| 1 | The resurgent landscape of xenotransplantation of pig organs in nonhuman primates显示文摘Organ shortage is a major bottleneck in allotransplantation and causes many wait-listed patients to die or become too sick for transplantation.Genetically engineered pigs have been discussed as a potential alternative to allogeneic donor organs.Although xenotransplantation of pig-derived organs in nonhuman primates(NHPs)has shown sequential advances in recent years,there are still underlying problems that need to be completely addressed before clinical applications,including(i)acute humoral xenograft rejection;(ii)acute cellular rejection;(iii)dysregulation of coagulation and inflammation;(iv)physiological incompatibility;and(v)cross-species infection.Moreover,various genetic modifications to the pig donor need to be fully characterized,with the aim of identifying the ideal transgene combination for upcoming clinical trials.In addition,suitable pretransplant screening methods need to be confirmed for optimal donor-recipient matching,ensuring a good outcome from xenotransplantation.Herein,we summarize the understanding of organ xenotransplantation in pigs-to-NHPs and highlight the current status and recent progress in extending the survival time of pig xenografts and recipients.We also discuss practical strategies for overcoming the obstacles to xenotransplantation mentioned above to further advance transplantation of pig organs in the clinic. | Xuan Zhang Quancheng Wang Jingjun Zhao Xiao Li Wei Peng Zhaoxu Yang Zhibin Lin Long Yang Rui Ding Kaishan Tao Kefeng Dou | 2021 | Science China(Life Sciences)2021,64,5: | 10 |
| 2 | A unified approach to type synthesis of both rigid and flexure parallel mechanisms显示文摘Type synthesis of both rigid and compliant parallel mechanisms has become a hot issue in the field of mechanisms and robotics research in recent years. A unified approach to type synthesis of the two classes of mechanisms, however, has not been referred and investigated up to date. Based on the state-of-art analysis for several major type synthesis approaches related to rigid and compliant mechanisms, respectively, it proves feasible to establish a unified methodology for type synthesis of these two classes of mechanisms. That is a synthesis philosophy in terms of the hierarchy mapping between mathematic, physical, and mechanical building blocks in the framework of screw theory, as addressed in this paper. The key point of the proposed method lies in establishing the mapping among three different building blocks (i.e. geometric building block, kinematic or constraint building block, and mechanical building block). As a result, it makes the whole type synthesis process simple and visible. By using the proposed method, two examples are taken to verify the effectiveness for the type synthesis of both rigid and flexure mechanisms. | 3 | Intensive phototherapy vs.exchange transfusion for the treatment of neonatal hyperbilirubinemia:a multicenter retrospective cohort study显示文摘Background:Intensive phototherapy(IPT)and exchange transfusion(ET)are the main treatments for extreme hyperbilirubinemia.However,there is no reliable evidence on determining the thresholds for these treatments.This multicenter study compared the effectiveness and complications of IPT and ET in the treatment of extreme hyperbilirubinemia.Methods:This retrospective cohort study was conducted in seven centers from January 2015 to January 2018.Patients with extreme hyperbilirubinemia that met the criteria of ET were included.Patients were divided into three subgroups(low-,medium-,and high-risk)according to gestational week and risk factors.Propensity score matching(PSM)was performed to balance the data before treatment.Study outcomes included the development of bilirubin encephalopathy,duration of hospitalization,expenses,and complications.Mortality,auditory complications,seizures,enamel dysplasia,ocular motility disorders,athetosis,motor,and language development were evaluated during follow-up at age of 3 years.Results:A total of 1164 patients were included in this study.After PSM,296 patients in the IPT only group and 296 patients in the IPT plus ET group were further divided into the low-,medium-,and high-risk subgroups with 188,364,and 40 matched patients,respectively.No significant differences were found between the IPT only and IPT plus ET groups in terms of morbidity,complications,and sequelae.Hospitalization duration and expenses were lower in the low-and medium-risk subgroups in the IPT only group.Conclusions:In this study,our results suggest that IPT is a safe and effective treatment for extreme hyperbilirubinemia.The indication of ET for patients with hyperbilirubinemia could be stricter.However,it is necessary to have a contingency plan for emergency ET as soon as IPT is commenced especially for infants with risk factors.If IPT can be guaranteed and proved to be therapeutic,ET should be avoided as much as possible. | Meng Zhang Yang He Jun Tang Wenbin Dong Yong Zhang Benjin Zhang Hong Wan Quanmin Deng Lirong Guan Bin Xia Zhong Chen Min Ge Jing Zhao Wenxing Li Jingjun Pei Yi Qu Dezhi Mu | 2022 | Chinese Medical Journal2022,,5: | 7 |

