1Berberine reverses free-fatty-acid-induced insulin resistance in 3T3-L1 adipocytes through targeting IKKβ显示文摘AIM: To investigate the effects and molecular mechanisms of berberine on improving insulin resistance induced by free fatty acids (FFAs) in 3T3-L1 adipocytes. METHODS: The model of insulin resistance in 3T3-L1 adipocytes was established by adding palmic acid (0.5 mmol/L) to the culture medium. Berberine treatment was performed at the same time. Glucose uptake rate was determined by the 2-deoxy-[3H]-D-glucose method. The levels of IkB kinase beta (IKKβ) Ser181 phosphorylation, insulin receptor substrate-1(IRS-1) Ser307 phosphorylation, expression of IKKβ, IRS-1, nuclear transcription factor kappaB p65 (NF-κB p65), phosphatidylinositol-3-kinase p85 (PI-3K p85) and glucose transporter 4 (GLUT4) proteins were detected by Western blotting. The distribution of NF-κB p65 proteins inside the adipocytes was observed through confocal laser scanning microscopy (CLSM). RESULTS: After the intervention of palmic acid for 24 h, the insulin-stimulated glucose transport in 3T3-L1 adipocytes was inhibited by 67%. Meanwhile, the expression of IRS-1 and PI-3K p85 protein was reduced, while the levels of IKKβ Ser181 and IRS-1 Ser307 phosphorylation, and nuclear translocation of NF-κB p65 protein were increased. However, the above indexes, which indicated the existence of insulin resistance, were reversed by berberine although the expression of GLUT4, IKKβ and total NF-κB p65 protein were not changed during this study. CONCLUSION: Insulin resistance induced by FFAs in 3T3-L1 adipocytes can be improved by berberine. 2Estimation of internal nutrient release in large shallow Lake Taihu, China显示文摘Based on field investigation of wave, sediment suspension and the changes in nutrient concentration of the water column in Lake Taihu, China, we proposed two release models to quantify nutrient release under static and dynamic conditions, respectively. Under static conditions, nutrient release from sediments to the overlying water mainly depends on chemical diffusion induced by concentration gradient, in which the nutrient release is controlled by the temperature, dissolved oxygen concentration in the sediment-water interface, oxidation-reduction potential and the concentration difference between porewater and overlying water. Under dynamic condition (or disturbed condition),both dissolved and particulate nutrients in sediments are released into the water column because of wind-induced sediment suspension. The amount of nutrient release under dynamic conditions is larger than that under the static condition. The release of dissolved nutrients, however, does not increase because the wind induced turbulence made oxidation of metallic elements such as Fe (ferric iron), Mn which are capable of precipitating soluble reactive phosphate (SRP). Under dynamic conditions, therefore, the release of total phosphorus (TP) increases dramatically but the release of SRP is close to those under static conditions. In sediments of Lake Taihu, high Fe content leads to a high ratio of Fe to P contents in sediments (Fe:P ratio). Under dynamic conditions, therefore, nutrient release is controlled by the intensity of disturbance, sediment consolidation and nutrient content in sediments.As for dissolved nutrients, especially SRP, the release is also controlled by the intensity of dynamic re-oxidation, Fe content in sediments and nutrient concentration gradient between porewater and overlying water. Based on these two release modes, the release flux in Lake Taihu has been estimated. In the static condition (i.e. laboratory experimental condition), total release of NH4+-N for whole lake is ca. 10,000 ton/a, and PO43--P is ca. 900 ton/a. In the dynamic condition, nutrient release following sediment suspension was estimated according to three different intensities of wind forcing which were defined as 'calm' (wind speed is less than 2 m/s), 'gentle' (wind speed is greater than 2 m/s and less than 6 m/s) and 'gust' (wind speed is greater than 6 m/s). The release rate in the condition of 'calm' was estimated in terms of the nutrient release in the laboratory experimental static condition; whereas the release rate in conditions of 'gentle' and 'gust' was estimated in terms of measurement during sediment resuspension conducted in flume experiments. With the observation of wind velocity and frequency in 2001, each type of wind forcing took the frequency of 12%, 82% and 6% for 'calm', 'gentle' and 'gust', respectively. 