1Galangin induces apoptosis of hepatocellular carcinoma cells via the mitochondrial pathway显示文摘AIM:To investigate the mechanism by which galangin,a polyphenolic compound derived from medicinal herbs,induces apoptosis of hepatocellular carcinoma(HCC) cells.METHODS:The 3-(4,5-Dimethyl-thiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay was used to measure cell viability.Apoptosis was evaluated by in situ uptake of propidium iodide and Hoechst 33258 and was then detected by fluorescence microscopy.Protein expressions were detected by Western blotting.To confirm the apoptotic pathway mediated by galangin,cells were transfected by bcl-2 gene to overexpress Bcl-2 or siRNA to down-regulate Bcl-2 expression.RESULTS:Galangin(46.25-370.0 μmol/L) exerted an anti-proliferative effect,induced apoptosis,and decreased mitochondrial membrane potential in a dose and time-dependent manner.Treatment with galangin induced apoptosis by translocating the pro-apoptotic protein Bax to the mitochondria,which released apoptosis-inducing factor and cytochrome c into the cytosol.Overexpression of Bcl-2 attenuated galangin-induced HepG2 cell apoptosis,while decreasing Bcl-2 expression enhanced galangin-induced cell apoptosis.CONCLUSION:Our data suggests that galangin mediates apoptosis through a mitochondrial pathway,and may be a potential chemotherapeutic drug for the treatment of HCC.Hai-Tao Zhang Liu-Bo Lan Da-Hua Fan Kai-Dan Zhu Xiao-Yi Chen Min Wen Hui-Ming Liu Guangdong Province China Hui Luo 2010World Journal of Gastroenterology2010,16,27:32
2Expenditure and financial burden for the diagnosis and treatment of colorectal cancer in China:a hospital.based,multicenter,cross-sectional survey显示文摘Background: The increasing prevalence of colorectal cancer(CRC) in China and the paucity of information about relevant expenditure highlight the necessity of better understanding the financial burden and effect of CRC diagnosis and treatment. We performed a survey to quantify the direct medical and non-medical expenditure as well as the resulting financial burden of CRC patients in China.Methods: We conducted a multicenter, cross-sectional survey in 37 tertiary hospitals in 13 provinces across China between 2012 and 2014. Each enrolled patient was interviewed using a structured questionnaire. All expenditure data were inflated to the 2014 Chinese Yuan(CNY; 1 CNY = 0.163 USD). We quantified the overall expenditure and financial burden and by subgroup(hospital type, age at diagnosis, sex, education, occupation, insurance type, household income, clinical stage, pathologic type, and therapeutic regimen). We then performed generalized linear modeling to determine the factors associated with overall expenditure.Results: A total of 2356 patients with a mean age of 57.4 years were included, 57.1 % of whom were men; 13.9% of patients had stage I cancer; and the average previous-year household income was 54,525 CNY.The overall average direct expenditure per patient was estimated to be 67,408 CNY, and the expenditures for stage Ⅰ, Ⅱ, Ⅲ, and Ⅳ disease were 56,099 CNY, 59,952 CNY, 67,292 CNY, and 82,729 CNY, respectively. Non-medical expenditure accounted for 8.3%of the overall expenditure. The 1-year out-of-pocket expenditure of a newly diagnosed patient was 32,649 CNY, which accounted for 59.9% of their previous-year household income and caused 75.0% of families to suffer an unmanageable financial burden. Univariate analysis showed that financial burden and overall expenditure differed in almost all subgroups(P < 0.05), except for sex. Multivariate analysis showed that patients who were treated in specialized hospitals and those who were diagnosed with adenocarcinoma or diagnosed at a later stage were likely to spend more,whereas those with a lower household income and those who underwent surgery spent less(all P < 0.05).Conclusions: For patients in China, direct expenditure for the diagnosis and treatment of CRC seemed catastrophic,and non-medical expenditure was non-ignorable. 4Evaluating effective swath width and droplet distribution of aerial spraying systems on M-18B and Thrush 510G airplanes显示文摘Aerial spraying plays an important role in promoting agricultural production and protecting the biological environment due to its flexibility,high effectiveness,and large operational area per unit of time.In order to evaluate the performance parameters of the spraying systems on two fixed wing airplanes M-18B and Thrush 510G,the effective swath width and uniformity of droplet deposition under headwind flight were tested while the planes operated at the altitudes of 5 m and 4 m.The results showed that although wind velocities varied from 0.9 m/s to 4.6 m/s,and the directions of the atomizer switched upward