| 5 | Effect of Yupingfeng granules on the skin barrier in atopic dermatitis mice models显示文摘OBJECTIVE: To investigate effect of Yupingfeng granules, prepared with Chinese Medicines, on the wound healing and on the expression of aquaporin3(AQP3) and the skin barrier in the animal models of atopic dermatitis(AD).METHODS: Acute skin lesions of AD models were prepared using 2,4-dinitrochlorobenzo(DNCB) in mice and animals were treated with either Yupingfeng granules or placebo for two weeks. Skin wound healing outcome was assessed by measuring skin thickness, weight(quality) of the skin, and trans-epidermal water loss(TEWL). Expression of AQP3 mRNA and protein was assessed by reverse transcriotion polymerase chain reaction(RT-PCR)and immunoblotting, respectively.RESULTS: Yupingfeng granule treatment resulted in significant acceleration of wound healing with63.64% efficiency, which was significantly higher than that of placebo granule treatment(31.82%,P < 0.01 by Wilcoxon Rank-sum test). Skin thickness, weight of the wounded skin, and TEWL were significantly higher in the AD models compared to that of normal animals. Treatment with Yupingfeng granules resulted in significant decrease in skin thickness [(937 ± 31) vs(360 ± 21) μm, P < 0.01],weight of the wounded skin [(42 ± 4) vs(24 ± 5)mg, P < 0.01], and TEWL [(30 ± 4) vs(13 ± 4) g·h^(-1)·m-2, P < 0.01]. | 6 | Periodically poled lithium niobate whispering gallery mode microcavities on a chip显示文摘In this study, we investigate the fabrication of periodically poled lithium niobate(PPLN) microdisk cavities on a chip. These resonators are fabricated from a PPLN film with a 16 μm poling period on insulator using conventional microfabrication techniques.The quality factor of the PPLN microdisk resonators with a 40-μm radius and a 700-nm thickness is 6.7×10~5. Second harmonic generation(SHG) with an efficiency of 2.2×10^(-6) mW(-1) is demonstrated in the fabricated PPLN microdisks. | 7 | Experimental Study of Waste Tire Rubber,Wood-Plastic Particles and Shale Ceramsite on the Performance of Self-Compacting Concrete显示文摘In recent decades,the utilization of waste tires,plastic and artificial shale ceramsite as alternative fine aggregate to make self-compacting concrete(SCC)has been recognized as an eco-friendly and sustainable method to manufacture renewable construction materials.In this study,three kinds of recycled aggregates:recycled tire rubber particles,wood-plastic particles,artificial shale ceramsite were used to replace the sand by different volume(5%,10%,20%and 30%),and their effects on the fresh and hardened properties of SCC were investigated.The slump flow and V-funnel tests were conducted to evaluate the fresh properties of modified-SCC mixtures.The hardened properties include 3,7 and 28-day compressive strengths,axial compressive strength,static elastic modulus,and compressive stress-strain behavior at 28 days.The test results showed that the incorporation of these three kinds of alternative aggregates had a negative impact on the fresh properties of SCC.Besides,the 28-day compressive strength and axial compressive strength decreased with the increase of rubber and wood-plastic particles content.In this experiment,all the three kinds of recycled aggregates can improve the ductility and deformability of SCC,and the most excellent performance comes from SCC with recycled rubber particles. | Lei Tian Liuchao Qiu Jingjun Li Yongsen Yang | 2020 | Journal of Renewable Materials2020,8,2: | 5 |
| 8 | 7,8-Dihydroxyflavone suppresses proliferation and induces apoptosis of human osteosarcoma cells显示文摘Recent studies suggest that 7,8-dihydroxyflavone(7,8-DHF)inhibits the development of several tumors.However,its role in osteosarcoma(OS)remains unknown.This study was designed to investigate the effects and underlying mechanisms of 7,8-DHF that may influence OS development.Human OS cell lines(U2OS and 143B)were treated with 7,8-DHF;cell viability and cell migration were assessed by methylthiazolyldiphenyl-tetrazolium bromide(MTT)assay and wound-healing assay,respectively;and cell death and apoptosis were evaluated by LIVE/DEAD staining and terminal deoxynucleotidyl transferase(TdT)dUTP nick-end labeling(TUNEL)assay,respectively.Reactive oxygen species production was measured using 2,7-dichlorodihydrofluorescein diacetate probe.Akt,Bcl-xL/Bcl-2 asociated death promoter(Bad),p38 mitogen-activated protein kinase(MAPK),extracellular regulated protein kinase(ERK),and c-Jun N-terminal kinase(JNK)expression and their respective phosphorylation levels were detected by western blot analysis.We found that 7,8-DHF reduced cell viability in a dose-dependent manner and also promoted apoptosis,inhibited migration,and induced oxidative stress in OS cells.Moreover,7,8-DHF inhibited Akt,Bad,and p38MAPK,but activated ERK and JNK signals.In summary,our results suggest that 7,8-DHF inhibits OS progression,possibly by regulating Akt/Bad and MAPK signaling.These findings provide new evidence for the pharmacological effects of 7,8-DHF that may improve drug therapy for OS patients. | Jing Zhao Peifeng Li Hua Zhu Fengqin Ge Jie Liu Jingjun Xia Pengzhou Hang | 2021 | Acta Biochimica et Biophysica Sinica2021,53,7: | 3 |
