2根癌农杆菌介导的转录因子DREB1A基因在地被菊花中的遗传转化显示文摘以地被菊花(Dendranthema grandiflorum cv.White Snow)无菌苗叶片为外植体,在附加不同浓度植物生长调节剂的MS基础培养基上诱导培养,通过器官直接发生途径获得再生植株。结果表明,2.0mg/L6-BA和0.2mg/LNAA组合可获得93.8%的再生芽。将含拟南芥(Arabidopsis thaliana)逆境诱导转录因子DREB1A基因的植物表达载体pBI-DREB1A导入根癌农杆菌(Agrobacterium tumefaciens)LBA4404,农杆菌介导法遗传转化地被菊花White Snow叶盘,经PCR和PCR-Southern检测,DREB1A基因已整合到转化植株的基因组中。遗传转化过程中,预培养2d后,将OD600=0.5~0.7的根癌农杆菌稀释30倍后侵染叶盘10min,再共培养2d,有利于获得最高遗传转化效率。洪波 张常青 李邱华 高俊平 KASUGA Mie SHINOZAKI-YAMAGUCHI Kazuko 2005农业生物技术学报2005,13,3:20
3Heterologous expression of the AtDREB1A gene in chrysanthemum increases drought and salt stress tolerance显示文摘DNA cassette containing an AtDREB1A cDNA and a nos terminator,driven by a cauli- flower mosaic 35S promoter,or a stress-inducible rd29A promoter,was transformed into the ground cover chrysanthemum(Dendranthema grandiflorum)'Fall Color'genome.Compared with wild type plants,severe growth retardation was observed in 35S:DREB1A plants,but not in rd29A:DREB1A plants.RT-PCR analysis revealed that,under stress conditions,the DREB1A gene was over-expressed constitutively in 35S:DREB1A plants,but was over-expressed inductively in rd29A:DREB1A plants.The transgenic plants exhibited tolerance to drought and salt stress,and the tolerance was significantly stronger in rd29A:DREB1A plants than in 35S:DREB1A plants.Proline content and SOD activity were increased inductively in rd29A:DREB1A plants than in 35S:DREB1A plants under stress conditions.These results indicate that heterologous AtDREB1A can confer drought and salt tolerance in transgenic chrysanthemum,and improvement of the stress tolerance may be related to enhancement of proline content and SOD activity.KASUGA Mie YAMAGUCHI-SHINOZAKI Kazuko 2006Science China(Life Sciences)2006,49,5:18

5Differential effects of physical activity and sleep duration on cognitive function in young adults显示文摘Purpose: Although exercise and sleep duration habits are associated with cognitive function, their beneficial effects on cognitive function remain unclear. We aimed to examine the effect of sleep duration and daily physical activity on cognitive function, elucidating the neural mechanisms using near-infrared spectroscopy(NIRS).Methods: A total of 23 healthy young adults(age 22.0 ± 2.2 years) participated in this study. Exercise amount was assessed using a uniaxial accelerometer. We evaluated total sleep time(TST) and sleep efficiency by actigraphy. Cognitive function was tested using the N-back task, the Wisconsin Card Sorting Test(WCST), and the Continuous Performance Test—Identical Pairs(CPT-IP), and the cortical oxygenated hemoglobin levels during a word fluency task were measured with NIRS.Results: Exercise amount was significantly correlated with reaction time on 0- and 1-back tasks(r = —0.602, p = 0.002; r = —0.446, p = 0.033,respectively), whereas TST was significantly correlated with % corrects on the 2-back task(r = 0.486, p = 0.019). Multiple regression analysis,including exercise amount, TST, and sleep efficiency, revealed that exercise amount was the most significant factor for reaction time on 0- and 1-back tasks(b = —0.634, p = 0.002; b = —0.454, p = 0.031, respectively), and TST was the most significant factor for % corrects on the 2-back task(b = 0.542, p = 0.014). The parameter measured by WCST and CPT-IP was not significantly correlated with TST or exercise amount. Exercise amount, but not TST, was significantly correlated with the mean area under the NIRS curve in the prefrontal area(r = 0.492, p = 0.017).Conclusion: Exercise amount and TST had differential effects on working memory and cortical activation in the prefrontal area. 7Impairment of IFN-a production capacity in patients with hepatitis C virus and the risk of the development of hepatocellular carcinoma显示文摘AIM: To determine the utility of interferon (IFN) -αproduction capacity in patients with hepatitis C virus (HCV) infection for the measurement of immunosurveillance potential and for the early detection of hepatocellular carcinoma (HCC) by investigating the Sendai virus (HVJ) stimulated IFN-α production capacity of patients with HCV infection.METHODS: HVJ stimulated IFN-α production was determined in a large number of patients with HCV infection and the development of HCC was monitored for 3 years in patients with liver cirrhosis (LC).RESULTS: IFN-α production capacity decreases gradually with the progression of liver disease from chronic hepatitis (CH) to HCC. A significant correlation between the duration of HCV infection and impaired IFN-α production capacity was observed. 