Platelets in Kawasaki disease:Is this only a numbers game or something beyond?显示文摘Kawasaki disease(KD)is a medium vessel vasculitis with predilection to cause coronary artery abnormalities.KD is now the most common cause of acquired heart disease in developed countries.Thrombocytosis is consistently found in patients with KD,usually in 2nd to 3rd week of illness.Thrombocytopenia has occasionally been reported in the acute phase of KD.An increase or decrease in platelet number in patients with KD was initially considered to be a benign phenomenon. However, recent literature on platelet biology in KD has suggested that platelets are not only increasing but are rather activated. This phenomenon has been found to increase the risk of thrombosis in these patients.Similarly a fall in platelet counts during acute stage of KD has also been found to be associated with increased severity of disease. In this review,we update on the current best understanding about pathogenic role of platelets in patients with KD. | Kanika Arora Sandesh Guleria Ankur Kumar Jindal Amit Rawat Surjit Singh | 2020 | Genes & Diseases2020,7,1: | 9
Analysis of isoquinoline alkaloids from Mahonia leschenaultia and Mahonia napaulensis roots using UHPLC-Orbitrap-MSn and UHPLC-QqQ LIT-MS/MS显示文摘Mahonia leschenaultia(ML) and Mahonia napaulensis(MN) are less known and unexplored medicinal plants of the family Berberidaceae. They are used by the Todas of Nilgiris in their religious and medical practices but chemically less identified. Hence, we decided to do extensive phytochemical analysis to explore the potential of these plant extracts. An ultrahigh performance electrospray tandem mass spectrometry(UHPLC–ESI–MS/MS)method was successfully developed for qualitative analysis of the bioactive components in Mahonia species using Orbitrap Velos Pro mass spectrometer. Sixteen compounds were identified by comparison of their retention times and mass spectra(MS) with authentic standards and reported literature. Multi-stage mass spectra(MS^(2–8)) for the identification of protoberberine and aporphine alkaloids showed the sequential expulsion of all the substituents attached with their basic skeleton followed by CO loss. Eight of the identified compounds(berberine, jatrorrhizine, palmatine, magnoflorine, isocorydine, glaucine, tetrahydropalmatine and tetrahydroberberine) were simultaneously determined by another UHPLC–ESI–MS/MS method under the multiple reactions monitoring(MRM) mode quantitatively using triple quadrupole linear ion trap mass spectrometer. The analytical method was validated for 8 bioactive compounds with overall recovery in the range98.5%–103.6%(RSD≤2.2%), precise(RSD≤2.07%) and linear(r≥0.9995) over the concentration range of 0.5–1000 ng/mL and successfully applied in ML and MN roots, which suggests the suitability of the proposed approach for the routine analysis of Mahonia species and their quality control. | Awantika Singh Vikas Bajpai Sunil Kumar Ajay Kumar Singh Rawat Brijesh Kumar | 2017 | Journal of Pharmaceutical Analysis2017,7,2: | 9
Percutaneous endoscopic gastrostomy and gastrooesophageal reflux in neurologically impaired children显示文摘AIM:To investigate the effects of percutaneous endoscopic gastrostomy(PEG) feeding on gastro-oesophageal reflux(GOR) in a group of these children using combined intraluminal pH and multiple intraluminal impedance(pH/MII) . METHODS:Ten neurologically impaired children underwent 12 h combined pH/MII procedures at least 1 d before and at least 12 d after PEG placement. METHODS:Prior to PEG placement(pre-PEG) a total of 183 GOR episodes were detected,156(85.2%) were non-acidic.After PEG placement(post-PEG) a total of 355 episodes were detected,182(51.3%) were nonacidic. The total number of distal acid reflux events statistically significantly increased post-PEG placement(prePEG total 27,post-PEG total 173,P=0.028) and themean distal pH decreased by 1.1 units. The distal reflux index therefore also significantly increased post-PEG [pre-PEG 0.25(0-2) ,post-PEG 2.95(0-40) ].Average proximal pH was lower post-PEG but the within subject difference was not statistically significant(P=0.058) . Median number of non-acid GOR,average reflux height, total acid clearance time and total bolus clearance time were all lower pre-PEG,but not statistically significant. CONCLUSION:PEG placement increases GOR episodes in neurologically impaired children. | Mike Thomson Prithviraj Rao David Rawat Tobias G Wenzl | 2011 | World Journal of Gastroenterology2011,17,2: | 8
