| 2 | Fingolimod alters inflammatory mediators and vascular permeability in intracerebral hemorrhage显示文摘Intracerebral hemorrhage(ICH) leads to high rates of death and disability. The pronounced infl ammatory reactions that rapidly follow ICH contribute to disease progression. Our recent clinical trial demonstrated that oral administration of an immune modulator fingolimod restrained secondary injury derived from initial hematoma,but the mechanisms remain unknown. In this study,we aim to investigate the effects of fingolimod on inflammatory mediators and vascular permeability in the clinical trial of oral fingolimod for intracerebral hemorrhage(ICH). The results showed that fingolimod decreased the numbers of circulating CD4^+ T,CD8^+ T,CD(19)^+B,NK,and NKT cells and they recovered quickly after the drug was stopped. The plasma ICAM level was decreased and IL-10 was increased by fingolimod. Interestingly,fingolimod protected vascular permeability as indicated by a decreased plasma level of MMP9 and the reduced r T1%. In conclusion,modulation of systemic inflammation by fingolimod demonstrates that it is an effective therapeutic agent for ICH. | 4 | Genome editing in Drosophila melanogaster: from basic genome engineering to the multipurpose CRISPR-Cas9 system显示文摘Nowadays, genome editing tools are indispensable for studying gene function in order to increase our knowledge of biochemical processes and disease mechanisms. The extensive availability of mutagenesis and transgenesis tools make Drosophila melanogaster an excellent model organism for geneticists. Early mutagenesis tools relied on chemical or physical methods,ethyl methane sulfonate(EMS) and X-rays respectively, to randomly alter DNA at a nucleotide or chromosomal level. Since the discovery of transposable elements and the availability of the complete fly genome, specific genome editing tools, such as P-elements, zinc-finger nucleases(ZFNs) and transcription activator-like effector nucleases(TALENs), have undergone rapid development. Currently, one of the leading and most effective contemporary tools is the CRISPR-cas9 system made popular because of its low cost, effectiveness, specificity and simplicity of use. | 7 | High-throughput prediction of the ground-state collinear magnetic order of inorganic materials using Density Functional Theory显示文摘We present a robust,automatic high-throughput workflow for the calculation of magnetic ground state of solid-state inorganic crystals,whether ferromagnetic,antiferromagnetic or ferrimagnetic,and their associated magnetic moments within the framework of collinear spin-polarized Density Functional Theory.This is done through a computationally efficient scheme whereby plausible magnetic orderings are first enumerated and prioritized based on symmetry,and then relaxed and their energies determined through conventional DFT+U calculations.This automated workflow is formalized using the atomate code for reliable,systematic use at a scale appropriate for thousands of materials and is fully customizable.The performance of the workflow is evaluated against a benchmark of 64 experimentally known mostly ionic magnetic materials of non-trivial magnetic order and by the calculation of over 500 distinct magnetic orderings.A non-ferromagnetic ground state is correctly predicted in 95% of the benchmark materials,with the experimentally determined ground state ordering found exactly in over 60% of cases.Knowledge of the ground state magnetic order at scale opens up the possibility of high-throughput screening studies based on magnetic properties,thereby accelerating discovery and understanding of new functional materials. | Matthew Kristofer Horton Joseph Harold Montoya Miao Liu Kristin Aslaug Persson | 2019 | npj Computational Materials2019,,1: | 3 |

It crystallizes in the triclinic space group P1 with a = 9.3767(14), b = 10.1451(15), c = 12.1488(17), α = 102.6450(10), β = 108.856(2), γ = 98.807(2)°, V = 1035.3(3)3, Z = 2, C22H17 Co N5O6, Mr = 506.34, Dc = 1.624 g/cm3, μ = 0.882 mm-1 and F(000) = 518. In the complex, the nph2- ligands connect neighbouring cobalt atoms to form binuclear [Co(nph)]2 subunits, which are linked by pairs of bix ligands to form a 2D honeycomb-like(6,3) network. In addition, the compound is further extended into a 3D supramolecular architecture by π···π stacking interactions. | 10 | Past,present and future of cyanide antagonism research:From the early remedies to the current therapies显示文摘This paper reviews milestones in antidotal therapies for cyanide(CN) spanning early remedies,current antidotal systems and research towards next generation therapies.CN has been a part of plant defense mechanisms for millions of years.It became industrially important in the nineteenth century with the advent of CN assisted gold mining and the use of CN as a pest control agent.The biochemical basis of CN poisoning was actively studied and key mechanisms were understood as early as 1929.These fundamental studies led to a variety of antidotes,including indirect CN binders that generate methemoglobin,direct CN binders such as hydroxocobalamin,and sulfur donors that convert CN to the less toxic thiocyanate.Research on blood gases at the end of the twentieth century shed new light on the role of nitric oxide(NO) in the body.The discovery of NO's ability to compete with CN for enzymatic binding sites provided a previously missed explanation for the rapid efficacy of NO generating antidotes such as the nitrites.Presently used CN therapies include:methemoglobin/NO generators(e.g.,sodium nitrite,amyl nitrite,and dimethyl aminophenol),sulfur donors(e.g.,sodium thiosulfate and glutathione),and direct binding agents [(e.g.,hydroxocobalamin and dicobalt salt of ethylenediaminetetraacetic acid(dicobalt edetate)].A strong effort is being made to explore novel antidotal systems and to formulate them for rapid administration at the point of intoxication in mass casualty scenarios.New antidotes,formulations,and delivery systems are enhancing bioavailability and efficacy and hold promise for a new generation of improved CN countermeasures. | Ilona Petrikovics Marianna Budai Kristof Kovacs David E Thompson | 2015 | World Journal of Methodology2015,5,2: | 3 |
