1Biomechanical and biological responses of periodontium in orthodontic tooth movement:up-date in a new decade显示文摘Nowadays,orthodontic treatment has become increasingly popular.However,the biological mechanisms of orthodontic tooth movement(OTM)have not been fully elucidated.We were aiming to summarize the evidences regarding the mechanisms of OTM.Firstly,we introduced the research models as a basis for further discussion of mechanisms.Secondly,we proposed a new hypothesis regarding the primary roles of periodontal ligament cells(PDLCs)and osteocytes involved in OTM mechanisms and summarized the biomechanical and biological responses of the periodontium in OTM through four steps,basically in OTM temporal sequences,as follows:(1)Extracellular mechanobiology of periodontium:biological,mechanical,and material changes of acellular components in periodontium under orthodontic forces were introduced.(2)Cell strain:the sensing,transduction,and regulation of mechanical stimuli in PDLCs and osteocytes.(3)Cell activation and differentiation:the activation and differentiation mechanisms of osteoblast and osteoclast,the force-induced sterile inflammation,and the communication networks consisting of sensors and effectors.(4)Tissue remodeling:the remodeling of bone and periodontal ligament(PDL)in the compression side and tension side responding to mechanical stimuli and root resorption.Lastly,we talked about the clinical implications of the updated OTM mechanisms,regarding optimal orthodontic force(OOF),acceleration of OTM,and prevention of root resorption.Yuan Li Qi Zhan Minyue Bao Jianru Yi Yu Li 2021International Journal of Oral Science2021,13,2:15
2The Performance of Whole Genome Amplification Methods and Next-Generation Sequencing for Pre-Implantation Genetic Diagnosis of Chromosomal Abnormalities显示文摘Reliable and accurate pre-implantation genetic diagnosis(PGD) of patient's embryos by next-generation sequencing(NGS) is dependent on efficient whole genome amplification(WGA) of a representative biopsy sample. However, the performance of the current state of the art WGA methods has not been evaluated for sequencing. Using low template DNA(15 pg) and single cells, we showed that the two PCR-based WGA systems Sure Plex and MALBAC are superior to the REPLI-g WGA multiple displacement amplification(MDA) system in terms of consistent and reproducible genome coverage and sequence bias across the 24 chromosomes, allowing better normalization of test to reference sequencing data. When copy number variation sequencing(CNV-Seq) was applied to single cell WGA products derived by either Sure Plex or MALBAC amplification, we showed that known disease CNVs in the range of 3e15 Mb could be reliably and accurately detected at the correct genomic positions. 3Developing Agroforestry in Slopelands to Combat Non-point Pollution in China显示文摘The status of agricultural non-point source (NPS) pollution in China is quite austere owing to lots of factors, the characteristics of which include severe N, P eutrophications in water bodies, unsuitable structure of agriculture and the lack of reasonable management. It was well known that forest could play an important role to control soil & water erosion, uptake extra soil nutrients as well. However forest coverage is rather low in the headwaters region. The research on the causes of NSP pollution and the significance by developing agroforestry to deal with the problem is presented in the paper, as well as the function of agroforestry and the related measures of pollution controlling, e.g., riparian forest buffer zones building. As a conclusion, controlling the loss of soil and water is regarded as an important crucial action and major research direction in the field. 4Effects of Carboxymethyl-Chitosan on Wound Healing in vivo and in vitro显示文摘Effects of carboxymethyl-chitosan with different molecular weights on wound healing were investigated in vivo and in vitro.A second degree burn model was performed in rats and the accelerative effects of carboxymethyl-chitosan on wound repair were observed.Contents of transforming growth factor(TGF)-β1,interleukin(IL)-6 and matrix metalloproteinase(MMP)-1 in wounds were determined by enzyme-linked immunosorbent assay(ELISA).In vitro study evaluated the influence of carboxy-methyl-chitosan on cytokines secretion of fibroblasts and macrophages.In vivo results showed that carboxymethyl-chitosan effec-tively accelerated the wound healing process in burned rats(P<0.05).Levels of TGF-β1,IL-6 and MMP-1 in carboxymethyl-chitosan groups were