| 1 | Blocking FSH inhibits hepatic cholesterol biosynthesis and reduces serum cholesterol显示文摘Menopause is associated with dyslipidemia and an increased risk of cardio-cerebrovascular disease.The classic view assumes that the underlying mechanism of dyslipidemia is attributed to an insufficiency of estrogen.In addition to a decrease in estrogen, circulating follicle-stimulating hormone (FSH)levels become elevated at menopause.In this study,we find that blocking FSH reduces serum cholesterol via inhibiting hepatic cholesterol biosynthesis.First,epidemiological results show that the serum FSH levels are positively correlated with the serum total cholesterol levels,even after adjustment by considering the effects of serum estrogen,in addition,the prevalence of hypercholesterolemia is significantly higher in peri-menopausal women than that in premenopausal women.Furthermore,we generated a mouse model of FSH elevation by intraperitoneally injecting exogenous FSH into ovariectomized (OVX)mice,in which a normal level of estrogen (E2)was maintained by exogenous supplementation. Consistently,the results indicate that FSH,independent of estrogen,increases the serum cholesterol level in this mouse model. | 4 | p53-dependent upregulation of PIG3 transcription by γ-ray irradiation and its interaction with KAP1 in responding to DNA damage显示文摘PIG3 (p53-inducible gene 3), originally identified as one of a set of genes induced by p53 before the onset of apoptosis, was assumed to contribute to early cellular response to DNA damage. Here, we studied the relation between p53 status and the increased expression of PIG3 by ionizing radiation (IR), and the related clues regarding the involvement of PIG3 in the cellular response to IR-induced DNA damage signaling. We demonstrated that the pentanucleotide microsatellite sequence was responsible for the p53-dependent induction of PIG3 transcription after irradiation, while sequence upstream of PIG3 promoter could maintain the basal level of expression which was not inducible by irradiation. The interaction of PIG3 and the KRAB-ZFP-associated protein 1 (KAP1), a DNA damage response protein, was revealed. PIG3 nucleus foci were formed 15 min after γ-ray irradiation, and which were found to partially colocalize with the phospho-KAP-1 foci as well as γ-H2AX foci. Although the lac operator tagged EGFP based reporter system revealed that PIG3 does not remodel chromatin in large scale in the cells under normal growing condition, it indeed prompted the chromatin relaxation in the cellular response to DNA damage signaling. | 6 | Intrauterine growth restriction alters nutrient metabolism in the intestine of porcine offspring显示文摘Background:Intrauterine growth restriction(IUGR)has negative impacts on the postnatal survival,growth and development of humans and animals,with not only on newborns but also adulthood.However,the characteristics for nutrient digestion and absorption in IUGR offspring are still largely unknown.Therefore,the normal birth weight(NBW)and IUGR growing pigs were used in this study to investigate their differences in nutrient utilization,with an expectition for further nutritional optimization of the IUGR offspring during their later life.Methods:Twelve IUGR and 12 NBW growing pigs were fitted with catheters in their portal vein to measure blood flow rate as well as nutrients and metabolites in plasma.The digestibilities of nutrients in different intestinal segments,and bacterial fermentation in the large intestine were examined to reveal the characteristics of nutrients utilization in IUGR versus NBW pigs.Results:The rate of portal venous blood flow did not differ beween IUGR and NBW pigs.Plasma concentrations of total cholesterol,triglycerides and glucose were much lower but those of urea were higher in the portal vein of IUGR pigs,compared with the NBW pigs.The ileal digestibility of dry matter,gross energy and starch were lower in IUGR pigs than in NBW pigs.IUGR increased hindgut microbial diversity and bacterial fermentation activity in the caecum.In vitro cross-fermentation of ileal digesta by caecal microbes of NBW and IUGR pigs showed that gas production was much higher for IUGR ileal digesta regardless of the source of caecal inocula.Conclusion:IUGR impairs the nutrient digestion and absorption in small intestine,reduces caecal microbial diversity and promotes bacterial fermentation in the large intestine during the growing phase.These findings aid in our understanding of nutrient metabolism in IUGR pigs and provide the basis for future nutritional interventions. | Tiantian Li Shimeng Huang Long Lei Shiyu Tao Yi Xiong Guoyao Wu Jie Hu Xiongkun Yuan Shengjun Zhao Bin Zuo Hongjian Yang Yingping Xiao Gang Lin Junjun Wang | 2021 | Journal of Animal Science and Biotechnology2021,12,2: | 2 |