4Drug resistance and new therapies in colorectal cancer显示文摘Colorectal cancer(CRC) is often diagnosed at an advanced stage when tumor cell dissemination has taken place. Chemo-and targeted therapies provide only a limited increase of overall survival for these patients. The major reason for clinical outcome finds its origin in therapy resistance. Escape mechanisms to both chemo-and targeted therapy remain the main culprits. Here, we evaluate major resistant mechanisms and elaborate on potential new therapies. Amongst promising therapies is α-amanitin antibodydrug conjugate targeting hemizygous p53 loss. It becomes clear that a dynamic interaction with the tumor microenvironment exists and that this dictates therapeutic outcome. In addition, CRC displays a limited response to checkpoint inhibitors, as only a minority of patients with microsatellite instable high tumors is susceptible. 6Heart failure and left ventricular dysfunction in older patients with chronic kidney disease: the China Hypertension Survey (2012‒2015)显示文摘Background Heart failure(HF)is a leading cause of hospitalization and mortality for older chronic kidney disease(CKD)patients.However,the epidemiological data is scarce.We aimed to determine the prevalence of left ventricular(LV)dysfunction and HF,and to explore the risk factors for HF among those patients.Methods This is a cross-sectional analysis of the China Hypertension Survey conducted between October 2012 and December 2015.A total of 5,808 participants aged≥65 years were included in the analysis.Self-reported history of HF and any other cardiovascular diseases was acquired.2-D and Doppler echocardiography were used to assess LV dysfunction.CKD was defined as either estimated glomerular filtration rate(eGFR)<60 mL/min per 1.73 m2 or urinary albumin to creatinine ratio(ACR)≥30 mg/g.Results Among CKD patients aged≥65 years,the weighted prevalence of HF,heart failure with preserved ejection fraction(HFpEF),heart failure with mid-range ejection fraction(HFmrEF),and heart failure with reduced ejection fraction(HFrEF)was 4.8%,2.5%,0.8%,and 1.7%,respectively.The weighted prevalence of HF was 5.0%in patients with eGFR<60 mL/min per 1.73 m2,and was 5.9%in patients with ACR≥30 mg/g.The prevalence of LV systolic dysfunction was 3.1%,and while it was 8.9%for moderate/severe diastolic dysfunction.Multivariate analysis showed that smoking was significantly associated with the risk of HF.Furthermore,age,smoking,and residents in rural areas were significantly associated with a risk of LV diastolic dysfunction.Conclusions The prevalence of HF and LV dysfunction was high in older patients with CKD,suggesting that particular strategies will be required.Xin WANG Guang HAO Lu CHEN Lin-Feng ZHANG Zuo CHEN Yu-Ting KANG Ying YANG Cong-Yi ZHENG Hao-Qi ZHOU Zeng-Wu WANG Run-Lin GAO 2020Journal of Geriatric Cardiology2020,17,10:19

8Facilitating effects of berberine on rat pancreatic islets through modulating hepatic nuclear factor 4 alpha expression and glucokinase activity显示文摘AIM: To observe the effect of berberine on insulin secretion in rat pancreatic islets and to explore its possible molecular mechanism. METHODS: Primary rat islets were isolated from male Sprague-Dawley rats by collagenase digestion and treated with different concentrations (1,3,10 and 30 μmol/L) of berberine or 1 μmol/L Glibenclamide (GB) for 24 h. Glucose-stimulated insulin secretion (GSIS) assay was conducted and insulin was determined by radioimmunoassay. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was performed to evaluate cytotoxicity. The mRNA level of hepatic nuclear factor 4 alpha (HNF4α) was determined by reverse transcription polymerase chain reaction (RT-PCR). Indirect immunofluorescence staining and Western blot analysis were employed to detect protein expression of HNF4α in the islets. Glucokinase (GK) activity was measured by spectrophotometric method. RESULTS: Berberine enhanced GSIS rather than basal insulin secretion dose-dependently in rat islets and showed no significant cytotoxicity on islet cells at the concentration of 10 μmol/L. Both mRNA and protein expressions of HNF4α were up-regulated by berberine in a dose-dependent manner,and GK activity was also increased accordingly. However,GB demonstrated no regulatory effects on HNF4α expression or GK activity. 9Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 12Chinese Medicine Sequential Therapy Improves Pregnancy Outcomes after Surgery for Endometriosis-Associated Infertility: A Multicenter Randomized Double-blind Placebo Parallel Controlled Clinical Trial显示文摘Objective:To evaluate the efficacy and safety of Chinese medicine(CM)improving pregnancy outcomes after surgery for endometriosis-associated infertility.Methods:A multicenter,randomized,doubleblind placebo parallel controlled clinical trial was designed.A total of 202 patients who had laparoscopy for endometriosis-associated infertility with qi stagnation and blood stasis syndrome were included and randomly divided into the CM treatment group and placebo control group at a ratio of 1:1 using a central block randomization from May 2014 to September 2017,101 patients in each group.The two groups received continuous intervention at 1–5 days after surgery,for 6 menstrual cycles.Before ovulation,the CM group was treated Huoxue Xiaoyi Granule(活血消异颗粒);after