10DIFFERENTIAL CYTOTOXIC SENSITIVITY AMONG MTT,NR AND ALPASE ACTIVITY ASSAYS IN HUMAN PULP CELLS EXPOSED TO DENTAL RESIN MONOMERS显示文摘A transformed human dental pulp cell(LSC) was made to assess thecytotoxicity of dental resin monomers with MTT assay,NR assay and ALPase activity assay. L- 929 cells were used as comparative cell line. The sensitivity amongMTT,NR and ALPase activity assays and between two types of cells were investigated. The relationship of ALPase activity to two colorimetric assays was also evaluated. According to IC50 and statistics analysis on cytotoxicity curve,there were significance differences between MTT and NR assay or both cell lines for TECDMAmonomer (P<0.005). MTT assay showed lower sensitivity than NR assay in twokinds of cell for both monomer,and LSC cells presented less sensitivity than L-929cells for TEGDMA monomer. The ALPase activity assay revealed the highest sensitivity among the three tested assays. There was a positive relationship between thecontent of ALPase activity and cytotoxic response of the other two assays. The results indicated that the sensitive degree of test assay or cell line may differ from thetest chemicals. 11Systemic hemodynamics in advanced cirrhosis: Concerns during perioperative period of liver transplantation显示文摘Advanced liver cirrhosis is usually accompanied by portal hypertension. Long-term portal hypertension results in various vascular alterations. The systemic hemodynamic state in patients with cirrhosis is termed a hyperdynamic state. This peculiar hemodynamic state is characterized by an expanded blood volume, high cardiac output, and low total peripheral resistance. Vascular alterations do not disappear even long after liver transplantation(LT), and recipients with cirrhosis exhibit a persistent systemic hyperdynamic state even after LT. Stability of optimal systemic hemodynamics is indispensable for adequate portal venous flow(PVF) and successful LT, and reliable parameters for optimal systemic hemodynamics and adequate PVF are required. Even a subtle disorder in systemic hemodynamics is precisely indicated by the balance between cardiac output and blood volume. The indocyanine green(ICG) kinetics reflect the patient's functional hepatocytes and effective PVF, and PVF is a major determinant of the ICG elimination constant(kICG) in the well-preserved allograft. The kICG value is useful to set the optimal PVF during living-donor LT and to evaluate adequate PVF after LT. Perioperative management has a large influence on the postoperative course and outcome; therefore, key points and unexpected pitfalls for intensive management are herein summarized. 13Screening of agonistic activities against four nuclear receptors in wastewater treatment plants in Japan using a yeast two-hybrid assay显示文摘To assess the potential endocrine disruptive effects through multiple nuclear receptors(NRs),especially non-steroidal NRs,in municipal wastewater,we examined the agonistic activities on four NRs(estrogen receptor α,thyroid hormone receptor α,retinoic acid receptor α and retinoid X receptor α) of untreated and treated wastewater from municipal wastewater treatment plants(WWTPs) in Japan using a yeast two-hybrid assay.Investigation of the influent and effluent of seven WWTPs revealed that agonistic activities against steroidal and non-steroidal NRs were always detected in the influents and partially remained in the effuents.Further investigation of four WWTPs employing conventional activated sludge,pseudo-anoxic-oxic,anoxic-oxic and anaerobic-anoxic-oxic processes revealed that the ability to reduce the agonistic activity against each of the four NRs varies depending on the treatment process.These results indicated that municipal wastewater in Japan commonly contains endocrine disrupting chemicals that exert agonistic activities on steroidal and non-steroidal NRs,and that some of these chemicals are released into the natural aquatic environment.Although the results obtained in yeast assays suggested that measured levels of non-steroidal NR agonists in the effluent of WWTPs were not likely to cause any biological effect,further study is required to assess their possible risks in detail.Daisuke Inoue Koki Nakama Kazuko Sawada Taro Watanabe Hisae Matsui Kazunari Sei Tsuyoshi Nakanishi Michihiko Ike 2011Journal of Environmental Sciences2011,23,1:2

15Antibody to eosinophil cationic protein suppresses dextran sulfate sodium-induced colitis in rats显示文摘AIM: To produce an antibody against rat eosinophil cationic protein (ECP) and to examine the effects of the antibody in rats with dextran sulfate sodium (DSS)-induced colitis.METHODS: An antibody was raised against rat ECP. Rats were treated with 3% DSS in drinking water for 7 d and received the antibody or normal serum. The colons were exarmined histologically and correlated with clinical symptoms.Immunohistochemistry and Western blot analysis were estimated as a grade of inflammation.RESULTS: The ECP antibody stained the activated eosinophils around the injured crypts in the colonic mucosa.Antibody treatment reduced the severity of colonic ulceration and acute clinical symptoms (diarrhea and/or blood-stained stool). Body weight gain was significantly greater and the colon length was significantly longer in anti-ECP-treated rats than in normal serum-treated rats. Expression of ECP in activated eosinophils was associated with the presence of erosions and inflammation. The number of Ki-67-positive cells in the regenerated surface epithelium increased in anti-ECP-treated rats compared with normal serum-treated rats.