and downward in eight flights,the effective swath widths were kept approximately at 27 m and 15 m for the M-18B and Thrush 510G,respectively,and the latter was more stable.In addition,through analyzing the coefficients of variation(CVs)of droplet distribution,it was found that the CVs of the M-18B were 39.57%,33.54%,47.95%,and 59.04% at wind velocities of 0.9,1.1,1.4 and 4.6 m/s,respectively,gradually enhancing with the increasing of wind speed;the CVs of Thrush 510G were 79.12%,46.19%,14.90%,and 48.69% at wind velocities of 1.3,2.3,3.0 and 3.4 m/s,respectively,which displayed the irregularity maybe due to change of instantaneous wind direction.Moreover,in terms of the CVs and features of droplet distribution uniformity for both airplanes in the spray swath,choosing smaller CV(20%-45%)as the standard of estimation,it was found that the Thrush 510G had a better uniform droplet distribution than the M-18B.The results provide a research foundation for promoting the development of aerial spraying in China.Zhang Dongyan Chen Liping Zhang Ruirui Xu Gang Lan Yubin Wesley Clint Hoffmann Wang Xiu Xu Min 2015International Journal of Agricultural and Biological Engineering2015,8,2:18
5Fine mapping and marker-assisted selection (MAS) of a low glutelin content gene in rice显示文摘Rice with low glutelin content is suitable as functional food for patients affected with diabetes and kidney failure. The fine mapping of the gene(s) responsible for low glutelin content will provide information regarding the distribution of glutelin related genes in rice genome and will generate markers for the selection of low glutelin rice varieties. Following an SDS-PAGE screen of rice germplasm from Taihu Valley of China, Japonica selection W3660 is identified to be a novel mutant characterized with low glutelin content. For fine mapping the mutant gene for low glutelin content, F2 and F3 populations were derived from a cross between W3660 and Jingrennuo. SDS-PAGE analysis of the total endosperm protein showed that the low glutelin content trait was controlled by a single dominant nuclear gene. Genetic mapping, using SSRs, located this gene to chromosome 2, in the region between SSR2-001/SSR2-004 and RM1358. The dis- tances of the two markers to the target gene were 1.1 cM and 3.8 cM respectively. By semi-quantitative RT-PCR analysis, the transcripts of GluB4/GluB5 genes located within the region do not change. However, GluB5 gene located proximal to SSR2-001/SSR2-004 was specifically reduced. SSR profiles of seven Japonica varieties were compared with that of W3660 for loci in the relevant genetic region. The markers SSR2-004 and RM1358 were used for marker- assisted selection. The selection efficiencies of SSR2-004 and RM1358 were 96.8% and 92.7% respectively. 8Milling surface roughness for 7050 aluminum alloy cavity influenced by nozzle position of nanofluid minimum quantity lubrication显示文摘In nanofluid minimum quantity lubrication(NMQL)milling of aviation aluminum alloy,it is the bottleneck problem to adjust the position parameters(target distance,incidence angle,and elevation angle)of the nozzle to improve the surface roughness of milling,which has large and uncontrollable errors.In this paper,the influence law of milling cutter speed,helical angle,and cavity shape on the flow field around the milling cutter was studied,and the optimal nozzle profile parameters were obtained.Using 7050 aluminum alloy as the workpiece material,the milling experiment of the NMQL cavity was conducted by utilizing cottonseed oil-based Al2 O3 nanofluid.Results show that the high velocity of the surrounding air flow field and the strong gas barrier could be attributed to high rotating velocities of the milling cutter.The incidence angle of the nozzle was consistent with the helical angle of the milling cutter,the target distance was appropriate at 25–30 mm,and the elevation angle was suitable at 60°–65°.The range and variance analyses of the signal-to-noise ratio of milling force and roughness were performed,and the chip morphology was observed and analyzed.The results show that the optimal combination of nozzle position parameters was the target distance of 30 mm,the incidence angle of 35°,and the elevation angle of 60°.Among these parameters,target distance had the largest impact on cutting performance with a contribution rate of more than 55%,followed by incidence angle and elevation contribution rate.Analysis by orthogonal experiment revealed that the nozzle position parameters were appropriate,and Ra(0.087 lm)was reduced by 30.4%from the maximum value(0.125 lm).Moreover,Rsm(0.05 mm)was minimum,which was 36%lower than that of the seventh group(Rsm=0.078 mm).Zhenjing DUAN Changhe LI Yanbin ZHANG Lan DONG Xiufang BAI Min YANG Dongzhou JIA Runze LI Huajun CAO Xuefeng XU 2021Chinese Journal of Aeronautics2021,34,6:10