significantly elevated,compared with control group(P<0.05).In vitro results indicated that carboxymethyl-chitosan significantly promoted the proliferation of fibroblasts and stimulated its IL-8 and IL-10 secretion at different incubation time,but it did not affect collagen secretion of fibroblasts.Carboxymethyl-chitosan enhanced phagocytosis ability of macrophages,and in-creased its tumor necrosis factor(TNF)-α secretion.In conclusion,carboxymethyl-chitosan promoted wound healing by activating macrophages,accelerating fibroblasts growth and exerting considerable effects on the secretion of a series of cytokines.PENG Sikai LIU Wanshun HAN Baoqin CHANG Jing LI Minyu ZHI Xuan 2011Journal of Ocean University of China2011,10,4:7
5Effects of grazing disturbance of spatial distribution pattern and interspecies relationship of two desert shrubs显示文摘Grazing significantly affects the distribution,growth,and productivity of shrubs.In this study,we evalu-ated the effects of grazing disturbance on the spatial distri-bution patterns and interspecific relationships of two desert shrubs,Ammopiptanthus mongolicus and Sarcozygium xan-thoxylon.Three types of grazing conditions were considered,including enclosed area(EA),seasonal rotational grazing area(SRGA),and grazing area(GA)(100 m×100 m),in the West Ordos Nature Reserve of Inner Mongolia,China.The results showed that A.mongolicus and S.xanthoxylon populations were uniformly distributed at a small scale,and the distribution in EA and SRGA became gradually ran-dom.In GA,A.mongolicus population showed aggregated distribution but S.xanthoxylon population showed random distribution at a small scale. Moreover, both A. mongolicus and S. xanthoxylon populations at the 5-7 m scale showed random distribution. At the small and intermediate scales, the two species showed positive interspecific relationships of GA. However, no interspecific relationship was noted between the two species in EA and SRGA. A significant positive relationship (P < 0.01) was noted between the two species at 2-9 m and a negative relationship (P < 0.01) at 13-17 m scales in GA. Positive relationship (P < 0.01) was noted between the two species at 6-13 m scales and a sig-nificant negative relationship (P < 0.01) at 14-24 m scales in SRGA. The two species of desert shrubs showed posi-tive interspecific relationships at the small scale, and they showed negative relationships as the interspecific compe-tition intensified in the presence of grazing disturbance. When the grazing intensity exceeds a certain threshold, the interspecific relationships become weak. 7A strategy of screening and binding analysis of bioactive components from traditional Chinese medicine based on surface plasmon resonance biosensor显示文摘Elucidating the active components of traditional Chinese medicine(TCM)is essential for understanding the mechanisms of TCM and promote its rational use as well as TCM-derived drug development.Recent studies have shown that surface plasmon resonance(SPR)technology is promising in this field.In the present study,we propose an SPR-based integrated strategy to screen and analyze the major active components of TCM.We used Radix Paeoniae Alba(RPA)as an example to identify the compounds that can account for its anti-inflammatory mechanism via tumor necrosis factor receptor type 1(TNF-R1).First,RPA extraction was analyzed using an SPR-based screening system,and the potential active ingredients were collected,enriched,and identified as paeoniflorin and paeonol.Next,the affinity constants of paeoniflorin and paeonol were determined as 4.9 and 11.8 mM,respectively.Then,SPR-based competition assays and molecular docking were performed to show that the two compounds could compete with tumor necrosis factor-a(TNF-a)while binding to the subdomain 1 site of TNF-R1.Finally,in biological assays,the two compounds suppressed cytotoxicity and apoptosis induced by TNF-a in the L929 cell line.These findings prove that SPR technology is a useful tool for determining the active ingredients of TCM at the molecular level and can be used in various aspects of drug development.The SPR-based integrated strategy is reliable and feasible in TCM studies and will shed light on the elucidation of the pharmacological mechanism of TCM and facilitate its modernization.Diya Lv Jin Xu Minyu Qi DongyaoWang Weiheng Xu Lei Qiu Yinghua Li Yan Cao 2022Journal of Pharmaceutical Analysis2022,12,3:3

