1Accumulation and Dynamic Trends of Triterpenoid Saponin in Vegetative Organs of Achyranthus bidentata显示文摘The relationship between structural features of various vegetative organs and triterpenoid saponin accumulation in Achyranthus bidentata Blume was investigated using anatomy, histochemistry and phytochemistry. The results showed that the primary and secondary structures of roots, and the structures of stems and leaves of A. bidentata, were similar to those of ordinary dicotyledonous plants. The enlargement of its roots, however, was primarily associated with growth and differentiation of tertiary structures. There were collateral medullary vascular bundles in addition to the normal vascular bundles in the stem. The tertiary structure was not only main parts in the roots of A. bidentata, but also important storage region of triterpenoid saponin in its growth and development. The stem may be the essential transport organ of triterpenoid saponin, while palisade parenchyma may be the primary synthesis location. In November, the total quantity of triterpenoid saponin and overall biomass in the roots reach a maximum level. This was the best time, therefore, to harvest the roots and corresponded to the traditional harvest period. Despite the withered appearance of leaves, stems also contained substantial amounts of triterpenoid saponin, and it was recommended that the stems of A. bidentata should be used.Jinting Li Zhenghai Hu 2009Journal of Integrative Plant Biology2009,51,2:9
2Activin A prevents neuron-like PC12 cell apoptosis after oxygen-glucose deprivation显示文摘In this study, PC12 cells were induced to differentiate into neuron-like cells using nerve growth facto and were subjected to oxygen-glucose deprivation. Cells were treated with 0, 10, 20, 30, 50, 100 ng/mL exogenous Activin A. The 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl tetrazolium bromide assay and Hoechst 33324 staining showed that the survival percentage of PC12 cells significantly decreased and the rate of apoptosis significantly increased after oxygen-glucose deprivation. Exogenous Activin A significantly increased the survival percentage of PC12 cells in a dose-dependent manner. Reverse transcription-PCR results revealed a significant increase in Activin receptor IIA, Smad3 and Smad4 mRNA levels, which are key sites in the Activin A/Smads signaling pathway, in neuron-like cells subjected to oxygen-glucose deprivation, while mRNA expression of the apoptosis-regulation gene caspase-3 decreased. 3Preparation of an anti-diethylstilbestrol monoclonal antibody and development of an indirect competitive ELISA to detect diethylstilbestrol in biological samples显示文摘Based on the preparation of an anti-diethylstilbestrol(DES) monoclonal antibody,a simple and convenient indirect competitive enzyme-linked immunosorbent assay(ELISA) method for DES detection has been developed.The monoclonal antibody demonstrated high sensitivity to DES with an IC50 value of 275 pg mL-1 and detection limit(LOD) of 90 pg mL-1.The specificity of the assay was studied by measuring cross-reactivity of the antibody with structurally related compounds of ethinyl estradiol(<7%),estrone(<0.1%),estriol(<0.1%),and diethylstilbestrol benzoate(<0.1%).Chicken,fish,shrimp,urine and bile spiked with different concentration of DES were detected by the developed method,and the recovery rates were greater than 79.5%.Intra-and inter-assay variations were about 6%.This method exhibited high stability with a coefficient of variation less than 10% in buffer and in real samples.The LODs in fish/shrimp,liver,feed and urine spiked with DES were 600,600,4800 and 600 pg mL-1,respectively.These results confirmed that the antibody to DES was successfully produced and could be used to establish ELISA methods for DES detection in food producing animals.LI WeiHua MENG Meng HE FangYang WAN YuPing XUE HuYin LIU Wei YIN WeiWei XU Jing FENG CaiWei WANG ShanLiang LU Xiao LIU JinTing XI RiMo 2011Chinese Science Bulletin2011,56,8:4
4Enhancing nitrogen removal from low carbon to nitrogen ratio wastewater by using a novel sequencing batch biofilm reactor显示文摘Removing nitrogen from wastewater with low chemical oxygen demand/total nitrogen(COD/TN) ratio is a difficult task due to the insufficient carbon source available for denitrification. Therefore, in the present work, a novel sequencing batch biofilm reactor(NSBBR) was developed to enhance the nitrogen removal from wastewater with low COD/TN ratio. The NSBBR was divided into two units separated by a vertical clapboard. Alternate feeding and aeration was performed in the two units, which created an anoxic unit with rich substrate content and an aeration unit deficient in substrate simultaneously.Therefore, the utilization of the influent carbon source for denitrification was increased,leading to higher TN removal compared to conventional SBBR(CSBBR) operation. The results show that the CSBBR removed up to 76.8%, 44.5% and 10.4% of TN, respectively, at three tested COD/TN ratios(9.0, 4.8 and 2.5). In contrast, the TN removal of the NSBBR could reach 81.9%, 60.5% and 26.6%, respectively, at the corresponding COD/TN ratios. Therefore,better TN removal performance could be achieved in the NSBBR, especially at low COD/TN ratios(4.8 and 2.5). 