1First-generation EGFR tyrosine kinase inhibitor therapy in 106 patients with compound EGFR-mutated lung cancer: a single institution’s clinical practice experience显示文摘Background:The antitumour efficacy of tyrosine kinase inhibitors(TKIs)in lung cancer patients with compound epidermal growth factor receptor(EGFR)mutations has not been resolved.Our study summarizes a single institutional experience of first-generation TKI therapy for lung cancers with compound EGFR mutations.Methods:A total of 106 consecutive patients with tumours bearing compound EGFR mutations were identified between January 2012 and May 2016;all patients received first-generation TKI therapy.Deletions in exon 19 and the L858R point mutation in exon 21 were considered common mutations;T790M was considered separately because of its association with TKIs resistances.Any other mutation was defined as a rare mutation.Patients were divided as follows:double common mutations(group A);common plus T790M mutations(group B);common plus rare muta-tions(group C);double rare mutations(group D);and rare plus T790M mutations(group E).A separate group of 115 consecutive patients with a single common mutation was created for comparative analysis(group F).Results:The frequency of patients with compound EGFR was 2.9%(114/3925)and their response rate to first-genera-tion TKIs was 50.9%,which was not significantly different from group F(67.0%,P=0.088).The progression-free survival(PFS)of the 106 patients receiving TKI therapy was worse than that of group F(median,9.1 vs.13.0 months,respec-tively;P<0.001).The PFS of the compound mutation group was shorter than that of the single common mutation group(median,10.1 months in group A,P=0.240;9.1 months in group B,P<0.001;9.6 months in group C,P=0.010;6.5 months in group D,P=0.048;5.4 months in group E,P=0.017).Patients with a co-occurring mutation in exon 20(excluding T790M)exhibited significantly worse PFS than the patients with other compound mutations or with a single common mutation(median,6.5 vs.9.1 vs.13.0 months,respectively,P=0.002).Conclusions:There was significant heterogeneity among the compound EGFR mutations and their response to first-generation TKIs.Individualized treatment in clinical practice should be considered for each case.Xiangyang Yu Xuewen Zhang Zichen Zhang Yongbin Lin Yingsheng Wen Yongqiang Chen Weidong Wang Lanjun Zhang 2018Cancer Communications2018,38,1:5

4Microwave-assisted acid-induced formation of linker vacancies within Zr-based metal organic frameworks with enhanced heterogeneous catalysis显示文摘Herein,we report a microwave-assisted acid-induced post-treatment method for the formation of linker vacancies within Zr-based metal organic frameworks(Zr-MOFs).The number of linker vacancies can be easily regulated with this method by changing the concentration of the HCl solution and the duration of microwave irradiation.The optimized defective UiO-66 showed higher linker defects with a higher specific surface area and thermal stability.The results of the catalytic cyclization of citronella showed that the Zr-MOFs with more defects exhibited enhanced catalytic performance.This work may provide a new method for the creation of defective MOFs with high activity and stability.Yu Liang Chenhui Li Lanjun Chen Jia Huo Mohammed Loubidi Yangyang Zhou Yanbo Liu 2021Chinese Chemical Letters2021,32,2:1

