| 2 | The Sunway TaihuLight supercomputer: system and applications显示文摘The Sunway Taihu Light supercomputer is the world's first system with a peak performance greater than 100 PFlops. In this paper, we provide a detailed introduction to the Taihu Light system. In contrast with other existing heterogeneous supercomputers, which include both CPU processors and PCIe-connected many-core accelerators(NVIDIA GPU or Intel Xeon Phi), the computing power of Taihu Light is provided by a homegrown many-core SW26010 CPU that includes both the management processing elements(MPEs)and computing processing elements(CPEs) in one chip. With 260 processing elements in one CPU, a single SW26010 provides a peak performance of over three TFlops. To alleviate the memory bandwidth bottleneck in most applications, each CPE comes with a scratch pad memory, which serves as a user-controlled cache. To support the parallelization of programs on the new many-core architecture, in addition to the basic C/C++and Fortran compilers, the system provides a customized Sunway Open ACC tool that supports the Open ACC2.0 syntax. | 7 | Rapid generation of genetic diversity by multiplex CRISPR/Cas9 genome editing in rice显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated endonuclease 9(CRISPR/Cas9) system has emerged as a promising technology for specific genome editing in many species. Here we constructed one vector targeting eight agronomic genes in rice using the CRISPR/Cas9 multiplex genome editing system. By subsequent genetic transformation and DNA sequencing, we found that the eight target genes have high mutation efficiencies in the T_0 generation. Both heterozygous and homozygous mutations of all editing genes were obtained in T_0 plants. In addition, homozygous sextuple, septuple, and octuple mutants were identified. As the abundant genotypes in T_0 transgenic plants, various phenotypes related to the editing genes were observed. | 8 | miR-21 promotes the differentiation of hair folliclederived neural crest stem cells into Schwann cells显示文摘Hair follicle-derived neural crest stem cells can be induced to differentiate into Schwann cells in vivo and in vitro. However, the underlying regulatory mechanism during cell differentiation remains poorly understood. This study isolated neural crest stem cells from human hair follicles and induced them to differentiate into Schwann cells. Quantitative RT-PCR showed that microRNA(miR)-21 expression was gradually increased during the differentiation of neural crest stem cells into Schwann cells. After transfection with the miR-21 agonist(agomir-21), the differentiation capacity of neural crest stem cells was enhanced. By contrast, after transfection with the miR-21 antagonist(antagomir-21), the differentiation capacity was attenuated. Further study results showed that SOX-2 was an effective target of miR-21. Without compromising SOX2 mRNA expression, miR-21 can down-regulate SOX protein expression by binding to the 3′-UTR of miR-21 mRNA. Knocking out the SOX2 gene from the neural crest stem cells significantly reversed the antagomir-21 inhibition of neural crest stem cells differentiating into Schwann cells. | 11 | Treatment of Donor-derived Carbapenem-resistant Klebsiella pneumoniae Infection after Renal Transplantation with Tigecycline and Extended-infusion Meropenem显示文摘Objective Donor-derived carbapenem-resistant Klebsiella pneumoniae(CRKP)infection has recently emerged as a critical early complication after renal transplantation.Although CRKP is usually sensitive to tigecycline,monotherapy with this drug is often less than effective.We investigated the efficacy of a combined regimen of tigecycline with high-dose,extended-infusion meropenem in the treatment of donor-derived CRKP infection after kidney transplantation.Methods From Jan.2016 to Dec.2017,a total of 12 CRKP isolates were detected from cultures of the organ preservation solution used for soaking the donor kidneys at our institute.Probable or possible donor-derived infection(DDI)was identified in 8 transplant recipients.Clinical data were retrospectively analyzed.Results Klebsiella pneumoniae carbapenemase-2(KPC-2)-producing CRKP was reported to be positive in organ preservation solution cultures at 3.5±0.9 days after transplantation,leading to surgical site(n=3),urinary tract(n=4),and/or bloodstream(n=2)infections in 8 recipients.The