1Environmental application of three-dimensional graphene materials as adsorbents for dyes and heavy metals: Review on ice-templating method and adsorption mechanisms显示文摘The remediation of wastewater requires treatment technologies which are robust, efficient, simple to operate and affordable such as adsorption. Lately, three-dimensional(3D)graphene based materials have attracted significant attention as effective adsorbents for wastewater treatment. The intrinsic properties of 3D graphene structure such as large surface area and interconnected porous structure can facilitate the transport of pollutants into the 3D network and provide abundant active sites for trapping the pollutants. For the synthesis of 3D graphene structure, ice-templating is commonly practiced due to its facile steps, cost effectiveness and high scalability potential. This review covers the icetemplating fabrication technique for 3D graphene based materials and their application as adsorbents in eliminating dyes and heavy metals from aqueous media. The assembly mechanisms of the ice-templating fsynthesis are comprehensively discussed. Further discussion on the fundamental principles, critical process parameters and characteristics of ice-templated 3D graphene structures is also included. A thorough review on the mechanisms for batch adsorption of dyes and heavy metals is presented based on the structures and properties of the 3D graphene materials. The review further evaluates the dynamic adsorption in packed columns and the regeneration of 3D graphene based materials.Kar Chiew Lai Lai Yee Lee Billie Yan Zhang Hiew Suchithra Thangalazhy-Gopakumar Suyin Gan 2019Journal of Environmental Sciences2019,31,5:13
2Short-term entecavir versus lamivudine therapy for HBeAg-negative patients with acute-on-chronic hepatitis B liver failure显示文摘BACKGROUND: Selection of drugs for antiviral therapy of patients with hepatitis B virus (HBV)-related acute-on-chronic liver failure (ACLF) remains difficult. This study was undertaken to evaluate the short-term efficacy of entecavir versus lamivudine on hepatitis B e antigen (HBeAg)-negative patients with ACLF. METHODS: The data of 182 HBeAg-negative patients with ACLF were retrospectively collected from patient profiles of the hospital. In these patients, 93 HBeAg-negative patients with ACLF were treated orally with 0.5 mg of entecavir and 89 were treated orally with 100 mg of lamivudine every day. The gender and age were matched between the two groups. Biochemical items, the model for end-stage liver disease (MELD) score, and HBV DNA level were matched at baseline between the two groups and monitored during treatment. The 3-month mortalities of the two groups were compared. RESULTS: No significant differences were found in biochemical items, MELD score, and HBV DNA level at baseline (P>0.05). HBV DNA level decreased within 3 months in both groups (P<0.05), regardless of the pretreatment MELD score. In patients with the same range of pretreatment MELD scores, treatment duration, posttreatment HBV DNA levels, percentage of HBV DNA level <2.7 lg copies/mL, biochemical items, MELD scores and 3-month mortality were similar in the two groups (all P>0.05). Pretreatment MELD score was not related to posttreatment HBV DNA levels (P>0.05), but related to a 3-month mortality in both groups (both P<0.001).CONCLUSIONS: In HBeAg-negative patients with ACLF, the short-term efficacy of entecavir versus lamivudine was similar. The degree of pretreatment liver failure significantly affected the outcome of treatment.Jing Lai Ying Yan Li Mai Yu-Bao Zheng Wei-Qiang Gan Wei-Min Ke 2013Hepatobiliary & Pancreatic Diseases International2013,12,2:12
3Modeling and simulation of urea-water-solution droplet evaporation and thermolysis processes for SCR systems显示文摘A reliable mathematical model of urea-water-solution(UWS) droplet evaporation and thermolysis is developed. The well known Abramzon–Sirignano evaporation model is corrected by introducing an adjustment coefficient considering the different evaporation behaviors of UWS droplet at different ambient temperatures. A semidetailed kinetic scheme of urea thermolysis is developed based on Ebrahimian's work. Sequentially, the evaporation characteristics, decomposition efficiency of a single UWS droplet and deposit formation are simulated. As a result, the relation of evaporation time, relative velocity, exhaust temperature and droplet initial diameter is presented. Synchronously, it indicates that temperature is the decisive factor for urea thermolysis. Different temperatures result in different deposit components, and deposit yield is significantly influenced by temperature and decomposition time. The current work can provide guidance for designing urea injection strategy of SCR systems.Xubo Gan Dongwei Yao Feng Wu Jiawei Dai Lai Wei Xingwen Li 2016Chinese Journal of Chemical Engineering2016,24,8:7
