1Canopy interception loss in a Pinus sylvestris var. mongolica forest of Northeast China显示文摘Pinus sylvestris var. mongolica is one of the main species to be afforested in deserts of China. But little work has been carried out on the canopy interception loss of this plant species. For researching the canopy interception loss of a natural P. sylvestris forest, we observed the gross precipitation, gross snowfall, throughfall and stemflow in a sample plot at the Forest Ecosystem Research Station of Mohe in the Great Khingan Mountains of Northeast China from July 2012 to September 2013. Considering the spatial variability of the throughfall, we increased the area rather than the number of collector and randomly relocated them once a week. The results demonstrated that the throughfall, stemflow, and derived estimates of rainfall and snowfall interception loss during the main rainy and snowy seasons were 77.12%±5.70%, 0.80%, 22.08%±5.51% and 21.39%±1.21% of the incident rainfall or snowfall, respectively. The stemflow didn't occur unless the accumulated rainfall reached up to 4.8 mm. And when the gross precipitation became rich enough, the stemflow increased with increasing tree diameters. 3A transgene-free method for rapid and efficient generation of precisely edited pigs without monoclonal selection显示文摘Gene-edited pigs for agricultural and biomedical applications are typically generated using somatic cell nuclear transfer(SCNT).However, SCNT requires the use of monoclonal cells as donors, and the time-consuming and laborious monoclonal selection process limits the production of large populations of gene-edited animals. Here, we developed a rapid and efficient method named RE-DSRNP(reporter RNA enriched dual-sg RNA/CRISPR-Cas9 ribonucleoproteins) for generating gene-edited donor cells. RE-DSRNP takes advantage of the precise and efficient editing features of dual-sg RNA and the high editing efficiency, low off-target effects, transgene-free nature, and low cytotoxic characteristics of reporter RNA enriched RNPs(CRISPR-Cas9ribonucleoproteins), thus eliminating the need for the selection of monoclonal cells and thereby greatly reducing the generation time of donor cells from 3–4 weeks to 1 week, while also reducing the extent of apoptosis and chromosomal aneuploidy of donor cells. We applied RE-DSRNP to produce cloned pigs bearing a deletion edit of the wild-type p53-induced phosphatase 1(WIP1)gene: among 32 weaned cloned pigs, 31(97%) carried WIP1 edits, and 15(47%) were homozygous for the designed fragment deletion, and no off-target event was detected. The WIP1 knockout(KO) pigs exhibited male reproductive disorders, illustrating the utility of RE-DSRNP for rapidly generating precisely edited animals for functional genomics and disease research. 4Molecular mapping of QTLs for root response to phosphorus deficiency at seedling stage in wheat (Triticum aestivum L.)显示文摘Phosphorus (P) deficiency in the soil is one of the major abiotic stresses that limit plant growth and crop productivity throughout the world. Development of cultivars with improved P-deficiency tolerance is an efficient strategy for sustainable agriculture. Plant roots play an important role in crop growth and development, especially in nutrient uptake and improvement of P-efficiency. Mapping quantitative trait loci (QTLs) for root traits and their response to low P stress at seedling stage will facilitate the development of P-efficient wheat cultivars. In this study, 30 QTLs (LOD>2.0) were mapped for the three root traits, such as root length, root number and root dry matter under different P supply conditions and their response to P-stress. These QTLs were distributed on 14 chromosomes, with each of the 5 QTLs explaining more than 10% phenotype variance. Analyses showed that root traits and their response to P-deficiency were controlled by different QTLs. 6Natural continuous influent nitrifier immigration effects on nitrification and the microbial community of activated sludge systems显示文摘Two sequencing batch reactors(SBRs) were operated for 100 days under aerobic conditions,with one being fed with unsterilized municipal wastewater(USBR), and the other fed with sterilized municipal wastewater(SSBR). Respirometric assays and fluorescence in situ hybridization(FISH) results show that active nitrifiers were present in the unsterilized influent municipal wastewater. The maximum ammonia utilization rate(AUR) and nitrite utilization rate(NUR) of the unsterilized influent were 0.32 ± 0.12 mg NH4+-N/(L·hr) and0.71 ± 0.18 mg NO2--N/(L·hr). Based on the maximum utilization rates, the estimated seeding intensity for the ammonia oxidizing bacteria(AOB) and nitrite oxidizing bacteria(NOB) of the USBR was 0.08 g AOB/(g AOB·day) and 0.20 g NOB/(g NOB·day) respectively. The fraction of nitrifiers/total bacteria in the influent was 5.35% ± 2.1%, the dominant AOB was Nitrosomonas spp., Nitrosococcus mobilis hybridizated with Nsm156, and the dominant NOB was Nitrospira hybridizated with Ntspa662. The influent nitrifiers potentially seeded the activated sludge of the bioreactor and hence demonstrated a mitigation of the acclimatization times and instability during start-up and early operation. The AUR and NUR in the USBR was 15% and 13% higher than the SSBR respectively during the stable stage, FISH results showed that nitrifiers population especially the Nitrospira in the USBR was higher than that in the SSBR. 7Speciation of organic phosphorus in a sediment profile of