| 7 | Dynamic changes of postprandial plasma metabolites after intake of corn-soybean meal or casein-starch diets in growing pigs显示文摘Background: Postprandial nutrients utilization and metabolism of a certain diet is a complicated process. The metabolic feature of pigs after intake of corn-soybean meal or casein-starch diets are largely unknown. Therefore,this study was conducted to investigate the dynamic postprandial changes of plasma metabolic profile using growing pigs using metabolomics.Methods: Twenty-four growing pigs with average initial body weight(BW) about 30 kg were placed in metabolic cages and then fitted with precaval vein catheters. Pigs were fed daily 4% of initial body weight. Two experimental diets were included:(i) a starch-casein based purified diet(PD) and(ii) a common corn-soybean meal diet(CD).Plasma was collected before feeding and 0.5 h, 1 h, 2 h, 4 h, 8 h after feeding.Results: In both diets, compared to prior to feeding, the concentrations of glucose, most amino acids, metabolites such as 5-aminopentanoic acid, pipecolic acid, ornithine and 5-hydroxy-L-tryptophan were significantly increased in plasma during the first hour, whereas the concentrations of plasma triglycerides, glutamate, glycine, palmitelaidic acid, 13-HODE and oleic acid were decreased in the first hour. Compared with PD group, concentration of plasma leucine and isoleucine declined at 30 min in CD group. Plasma linoleic acid, sphingosine and many dipeptides were significantly higher in pigs fed CD.Conclusion: Most significant metabolic changes occurred during the first hour after feeding and then became relatively stable after 2 h in both diets. | 8 | DARPA Robotics Grand Challenge Participation and Ski-Type Gait for Rough-Terrain Walking显示文摘In this paper, we briefly introduce the history of the Defense Advanced Research Projects Agency(DARPA) Grand Challenge programs with particular focus on the 2012 Robotics Challenge. As members of team DRC-HUBO, we propose different approaches for the Rough-Terrain task, such as enlarged foot pedals and a transformation into quadruped walking. We also introduce a new gait for humanoid robot locomotion to improve stability performance, called the Ski-Type gait. We analyze the stability performance of this gait and use the stability margin to choose between two candidate step sequences, Crawl-1 and Crawl-2. Next, we perform a force/torque analysis for the redundant closedchain system in the Ski-Type gait, and determine the joint torques by minimizing the total energy consumption. Based on the stability and force/torque analysis, we design a cane length to support a feasible and stable Crawl-2 gait on the HUBO2 humanoid robot platform. Finally, we compare our experimental results with biped walking to validate the SkiType gait. | 10 | Recent progress on the hydrogen storage properties of ZrCo-based alloys applied in International Thermonuclear Experimental Reactor(ITER)显示文摘Under the development of International Thermonuclear Experimental Reactor(ITER) system aimed at realizing the controllable fusion reaction to solve the energy crisis fundamentally, there is an urgent need to find an appropriate material for tritium handling. ZrCo alloy is considered to be a promising candidate for the storage and delivery of hydrogen isotopes due to the favorable characteristics such as low plateau pressure for absorption, high dissociation pressure at moderate temperature and better ability of trapping ~3He. However, the hydrogen induced disproportionation and the slower recovery/deliverty rate of ZrCo-based alloys have limited their further application in ITER system. This review summarizes the efforts towards enhancing the hydrogen storage properties of ZrCo-based alloys including element substitution, surface modification, disproportionation mechanism investigation and the isotope effect study. Element substitution and surface modification play positive role to improve the anti-disproportionation ability and kinetic property of the alloys. However, the ZrCo-based alloys require to be further modified by more attempts such as new composition, novelty modification method or catalyst addition in order to better satisfy the application demands for tritium handling.