9Determination of Lignin in Marine Sediment Using Alkaline Cupric Oxide Oxidation-Solid Phase Extraction-on-Column Derivatization-Gas Chromatography显示文摘Lignin serves as one of the most important molecular fossils for tracing Terrestrial Organic Matters (TOMs) in marine environment. Extraction and derivatization of lignin oxidation products (LOPs) are crucial for accurate quantification of lignin in marine sediment. Here we report a modification of the conventional alkaline cupric oxide (CuO) oxidation method, the modification consisting in a solid phase extraction (SPE) and a novel on-column derivatization being employed for better efficiency and reproducibility. In spiking blanks, recoveries with SPE for the LOPs are between 77.84% and 99.57% with relative standard deviations (RSDs) ranging from 0.57% to 8.04% (n=3), while those with traditional liquid-liquid extraction (LLE) are from 44.52% to 86.16% with RSDs being from 0.53% to 13.14% (n=3). Moreover, the reproducibility is greatly improved with SPE, with less solvent consumption and shorter processing time. The average efficiency of on-column derivatization for LOPs is 100.8% ± 0.68%, which is significantly higher than those of in-vial or in-syringe derivatization, thus resulting in still less consumption of derivatizing reagents. Lignin in the surface sediments sampled from the south of Yangtze River estuary, China, was determined with the established method. Recoveries of 72.66% to 85.99% with standard deviation less than 0.01mg/10g dry weight are obtained except for p-hydroxyben-zaldehyde. The lignin content Σ8 (produced from 10 g dry sediment) in the research area is between 0.231 and 0.587 mg. 10Anti-diabetic vanadyl complexes reduced Alzheimer's disease pathology independent of amyloid plaque deposition显示文摘Association of Alzheimer's disease(AD) with cerebral glucose hypometabolism, likely due to impairments of insulin signaling,has been reported recently, with encouraging results when additional insulin is provided to AD patients. Here, we tested the potential effects of the anti-diabetic vanadium, vanadyl(IV) acetylacetonate(VAC), on AD in vitro and in vivo models. The experimental results showed that VAC at sub-micromolar concentrations improved the viability of neural cells with or without increased β-amyloid(Aβ) burden; and in APP/PS1 transgenic mice, VAC treatment(0.1 mmol kg-1 d-1) preserved cognitive function and attenuated neuron loss, but did not reduce brain Aβ plaques. Further studies revealed that VAC attenuated Aβpathogenesis by(i) activation of the PPARγ-AMPK signal transduction pathway, leading to improved glucose and energy metabolism;(ii) up-regulation of the expression of glucose-regulated protein 75(Grp75), thus suppressing p53-mediated neuronal apoptosis under Aβ-related stresses; and(iii) decreasing toxic soluble Aβ peptides. 11Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis.However,little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins,including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types,and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels.The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies.Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry,providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations.Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu 2022Cell Research2022,32,1:8

13Strong ozone production at a rural site in the North China Plain: Mixed effects of urban plumes and biogenic emissions显示文摘Regional ozone(O_3) pollution has drawn increasing attention in China over the recent decade, but the contributions from urban pollution and biogenic emissions have not been clearly elucidated. To better understand the formation of the regional O_3 problem in the North China Plain(NCP), intensive field measurements of O_3 and related parameters were conducted at a rural site downwind of Ji'nan, the capital city of Shandong province, in the summer of 2013. Markedly severe O_3 pollution was recorded, with the O_3 mixing ratios exceeding the Chinese national ambient air quality standard on 28 days(a frequency of 78%) and with a maximum hourly value of 198 ppbv. Extensive regional transport of wellprocessed urban plumes to the site was identified. An observation-constrained chemical box model was deployed to evaluate in situ photochemical O_3 production on two episodes.The results show that the in situ formation accounted for approximately 46% of the observed O_3 accumulation, while the remainder(~54%) was contributed by regional transport of the O_3-laden urban plumes. The in situ ozone production was in a mixed controlled regime that reducing either NOx or VOCs would lead to a reduction of ozone formation. Biogenic VOCs played an important role in the local ozone formation. 14Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 15Source apportionment of PM_(2.5)light extinction in an urban atmosphere in China显示文摘Haze in China is primarily caused by high pollution of atmospheric fine particulates(PM2.5).However, the detailed source structures of PM2.5 light extinction have not been well established, especially for the roles of various organic aerosols, which makes haze management lack specified targets. This study obtained the mass concentrations of the chemical compositions and the light extinction coefficients of fine particles in the winter in Dongguan, Guangdong Province, using high time resolution aerosol observation instruments. We combined the positive matrix factor(PMF) analysis model of organic aerosols and the multiple linear regression method to establish a quantitative relationship model between the main chemical components, in particular the different sources of organic aerosols and the extinction coefficients of fine particles with a high goodness of fit(R^2= 0.953). The results show that the contribution rates of ammonium sulphate,ammonium nitrate, biomass burning organic aerosol(BBOA), secondary organic aerosol(SOA) and black carbon(BC) were 48.1%, 20.7%, 15.0%, 10.6%, and 5.6%, respectively. It can be seen that the contribution of the secondary aerosols is much higher than that of the primary aerosols(79.4% versus 20.6%) and are a major factor in the visibility decline. BBOA is found to have a high visibility destroying potential, with a high mass extinction coefficient, and was the largest contributor during some high pollution periods. 16Medical expenditure for esophageal cancer in China:a 10-year multicenter retrospective survey(2002-2011)显示文摘Background: Esophageal cancer is associated with substantial disease burden in China, and data on the economic burden are fundamental for setting priorities in cancer interventions. The medical expenditure for the diagnosis and treatment of esophageal cancer in China has not been fully quantified. This study aimed to examine the medical expenditure of Chinese patients with esophageal cancer and the associated trends.Methods: From 2012 to 2014, a hospital-based multicenter retrospective survey was conducted in 37 hospitals in 13 provinces/municipalities across China as a part of the Cancer Screening Program of Urban China. For each esophageal cancer patient diagnosed between 2002 and 2011, clinical information and expense data were extracted by using structured questionnaires. All expense data were reported in Chinese Yuan(CNY; 1 CNY = 0.155 USD) based on the2011 value and inflated using the year-specific health care consumer price index for China.Results: A total of 14,967 esophageal cancer patients were included in the analysis. It was estimated that the overall average expenditure per patient was 38,666 CNY, and an average annual increase of 6.27% was observed from 2002(25,111 CNY) to 2011(46,124 CNY). The average expenditures were 34,460 CNY for stage Ⅰ,39,302 CNY for stage Ⅱ,40,353 CNY for stage Ⅲ, and 37,432 CNY for stage IV diseases(P < 0.01). 18Sensory and chemical assessment of silver pomfret (Pampus argenteus) treated with Ginkgo biloba leaf extract treatment during storage in ice显示文摘This study investigated the physical(L*,a*,b*,texture profile analyses,pH),chemical(TVB-N,K value and TBA),microbiological,amino acid content,and flavor effects that Gingko biloba leaf extract(GBLE)had on silver pomfret(Pampus argenteus)stored at 4±1C in ice for 18 days.Fresh pomfret samples were obtained directly from the local fish market and transported to the laboratory with ice immediately.After being gutted,washed,filleted and trimmed in a water-ice mixture,samples were treated with different concentrations of GBLE(0.0 mg/mL,2.5 mg/mL,5.0 mg/mL,10.0 mg/mL)and packaged in Polyethylene bag,then stored in a refrigerator at 4±1C with ice.The results show that the shelf-life of untreated(0.0 mg/mL)pomfret samples was 8e9 days compared to 14e15 days for the GBLE1(2.5 mg/mL)treated group.The assessment results showed that different concentrations of GBLE had variable effects on preserving the texture parameters of acceptability limit,inhibit lipid oxidation,protein degradation,and microorganism growth.2.5 mg/mL of GBLE was the best for the preservation of pomfret during storage in ice.Therefore,there is potential use for GBLE as a preservative to extend the shelf-life of pomfret during chilled storage in ice.Weiqing Lan Xu Che Qiaoling Xu Ting Wang Ruoyuan Du Jing Xie Min Hou Han Lei 2018Aquaculture and Fisheries2018,3,1:5
