Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance, essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders. The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells, along with the signaling mechanisms governing stem cell self-renewal and differentiation. In this review,we first summarize the biological characteristics of seven types of dental stem cells, including dental pulp stem cells, stem cells from apical papilla, stem cells from human exfoliated deciduous teeth, dental follicle precursor cells, periodontal ligament stem cells, alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva. We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways. We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications. Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade. | Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji | 2016 | Genes & Diseases2016,3,4: | 16
Joint contractures in severe burn patients with early rehabilitation intervention in one of the largest burn intensive care unit in China:a descriptive analysis显示文摘Background:Joint contracture is the major clinical complication in burn patients, especially, the severe burn patients. This study aimed to investigate the number and severity of joint contractures in patients with burns affecting greater than or equal to 50%of the total body surface area(TBSA)undergoing early rehabilitation in a burn intensive care unit(BICU).Methods:We analyzed burn patients with burns affecting greater than or equal to 50%of the TBSA admitted to a BICU who received early rehabilitation within 7 days post-injury from January 2011 to December 2015.Demographic and medical information was collected. The range of motion(ROM)of different joints was measured 1 month post-admission.Spearman’s correlation coefficient and logistic regression analysis was used to determine predictors of the presence and severity of contractures.Result:The average affected TBSA of the included burn patients was 67.4%,and the average length of stay in the BICU was 46.2±28.8 days. One hundred and one of 108 burn patients(93.5%)developed at least one joint contracture. The ROM in 67.9%of the affected joints was mildly limited. The majority of contractures in severe burn patients were mild(37.7%)or moderate(33.2%).The wrist was the most commonly affected joint(18.2%),followed by the shoulder, ankle, hip, knee, and elbow.A predictor of the presence of contractures was the length of hospital stay(p=0.049).The severe contracture was related to the area of full-thickness burns, the strict bed rest time, and the duration of rehabilitation in BICU.The length of rehabilitation stay(days)in patients with moderate contracture is 54.5%longer than that in severe contracture(p=0.024)Conclusion:During the long stay in BICU,the length of rehabilitation stay in a BICU could decrease the severity of contractures from severe to moderate in the patients with equal to 50%of the TBSA.Hence, this research reveals the important role of early rehabilitation interventions in severe burn patients. | Jianglin Tan Jian Chen Junyi Zhou Huapei Song Huan Deng Ming Ao Gaoxing Luo Jun Wu | 2019 | Burns & Trauma2019,7,1: | 9
The use of fire at Zhoukoudian:evidence from magnetic susceptibility and color measurements显示文摘In order to provide direct evidence for the use of fire by humans at Locality 1,Zhoukoudian,we examine the burnt and unburnt sediments of newly excavated area in Layer 4 by detailed measurements of magnetic susceptibility, color, and diffuse reflectance spectrum. Results show that the magnetic susceptibility and redness of the burnt sediments are remarkably higher than those of other areas on the same level:up to*22 times for magnetic susceptibility and*3 times for redness of those of the adjacent unburnt sediments.Fine-grained(superparamagnetic/stable single-domain)magnetite and hematite grains make dominant contributions to the distinctly high values of magnetic susceptibility and redness in the burnt sediments.Diffuse reflectance spectroscopy results show that the burnt sediments contain more hematite than those of other areas and localities 2 and 3.High-temperature magnetic susceptibility measurements demonstrate that the burnt sediments have been heated above 700°C.Those changes in low-frequency magnetic susceptibility and redness are impossibly resulted from natural fires, thus most likely signaling the human activities of controlled use of fire. However, further work is needed to confirm whether or not these heat-affected sediments were produced in situ. | Yan Zhang Zhengtang Guo Chenglong Deng Shuangquan Zhang Haibin Wu Chunxia Zhang Junyi Ge Deai Zhao Qin Li Yang Song Rixiang Zhu | 2014 | Chinese Science Bulletin2014,59,10: | 6
