| 1 | Targeted therapy of gastrointestinal stromal tumours显示文摘Gastrointestinal stromal tumours(GISTs) are mesen-chymal neoplasms originating in the gastrointestinal tract, usually in the stomach or the small intestine, and rarely elsewhere in the abdomen. The malignant potential of GISTs is variable ranging from small lesions with a benign behaviour to fatal sarcomas. The majo-rity of the tumours stain positively for the CD-117(KIT) and discovered on GIST-1(DOG-1 or anoctamin 1) expression, and they are characterized by the presence of a driver kinase-activating mutation in either KIT or platelet-derived growth factor receptor α. Although surgery is the primary modality of treatment, almost half of the patients have disease recurrence following surgery, which highlights the need for an effective adjuvant therapy. Traditionally, GISTs are considered chemotherapy and radiotherapy resistant. With the advent of targeted therapy(tyrosine kinase inhibitors), there has been a paradigm shift in the management of GISTs in the last decade. | 8 | Antioxidant activity of newly discovered lineage of marine actinobacteria显示文摘Objective:To investigate the antioxidant activity of marine actinobacteria.Methods:The content of total phenolics,the level of antioxidant potential by DPPH radical scavenging activity,metal chelating activity,FRAP method,βcarotene assay and NO scavenging activity in extract were determined.Results:In all the methods the extract exhibited good scavenging activity except NO scavenging activity.The IC50 values of marine actinobacteria extract on DPPH radical were found to be 41.09μg/mL.The zone of color retention was 12 mm inβ-carotene bleaching assay. | 9 | Development of a panel of unigene-derived polymorphic EST–SSR markers in lentil using public database information显示文摘Lentil(Lens culinaris Medik.), a diploid(2n = 14) with a genome size greater than 4000 Mbp, is an important cool season food legume grown worldwide. The availability of genomic resources is limited in this crop species. The objective of this study was to develop polymorphic markers in lentil using publicly available curated expressed sequence tag information(ESTs). In this study, 9513 ESTs were downloaded from the National Center for Biotechnology Information(NCBI) database to develop unigene-based simple sequence repeat(SSR) markers. The ESTs were assembled into 4053 unigenes and then analyzed to identify 374 SSRs using the MISA microsatellite identification tool. Among the 374 SSRs, 26 compound SSRs were observed.Primer pairs for these SSRs were designed using Primer3 version 1.14. To classify the functional annotation of ESTs and EST–SSRs, BLASTx searches(using E-value 1 × 10-5) against the public UniP rot(http://gffzzc0d2db89b939488bh6v0w6bffbok96qqx.ffgz.tsg.suse.edu.cn/) and NCBI(http://gffzz5b533c64939a43b3h6v0w6bffbok96qqx.ffgz.tsg.suse.edu.cn/) databases were performed. Further functional annotation was performed using PLAZA(version3.0) comparative genomics and GO annotation was summarized using the Plant GO slim category. Among the synthesized 312 primers, 219 successfully amplified Lens DNA. A diverse panel of 24 Lens genotypes was used to identify polymorphic markers. A polymorphic set of 57 markers successfully discriminated the test genotypes. | 10 | Understanding the molecular mechanisms of cancer prevention by dietary phytochemicals:From experimental models to clinical trials显示文摘Chemoprevention is one of the cancer prevention approaches wherein natural/synthetic agent(s) are prescribed with the aim to delay or disrupt multiple pathways and processes involved at multiple steps, i.e., initiation, promotion, and progression of cancer. Amongst environmental chemopreventive compounds, diet/beverage-derived components are under evaluation, because of their long history of exposure to humans, high tolerability, low toxicity, and reported biological activities. This compilation briefly covers and compares the available evidence on chemopreventive efficacy and probable mechanism of chemoprevention by selected dietary phytochemicals(capsaicin, curcumin, diallyl sulphide, genistein, green/black tea polyphenols, indoles, lycopene, phenethyl isocyanate, resveratrol, retinoids and tocopherols) in experimental systems and clinical trials. All the dietary phytochemicals covered in this review have demonstrated chemopreventive efficacy against spontaneous or carcinogen-induced experimental tumors and/or associated biomarkers and processes in rodents at several organ sites. The observed anti-initiating, anti-promoting and anti-progression activity of dietary phytochemicals in carcinogen-induced experimental models involve phytochemical-mediated redox changes, modulation of enzymes and signaling kinases resulting to effects on multiple genes and cell signaling pathways. Results from clinical trials using these