9Intercalation Assembly, Characterization and Luminescent Properties of Novel Supramolecular Composite Materials, Tb(Ⅲ) Complex with Tetrapodal Ligand-Montmorillonite显示文摘Two kinds of Tb(Ⅲ) complexes with tetrapodal ligand, [TbL(NO3)] 3+ and [TbL] 3+ (L: 1,1,1′,1′-tera(2-pyridinecarboxylester)-di(trimethylpropane)) were intercalated into the interlayer space of montmorillonite (MT) by ion exchange and coordination reaction of L with the Tb 3+ ion existing in the interlayer space of Tb-MT respectively. The obtained luminescent supramolecular composite materials, [TbL(NO3)] 2+ -MT and [TbL] 3+ -MT were characterized by elemental analysis, XRD, FT-IR, UV-vis and thermal analysis. At the same time, the luminescent properties of the materials were also studied. The results show that the intercalated materials with regular layered structure, good thermal stability and the interlayer spacing (d 001 ) approximates to the size of the complex ions which are located in the interlayer space of MT in the form of a monolayer. The material emit characteristic green emission of Tb 3+ strongly under excitation of ultraviolet light. It is also found that the thermal stabilities and relative luminescence intensities of the Tb 3+ species are distinctly improved after the Tb(Ⅲ) complexes are intercalated into the MT. 16Generation of patient-specific pluripotent stem cells and directed differentiation of embryonic stem cells for regenerative medicine显示文摘Embryonic stem(ES) cells are pluripotent cells that can give rise to derivatives of all three embryonic germ layers. Due to its characteristics, the patient-specific ES cells are of great potential for transplantation therapies. Several strategies can reprogramme somatic cells back to pluripotent stem cells: nuclear transfer, fusion with ES cells, treatment with cell extract and induction by specific factors. Considering the future clinical use, the differentiation from ES to neurons, cardiomyocytes and many other types of cells currently provide basic cognition and experience to regenerative medicine. 18Quasi-solid state dye-sensitized solar cells based on pyridine or imidazole containing copolymer chemically crosslinked gel electrolytes显示文摘Quasi-solid state dye-sensitized solar cells based on chemically crosslinking with backbone polymers of poly(vinylpyridine-co-acrylonitrile) (P(VP-co-AN)) or poly(vinylimidazole-co-acrylonitrile) (P(VIM-co- AN)) and diiodide compounds of I(CH2)6I or I(CH2CH2O)nCH2CH2I solidified EC/PC/KI/I2 gel electrolytes have been fabricated. The ionic conductivities and apparent diffusion coefficients of I3? of the electro- lytes and cell performances have been investigated. Providing chemically crosslinking points, pyridine or imidazole from the backbone polymers benefited the open circuit voltage and fill factor of the cells. Consequently, the overall energy conversion efficiencies of the quasi-solid DSSCs improved over 10% even near 20% from that of the liquid electrolyte before solidification. 20Comparative transcriptome analysis on candidate genes involved in lipid biosynthesis of developing kernels for three walnut cultivars in Xinjiang显示文摘Walnut(Juglans regia L.)is a good source of lipids and polyunsaturated fatty acids(PUFAs).In order to explore the biosynthesis molecular mechanism of oil accumulation and fatty acid(FA)synthesis in walnut,the samples at different development periods of three walnut cultivars,’Zhipi’(ZP),’Xinwu 417’(W417)and’Xinwen 81’(W81)were collected for transcriptomic analysis.The analysis of oil accumulation and FA profiles showed that the oil content in mature walnut kernel was nearly 70%,and over 90%of FAs were PUFAs.We identified 126 candidate genes including 64 genes for FA de novo synthesis,45 genes for triacylglycerol assembly,and 17 genes for oil bodies involved in lipid biosynthesis by RNA-sequencing.Ten key enzymes including ACCase,LACS6,LACS8,SAD,FAD2,FAD3,LPAAT1,DGAT2,PDAT2,and PLC encoded by 19 genes were highly associated with lipid biosynthesis.Quantitative PCR analysis further validated 9 important genes,and the results were well consistent with our transcriptomic data.Finally,5 important transcription factors including WRI1,ABI3,FUS3,PKL and VAL1 were identified,and their main regulatory genes might contain ACCase,KASII,LACS,FAD3 and LPAAT which were determined through correlation analysis of expression levels for 27 walnut samples.These findings will provide a comprehensive understanding and valuable information on the genetic engineering and molecular breeding in walnut.Wenqiang Wang Hao Wen Qiang Jin Wenjun Yu Gen Li Minyu Wu Hongjin Bai Lirong Shen Cuiyun Wu 2022Food Science and Human Wellness2022,11,5:0