ovulation,Bushen Zhuyun Granule(补肾助孕颗粒)was involved.The control group was treated with placebo.Transvaginal ultrasonography was performed every menstrual cycle during the treatment,and female hormone levels in the follicular and luteal phases were measured during the 1 st,3 rd and 6 th menstrual cycles.The analysis was continued until pregnancy.The primary outcomes were clinical pregnancy rate and pregnancy outcome,and the secondary outcomes were follicular development and endometrial receptivity.Safety evaluations were performed before and after treatment.Results:(1)Clinical pregnancy and live birth rates:the clinical pregnancy and live birth rates of the CM group were significantly higher than those of the placebo group[44.6%(45/101)vs.29.7%(30/101),34.7%(35/101)vs.20.8%(21/101),both P<0.05].(2)Follicle development:the incidence of dominant follicles,rate of cumulative cycle ovulation,and rate of cumulative cycle mature follicle ovulation were significantly higher in the CM group than those in the placebo group[93.8%(350/373)vs.89.5%(341/381),80.4%(275/342)vs.69.1%(253/366),65.8%(181/275)vs 56.1%(142/253),P<0.05 or P<0.01]).The incidence of cumulative cycle luteinized unruptured follicle syndrome was significantly lower in the CM group than in the placebo group[11.7%(40/342)vs.17.8%(65/366),P<0.05).(3)Endometrial receptivity:after treatment,both endometrial types and endometrial blood flow types in the CM group were mainly types A and B,while those in the placebo group were mainly types B and C,with a significant difference between the two groups(both P<0.05).(4)Adverse events:the incidence of adverse events between the two groups was not significantly different(P>0.05).Conclusion:Strategies for activating blood circulation-regulating Gan(Liver)-tonifying Shen(Kidney)sequential therapy can effectively improve the clinical pregnancy rate and live birth rate of endometriosis-associated infertility with qi stagnation and blood stasis after laparoscopy,improve follicular development,promote ovulation,improve endometrial receptivity,while being a safe treatment option.(Trial registration No.NCT02676713).ZHAO Rui-hua LIU Yong LU Dan WU Ying WANG Xiao-yun LI Wei-li ZENG Cheng MENG Qing-wei LIAN Feng-mei ZHOU Jun SHI Yun SUN Wei-wei HAN Qian TANG Yi SHI Guang 2020Chinese Journal of Integrative Medicine2020,26,2:13

16Lithofacies paleogeography mapping and reservoir prediction in tight sandstone strata: A case study from central Sichuan Basin, China显示文摘Sand-rich tight sandstone reservoirs are potential areas for oil and gas exploration. However, the high ratio of sandstone thickness to that of the strata in the formation poses many challenges and uncertainties to traditional lithofacies paleogeography mapping. Therefore, the prediction of reservoir sweet spots has remained problematic in the field of petroleum exploration. This study provides new insight into resolving this problem, based on the analyses of depositional characteristics of a typical modern sand-rich formation in a shallow braided river delta of the central Sichuan Basin, China. The varieties of sand-rich strata in the braided river delta environment include primary braided channels,secondary distributary channels and the distribution of sediments is controlled by the successive superposed strata deposited in paleogeomorphic valleys. The primary distributary channels have stronger hydrodynamic forces with higher proportions of coarse sand deposits than the secondary distributary channels. Therefore, lithofacies paleogeography mapping is controlled by the geomorphology, valley locations, and the migration of channels. We reconstructed the paleogeomorphology and valley systems that existed prior to the deposition of the Xujiahe Formation. Following this, rock-electro identification model for coarse skeletal sand bodies was constructed based on coring data. The results suggest that skeletal sand bodies in primary distributary channels occur mainly in the valleys and low-lying areas,whereas secondary distributary channels and fine deposits generally occur in the highland areas. The thickness distribution of skeletal sand bodies and lithofacies paleogeography map indicate a positive correlation in primary distributary channels and reservoir thickness. A significant correlation exists between different sedimentary facies and petrophysical properties. In addition, the degree of reservoir development in different sedimentary facies indicates that the mapping method reliably predicts the distribution of sweet spots. 17Yushu earthquake synergic analysis using multimodal SAR datasets显示文摘The large earthquake measuring 7.1 on the Richter scale that occurred in Yushu County,Qinghai Province,China,on April 14,2010,caused a huge loss of life and major damage to people's properties.Using three modal synthetic-aperture radar(SAR) datasets,wide-swath SAR,interferometric SAR and polarimetric SAR,the following studies were conducted:(1) From Radarsat-2 wide swath SAR data,the regional geological structures and lithologic distribution characteristics of the Yushu earthquake were analyzed.