scanner bed. The wrist was wrapped with a flexible surface coil. MRI investigations were performed at 3T(Verio-Siemens) using volume interpolated breath hold examination(VIBE) and dual echo steady state(DESS) MRI sequences. Cartilage cross sectional area(CSA) was measured on a slice of interest selected from a 3D dataset of the entire carpus and metacarpalphalangeal areas on the basis of anatomical criteria using conventional image processing radiology software. Cartilage cross-sectional areas between opposite bones in the carpal region were manually selected and quantified using a thresholding method.RESULTS: Cartilage CSA measurements performed on a selected predefined slice were 292.4 ± 39 mm2 using the VIBE sequence and slightly lower, 270.4 ± 50.6 mm2, with the DESS sequence. The inter(14.1%) and intra(2.4%) subject variability was similar for both MRI methods. The coefficients of variation computed for the repeated measurements were also comparable for the VIBE(2.4%) and the DESS(4.8%) sequences. The carpus length averaged over the group was 37.5 ± 2.8 mm with a 7.45% between-subjects coefficient of variation. Of note, wrist cartilage CSA measured with either the VIBE or the DESS sequences was linearly related to the carpal bone length.