Efficacy of fermented milk and whey proteins in Helicobacter pylori eradication:A review显示文摘Helicobacter pylori(H.pylori)eradication is considered a necessary step in the management of peptic ulcer disease, chronic gastritis, gastric adenocarcinoma and mucosa associated lymphoid tissue lymphoma.Standard triple therapy eradication regimens are inconvenient and achieve unpredictable and often poor results.Eradication rates are decreasing over time with increase in antibiotic resistance.Fermented milk and several of its component whey proteins have emerged as candidates for complementary therapy. In this context the current review seeks to summarize the current evidence available on their role in H.pylori eradication.Pertinent narrative/systematic reviews, clinical trials and laboratory studies on individual components including fermented milk, yogurt, whey proteins,lactoferrin,α-lactalbumin(α-LA),glycomacropeptide and immunoglobulin were comprehensively searched and retrieved from Medline,Embase,Scopus,Cochrane Controlled Trials Register and abstracts/proceedings of conferences up to May 2013.A preponderance of the evidence available on fermented milk-based probiotic preparations and bovine lactoferrin suggests a beneficial effect in Helicobacter eradication.Evidence forα-LA and immunoglobulins is promising while that for glycomacropeptide is preliminary and requires substantiation. The magnitude of the potential benefit documented so far is small and the precise clinical settings are ill defined. This restricts the potential use of this group as a complementary therapy in a nutraceutical setting hinging on better patient acceptability/compliance.Further work is necessary to identify the optimal substrate, fermentation process, dose and the ideal clinical setting(prevention/treatment,first line therapy/recurrence,symptomatic/asymptomatic,gastritis/ulcer diseases etc.).The potential of this group in high antibiotic resistance or treatment failure settings presents interesting possibilities and deserves further exploration. | Aarti Sachdeva Swapnil Rawat Jitender Nagpal | 2014 | World Journal of Gastroenterology2014,20,3: | 7
Recent advances in chronic granulomatous disease显示文摘Chronic granulomatous disease(CGD)is an inherited defect of phagocyte function due to defective NADPH oxidase.Patients with CGD are not able to effectively clear the infections because of the defect in the phagocyte production of oxygen free radicals and are prone to recurrent bacterial and fungal infections.Inflammatory complications are also noted in CGD such as colitis,non-infective granulomas causing gastrointestinal or urinary tract obstruction, hemophagocytic lymphohistiocytosis, and arthritis.Studies on toll-like receptor pathways and neutrophil extracellular traps in CGD have shed light on the role of NADPH oxidase in the innate immunity and pathogenesis of infections in CGD.Some reports also indicate a reduction of memory B cells and defective production of functional antibodies in CGD.Though the exact mechanisms for non-infective inflammatory complications in CGD are not yet clear, studies on efferocytosis and defective autophagy with inflammasome activation have made a substantial contribution to our understanding of the pathogenesis of inflammation in CGD.We also discuss the clinical and molecular features of p40phox defects and a newer genetic defect,EROS.Clinical phenotypes of X-linked carriers of CYBB are also discussed. | Gummadi Anjani Pandiarajan Vignesh Vibhu Joshi Jitendra Kumar Shandilya Dharmagat Bhattarai Jyoti Sharma Amit Rawat | 2020 | Genes & Diseases2020,7,1: | 6
