1Histone methyltransferase NSD2 mediates the survival and invasion of triple-negative breast cancer cells via stimulating ADAM9-EGFR-AKT signaling显示文摘Triple-negative breast cancer (TNBC) is a heterogeneous disease with a poor prognosis due to the lack of an effective targeted therapy. Histone lysine methyltransferases (KMTs) have emerged as attractive drug targets for cancer therapy. However, the function of the majority of KMTs in TNBC has remained largely unknown. In the current study, we found that KMT nuclear receptor binding SET domain protein 2 (NSD2) is overexpressed in TNBC tumors and that its overexpression is associated with poor survival of TNBC patients. NSD2 regulates TNBC cell survival and invasion and is required for tumorigenesis and tumor growth. Mechanistically, NSD2 directly controls the expression of EGFR and ADAM9, a member of the ADAM (a disintegrin and metalloproteinase) family that mediates the release of growth factors, such as HB-EGF. Through its methylase activity, NSD2 overexpression stimulates EGFR-AKT signaling and promotes TNBC cell resistance to the EGFR inhibitor ge?tinib. Together, our results identify NSD2 as a major epigenetic regulator in TNBC and provide a rationale for targeting NSD2 alone or in combination with EGFR inhibitors as a targeted therapy for TNBC.Jun-Jian Wang June X. 4Establishment of adult right ventricle failure in ovine using a graded, animal-specific pulmonary artery constriction model显示文摘Background: Right ventricle failure(RVF) is associated with serious cardiac and pulmonary diseases that contribute significantly to the morbidity and mortality of patients. Currently, the mechanisms of RVF are not fully understood and it is partly due to the lack of large animal models in adult RVF. In this study, we aim to establish a model of RVF in adult ovine and examine the structure and function relations in the RV.Methods: RV pressure overload was induced in adult male sheep by revised pulmonary artery constriction(PAC). Briefly, an adjustable hydraulic occluder was placed around the main pulmonary artery trunk. Then, repeated saline injection was performed at weeks 0, 1, and 4, where the amount of saline was determined in an animal-specific manner. Healthy, age-matched male sheep were used as additional controls. Echocardiography was performed bi-weekly and on week 11 post-PAC, hemodynamic and biological measurements were obtained.Results: This PAC methodology resulted in a marked increase in RV systolic pressure and decreases in stroke volume and tricuspid annular plane systolic excursion, indicating signs of RVF. Significant increases in RV chamber size, wall thickness, and Fulton's index were observed. Cardiomyocyte hypertrophy and collagen accumulation(particularly type III collagen) were evident, and these structural changes were correlated with RV dysfunction.Conclusion: In summary, the animal-specific, repeated PAC provided a robust approach to induce adult RVF, and this ovine model will offer a useful tool to study the progression and treatment of adult RVF that is translatable to human diseases.Michael Nguyen-Truong Wenqiang Liu June Boon Brad Nelson Jeremiah Easley Eric Monnet Zhijie Wang 2020Animal Models and Experimental Medicine2020,3,2:1

8A case study of digital-twin-modelling analysis on power-plant-performance optimizations显示文摘China Energy’s National Institute of Clean-and-Low-Carbon Energy(NICE)is developing a Power Plant Smart Management(PPSM)platform that employs digital-twin technology to undertake techno-economic modelling analysis on China Energy’s existing coal-fired power-plant units and explore cost-effective solutions to improve those plant units’thermal efficiencies and operating performance.This paper presents a case study of PPSM on a 320-MWe coal-fired thermal power-plant unit,demonstrating how the digital-twin technology was employed to explore and analyse optimization solutions.Various optimization solutions and their cost-effectiveness were assessed using the digital-twin-modelling analysis;the results indicated the optimization solutions are expected to improve the plant unit’s operating efficiency and reduce its current electricity-generation coal consumption by up to 3.5 g/kWh standard coal equivalent(sce),worth annual fuel-cost savings of approximately 4 million RMB for a single unit or 8 million RMB for the two identical 320-MWe units that the power plant currently operates.The digital twin was also employed to assess the power-plant unit’s operating economics during both summer and winter.In summer,when the unit operates in electricity-generation-only mode,the unit’s operating thermal efficiency could drop by up to 6%points following the grid demand of load changes from 100%maximum continuous rating(MCR)down to 30% MCR,resulting in an~45 RMB/MWh increase of electricity-generation cost.In winter,when the unit operates in combined heat and power(CHP)cogeneration mode,for the same boiler load,the CHP operation increases the plant unit’s operating profit with increasing district-heating duty,although the relative profit gain from the CHP cogeneration could start to decrease when the district-heating steam-extraction flow increases to a certain point that varies depending on the market prices of heat and electricity,while the fuel cost was found to be equivalent to~50% of the unit’s total CHP income cogenerated from its electricity and district heat outputs.Bin Xu June Wang Xinping Wang Zhihong Liang Liming Cui Xiao Liu Anthony Y.Ku 2019Clean Energy2019,3,3:1

11Probing proton halo of the exotic nucleus ^(28)S by elastic electron scattering显示文摘Elastic electron scattering on the exotic light nucleus 28S is investigated in the plane wave Born approximation. The variation of the squared form factors of 28S with momentum transfer is compared with that of 32S. It is found that the behavior of the form factors near the second minimum (with a moderate momentum transfer) is sensitive to the alteration of the charge density distribution of halo protons in 28S. 14Development of Mg-based Hydrogen Storage Alloy显示文摘Mg-based hydrogen storage alloys are considered as a promising candidate for hydrogen system because of its lightweight, high storage capacity, low price and rich mineral resources. In detail, we reviewed the preparation and properties of Mg-Ni-based hydrogen storage alloys. All kinds of attempts have been done to improve the hydriding and dehydriding behaviors. It is found that the partial substitution of foreign elements can decrease the hydrogen absorption temperature, especially the substitution of a more electronegative element, such as Al and Mn. Mechanical alloying (MA) and mechanical grinding (MG) are the most effective methods to improve the hydriding/dehydriding kinetics and electrochemical capacity, and decrease the desorption tem- perature, but the corrosion resistance is so poor that the 80% of maximum capacity is lost within ten cycles. Microencapsulation is a useful measurement for improving the corrosion resistance and electrocatalytic activity. In order to improve the properties of the alloys for practical applica- tion, the alloys should have a large number of defects, which give activated sites, subsequently, MA, MG and electroless plating should be used to improve the hydriding/dehydriding kinetics and protect the surface of alloys, respectively. The new composite Mg-based alloys give a new way for the hydrogen storage material to practical application.