compounds have not shown them to be chemopreventive. This may be due to our:(1) inability to reproduce the exposure conditions, i.e., levels, complexity, other host and lifestyle factors; and(2) lack of understanding about the mechanisms of action and agent-mediated toxicity in several organs and physiological processes in the host. FMD virus serotypes O, A and Asia1 are prevalent in India and systematic efforts are on to control and eventually eradicate the disease from the country. FMD epidemiology is complex due to factors like co-circulation, extinction, emergence and re-emergence of genotypes/lineages within the three serotypes, animal movement, diverse farm practices and large number of susceptible livestock in the country. Systematic vaccination, prompt diagnosis, strict biosecurity measures, and regular monitoring of vaccinal immunity and surveillance of virus circulation are indispensible features for the effective implementation of the control measures. | 16 | Quaternary tectonic history of seismically active intraplate Kachchh Rift Basin,western India:A review显示文摘Since the recorded historical period,the Kachchh Rift Basin(KRB)has encountered numerous moderate to large magnitude earthquakes.According to the series of seismicity research so far,there are several important points of debate regarding the tectonic history and evolution of the KRB,especially during the Quaternary period.Therefore,the main objective of the present research is to inspect and perceive the association amongst the strain build-up,earthquake provenance,landform evolution and progression as archived by the Quaternary deposits of the KRB.The previous studies demonstrated the evolutions of various landforms,such as the uplifted fluvial terraces,formation of the gorges,uplifted alluvial fan sequences,which can be ideally used to reconstruct the neotectonic history along active faults of KRB.Considering this,the analysis of the accessible and supportive data,including geochronology provided by earlier studies along with some new dataset for a superior knowledge on the Quaternary tectonic forces prevailing in the KRB,have been carried out.Furthermore,we also emphasized the differences and directions for future potential research issues.The observations of variability in uplift rates across the various active faults in the KRB suggest a complex geological history during the Quaternary period.The results show that the vertical uplift rate along the significant active faults range from 0.8 to 2.8 mm/yr,demonstrating the variable tectonic stress regime prevailing in the KRB.The uplift rates constricted from geomorphic and chronological aspects suggest that the tectonic movements within the Kachchh intraplate region is regulated by the fault segments and the present tectonic stress field is in accordance with the encompassing tectonic stress field associated with the northward movement of the Indian plate corresponding to the Eurasian plate. | Girish Ch.Kothyari Raj Sunil Kandregula Rakesh Dumka Gaurav Chauhan Ajay Kumar Taloor | 2022 | Geodesy and Geodynamics2022,13,3: | 2 |

| 18 | Hepatoprotective and antioxidant activity of methanolic extract of flowers of Nerium oleander against CCl_4-induced liver injury in rats显示文摘Objective:To investigate the antioxidant and hepatoproteetive activity of methanolic flower extract of Nerium oleander against CCl4—induced hepatotoxicity in rats.Methods:In vitro antioxidant activity of methanolic extract of dowers of Nerium oleander(MENO-F) was evaluated by various assays,including reducing power,lipid peroxidation.DPPH.ARTS,superoxide anion,hydroxyl radicals and metal chelation.The hepatoproteetive and in vivo antioxidant activity of MENO-F were evaluated against CCl4-induced hepatic damage in rats.The MENO-F at dose of 100.200 and 400 mg/kg were administered orally once daily for seven days.Serum enzymatic levels of serum glutamate oxaloacetate transaminase(AST),serum glutamate pyruvate transaminase(ALT),serum alkaline phosphatase(ALP) and total bilirubin were estimated along with estimation of superoxide dismutase(SOD) and malondialdehvde(MDA) levels in liver tissues.Further histopathological examination of the liver sections was carried out to support the induction of hepalotoxicity and hepatoproteetive efficacy.Results:The extract showed potent activities on reducing power,lipid peroxide.DPPH.ABTS.superoxide anion,hydroxyl radical and metal chelation.The substantially elevated serum enzymatic levels of AST,ALT.ALP and total bilirubin were found to he restored towards normalization significantly by the MENO-F in a dose dependent manner with maximum hepatoprotection at 400 mg/kg dose level.The histopathological observations supported the biochemical evidences of hepatoprotection.Elevated level of SOD and decreased level of MDA further strengthen the hepatoproteetive observations.