An all-round AI-Chemist with a scientific mind显示文摘The realization of automated chemical experiments by robots unveiled the prelude to an artificial intelligence(AI)laboratory.Several AI-based systems or robots with specific chemical skills have been demonstrated, but conducting all-round scientific research remains challenging. Here,we present an all-round AI-Chemist equipped with scientific data intelligence that is capable of performing basic tasks generally required in chemical research.Based on a service platform, the AI-Chemist is able to automatically read the literatures from a cloud database and propose experimental plans accordingly. It can control a mobile robot in-house or online to automatically execute the complete experimental process on 14 workstations, including synthesis, characterization and performance tests. The experimental data can be simultaneously analysed by the computational brain of the AI-Chemist through machine learning and Bayesian optimization, allowing a new hypothesis for the next iteration to be proposed. The competence of the AI-Chemist has been scrutinized by three different chemical tasks. In the future, the more advanced all-round AI-Chemists equipped with scientific data intelligence may cause changes to the landscape of the chemical laboratory. | Qing Zhu Fei Zhang Yan Huang Hengyu Xiao LuYuan Zhao XuChun Zhang Tao Song XinSheng Tang Xiang Li Guo He BaoChen Chong JunYi Zhou YiHan Zhang Baicheng Zhang JiaQi Cao Man Luo Song Wang GuiLin Ye WanJun Zhang Xin Chen Shuang Cong Donglai Zhou Huirong Li Jialei Li Gang Zou WeiWei Shang Jun Jiang Yi Luo | 2022 | National Science Review2022,9,10: | 5

Prevalence and clinical significance of pathogenic germline BRCA1/2 mutations in Chinese non-small cell lung cancer patients显示文摘Objective: Germline alterations in the breast cancer susceptibility genes type 1 and 2, BRCA1 and BRCA2, predispose individuals to hereditary cancers, including breast, ovarian, prostate, pancreatic, and stomach cancers.Accumulating evidence suggests inherited genetic susceptibility to lung cancer. The present study aimed to survey the prevalence of pathogenic germline BRCA mutations(gBRCAm) and explore the potential association between gBRCAm and disease onset in Chinese advanced non-small cell lung cancer(NSCLC) patients. Methods: A total of 6,220 NSCLC patients were screened using capture-based ultra-deep targeted sequencing to identify patients harboring germline BRCA1/2 mutations. Results: Out of the 6,220 patients screened, 1.03%(64/6,220) of the patients harbored the pathogenic gB RCAm, with BRCA2 mutations being the most predominant mutations(49/64, 76.5%).Patients who developed NSCLC before 50 years of age were more likely to carry gBRCAm(P = 0.036).Among the patients harboring classic lung cancer driver mutations, those with concurrent gBRCAm were significantly younger than those harboring the wild-type gBRCA(P = 0.029).By contrast, the age of patients with or without concurrent gBRCAm was comparable to those of patients without the driver mutations(P = 0.972).In addition, we identified EGFR-mutant patients with concurrent gBRCAm who showed comparable progression-free survival but significantly longer overall survival(P = 0.002) compared to EGFR-mutant patients with wild-type germline BRCA.Conclusions: Overall, our study is the largest survey of the prevalence of pathogenic gBRCAm in advanced Chinese NSCLC patients. Results suggested a lack of association between germline BRCA status and treatment outcome of EGFR-TKI.In addition, results showed a positive correlation between pathogenic gB RCAm and an early onset of NSCLC. | Xingsheng Hu Dongyong Yang Yalun Li Li Li Yan Wang Peng Chen Song Xu Xingxiang Pu Wei Zhu Pengbo Deng Junyi Ye Hanhan Zhang Analyn Lizaso Hao Liu Xinru Mao Hai Huang Qian Chu Chengping Hu | 2019 | Cancer Biology & Medicine2019,16,3: | 4