Hepatoprotective potential of ethanolic extract of Ziziphus oenoplia(L.)Mill roots against antitubercular drugs induced hepatotoxicity in experimental models显示文摘Objective:To evaluate the hepatoprotective potential of ethanolic(50%) extract of Ziziphus oenoplia(L.) Mill(Z.oenoplia) root against isoniazid(INH) and rifampicin(RIF) induced liver damage in animal models. Methods:Five groups of six rats each were selected for the study.Ethanolic extract at a dose of 150 and 300 mg/kg as well as silymarin(100 mg/kg) were administered orally once daily for 21 d in INH + RIF treated groups. The serum levels of glutamic oxaloacetic transaminase(SGOT),glutamate pyruvate transaminase(SGPT),alkaline phosphatase (SALP),and bilirubin were estimated along with activities of superoxide dismutase, catalase, glutathione S-transferase,glutathione peroxidase, and hepatic melondialdehyde formation. Histopathological analysis was carried out to assess injury to the liver.Result:The considerably elevated serum enzymatic activities of glutamic oxaloacetic transaminase, glutamate pyruvate transaminase, alkaline phosphatase and bilirubin due to INH + RIF treatment were restored towards norma) in a dose dependent manner after the treatment with ethanolic extract of Z.oenoplia roots.Meanwhile, the decreased activities of superoxide dismutase, catalase, glutathione S-transferase and glutathione peroxidase were also restored towards normal dose dependency. In addition, ethanolic extract also significantly prevented the elevation of hepatic melondialdehyde formation in the liver of INH + RIF intoxicated rats in a dose dependent manner. The biochemical observations were supplemented with histopathological examination of rat liver sections. Conclusions:The results of this study slrongly indicate that ethanolic extract of Z.oenoplia has a potent hepatoprotective action against INH + RIF induced hepatic damage in rats. | Ch V Rao AKS Rawat Anil P Singh Arpita Singh Neeraj Verma | 2012 | Asian Pacific Journal of Tropical Medicine2012,5,4: | 6
Genome resequencing and transcriptome profiling reveal structural diversity and expression patterns of constitutive disease resistance genes in Huanglongbing-tolerant Poncirus trifoliata and its hybrids显示文摘Huanglongbing(HLB)is the most destructive bacterial disease of citrus worldwide. While most citrus varieties are susceptible to HLB,Poncirus trifoliata,a close relative of Citrus, and some of its hybrids with Citrus are tolerant to HLB.No specific HLB tolerance genes have been identified in P.trifoliata but recent studies have shown that constitutive disease resistance(CDR)genes were expressed at much higher levels in HLB-tolerant Poncirus hybrids and the expression of CDR genes was modulated by Candidatus Liberibacter asiaticus(CLas),the pathogen of HLB.The current study was undertaken to mine and characterize the CDR gene family in Citrus and Poncirus and to understand its association with HLB tolerance in Poncirus. We identified 17 CDR genes in two citrus genomes, deduced their structures, and investigated their phylogenetic relationships. We revealed that the expansion of the CDR family in Citrus seems to be due to segmental and tandem duplication events.Through genome resequencing and transcriptome sequencing,we identified eight CDR genes in the Poncirus genome(PtCDR1-PtCDR8).The number of SNPs was the highest in PtCDR2 and the lowest in PtCDR7.Most of the deletion and insertion events were observed in the UTR regions of Citrus and Poncirus CDR genes.PtCDR2 and PtCDR8 were in abundance in the leaf transcriptomes of two HLB-tolerant Poncirus genotypes and were also upregulated in HLB-tolerant,Poncirus hybrids as revealed by real-time PCR analysis. These two CDR genes seem to be good candidate genes for future studies of their role in citrus-CLas interactions. | Nidhi Rawat Brajendra Kumar Ute Albrecht Dongliang Du Ming Huang Qibin Yu Yi Zhang Yong-Ping Duan Kim D Bowman Fred G Gmitter Jr Zhanao Deng | 2017 | Horticulture Research2017,4,1: | 5

Characteristics of life-form and growth-form of plant species in an alpine ecosystem of North-West Himalaya显示文摘The present study was conducted in the alpine pastures of Tungnath (30° 14' N and 79° 13' E) to observe life-form and growth-form patterns of alpine plant species under grazed and ungrazed conditions and to work out the plant life form spectrum. Species were categorized as plant habit, height and length of growth-cycle and life-form classes according to Raunkiaer's system. The results show that in total of 68 species at grazed site, hemicryptophytes (He) accounted for 50.00% species, followed by cryptophytes (26.47%), chamaephytes (16.18%), phanerophytes (4.41%) and therophytes (2.94%). At the ungrazed site in 65 plant species, hemicryptophytes (He) accounted for 49.23% species, cryptophytes (26.15%), chamaephytes (15.38%), phanerophytes (6.15%) and therophytes (3.08 % species). In general, hemicrptophyte are dominant in both sites i.e. graged and ungrazed. Growth form categories were classified as forbs, shrubs, grasses and sedges and undershrubs, according to plant habit and height. On the basis of length of the growth cycle, species were categorized as plant species of short growth cycle, intermediate growth cycle and long growth cycle. The short forbs of plant habit and height, had the highest emergence, and grasses and sedges had the lowest emergence in representative species. Percentage of species with long growth cycle was highest in both sites. | R. K. Vashistha Neelam Rawat A. K. Chaturvedi B. P. Nautiyal P. Prasad M. C. | 11 | Recent advances in elucidating the genetics of common variable immunodeficiency显示文摘Common variable immunodeficiency disorders(CVID),a heterogeneous group of inborn errors of immunity,is the most common symptomatic primary immunodeficiency disorder.Patients with CVID have highly variable clinical presentation. With the advent of whole genome sequencing and genome wide association studies(GWAS),there has been a remarkable improvement in understanding the genetics of CVID.This has also helped in understanding the pathogenesis of CVID and has drastically improved the management of these patients.A multiomics approach integrating the DNA sequencing along with RNA sequencing, proteomics, epigenetic and metabolomics profile is the need of the hour to unravel specific CVID associated disease pathways and novel therapeutic targets. In this review,we elaborate various techniques that have helped in understanding the genetics of CVID. | Vaishali Aggarwal Aaqib Zaffar Banday Ankur Kumar Jindal Jhumki Das Amit Rawat | 2020 | Genes & Diseases2020,7,1: | 4
Genetics on early onset inflammatory bowel disease:An update显示文摘Inflammatory bowel disease(IBD)is more common in adults than in children.Onset of IBD before 17 years of age is referred as pediatric onset IBD and is further categorized as very early onset IBD(VEO-IBD)for children who are diagnosed before 6 years of age, infantile IBD who had the disease before 2 years of age and neonatal onset IBD for children less than 28 days of life.Children presenting with early onset disease may have a monogenic basis.Knowledge and awareness of the clinical manifestations facilitates early evaluation and diagnosis.Next generation sequencing is helpful in making the genetic diagnosis.Treatment of childhood IBD is difficult;targeted therapies and hematopoietic stem cell transplantation form the mainstay. In this review we aim to summarize the genetic defects associated with IBD phenotype. We describe genetic location and functions of various genetic defect associated with VEO-IBD with their key clinical manifestations. We also provide clinical clues to suspect these conditions and approaches to the diagnosis of these disorders and suitable treatment options. | Johnson Nameirakpam Rashmi Rikhi Sanjay Singh Rawat Jyoti Sharma Deepti Suri | 2020 | Genes & Diseases2020,7,1: | 3
Enhancing bifunctionality of CoN nanowires by Mn doping for long-lasting Zn-air batteries显示文摘Tailoring the nanostructure and composition of transition metal nitrides is highly important for their use as potent low-cost electrocatalysts. Cobalt nitride(CoN) exhibits strong catalytic activity for oxygen evolution reaction(OER). However, its poor catalytic efficiency for oxygen reduction reaction(ORR) hinders its application in rechargeable zinc-air batteries(ZABs) as the air cathode. In this work, we deploy the effective strategy of Mn doping to improve both OER and ORR activity of CoN nanowires as the cathode material for ZAB. Theoretical calculation predicts that moderate Mn doping in cobalt nitride results in a downshift of the d-band center and reduces the adsorption energy of reaction intermediates. With ~10 at% Mn dopants, stronger catalysis activities for both OER and ORR are achieved compared to pure CoN nanowires. Subsequently, both aqueous and flexible quasi-solid-state ZABs are constructed using the Mn-doped CoN nanowires array as additive-free air cathode. Both types of devices present large open circuit potential, high power density and long-cycle stability. This work pushes forward the progress in developing cost-effective ZABs. | Yongqi Zhang Bo Ouyang Guankui Long Hua Tan Zhe Wang Zheng Zhang Weibo Gao Rajdeep Singh Rawat Hong Jin Fan | 2020 | Science China Chemistry2020,63,7: | 3