4Nonequilibrium and morphological characterizations of Kelvin-Helmholtz instability in compressible flows显示文摘We investigate the effects of viscosity and heat conduction on the onset and growth of Kelvin-Helmholtz instability (KHI) via an efficient discrete Boltzmann model.Technically, two effective approaches are presented to quantitatively analyze and understand the configurations and kinetic processes. One is to determine the thickness of mixing layers through tracking the distributions and evolutions of the thermodynamic nonequilibrium (TNE) measures;the other is to evaluate the growth rate of KHI from the slopes of morphological functionals.Physically,it is found that the time histories of width of mixing layer,TNE intensity, and boundary length show high correlation and attain their maxima simultaneously. The viscosity effects are twofold, stabilize the KHI,and enhance both the local and global TNE intensities.Contrary to the monotonically inhibiting effects of viscosity, the heat conduction effects firstly refrain then enhance the evolution afterwards. The physical reasons are analyzed and presented.Yan-Biao Gan Ai-Guo Xu Guang-Cai Zhang Chuan-Dong Lin Hui-Lin Lai Zhi-Peng Liu 2019Frontiers of physics2019,14,4:7
5Dalbavancin binds ACE2 to block its interaction with SARS-CoV-2 spike protein and is effective in inhibiting SARS-CoV-2 infection in animal models显示文摘Infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has caused a pandemic worldwide.Currently, however,no effective drug or vaccine is available to treat or prevent the resulting coronavirus disease 2019(COVID-19).Here,we report our discovery of a promising an ti-COVID-19 drug can didate, the lipoglycopeptide antibiotic dalbava ncin, based on virtual screening of the FDA-approved peptide drug library combined with in vitro and in vivo functional antiviral assays. Our results showed that dalbavancin directly binds to human angiotensin-converting enzyme 2(ACE2)with high affinity, thereby blocking its interaction with the SARS-CoV-2 spike protein.Furthermore, dalbavancin effectively prevents SARS-CoV-2 replication in Vero E6 cells with an EC50 of~12 nM.In both mouse and rhesus macaque models, viral replication and histopathological injuries caused by SARS-CoV-2 infection are significantly inhibited by dalbavancin administration.Given its high safety and long plasma half-life(8-10 days)shown in previous clinical trials, our data indicate that dalbavancin is a promising anti-COVID-19 drug candidate.Gan Wang Meng-Li Yang Zi-Lei Duan Feng-Liang Liu Lin Jin Cheng-Bo Long Min Zhang Xiao-Peng Tang Ling Xu Ying-Chang Li Peter Muiruri Kamau Lian Yang Hong-Qi Liu Jing-Wen Xu Jie-Kai Chen Yong-Tang Zheng Xiao-Zhong Peng Ren Lai 2021Cell Research2021,31,1:6
63’-Methoxydaidzein exerts analgesic activity by inhibiting voltage-gated sodium channels显示文摘Isoflavones are widely consumed by people around the world in the form of soy products, dietary supplements and drugs. Many isoflavones or related crude extracts have been reported to exert pain-relief activities, but the mechanism remains unclear. Voltage-gated sodium channels(VGSCs) play important roles in excitability of pain sensing neurons and many of them are important nociceptors. Here, we report that several isoflavones including 3’-methoxydaidzein(3 MOD), genistein(GEN) and daidzein(DAI) show abilities to block VGSCs and thus to attenuate chemicals and heat induced acute pain or chronic constriction injury(CCI) induced pain hypersensitivity in mice. Especially, 3 MOD shows strong analgesic potential without inducing addiction through inhibiting subtypes NaV1.7, NaV1.8 and NaV1.3 with the IC50 of 181 ± 14, 397 ± 26, and 505 ± 46 nmol·L–1, respectively, providing a promising compound or parent structure for the treatment of pain pathologies. This study reveals a pain-alleviating mechanism of dietary isoflavones and may provide a convenient avenue to alleviate pain.XU Run-Jia FEI Shuo-Han CHEN Lin-Yan WANG Gan LIU Ming ZHANG Wen-Sheng YAN Xiu-Wen LAI Ren SHEN Chuan-Bin 2019Chinese Journal of Natural Medicines2019,17,6:4