5Automatic modal parameter identification of high arch dams:feasibility verification显示文摘Modal parameters, including fundamental frequencies, damping ratios, and mode shapes, could be used to evaluate the health condition of structures. Automatic modal parameter identification, which plays an essential role in realtime structural health monitoring, has become a popular topic in recent years. In this study, an automatic modal parameter identification procedure for high arch dams is proposed. The proposed procedure is implemented by combining the densitybased spatial clustering of applications with noise(DBSCAN) algorithm and the stochastic subspace identification(SSI). The 210-m-high Dagangshan Dam is investigated as an example to verify the feasibility of the procedure. The results show that the DBSCAN algorithm is robust enough to interpret the stabilization diagram from SSI and may avoid outline modes. This leads to the proposed procedure obtaining a better performance than the partitioned clustering and hierarchical clustering algorithms. In addition, the errors of the identified frequencies of the arch dam are within 4%, and the identified mode shapes are in agreement with those obtained from the finite element model, which implies that the proposed procedure is accurate enough to use in modal parameter identification. 7Upgrade of three municipal wastewater treatment lagoons using a high surface area media显示文摘Abstract Lagoon-based municipal wastewater treatment plants (WWTPs) are facing difficulties meeting the needs of rapid population growth as well as the more stringent requirements of discharge permits. Three municipal WWTPs were modified using a high surface area media with upgraded fine-bubble aeration systems. Performance data collected showed very promising results in terms of five-day biochemical oxygen demand (BOD5), ammonia (NH3) and total suspended solids (TSS) removal. Two-year average ammonia effluents were 4.1 mg. L-1 for Columbia WWTP, 4 mg. L-1 for Larchmont WWTP and 2.1 mg. L-1 for Laurelville WWTE respectively. 9CDK7-YAP-LDHD axis promotes D-lactate elimination and ferroptosis defense to support cancer stem cell-like properties显示文摘Reprogrammed cellular metabolism is essential for maintaining cancer stem cells(CSCs)state.Here,we report that mitochondrial D-lactate catabolism is a necessary initiating oncogenic event during tumorigenesis of esophageal squamous cell carcinoma(ESCC).We discover that cyclin-dependent kinase 7(CDK7)phosphorylates nuclear Yes-associated protein 1(YAP)at S127 and S397 sites and enhances its transcription function,which promotes D-lactate dehydrogenase(LDHD)protein expression.Moreover,LDHD is enriched significantly in ESCC-CSCs rather than differentiated tumor cells and high LDHD status is connected with poor prognosis in ESCC patients.Mechanistically,the CDK7-YAP-LDHD axis helps ESCC-CSCs escape from ferroptosis induced by D-lactate and generates pyruvate to satisfy energetic demands for their elevated self-renewal potential.Hence,we conclude that esophageal CSCs adopt a D-lactate elimination and pyruvate accumulation mode dependent on CDK7-YAP-LDHD axis,which drives stemness-associated hallmarks of ESCC-CSCs.Reasonably,targeting metabolic checkpoints may serve as an effective strategy for ESCC therapy.Mengzhu Lv Ying Gong Xuesong Liu Yan Wang Qingnan Wu Jie Chen Qingjie Min Dongyu Zhao Xianfeng Li Dongshao Chen Di Yang Danna Yeerken Rui Liu Jinting Li Weimin Zhang Qimin Zhan 2023Signal Transduction and Targeted Therapy2023,8,9:1

11Defect of SLC38A3 promotes epithelial-mesenchymal transition and predicts poor prognosis in esophageal squamous cell carcinoma显示文摘Objective: Solute carrier family 38(SLC38 s) transporters play important roles in amino acid transportation and signaling transduction. However, their genetic alterations and biological roles in tumors are still largely unclear.This study aimed to elucidate the genetic signatures of SLC38 s transporters and their implications in esophageal squamous cell carcinoma(ESCC).Methods: Analyses on somatic mutation and copy number alterations(CNAs) of SLC38 A3 were performed as described. Immunohistochemistry(IHC) assay and Western blot assay were used to detect the protein expression level. MTS assay, colony formation assay, transwell assay and wound healing assay were used to explore the malignant phenotypes of ESCC cells. Immunofluorescence assay was used to verify the colocalization of two indicated proteins and immunopreciptation assay was performed to confirm the interaction of proteins.Results: Our findings revealed that SLC38 s family was significantly disrupted in ESCC, with high frequent CNAs and few somatic mutations. SLC38 A3 was the most frequent loss gene among them and was linked to poor survival and lymph node metastasis. The expression of SLC38 A3 was lower in tumor tissues compared to that in normal tissues, which was also significantly associated with worse clinical outcome. Further experiments revealed that depletion of SLC38 A3 could promote EMT in ESCC cell lines, and the interaction of SLC38 A3 and SETDB1 might lead to the reduced transcription of Snail. 