drug susceptibility tests showed that CRKP was sensitive to tigecycline,but resistant to meropenem.In 7 patients who received tigecycline combined with high-dose extended-infusion meropenem,DDIs were successfully cured.The length of hospital stay was 31(18–129)days,and the serum creatinine at discharge was 105.8±16.7µmol/L.The one remaining patient who received tigecycline combined with intravenous-drip meropenem died of septic shock.A median follow-up of 43 months(33–55)showed no recurrence of new CRKP infection in the 7 surviving recipients.Conclusion It was suggested that a prompt and appropriate combination therapy using tigecycline with high-dose extended-infusion meropenem is effective in treating donor-derived KPC-2-producing CRKP infection after renal transplantation. | Zhi-qiang WANG Zhi-liang GUO Hao FENG Cheng FU Guang-yuan ZHAO Ke MA Lan ZHU Gang CHEN | 2021 | Current Medical Science2021,41,4: | 8 |
| 12 | Pulsed radiofrequency inhibits expression of P2X3 receptors and alleviates neuropathic pain induced by chronic constriction injury in rats显示文摘Background:Pulsed radiofrequency (PRF) is a minimally invasive interventional technique that provides a novel and effective treatment strategy for neuropathic pain (NP).PRF is advantageous because it does not damage nerves and avoids sensory loss after treatment.At present,animal studies have demonstrated that PRF is safe and effective for relieving the NP associated with sciatic nerve damage in rats with chronic constriction injury (CCI).However,the mechanism through which this effect occurs is unknown.An increasing body of evidence shows that the expression of the P2X ligand-gated ion channel 3 (P2X3) receptor is closely related to NP;this study was to investigate whether the expression of this receptor is involved in NP relief due to PRF.Methods:A total of 36 healthy adult male Sprague-Dawley (SD) rats were randomly divided into three groups:Sham group,CCI group,and PRF group.The right sciatic nerve was ligated in CCI group and PRF group to establish a CCI model;the right sciatic nerve was separated but not ligated in Sham group.On day 14 after the operation,PRF was administered to the ligated sciatic nerve in PRF group (42℃,45 V,2 min).A non-live electrode was placed at the exposed sciatic nerve for the rats in Sham and CCI groups.The hindpaw withdrawal threshold (HWT) and thermal withdrawal latency (TWL) were measured at the right hindpaw at different time points before and after PRF or sham therapy.On day 28 after treatment,the dorsal root ganglion (DRG) and spinal dorsal horn of the right L4-6 were harvested from each group to determine the mRNA and protein levels of the P2X3 receptor.Results:On day 28 after PRF treatment,the HWT (8.33 ± 0.67 g vs.3.62 ± 0.48 g) and TWL (25.42 ± 1.90 s vs.15.10 ± 1.71 s) were significantly higher in PRF group as compared to CCI group (P < 0.05).The mRNA expression of the P2X3 receptor in the DRG in PRF group was 23.7% lower than that in CCI group (P < 0.05),in the spinal dorsal horns in PRF group was 22.7% lower than that in CCI group (P < 0.05).The protein expression of the P2X3 receptor in the DRG in PRF group was 27.8 % lower than that in CCI group (P < 0.05),in the spinal dorsal horns in PRF group was 35.6% lower than that in CCI group (P < 0.05).Conclusion:PRF possibly reduces NP in CCI rats by inhibiting the expression of the P2X3 receptor in the L4-6 DRG and spinal dorsal horns. | Miao Fu Lan Meng Hao Ren Fang Luo | 2019 | Chinese Medical Journal2019,,14: | 8 |

Earlier studies have shown that air particulate matter (PM), particularly fine particulate matter (PM2.5)[2], is an important factor triggering childhood asthma. Since national PM2.5 data were not available until 2013, the effects of PM2.5 on childhood asthma are less known, and most of the related research has been conducted primarily in some metropolises in China. Moreover, the majority of these studies focus on the impact of a unit increase (1 or 10 μg/m3) or an interquartile range increase in PM2.5 on childhood asthma based on the same pollutant background concentrations. | 16 | Simultaneous anaerobic-aerobic biodegradation of halogenated phenolic compound under oxygen-limited conditions显示文摘The successful application of co-immobilized aerobic-anaerobic biomass under limited aeration in wastewater treatment systems would eliminate the