8Transferrin plays a central role in coagulation balance by interacting with clotting factors显示文摘Coagulation balance is maintained through fine-tuned interactions among clotting factors, whose physiological concentrations vary substantially. In particular, the concentrations of coagulation proteases(pM to nM)are much lower than their natural inactivator antithrombin(AT,〜3μM),suggesting the existence of other coordinators. In the current study,we found that transferrin(norm plasma concentration〜40 μM)interacts with fibrinogen, thrombin, factor Xlla(FXlla),and AT with different affinity to maintain coagulation balance.Normally, transferrin is sequestered by binding with fibrinogen(normal plasma concentration〜10 μM)at a molar ratio of 4:1.In atherosclerosis, abnormally up-regulated transferrin interacts with and potentiates thrombin/FXlla and blocks ATs inactivation effect on coagulation proteases by binding to AT,thus inducing hypercoagulability. In the mouse model, transferrin overexpression aggravated atherosclerosis, whereas transferrin inhibition via shRNA knockdown or treatment with antitransferrin antibody or designed peptides interfering with transferrin-thrombin/FXlla interactions alleviated atherosclerosis.Collectively, these findings identify that transferrin is an important clotting regulator and an adjuster in the maintenance of coagulation balance and modifies the coagulation cascade.Xiaopeng Tang Zhiye Zhang Mingqian Fang Yajun Han Gan Wang Sheng Wang Min Xue Yaxiong Li Li Zhang Jian Wu Biqing Yang James Mwangi Qiumin Lu Xiaoping Du Ren Lai 2020Cell Research2020,30,2:2
9Phenolic acids isolated from the fungus Schizophyllum commune exert analgesic activity by inhibiting voltage-gated sodium channels显示文摘The present study was designed to search for compounds with analgesic activity from the Schizophyllum commune(SC), which is widely consumed as edible and medicinal mushroom world. Thin layer chromatography(TLC), tosilica gel column chromatography, sephadex LH 20, and reverse-phase high performance liquid chromatography(RP-HPLC) were used to isolate and purify compounds from SC. Structural analysis of the isolated compounds was based on nuclear magnetic resonance(NMR). The effects of these compounds on voltage-gated sodium(NaV) channels were evaluated using patch clamp. The analgesic activity of these compounds was tested in two types of mouse pain models induced by noxious chemicals. Five phenolic acids identified from SC extracts in the present study included vanillic acid, m-hydroxybenzoic acid, o-hydroxybenzeneacetic acid, 3-hydroxy-5-methybenzoic acid, and p-hydroxybenzoic acid. They inhibited the activity of both tetrodotoxin-resistant(TTX-r) and tetrodotoxin-sensitive(TTX-s) NaV channels. All the compounds showed low selectivity on NaV channel subtypes. After intraperitoneal injection, three compounds of these compounds exerted analgesic activity in mice. In conclusion, phenolic acids identified in SC demonstrated analgesic activity, facilitating the mechanistic studies of SC in the treatment of neurasthenia.YAO Hui-Min WANG Gan LIU Ya-Ping RONG Ming-Qiang SHEN Chuan-Bin YAN Xiu-Wen LUO Xiao-Dong LAI Ren 2016Chinese Journal of Natural Medicines2016,14,9:2

20Anticarin-β shows a promising antiosteosarcoma effect by specifically inhibiting CCT4 to impair proteostasis显示文摘Unlike healthy, non-transformed cells, the proteostasis network of cancer cells is taxed to produce proteins involved in tumor development. Cancer cells have a higher dependency on molecular chaperones to maintain proteostasis. The chaperonin T-complex protein ring complex(TRiC) contains eight paralogous subunits(CCT1-8), and assists the folding of as many as 10% of cytosolic proteome.TRiC is essential for the progression of some cancers, but the roles of TRiC subunits in osteosarcoma remain to be explored. Here, we show that CCT4/TRiC is significantly correlated in human osteosarcoma, and plays a critical role in osteosarcoma cell survival. We identify a compound anticarin-β that can specifically bind to and inhibit CCT4. Anticarin-β shows higher selectivity in cancer cells than in normal cells. Mechanistically, anticarin-β potently impedes CCT4-mediated STAT3 maturation. Anticarin-β displays remarkable antitumor efficacy in orthotopic and patient-derived xenograft models of osteosarcoma.Collectively, our data uncover a key role of CCT4 in osteosarcoma, and propose a promising treatment strategy for osteosarcoma by disrupting CCT4 and proteostasis.Gan Wang Min Zhang Ping Meng Chengbo Long Xiaodong Luo Xingwei Yang Yunfei Wang Zhiye Zhang James Mwangi Peter Muiruri Kamau Zhi Daic Zunfu Ke Yi Zhang Wenlin Chen Xudong Zhao Fei Ge Qiumin Lv Mingqiang Rong Dongsheng Li Yang Jin Xia Sheng Ren Lai 2022Acta Pharmaceutica Sinica B2022,12,5:1