Deflection estimation of industrial robots with flexible joints显示文摘The application of industrial robots in manufacturing industries has received considerable concerns due to the high flexibility, multifunctionality, and cost-efficiency.It is well known that the robot positioning accuracy is susceptible to the load and motion of robots owing to the insufficient stiffness of robots. Therefore, the machining accuracy improvement has been a research focus in the robotic manufacturing industries in the last decade. To overcome the measurement difficulty of the joint torque and position as well as the complex dynamic coupling between rotors and links, two forward dynamics algorithms for the robot deflection estimation are proposed in this paper. The robot kinematics and dynamics algorithms considering the dynamic coupling between rotors and links are developed based on Lie theory. The forward dynamics equations of robots are solved via the proposed algorithms:the implicit numerical integration algorithm and numerical iterative estimation algorithm. When only the motor position is available, the implicit numerical integration algorithm is employed to solve the forward dynamics equations to estimate the joint torque and position. At the same time, when both the motor position and torque are available, the forward dynamics equations can be reorganized as algebraic equations and solved by the numerical iterative estimation algorithm.Simulations of a 6-DOF serial robot are performed to verify the accuracy of the proposed algorithms. | Huan Liu Yaguo Lei Xiao Yang Wenlei Song Junyi Cao | 2022 | Fundamental Research2022,2,3: | 1

Rewiring ERBB3 and ERK signaling confers resistance to FGFR1 inhibition in gastrointestinal cancer harbored an ERBB3-E928G mutation显示文摘Dear Editor,Recently,a large number of studies found that activation of ERBB3(Erb-B2 receptor tyrosine kinase 3,also known as HER3)may be one of the major mechanisms underlying resistance to therapies that target the EGFR(epidermal growth factor receptor),HER2,and other receptor tyrosine kinases(RTKs)(Chen et al.,2011;Choi et al.,2012;Ross et al.,2018).Interestingly,mutations in ERBB3 are commonly reported in gastrointestinal(Gl)cancer, with mutations identified in approximately 12%of stomach and colorectal cancer cases(Jaiswal et al.,2013). | Xiang Yang Hongxiao Wang Enjun Xie Biyao Tang Qingdian Mu Zijun Song Junyi Chen Fudi Wang Junxia Min | 2020 | Protein & Cell2020,11,12: | 1
Long noncoding RNA(lncRNA)H19:An essential developmental regulator with expanding roles in cancer, stem cell differentiation, and metabolic diseases显示文摘Recent advances in deep sequencing technologies have revealed that, while less than 2%of the human genome is transcribed into mRNA for protein synthesis, over 80%of the genome is transcribed, leading to the production of large amounts of noncoding RNAs(ncRNAs).It has been shown that ncRNAs,especially long non-coding RNAs(lncRNAs),may play crucial regulatory roles in gene expression. As one of the first isolated and reported lncRNAs,H19 has gained much attention due to its essential roles in regulating many physiological and/or pathological processes including embryogenesis,development,tumorigenesis,osteogen-esis,and metabolism.Mechanistically,H19 mediates diverse regulatory functions by serving as competing endogenous RNAs(CeRNAs),Igf2/H19 imprinted tandem gene, modular scaffold, cooperating with H19 antisense, and acting directly with other mRNAs or lncRNAs.Here,we summarized the current understanding of H19 in embryogenesis and development, cancer development and progression, mesenchymal stem cell lineage-specific differentiation, and metabolic diseases. We discussed the potential regulatory mechanisms underlying H19’s func-tions in those processes although more in-depth studies are warranted to delineate the exact molecular, cellular, epigenetic, and genomic regulatory mechanisms underlying the physiolog-ical and pathological roles of H19.Ultimately,these lines of investigation may lead to the development of novel therapeutics for human diseases by exploiting H19 functions. | Junyi Liao Bowen Chen Zhenglin Zhu Chengcheng Du Shengqiang Gao Guozhi Zhao Piao Zhao Yonghui Wang Annie Wang Zander Schwartz Lily Song Jeffrey Hong William Wagstaff Rex CHaydon Hue HLuu Jiaming Fan Russell RReid Tong-Chuan He Lewis Shi Ning Hu Wei Huang | 2023 | Genes & Diseases2023,10,4: | 1

