| 1 | Mitochondrial uncoupler BAM15 inhibits artery constriction and potently activates AMPK in vascular smooth muscle cells显示文摘Our previous studies found that mitochondrial uncouplers CCCP and niclosamide inhibited artery constriction and the mechanism involved AMPK activation in vascular smooth muscle cells. BAM15 is a novel type of mitochondrial uncoupler. The aim of the present study is to identify the vasoactivity of BAM15 and characterize the BAM15-induced AMPK activation in vascular smooth muscle cells(A10 cells). BAM15 relaxed phenylephrine(PE)-induced constricted rat mesenteric arteries with intact and denuded endothelium. Pretreatment with BAM15 inhibited PEinduced constriction of rat mesenteric arteries with intact and denuded endothelium. BAM15, CCCP,and niclosamide had the comparable IC50 value of vasorelaxation in PE-induced constriction of rat mesenteric arteries. BAM15 was less cytotoxic in A10 cells compared with CCCP and niclosamide.BAM15 depolarized mitochondrial membrane potential, induced mitochondrial fission, increased mitochondrial ROS production, and increased mitochondrial oxygen consumption rate in A10 cells.BAM15 potently activated AMPK in A10 cells and the efficacy of BAM15 was stronger than that of CCCP, niclosamide, and AMPK positive activators metformin and AICAR. In conclusion, BAM15 activates AMPK in vascular smooth muscle cells with higher potency than that of CCCP, niclosamide and the known AMPK activators metformin and AICAR. | 2 | Identification of maize seed varieties based on near infrared reflectance spectroscopy and chemometrics显示文摘False seeds can often be seen in the maize seed market,leading to a serious decline in maize yield.Those existing variety identification methods are expensive,time consuming,and destructive to seeds.The aim of this study is to develop a cheap,fast and non-destructive method which can robustly identify large amounts of maize seed varieties based on near-infrared reflectance spectroscopy(NIRS)and chemometrics.Because it is difficult to establish models for every variety in the market,this study mainly investigated the performance of models based on a large number of samples(more than 40 major varieties in the market).The reflectance spectra of maize seeds were collected by two modes(bulk kernels mode and single kernel mode).Both collection modes can be applied to identification,but only the single kernel mode can be applied to purity sorting.The spectra were pretreated with smoothing,the first derivative and vector normalization;and then principal component analysis(PCA),linear discriminant analysis(LDA)and biomimetic pattern recognition(BPR)were applied to establish identification models.The environmental factors such as producing areas and years have a significant influence on the performance of the models.Therefore,the method to improve the robustness of the models was investigated in this study.New indexes(correct acceptance degree(CAD),correct rejection degree(CRD)and correct degree(CD))were defined to analyze the performance of the models more accurately.Finally,the models obtained a mean correct discrimination rate of over 90%,and exhibited robust properties for samples harvested from different areas and years.The results showed that NIR technology combined with chemometrics methods such as PCA,LDA,and BPR could be a suitable and alternative technique to identify the authenticity of maize seed varieties. | Yongjin Cui Lanjun Xu Dong An Zhe Liu Jiancheng Gu Shaoming Li Xiaodong Zhang Dehai Zhu | 2018 | International Journal of Agricultural and Biological Engineering2018,11,2: | 3 |

| 6 | The role of human ADA2a in the regulation of p53 acetylation and stability显示文摘The tumor suppressor protein p53 is a well-known transcription factor that functions as a critical component of the genotoxic stress response via regulating the expression of effector proteins that control cellular fate following DNA damage. The human p300/CBP-associated factor(PCAF)-containing histone acetyltransferase(HAT) complex is important for the stability and activity of p53 via its acetylation. The human homolog of yeast alteration/deficiency in activation 2a(ADA2a) is a stable component of the human PCAF-containing HAT complex. In this study,we demonstrated that p53 and hADA2a physically interact with each other in human HEK 293T cells. Using overexpression and small interfering RNA-mediated knockdown,we demonstrated that hADA2a stabilizes p53 via promoting its acetylation at lysine 320 - a PCAF-dependent acetylation site. Furthermore,hADA2a can potentiate the transcriptional activity of p53 at the BAX and p21 promoters to induce cell apoptosis and cell cycle arrest. | 10 | SUMOylation-triggered ALIX activation modulates extracellular vesicles circTLCD4-RWDD3 to promote lymphatic metastasis of non-small cell lung cancer显示文摘Lymph node(LN)metastasis is one of the predominant metastatic routes of non-small cell lung cancer(NSCLC)and is considered as a leading cause for the unsatisfactory prognosis of patients.Although lymphangiogenesis is well-recognized as a crucial process in mediating LN metastasis,the regulatory mechanism involving lymphangiogenesis and LN metastasis in NSCLC remains unclear.In this study,we employed high-throughput sequencing to identify a novel circular RNA(circRNA),circTLCD4-RWDD3,which was significantly upregulated in extracellular vesicles(EVs)from LN metastatic NSCLC and was positively associated with deteriorated OS and DFS of patients with NSCLC from multicenter clinical cohort.Downregulating the expression of EV-packaged circTLCD4-RWDD3 inhibited lymphangiogenesis and LN metastasis of NSCLC both in vitro and in vivo.Mechanically,circTLCD4-RWDD3 physically interacted with hnRNPA2B1 and mediated the SUMO2 modification at K108 residue of hnRNPA2B1 by upregulating UBC9.Subsequently,circTLCD4-RWDD3-induced SUMOylated hnRNPA2B1 was recognized by the SUMO interaction motif(SIM)of ALIX and activated ALIX to recruit ESCRT-III,thereby facilitating the sorting of circTLCD4-RWDD3 into NSCLC cell-derived EVs.Moreover,EV-packaged circTLCD4-RWDD3 was internalized by lymphatic endothelial cells to activate the transcription of PROX1,resulting in the lymphangiogenesis and LN metastasis of NSCLC.Importantly,blocking EV-mediated transmission of circTLCD4-RWDD3 via mutating SIM in ALIX or K108 residue of hnRNPA2B1 inhibited the lymphangiogenesis and LN metastasis of NSCLC in vivo.Our findings reveal a precise mechanism underlying SUMOylated hnRNPA2B1-induced EV packaging of circTLCD4-RWDD3 in facilitating LN metastasis of NSCLC,suggesting that EV-packaged circTLCD4-RWDD3 could be a potential therapeutic target against LN metastatic NSCLC. | Xiayao Diao Chao Guo Hanhao Zheng Ke Zhao Yuming Luo Mingjie An Yan Lin Jiancheng Chen Yuanlong Li Yuting Li Xuehan Gao Jiaqi Zhang Mengxin Zhou Wenliang Bai Lei Liu Guige Wang Lanjun Zhang Xiaotian He Rusi Zhang Zhihua Li Changhao Chen Shanqing Li | 2023 | Signal Transduction and Targeted Therapy2023,8,12: | 0 |