(2) Using advanced land observing satellite/phased array L-band interferometric SAR data,the coseismic deformation field of the Yushu earthquake was obtained,and the size and spatial distribution of deformation were studied combining with the result of regional geological background analysis.(3) From Radarsat-2 polarimetric SAR data,the H-α-ρ method was used to extract the spatial distribution of building collapse caused by the earthquake in the Yushu urban area,and the reasons for the severe earthquake damage to buildings are also discussed and assessed.The study results demonstrate the effectiveness and important potential of using multimodal SAR data for synergic analysis and assessment of earthquake disasters.GUO HuaDong WANG XinYuan LI XinWu LIU Guang ZHANG Lu YAN ShiYong 2010Chinese Science Bulletin2010,55,31:9
18Synergistic effects of elevated homocysteine level and abnormal blood lipids on the onset of stroke显示文摘Hyperhomocysteinemia and abnormal blood lipids are independent risk factors for stroke.However,whether both factors exert a synergistic effect in the onset of stroke remains unclear.The present study is a retrospective analysis of 2 089 cases of stroke and 2 089 control cases of simple intervertebral disk protrusion using a paired multivariate logistic regression method.Adjusting for known confounding variables including the patients’age,gender,smoking status,alcohol consumption status,patient and family medical history,and clinical biochemical indices,elevated homocysteine level was related to the onset of stroke.Patients with elevated homocysteine levels and abnormal blood lipids showed a 40.9%increase in the risk for stroke compared to patients with normal homocysteine levels and blood lipids(odds ratio 1.409;95%confidence interval1.127–1.761).These results indicate that elevated homocysteine and abnormal blood lipids exert synergistic effects in the onset of stroke.Patients with elevated homocysteine levels and abnormal blood lipids are predisposed to stroke.Lu Hao Liming Chen Xiaoyong Sai Zhefeng Liu Guang Yang Rongzeng Yan Lili Wang Caiyun Fu Xuan Xu Zhenzhen Cheng Qiang Wu Shuzhang Li 2013Neural Regeneration Research2013,8,31:9
19Large-eddy simulation of shock-wave/turbulent boundary layer interaction with and without Spark Jet control显示文摘The efficiency and mechanism of an active control device ‘‘Spark Jet' and its application in shock-induced separation control are studied using large-eddy simulation in this paper.The base flow is the interaction of an oblique shock-wave generated by 8° wedge and a spatially-developing Ma = 2.3 turbulent boundary layer.The Reynolds number based on the incoming flow property and the boundary layer displacement thickness at the impinging point without shock-wave is20000.The detailed numerical approaches were presented.The inflow turbulence was generated using the digital filter method to avoid artificial temporal or streamwise periodicity.The numerical results including velocity profile,Reynolds stress profile,skin friction,and wall pressure were systematically validated against the available wind tunnel particle image velocimetry(PIV) measurements of the same flow condition.Further study on the control of flow separation due to the strong shock-viscous interaction using an active control actuator ‘‘Spark Jet' was conducted.The single-pulsed characteristic of the device was obtained and compared with the experiment.Both instantaneous and time-averaged flow fields have shown that the jet flow issuing from the actuator cavity enhances the flow mixing inside the boundary layer,making the boundary layer more resistant to flow separation.Skin friction coefficient distribution shows that the separation bubble length is reduced by about 35% with control exerted.Yang Guang Yao Yufeng Fang Jian Gan Tian Li Qiushi Lu Lipeng 2016Chinese Journal of Aeronautics2016,29,3:8
20Status and development of high-power laser facilities at the NLHPLP显示文摘In this paper, we review the status of the multifunctional experimental platform at the National Laboratory of High Power Laser and Physics(NLHPLP). The platform, including the SG-II laser facility, SG-II 9th beam, SG-II upgrade(SG-II UP) facility, and SG-II 5 PW facility, is operational and available for interested scientists studying inertial confinement fusion(ICF) and a broad range of high-energy-density physics. These facilities can provide important experimental capabilities by combining different pulse widths of nanosecond, picosecond, and femtosecond scales. In addition, the SG-II UP facility, consisting of a single petawatt system and an eight-beam nanosecond system, is introduced including several laser technologies that have been developed to ensure the performance of the facility.