problems associated with the intermediates mono-chlorophenol(MCP) and di-chlorophenol(DCP) accumulations. With low initial pentachlorophenol(PCP) concentration, all PCP could be completely removed under oxygen-limited strict anaerobic conditions, and the removal efficiencies with different initial headspace oxygen percentage(IHOP) were not obviously different from each other. While at high initial PCP concentration, under strictly anaerobic conditions PCP and their intermediates were clearly higher than that under other conditions, and produced obvious accumulation, the highest PCP reduction was achieved by the system receiving 30 IHOP, oxygen-limited system also exhibited lower residual TOC concentration and lower concentration of metabolic intermediates MCP and DCP. These results suggested that under strictly anaerobic condition the reductive dechlorination of low chlorinated compounds became rate limiting in the reductive dechlorination pathway, less chlorinated compounds be more amenable to aerobic degradation, and the aerobes of outer layers could function under limited oxygen. Whole cell patch-clamp techniques wereused in our experiment.RESULTS: The peak amplitude.of ICRAC was -508±115 pA(n=15), its reversal potential of ICRAC was about 0 mV. At thepotential of -100 mV, Tet inhibited the peak amplitude ofICRAC from -521±95 pA to -338±85 pA (P<0.01 vs control,n=5), with the inhibitory rate of 35 % at 10 μmol/L andfrom -504±87 pA to -247±82 pA (P<0.01 vscontrol, n=5),with the inhibitory rate of 49 % at 100 μmol/L, withoutaffecting its reversal potential. The amplitude of ICRAC wasdependent on extracellular Ca2+ concentration. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. | 19 | Characteristics of fault zones and their control on remaining oil distribution at the fault edge: a case study from the northern Xingshugang Anticline in the Daqing Oilfield, China显示文摘Most major oil zones in the Daqing Oilfield have reached a later,high water cut stage,but oil recovery is still only approximately 35%,and 50%of reserves remain to be recovered.The remaining oil is primarily distributed at the edge of faults,in poor sand bodies,and in insufficiently injected and produced areas.Therefore,the edge of faults is a major target for remaining oil enrichment and potential tapping.Based on the dynamic change of production from development wells determined by the injection-recovery relationship at the edge of faults,we analyzed the control of structural features of faults on remaining oil enrichment at the edge.Our results show that the macroscopic structural features and their geometric relationship with sand bodies controlled remaining oil enrichment zones like the edges of NNE-striking faults,the footwalls of antithetic faults,the hard linkage segments(two faults had linked together with each other to form a bigger through-going fault),the tips of faults,and the oblique anticlines of soft linkages.Fault edges formed two types of forward microamplitude structures:(1) the tilted uplift of footwalls controlled by inverse fault sections and(2) the hanging-wall horizontal anticlines controlled by synthetic fault points.The remaining oil distribution was controlled by microamplitude structures.Consequently,such zones as the tilted uplift of the footwall of the NNW-striking antithetic faults with a fault throw larger than 40 m,the hard linkage segments,the tips of faults,and the oblique anticlines of soft linkage were favorable for tapping the remaining oil potential.Multi-target directional drilling was used for remaining oil development at fault edges.Reasonable fault spacing was determined on the basis of fault combinations and width of the shattered zone.Well core and log data revealed that the width of the shattered zone on the side of the fault core was less than 15 m in general;therefore,the distance from a fault to the development target should be larger than 15 m.Vertically segmented growth faults should take the separation of the lateral overlap of faults into account.Therefore,the safe distance of remaining oil well deployment at the fault edge should be larger than the sum of the width of shattered zone in faults and the separation of growth faults by vertical segmentation. | Xiao-Fei Fu Xiao Lan Ling-Dong Meng Hai-Xue Wang Zong-Bao Liu Zhi-Qiang Guo Zai-He Chen | 2016 | Petroleum Science2016,13,3: | 7 |