Lake Taihu I:Chemical forms and their transformation显示文摘Organic phosphorus(nonreactive P,NRP)is a major component of P in sediments,but information about its chemical forms and dynamic transformation is limited.The chemical forms and dynamic behaviors of NRP in a sediment profile from Lake Taihu,a freshwater and eutrophic lake in China,were investigated.Five forms of NRP in the sediments were extracted based on a chemical fractionation technique,including easily labile NRP(NaHCO3-NRP),reactive metal oxide-bound NRP(HCl-NRP),humic acid-associated NRP(NaOH-NRPHA),fulvic acid-associated NRP(NaOH-NRPFA)and residual NRP(Res-TP).There were notable transformations with increasing sediment depth from the labile NaHCO3-NRP and NaOH-NRP pools to the recalcitrant HCl-NRP and Res-TP pools,which caused the NRP to become increasingly recalcitrant as the early diagenetic processes proceeded.Further analyses showed that the relative changes in contents of organic matter and reactive Fe oxides in the sediment profile triggered a competition for binding NRP fractions and led to the transformation of NRP.The results highlighted the importance of abiotic processes in regulating the diagenesis of organic P and its stability in sediments.Di Xu Shiming Ding Bin Li Xiuling Bai Chengxin Fan Chaosheng Zhang 2013Journal of Environmental Sciences2013,25,4:7
8Optimization of a new animal glue binder system cured by CO_2 for use in foundry显示文摘A new sand binder system cured by CO2 was prepared based on the animal bone glue.To overcome the disadvantages of animal glue such as agglomeration at room temperature,high energy consumption and low efficiency,an alkaline decomposition process was selected,and certain modifier was used to modify the performance of the animal glue binder.For the alkaline decomposition,NaOH was used as the catalyst with an addition of 4wt.% to the animal glue.A modifier was determined through the orthogonal experiment with a weight ratio of glycerin:glycol:dextrin:animal glue = 9:16:15:100,and the optimal modification reaction should be performed at 75 ℃ with a reaction time of 90 min.Ca(OH)2 was used as a promoter;the optimal CO2 gas flux blowing into the sand was 0.7 m3.h-1 for a duration of 60s under the experimental conditions.Results show that an original strength above 0.7 MPa and a final strength about 4.2 MPa can be achieved,which could meet the requirement of rapid moulding and core-making for foundry.The new binder was characterized and analyzed by means of IR,and the modification and CO2-cured mechanisms of this animal glue binder were also discussed.Liu Weihua Zhang Yilin Li Yingmin Liu Xiuling 2012China Foundry2012,9,4:5

12Human endogenous retrovirus W env increases nitric oxide production and enhances the migration ability of microglia by regulating the expression of inducible nitric oxide synthase显示文摘Human endogenous retrovirus W env(HERV-W env) plays a critical role in many neuropsychological diseases such as schizophrenia and multiple sclerosis(MS). These diseases are accompanied by immunological reactions in the central nervous system(CNS). Microglia are important immunocytes in brain inflammation that can produce a gasotransmitter – nitric oxide(NO). NO not only plays a role in the function of neuronal cells but also participates in the pathogenesis of various neuropsychological diseases. In this study, we reported increased NO production in CHME-5 microglia cells after they were transfected with HERV-W env. Moreover, HERV-W env increased the expression and function of human inducible nitric oxide synthase(hi NOS) and enhanced the promoter activity of hi NOS. Microglial migration was also enhanced. These data revealed that HERV-W env might contribute to increase NO production and microglial migration ability in neuropsychological disorders by regulating the expression of inducible NOS. 13Human leukemia antigen-A0201-restricted epitopes of humanendogenous retrovirus W family envelope(HERV-W env)inducestrong cytotoxic T lymphocyte responses显示文摘Human endogenous retrovirus W family(HERV-W) envelope(env) has been reported to be related to several human diseases, including autoimmune disorders, and it could activate innate immunity.However, there are no reports investigating whether human leukemia antigen(HLA)-A~*0201^+restriction is involved in the immune response caused by HERV-W env in neuropsychiatric diseases. In the present study, HERV-W env-derived epitopes presented by HLA-A~*0201 are described with the potential for use in adoptive immunotherapy. Five peptides displaying HLAA~*0201-binding motifs were predicted using SYFEPITHI and BIMAS, and synthesized. A CCK-8 assay showed peptides W, Q and T promoted lymphocyte proliferation. Stimulation of peripheral blood mononuclear cells from HLA-A~*0201^+ donors with each of these peptides induced peptidespecific CD8^+ T cells. High numbers of IFN-γ-secreting T cells were also detectable after several weekly stimulations with W, Q and T. Besides lysis of HERV-W env-loaded target cells, specific apoptosis was also observed. These data demonstrate that human T cells can be sensitized toward HERV-W env peptides(W, Q and T) and, moreover, pose a high killing potential toward HERV-W env-expressing U251 cells. In conclusion, peptides W Q and T, which are HERV-W env antigenic epitopes, have both antigenicity and immunogenicity, and can cause strong T cell immune responses. Our data strengthen the view that HERV-W env should be considered as an autoantigen that can induce autoimmunity in neuropsychiatric diseases, such as multiple sclerosis and schizophrenia. 