13Enhanced properties of solid solution (CeZr)O_2 modified with metal oxides for catalytic oxidation of low-concentration methane显示文摘The solid solution(Ce Zr)O_2 catalyst was synthesized, and it was modified with metal oxides by incipient impregnation. Morphology and structure were characterized by X-ray diffraction, transmission electron microscope, nitrogen ad/desorption and H_2-temperature program reduction techniques. The catalytic properties of methane oxidation were also investigated. The results showed that solid solution possessed a mesoporous structure and exhibited excellent catalytic performance. The activity of solid solution was improved effectively by nickel doping, and the optimal loading is 15 wt%. 14Enhanced hydrogen storage properties of NaBH_(4)–Mg(BH_(4))_(2) composites by NdF_(3) addition显示文摘As two important members of complex hydrides,Mg(BH_(4))2 and Na BH_(4) have a high gravimetric capacity(14.9 and10.8 wt%,respectively).In this study,the Mg(BH_(4))2 was synthesized by the ion exchange method.Afterwards,the Mg(BH_(4))_(2) and Na BH_(4) composites with different amounts(30,40 and 50 wt%)of NdF3 were prepared by mechanical milling.Effects of the Nd F3 on the microstructural evolution and hydrogen storage properties were investigated.The results show that Nd F3catalyst can significantly improve the dehydrogenation kinetics of the eutectic composites of Na BH_(4)–Mg(BH_(4))_(2).The onset hydrogen desorption temperature of the composites is about88.6C,which is about 110C lower than that of Mg(BH_(4))_(2)and Na BH_(4)composites.Mg(BH_(4))_(2)–Na BH_(4)-0.5NdF3composites can released 5.2 wt%H_(2)at 250C within 30 min,and the dehydrogenation capacity is significantly higher than that of Mg(BH_(4))_(2)–Na BH_(4)composites.The analysis of the dehydrogenation mechanism reveals that NdF3takes participate in the reaction to generate Na MgF3 to promote the dehydrogenation reaction process of the composites.Jianguang Yuan Jinting Chen Haixiang Huang Yujie Lv Bogu Liu Zhongyu Li Bao Zhang Wei Lv Ying Wu 2021Progress in Natural Science:Materials International2021,31,4:1

16Use of RNAi silencing to target preconditioned glial cell line-derived neurotrophic factor in neuronal apoptosis显示文摘Several studies have suggested that exogenous glial cell line-derived neurotrophic factor may pro-tect neurons from cerebral ischemic injury. However, the mechanisms underlying the neuroprotec-tive effects of endogenous glial cell line-derived neurotrophic factor remain unclear. The present experiments sought to elucidate the influence of various conditioned media on neuronal apoptosis, using a normal culture medium for astrocytes, an astrocyte medium highly expressing glial cell line-derived neurotrophic factor, and an astrocyte medium in which glial cell line-derived neurotro-phic factor expression was silenced using RNAi technology. The results confirmed that the use of RNAi silencing to target pretreated glial cell line-derived neurotrophic factor expression promoted neuronal apoptosis. 20High-efficiency genome editing of an extreme thermophile Thermus thermophilus using endogenous type I and type III CRISPR-Cas systems显示文摘Thermus thermophilus is an attractive species in the bioindustry due to its valuable natural products,abundant thermophilic enzymes,and promising fermentation capacities.However,efficient and versatile genome editing tools are not available for this species.In this study,we developed an efficient genome editing tool for T.thermophilus HB27 based on its endogenous type IB,I-C,and III-A/B CRISPR-Cas systems.First,we systematically characterized the DNA interference capabilities of the different types of the native CRISPR-Cas systems in T.thermophilus HB27.We found that genomic manipulations such as gene deletion,mutation,and in situ tagging could be easily implemented by a series of genome-editing plasmids carrying an artificial self-targeting mini-CRISPR and a donor DNA responsible for the recombinant recovery.We also compared the genome editing efficiency of different CRISPR-Cas systems and the editing plasmids with donor DNAs of different lengths.Additionally,we developed a reporter gene system for T.thermophilus based on a heat-stableβ-galactosidase gene TTP0042,and constructed an engineered strain with a high production capacity of superoxide dismutases by genome modification.Jinting Wang Junwei Wei Haijuan Li Yingjun Li 2022mLife2022,1,4:0