9Digital twins in smart farming:An autoware-based simulator for autonomous agricultural vehicles显示文摘Digital twins can improve the level of control over physical entities and help manage complex systems by integrating a range of technologies.The autonomous agricultural machine has shown revolutionary effects on labor reduction and utilization rate in field works.Autonomous vehicles in precision agriculture have the potential to improve competitiveness compared to current crop production methods and have become a research hotspot.However,the development time and resources required in experiments have limited the research in this area.Simulation tools in unmanned farming that are required to enable more efficient,reliable,and safe autonomy are increasingly demanding.Inspired by the recent development of an open-source virtual simulation platform,this study proposed an autoware-based simulator to evaluate the performance of agricultural machine guidance based on digital twins.Oblique photogrammetry using drones is used to construct threedimensional maps of fields at the same scale as reality.A communication format suitable for agricultural machines was developed for data input and output,along with an inter-node communication methodology.The conversion,publishing,and maintenance of multiple coordinate systems were completed based on ROS(Robot Operating System).Coverage path planning was performed using hybrid curves based on Bézier curves,and it was tested in both a simulation environment and actual fields with the aid of Pure Pursuit algorithms and PID controllers.Xin Zhao Wanli Wang Long Wen Zhibo Chen Sixian Wu Kun Zhou Mengyao Sun Lanjun Xu Bingbing Hu Caicong Wu 2023International Journal of Agricultural and Biological Engineering2023,16,4:0
10SUMOylation-triggered ALIX activation modulates extracellular vesicles circTLCD4-RWDD3 to promote lymphatic metastasis of non-small cell lung cancer显示文摘Lymph node(LN)metastasis is one of the predominant metastatic routes of non-small cell lung cancer(NSCLC)and is considered as a leading cause for the unsatisfactory prognosis of patients.Although lymphangiogenesis is well-recognized as a crucial process in mediating LN metastasis,the regulatory mechanism involving lymphangiogenesis and LN metastasis in NSCLC remains unclear.In this study,we employed high-throughput sequencing to identify a novel circular RNA(circRNA),circTLCD4-RWDD3,which was significantly upregulated in extracellular vesicles(EVs)from LN metastatic NSCLC and was positively associated with deteriorated OS and DFS of patients with NSCLC from multicenter clinical cohort.Downregulating the expression of EV-packaged circTLCD4-RWDD3 inhibited lymphangiogenesis and LN metastasis of NSCLC both in vitro and in vivo.Mechanically,circTLCD4-RWDD3 physically interacted with hnRNPA2B1 and mediated the SUMO2 modification at K108 residue of hnRNPA2B1 by upregulating UBC9.Subsequently,circTLCD4-RWDD3-induced SUMOylated hnRNPA2B1 was recognized by the SUMO interaction motif(SIM)of ALIX and activated ALIX to recruit ESCRT-III,thereby facilitating the sorting of circTLCD4-RWDD3 into NSCLC cell-derived EVs.Moreover,EV-packaged circTLCD4-RWDD3 was internalized by lymphatic endothelial cells to activate the transcription of PROX1,resulting in the lymphangiogenesis and LN metastasis of NSCLC.Importantly,blocking EV-mediated transmission of circTLCD4-RWDD3 via mutating SIM in ALIX or K108 residue of hnRNPA2B1 inhibited the lymphangiogenesis and LN metastasis of NSCLC in vivo.Our findings reveal a precise mechanism underlying SUMOylated hnRNPA2B1-induced EV packaging of circTLCD4-RWDD3 in facilitating LN metastasis of NSCLC,suggesting that EV-packaged circTLCD4-RWDD3 could be a potential therapeutic target against LN metastatic NSCLC.Xiayao Diao Chao Guo Hanhao Zheng Ke Zhao Yuming Luo Mingjie An Yan Lin Jiancheng Chen Yuanlong Li Yuting Li Xuehan Gao Jiaqi Zhang Mengxin Zhou Wenliang Bai Lei Liu Guige Wang Lanjun Zhang Xiaotian He Rusi Zhang Zhihua Li Changhao Chen Shanqing Li 2023Signal Transduction and Targeted Therapy2023,8,12:0

12Study on the Main Controlling Factors of the Early Ordovician Stromatolites Development in Liujiachang Area of Songzi City显示文摘The Early Ordovician stromatolites in the study area are mainly exposed at the bottom and middle and lower parts of the Second Member of Nanjinguan Formation,the top of the Fourth Member of Nanjinguan Formation,the middle of the Second Member of Fenxiang Formation and the bottom of the First Member of Honghuayuan Formation.In order to find out the controlling factors of Lower Ordovician stromatolites development in the study area,the analysis data of carbon,oxygen isotope samples,macro element samples as well as their variation characteristics are studied and then combined them with previous research results,which is eventually beneficial for reaching the conclusion:sea level change is one of the main factors controlling the formation of stromatolites.There are five sea level change cycles in the Early Ordovician period.The analysis of sea level change and accommodation space variation characteristics show that when the growth rate of the accommodation space is approximately equal to that of carbonate,the circulation of seawater is well,and the amount of light and oxygen is sufficient,so that the cyanobacteria organism can be multiplied in large quantities.The growth and development provide favorable conditions,and favorable conditions are provided for the growth and development of stromatolites;The peak changes in CaO content and CaO/MgO ratio indicate that the formation period of stromatolites is arid climate environment with high water temperature and large evaporation.Prokaryote reproduction is suitable for the environments with high seawater cleanliness.When the amount of land-based debris injected into the sea increases,the turbidity of water will lead to a large number of deaths of algae microorganisms that form stromatolites,and the stromatolitic microbial mats cannot be preserved.In addition,the growth and predation of macro-organisms play a restrictive role in the development of stromatolite-forming micro-organisms blue-green algae.According to the actual situa-tion of the development of the laminated rocks in the study area,what is the dominant function in the formation and decline of stromatolites between the evolution of metazoans and changing environments is discussed in the study.Lanjun Wu Chuantao Xiao Zhongzhen Cheng Yichen Bao Li Huang Kanglin Chen 2019Open Journal of Yangtze Oil and Gas2019,4,3:0