| 14 | Influence of impact energy on work hardening ability of austenitic manganese steel and its mechanism显示文摘To further understand the hardening mechanism of austenitic manganese steel under actual working conditions, the work hardening ability was studied and the microstructures of austenitic manganese steel were observed under different impact energies. The work hardening mechanism was also analyzed. The results show that the best strain hardening effect could be received only when the impact energy reaches or exceeds the critical impact energy. The microstructural observations reveal that dislocations, stacking faults and twins increase with raising impact energy of the tested specimens. The hardening mechanism changes at different hardening degrees. | 15 | Ectopic Overexpression of EuCHIT30.7 Improves Nicotiana tabacum Resistance to Powdery Mildew显示文摘Various strains of powdery mildew(PM),a notorious plant fungal disease,are prevalent and pose a significant threat to plant health.To control PM,transgenic technology can be used to cultivate more resistant plant varieties.In the present study,we utilized the rapid amplification of cDNA ends(RACE)technique to clone the full-length cDNA sequence of the EuCHIT30.7 gene to explore plant genes with disease resistance functions.Bioinformatics analysis revealed that this gene belongs to the GH18 family and is classified as a class III chitinase.The EuCHIT30.7 gene is expressed throughout the Eucommia ulmoides plant,with the most abundant expression in male flowers.Subcellular localization analysis indicated that the protein encoded by this gene was detected within both the cell membrane and cytoplasm.Upon PM inoculation,overexpression of EuCHIT30.7 in tobacco plants led to a significantly reduced relative lesion area and a decreased spore count compared to both wild-type and empty vector control plants.Activities of the protective enzymes,namely,peroxidase(POD),superoxide dismutase(SOD),catalase(CAT),and phenylalaninammo-nialyase(PAL),in tobacco plants overexpressing EuCHIT30.7 were significantly greater than those in wild-type and empty vector tobacco plants.Furthermore,the rate of increase in malondialdehyde(MDA)content was significantly lower in tobacco plants expressing EuCHIT30.7 compared to control tobacco plants.In EuCHIT30.7 transgenic tobacco,the expression of pathogen-related protein genes,namely,PR2,PR5,PR1a,PDF1.2,and MLP423,along with the tobacco PM negative regulatory gene,MLO2,were significantly higher compared to control tobacco plants.These findings suggested that EuCHIT30.7 significantly enhances the resistance of tobacco to PM. | Lanjun Li Degang Zhao | 2023 | Phyton-International Journal of Experimental Botany2023,92,11: | 0 |
| 16 | Study on the Main Controlling Factors of the Early Ordovician Stromatolites Development in Liujiachang Area of Songzi City显示文摘The Early Ordovician stromatolites in the study area are mainly exposed at the bottom and middle and lower parts of the Second Member of Nanjinguan Formation,the top of the Fourth Member of Nanjinguan Formation,the middle of the Second Member of Fenxiang Formation and the bottom of the First Member of Honghuayuan Formation.In order to find out the controlling factors of Lower Ordovician stromatolites development in the study area,the analysis data of carbon,oxygen isotope samples,macro element samples as well as their variation characteristics are studied and then combined them with previous research results,which is eventually beneficial for reaching the conclusion:sea level change is one of the main factors controlling the formation of stromatolites.There are five sea level change cycles in the Early Ordovician period.The analysis of sea level change and accommodation space variation characteristics show that when the growth rate of the accommodation space is approximately equal to that of carbonate,the circulation of seawater is well,and the amount of light and oxygen is sufficient,so that the cyanobacteria organism can be multiplied in large quantities.The growth and development provide favorable conditions,and favorable conditions are provided for the growth and development of stromatolites;The peak changes in CaO content and CaO/MgO ratio indicate that the formation period of stromatolites is arid climate environment with high water temperature and large evaporation.Prokaryote reproduction is suitable for the environments with high seawater cleanliness.When the amount of land-based debris injected into the sea increases,the turbidity of water will lead to a large number of deaths of algae microorganisms that form stromatolites,and the stromatolitic microbial mats cannot be preserved.In addition,the growth and predation of macro-organisms play a restrictive role in the development of stromatolite-forming micro-organisms blue-green algae.According to the actual situa-tion of the development of the laminated rocks in the study area,what is the dominant function in the formation and decline of stromatolites between the evolution of metazoans and changing environments is discussed in the study. | Lanjun Wu Chuantao Xiao Zhongzhen Cheng Yichen Bao Li Huang Kanglin Chen | 2019 | Open Journal of Yangtze Oil and Gas2019,4,3: | 0 |