15TEM and HRTEM Study of Microstructures in Omphacite from UHP Eclogites at Shima,Dabie Mountains,China显示文摘Transmission electron microscope (TEM) and high resolution transmission electron microscope (HRTEM) analyses have been performed on omphacite from ultra-high pressure (UHP) eclogites at the locality of Shima, Dabie Mountains, China. TEM reveals that the microstructures consist dominantly of dislocation substructures, including free dislocations, loops, tiltwalls, dislocation tangles and subboundaries. They were produced by high-temperature ductile deformation, of which the main mechanism was dislocation creep. Antiphase domain (APD) boundaries are common planar defects; an age of 470±6 Ma for UHP eclogite formation has been obtained from the equiaxial size of APDs in ordered omphacites from Shima, coincident with ages given by single-zircon U-Pb dating (471±2 Ma). HRTEM reveals C2/c and P2ln space groups in different parts of one single omphacite crystal, and no exsolution is observed in the studied samples, which is attributed to rapid cooling. 16Improving forest burn severity estimations with partial least squares regression and orthogonal signal correction methods in Daxing'an Mountains,China显示文摘Several indices and simple empirical models and ratios of single band from pre-and post-fire Landsat images have been developed to estimate and/or map burn severity.However,these models and indices are usually site-,time-and vegetation-dependent and their applications are limited.The Daxing'an Mountains range has the largest forested area in China and is prone to wildfires.Whether or not the existing models can effectively characterize the burn severity over a large region is unclear.In this study,we used the orthogonal signal correction method based on partial least squares regression(PLSR)to select those variables that better interpret the variance of burn severity.A new index and other commonly used indices were used to construct a new,multivariate PLSR model which was compared with the popular single variable models,according to three assessment indices:relative root mean square error(RMSE%),relative bias(R E%)and Nash–Sutcliffe efficiency(NSE%).The results indicate that the multivariate PLSR model performed better than the other single variable models with higher NSE%(68.2%vs.67.8%)and less RE%(3.7%vs.-8.7%),while achieving almost the same R MSE%.We also discuss the spectral characteristics of the four selected variables for constructing the multivariate PLSR model and their correlation with the field burn severity data.The new model developed from this study should help to better understand the patterns of forest burn severity and assist in vegetation restoration efforts in the region.Cunyong Ju Tijiu Cai Wenhong Li Ge Sun Chengliang Lei Xueying Di Xiuling Man 2021Journal of Forestry Research2021,32,3:2
17Transonic flow of moist air around an NACA 0012 airfoil with non-equilibrium condensation显示文摘The classical condensation model of water vapor is coupled with the Euler equations to calculate transonic flows of moist air with non-equilibrium condensation. By means of this model, numerical computations are implemented to investigate the aerodynamic characteristics of an NACA 0012 airfoil in transonic flows of moist air at various angles of attack and relative humidities, and the results are compared with those in dry air flows. For different angles of attack considered at 50% relative humidity, the lift decreases 30%—40%. The pressure drag increases when the angle of attack is smaller than 1.4° and decreases when higher than 1.4°. At zero angle of attack, with the relative humidity rising from zero to 90%, the pressure drag increases exponentially. At 90% relative humidity, the pressure drag increases 160%, and self-oscillation takes place periodically and alternately over the upper and lower surfaces of the airfoil. 19The oral microbiome of pregnant women facilitates gestational diabetes discrimination显示文摘The oral microbiota plays an important role in the development of various diseases,whereas its association with gestational diabetes mellitus (GDM) remains largely unclear.The aim of this study is to identify biomarkers from the oral microbiota of GDM patients by analyzing the microbiome of the saliva and dental plaque samples of 111 pregnant women.We find that the microbiota of both types of oral samples in GDM patients exhibits differences and significantly varies from that of patients with periodontitis or dental caries.Using bacterial biomarkers from the oral microbiota,GDM classification models based on support vector machine and random forest algorithms are constructed.The area under curve (AUC) value of the classification model constructed by combination of Lautropia and Neisseria in dental plaque and Streptococcus in saliva reaches 0.83,and the value achieves a maximum value of 0.89 by adding clinical features.These findings suggest that certain bacteria in either saliva or dental plaque can effectively distinguish women with GDM from healthy pregnant women,which provides evidence of oral microbiome as an informative source for developing noninvasive biomarkers of GDM.Xiaoqing Li Jiayong Zheng Xiuling Ma Bing Zhang Jinyang Zhang Wenhuan Wang Congcong Sun Yeping Wang Jianqiong Zheng Haiying Chen Jiejing Tao Hai Wang Fengyi Zhang Jinfeng Wang Hongping Zhang 2021Journal of Genetics and Genomics2021,48,1:2