Controllable design of high-efficiency triboelectric materials by functionalized metal–organic frameworks with a large electronwithdrawing functional group显示文摘Triboelectric nanogenerator(TENG)can directly convert mechanical energy into electric energy. However, the triboelectric materials are limited to the triboelectric series. Here, for the first time,we choose the isostructural UiO-66-X(X=H,NH_(2),NO_(2),and Br)family as triboelectric materials to investigate the underlying relationships between different functional groups and the triboelectric performance of TENG.Unlike traditional triboelectric material organic polymers, metal–organic frameworks(MOFs)can be oriented design synthesis and functionalized with various functional groups. The results demonstrate that the largest output voltage and current are from UiO-66-NO_(2) TENG,and are about 23.79 V and 0.29μA,which are 3.19 and 4.14 times over that of the UiO-66 TENG,respectively. The working mechanism of the MOF TENG was discussed in depth through experiments and theoretical calculations. This work proves a novel strategy to obtain high output properties by functionalized MOFs with large electron-withdrawing functional groups and promising guidance for the choice of high-efficiency triboelectric materials. | Rongmei Wen Rui Feng Bo Zhao Jiangfeng Song Liming Fan Junyi Zhai | 2022 | Nano Research2022,15,10: | 1
Measurements of the effective mass transfer areas for the gas–liquid rotating packed bed显示文摘Rotating packed bed(RPB) is one of the most effective gas–liquid mass transfer enhancement reactors, its effective specific mass transfer area(ae) is critical to understand the mass transfer process. By using the NaOH–CO_(2) chemical absorption method, the aevalues of three RPB reactors with different rotor sizes were measured under different operation conditions. The results showed that the high gravity factor and liquid flow rate were major affecting factors, while the gas flow rate exhibited minor influence. The radius of packing is the dominant equipment factor to affect aevalue. The results indicated that the contact area depends on the dispersion of the liquid phase, thus the centrifugal force of rotating packed bed greatly influenced the aevalue. Moreover, the measured ae/ap(effective specific mass transfer area/specific surface area of packing) values were fitted with dimensionless correlation formulas. The unified correlation formula with dimensionless bed size parameter can well predict the experimental data and the prediction errors were within 15%. | Wen Tian Junyi Ji Hongjiao Li Changjun Liu Lei Song Kui Ma Siyang Tang Shan Zhong Hairong Yue Bin Liang | 2023 | Chinese Journal of Chemical Engineering2023,55,3: | 0
Lysobacter hymeniacidonis sp. nov., Isolated from a Crude Oil-Contaminated Marine Sponge显示文摘An aerobic, Gram-negative bacterium, strain 2-5~T, was isolated from a crude oil-contaminated marine sponge collected near Dalian Bay, China, and subjected to a polyphasic taxonomic investigation. Cells of strain 2-5~T were non-spore forming, non-motile, rods 0.2–0.3 μm wide and 1.1–1.2μm long. Strain 2-5~T grew well on nutrient agar, ~TSA, R2 A agar and LB agar. Colonies of strain 2-5~T on LB agar were circular, smooth with entire margins, non-transparent and pale yellow after 3 d of incubation at 30℃. Growth of strain 2-5~T occurred in LN medium with 0–6% Na Cl; no growth occurred in the presence of 8.0% NaCl. Strain 2-5~T grew at 15–42℃ and at pH 6.0–8.0. Comparative 16 S r RNA gene sequence analysis showed that strain 2-5~T clustered with the species of the genus Lysobacter. Its closet neighbors were the type strains of Lysobacter concretionis KC^TC 12205~T(97% similarity), Lysobacter arseniciresistens ZS79~T(96%), and Lysobacter defluii APB-9 ~T(96%). ~The value for DNA-DNA relatedness between strain 2-5~~T and L. concretionis KC^TC 12205 ~T was 23%. Branched fatty acids iso-C16: 0, iso-C15: 0, iso-C11: 0 3-OH, iso-C17: 1ω9c and iso-C11: 0 were found to be predominant. ~The major polar lipids were diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. Strain 2-5~T had a DNA G+C content of 63.8 mol%. On the basis of the phenotypic, chemotaxonomic, DNA-DNA hybridization and phylogenetic data, strain 2-5~T represents a novel species of the genus Lysobacter, for which the name Lysobacter hymeniacidonis sp. nov. is proposed. ~The type strain is 2-5~T(=CGMCC 1.12190 ~T = JCM 18137~T). | XIN Yanjuan QU Junge XU Junyi WU Peichun CAO Xupeng XUE Song | 2015 | Journal of Ocean University of China2015,14,6: | 0
