| 1 | Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs. | Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu | 2020 | Cell Research2020,30,4: | 81 |

| 3 | Naringenin reduces lung metastasis in a breast cancer resection model显示文摘Metastasis is the main cause of death in cancer patients. To improve the outcomes of patients undergoing a surgery, new adjuvant therapies that can effectively inhibit metastases have to be developed. Studies have shown that flavonoid naringenin, a natural product that is mainly present in grapes and citrus, may contribute to cancer prevention. It has many advantages compared to traditional chemotherapeutic drugs, such as low toxicity. To determine whether naringenin can also inhibit metastases, a breast cancer resection model that mimics clinical situations was established. We found that orally administered naringenin significantly decreased the number of metastatic tumor cells in the lung and extended the life span of tumor resected mice. Flow cytometry analysis revealed that T cells displayed enhanced antitumor activity in naringenin treated mice, with an increased proportion of IFN-γ and IL-2 expressing T cells. In vitro studies further demonstrated that relief of immunosuppression caused by regulatory T cells might be the fundamental mechanism of metastasis inhibition by naringenin. These results indicate that orally administered naringenin can inhibit the outgrowth of metastases after surgery via regulating host immunity. | 6 | Liraglutide alleviates cardiac fibrosis through inhibiting P4hα-1 expression in STZ-induced diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy is an important contributor to morbidity and mortality of diabetic patients by causing heart failure.Interstitial and perivascular fibrosis plays a crucial role in diabetic cardiomyopathy.However,there is a lack of effective specific treatments available for diabetic cardiomyopathy.In the present study,we aim to explore the effects of Liraglutide,a GLP-1 analogue,on diabetic cardiomyopathy in STZ-induced diabetic rats fed with high-fat diet.A total of 60 male Wistar rats were randomly assigned to three groups,i.e.normal group,model group,and Liraglutide group,with 20 rats in each group.Serum levels of TC,TG,LDL-C,NEFA,and hydroxyproline were measured using commercial kits.Cardiac function was evaluated by QRS waves,LVEDd,LVESd,and LVEF.Myocardial fibrosis was measured by immunohistochemistry.Our results demonstrated that chronic administration of Liraglutide decreased the level of blood glucose and significantly alleviated lipid metabolic disturbance compared with the model group.Furthermore,Liraglutide was found to improve the damaged cardiac function.In line with this,we also found that the alleviation of cardiac dysfunction was associated with the decreased fibrosis in diabetic myocardial tissues,which was reflected by the decreased expressions of P4hα-1,COL-1,COL-3,MMP-1,and MMP-9.Our results thus suggest that Liraglutide might have a myocardial protective effect by inhibiting P4hα-1-mediated myocardial fibrosis. | Tong Zhao Huiqiang Chen Fei Xu Juan Wang Yusheng Liu Xiaowei Xing Linlin Guo Mingxiang Zhang Qinghua Lu | 2019 | Acta Biochimica et Biophysica Sinica2019,51,3: | 9 |

| 10 | Optimal power allocation for secure directional modulation networks with a full-duplex UAV user显示文摘This paper makes an investigation of a secure unmanned aerial vehicle(UAV)-aided communication network based on directional modulation(DM). In this network, ground base station(GBS) acts as a control center to transmit confidential message and artificial noise(AN). The UAV user, moving along a linear flight trajectory, is intended to receive the useful information from GBS. At the same time, it also sends AN signals to further interference eavesdropper’s channel. Aiming at maximizing secrecy rate during the UAV flight process, a joint optimization problem is formulated with respect to power allocation(PA)factors, beamforming vector and AN projection matrices. For simplicity, maximum rate transmission, nullspace projection and the leakage-based method are applied to form the transmit beamforming vector, AN projection matrix at GBS, and AN projection vector at UAV user, respectively. Following this, the optimization problem reduces to a bivariate optimization programme with two PA factors. An alternating iterative algorithm(AIA) is proposed to optimize the two PA factors. | 12 | High-yield preparation of robust gold nanoshells on silica nanorattles with good biocompatiblity显示文摘Although gold nanoshells are widely considered as one of the promising photothermal nanomaterials used for biomedicine, the high cost, low yield and poor stability severely limit their potential application in clinical trials.Herein, robust gold nanoshells on silica nanorattles(GSNs)were easily prepared in a high yield by an improved seedmediated method employing polyvinylpyrrolidone(PVP) as a stabilizing and capping agent. The present method is very simple, effective and reproducible and can well control the growth process of gold nanoshells. The as-prepared GSNs have a narrow size distribution(<10 % in standard deviation). Furthermore, the utilization rate of Au in the solution used for the growth of gold nanoshells increases by 70 %than that in previous method. The resultant GSNs have a good structural stability after placing over 6 months due to the protection of PVP. More importantly, in vivo and in vitro toxic studies indicate that the GSNs have good biocompatibility. In the present paper, diepoxycyclohexylethyl octamethyltetrasiloxane was synthesized, and the synthesized product was characterized by FTIR and 1 HMR. The synthesized product was compounded with some acrylates and an expoxide as well as photoinitiators to obtain a 3D printing stereolithography resin(3DSLR111). The properties of 3DSLR111 and its UV-cured samples were investigated by some instruments and equipments. The experimental results show that the critical exposure(Ec) of 3DSLR111 is 10.1 mJ/cm^2, its penetration depth(Dp) is 0.15 mm, and its viscosity at 30 ℃ is 319 mPa·s. Some samples were printed with 3DSLR111, and their linear shrinkage and warping factor were evaluated. | 19 | PSA controls hepatic lipid metabolism by regulating the NRF2 signaling pathway显示文摘The activity of proteinase is reported to correlate with the development and progression of nonalcoholic fatty liver disease(NAFLD).Puromycin-sensitive aminopeptidase(PSA/NPEPPS)is an integral nontransmembrane enzyme that functions to catalyze the cleavage of amino acids near the N-terminus of polypeptides.A previous study suggested that this enzyme acts as a regulator of neuropeptide activity;however,the metabolic function of this enzyme in the liver has not been explored.Here,we identified the novel role of PSA in hepatic lipid metabolism.Specifically,PSA expression was lower in fatty livers from NAFLD patients and mice(HFD,ob/ob,and db/db).PSA knockdown in cultured hepatocytes exacerbated diet-induced triglyceride accumulation through enhanced lipogenesis and attenuated fatty acid β-oxidation.Moreover,PSA mediated activation of the master regulator of antioxidant response,nuclear factor erythroid 2-related factor 2(NRF2),by stabilizing NRF2 protein expression,which further induced downstream antioxidant enzymes to protect the liver from oxidative stress and lipid overload.Accordingly,liver-specific PSA overexpression attenuated hepatic lipid accumulation and steatosis in ob/ob mice.Furthermore,in human liver tissue samples,decreased PSA expression correlated with the progression of NAFLD.Overall,our findings suggest that PSA is a pivotal regulator of hepatic lipid metabolism and its antioxidant function occurs by suppressing NRF2 ubiquitination.Moreover,PSA may be a potential biomarker and therapeutic target for treating NAFLD. | Bangliang Huang Xin Xiong Linlin Zhang Xiufei Liu Yuren Wang Xiaoli Gong Qian Sang Yongling Lu Hua Qu Hongting Zheng Yi Zheng | 2021 | Journal of Molecular Cell Biology2021,13,7: | 4 |