Determination of flavonoids, polyphenols and antioxidant activity of Tephrosia purpurea:a seasonal study显示文摘OBJECTIVE:Tephrosia purpurea(Linn.) Pers.is widely used in traditional medicine to treat liver disorders, febrile attacks, enlargement and obstruction of liver, spleen, and kidney. In the present study, investigations were carried out to determine the seasonal impact on the content of flavonoid glycosides and on antioxidant activities so as to identify the optimal time of harvesting.METHODS:The plant materials were collected in different seasons during 2013-2014.Air-dried,powdered plant materials were extracted with 95%ethanol and ethanokwater(1:1) by ultrasound-assisted extraction process.Their chemical composition in terms of total polyphenol and flavonoid contents(TPCs and TFCs) was determined using modified colorimetric Folin-Ciocalteu method and aluminum chloride colorimetric assay respectively. To determine the in vitro antioxidant activity,diphenyl-picryl hydrazyl(DPPH) radical-scavenging assay and total antioxidant capacity by phosphomolybdate antioxidant assay were carried out.High-performance liquid chromatography(HPLC)/photo-diode array(PDA) analysis was used to quantify the flavonoid glycosides in the samples collected in different seasons.Correlation studies were also carried out between antioxidant activities and TPCs.RESULTS:The highest TPC and TFC were found to be in the 95%ethanolic extract of the August sample and the lowest in the 50%hydro-alcoholic extract of the plant sample collected in winter season. It was observed that in both the assays used to determine the antioxidant activity, the 95%ethanolic extracts in all the seasons showed a higher activity than their respective 50%hydroalcoholic extracts with an increase in activity as we go from cold to hot to rainy seasons.Based on correlation analysis,DPPH radical-scavenging activities as well as the spectrophotometrically measured phosphomolybdenum complex were also strongly correlated with TPC of the extracts. The most abundant flavonoid glycoside was quercetin-3-O-rhamnoglucoside in all the seasons. The content of all flavonoids was observed highest in the 95%ethanolic extract of the plant collected in August(TP-3).The 50%hydro-alcoholic extract of the plant collected in December(TP-6) showed the lowest amount of flavonoids and antioxidant activity.CONCLUSION:The findings of this study confirmed that the metabolism and production of flavonoids in T.purpurea are vigorously affected by seasonal factors.Significant differences were observed in the TPC,TFC and flavonoid glycoside composition in the 95%ethanolic as well as 50%hydro-alcoholic extracts of T.purpurea collected in different seasons.Since the 95%ethanolic extract of the August sample(TP-3),which also happens to be its flowering season, exhibited the highest TPC.TFC and antioxidant activities in both DPPH and phosphomolybdate assays as well as contained the highest content of all flavonoids,it could be recommended as the optimal season of harvesting T.purpurea with respect to its pharmaceutically important constituents,i.e.,flavonoids. | Madan Mohan Pandey Sayyada Khatoon Subha Rastogi Ajay Kumar Singh Rawat | 2016 | Journal of Integrative Medicine2016,14,6: | 3

Effect of seed mass on emergence and seedling development in Pterocarpus marsupium Roxb.显示文摘We investigated the effect of seed mass on emergence, seedling survival and growth of Pterocarpus marsupium Roxb., a medium to large, commercially valuable and deciduous tree species. Among the three size classes viz. small(10-12 mm), medium(13-15 mm) and large(16-17 mm), the maximum proportion of seed by number(54.12%) and dry weight(51.87%) was recorded in the medium size seed class. Seed length and seed width were greatest in the large seed class(16.50 mm, 7.33 mm) followed by medium(13.50 mm, 5.60 mm) and small(11.37 mm, 3.66 mm). Similarly, hundred seed weight(100 sw) varied from a maximum of 12.92 g in the large seed class to intermediate 10.95 g in the medium seed class and minimum of 7.02 g in the small seed class. The large seed size showed maximum emergence and shoot length over the medium and small seed class. After six months of growth, significant variations due to seed size were also observed for the growth and dry weight of P. marsupium seedlings. Seedling vigour, expressed in terms of height, collar diameter, number of leaves and dry biomass, was significantly affected by seed class. Seedlings that emerged from large seeds showed better growth and produced heavier seedlings as compared to medium and small seeds. | Yogeshwar Mishra Rimi Rawat P. K. Rana M. K. | 17 | Genetics of severe combined immunodeficiency显示文摘Severe Combined Immunodeficiency(SCID)is an inherited group of rare, lifethreatening disorders due to the defect in T cell development and function.Clinical manifestations are characterised by recurrent and severe bacterial, viral, and fungal opportunistic infections that start from early infancy period.Haematopoietic stem cell transplantation(HSCT)is the treatment of choice. The pattern of inheritance of SCID may be X-linked or autosomal recessive.Though the diagnosis of SCID is usually established by flow cytometry-based tests, genetic diagnosis is often needed for genetic counselling, prognostication, and modification of pre-transplant chemotherapeutic agents. This review aims to highlight the genetic aspects of SCID. | Rajni Kumrah Pandiarajan Vignesh Pratap Patra Ankita Singh Gummadi Anjani Poonam Saini Madhubala Sharma Anit Kaur Amit Rawat | 2020 | Genes & Diseases2020,7,1: | 2
Leukocyte adhesion defect:Where do we stand circa 2019?显示文摘Migration of polymorphonuclear leukocytes from bloodstream to the site of inflammation is an important event required for surveillance of foreign antigens. This trafficking of leukocytes from bloodstream to the tissue occurs in several distinct steps and involves several adhesion molecules.Defect in adhesion of leukocytes to vascular endothelium affecting their subsequent migration to extravascular space gives rise to a group of rare primary immunodeficiency diseases(PIDs)known as Leukocyte Adhesion Defects(LAD).Till date, four classes of LAD are discovered with LAD I being the most common form.LAD I is caused by loss of function of common chain, cluster of differentiation(CD)18 of β2 integrin family. These patients suffer from life-threatening bacterial infections and in its severe form death usually occurs in childhood without bone marrow transplantation.LAD II results from a general defect in fucose metabolism. These patients suffer from less severe bacterial infections and have growth and mental retardation.Bombay blood group phenotype is also observed in these patients.LAD III is caused by abnormal integrin activation.LAD III patients suffer from severe bacterial and fungal infections.Patients frequently show delayed detachment of umbilical cord, impaired wound healing and increased tendency to bleed.LAD IV is the most recently described class. It is caused by defects in β2 and α4β1 integrins which impairs lymphocyte adhesion.LAD IV patients have monogenic defect in cystic-fibrosis-transmembraneconductance-regulator(CFTR)gene,resulting in cystic fibrosis.Pathophysiology and genetic etiology of all LAD syndromes are discussed in detail in this paper. | Jhumki Das Avinash Sharma Ankur Jindal Vaishali Aggarwal Amit Rawat | 2020 | Genes & Diseases2020,7,1: | 2
Froth flotation of refuse coal fines and process optimization using 2D surface plots显示文摘Refuse coal fines of size <500 μm was collected from a metallurgical coal preparation plant located in the eastern coalfield region of India. The coal was beneficiated using froth flotation technique to recover clean coal with ash content of about 20% with the highest possible yield. Diesel oil as collector and pine oil as frother were used. Box-Behnken statistical design was followed for analyzing the performance at varying pulp density, collector and frother dosage. Results were discussed using 2D surface plots. Response function predictions determined by the regression analysis show coefficient of correlation(R^2) for yield and the ash content as 0.72 and 0.86, respectively. The highest yield of 45.79% is obtained at pulp density 10%, collector dose 2 kg/t and frother dose 1.5 kg/t. The lowest ash content of 18.9% is obtained at pulp density 10%, collector dose 1 kg/t and frother dose 1 kg/t. | Shravan Kumar Veomesh Rawat | 2016 | Journal of Central South University2016,23,10: | 2