| 9 | First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. Particularly, various targets synthesized by hot pressing mixture of Cr, Al, and C powders at 650–1000°C were used. It was found that regardless of the phase compositions and density of the composite targets, when the molar ratio of Cr:Al:C in the starting materials was 2:1:1, phase-pure crystalline Cr_2AlC coatings were prepared by magnetron sputtering and post crystallization. The Cr_2AlC coatings were dense and crack-free and had a duplex structure. | 12 | Porphyromonas gingivalis bacteremia increases the permeability of the blood-brain barrier via the Mfsd2a/Caveolin-1 mediated transcytosis pathway显示文摘Bacteremia induced by periodontal infection is an important factor for periodontitis to threaten general health. P. gingivalis DNA/virulence factors have been found in the brain tissues from patients with Alzheimer’s disease(AD). The blood-brain barrier(BBB) is essential for keeping toxic substances from entering brain tissues. However, the effect of P. gingivalis bacteremia on BBB permeability and its underlying mechanism remains unclear. In the present study, rats were injected by tail vein with P. gingivalis three times a week for eight weeks to induce bacteremia. An in vitro BBB model infected with P. gingivalis was also established. We found that the infiltration of Evans blue dye and Albumin protein deposition in the rat brain tissues were increased in the rat brain tissues with P. gingivalis bacteremia and P. gingivalis could pass through the in vitro BBB model. Caveolae were detected after P. gingivalis infection in BMECs both in vivo and in vitro. Caveolin-1(Cav-1) expression was enhanced after P. gingivalis infection.Downregulation of Cav-1 rescued P. gingivalis-enhanced BMECs permeability. We further found P. gingivalis-gingipain could be colocalized with Cav-1 and the strong hydrogen bonding between Cav-1 and arg-specific-gingipain(RgpA) were detected.Moreover, P. gingivalis significantly inhibited the major facilitator superfamily domain containing 2a(Mfsd2a) expression. Mfsd2a overexpression reversed P. gingivalis-increased BMECs permeability and Cav-1 expression. | 13 | First results on ^(76)Ge neutrinoless double beta decay from CDEX-1 experiment显示文摘We report the first results on ^(76)Ge neutrinoless double beta decay from stage one of the China dark-matter experiment(CDEX).A p-type point-contact high-purity germanium detector with a mass of 994 g has been installed to detect neutrinoless double beta decay events, as well as to directly detect dark matter particles. An exposure of 304 kg d has been analyzed over a wide spectral band from 500 keV to 3 MeV. The average event rate obtained was about 0.012 counts per keV per kg per day over the 2.039 MeV energy range. The half-life of ^(76)Ge neutrinoless double beta decay derived based on this result is T^(0ν)1/2>6.4×10^(22) yr(90% C.L.). | 15 | Femtosecond laser-induced periodic surface structures on lithium niobate crystal benefiting from sample heating显示文摘Periodic surface structures were fabricated by irradiating lithium niobate(LN) crystals with femtosecond laser pulses at sample temperatures ranging from 28℃ to 800℃. Carrier density and conductivity of the samples were increased via heating LN, which inhibited coulomb explosion to obtain a uniform periodic surface structure. The periodic surface structures cover an area of 8 mm × 8 mm and have an average spacing of 174 5 nm.Meanwhile, the absorption of the processed sample is about 70% in the spectral range of 400–1000 nm, which is one order of magnitude higher than that of pure LN. | 17 | Predictive value of MGMT promoter methylation on the survival of TMZ treated IDH-mutant glioblastoma显示文摘Objective:O6 methylguanine-DNA methyltransferase(MGMT)promoter methylation is a biomarker widely used to predict the sensitivity of IDH-wildtype glioblastoma to temozolomide therapy.Given that the IDH status has critical effects on the survival and epigenetic features of glioblastoma,we aimed to assess the role of MGMT promoter methylation in IDH-mutant glioblastoma.Methods:This study included 187 IDH-mutant glioblastomas and used 173 IDH-wildtype glioblastomas for comparison.KaplanMeier curves and multivariate Cox regression were used to study the predictive effects.Results:Compared with IDH-wildtype glioblastomas,IDH-mutant glioblastomas showed significantly higher(P<0.0001)MGMT promoter methylation.We demonstrated that MGMT promoter methylation status,as determined by a high cutoff value(≥30%)in pyrosequencing,could be used to significantly stratify the survival of 50 IDH-mutant glioblastomas receiving temozolomide therapy(cohort A);this result was validated in another cohort of 25 IDH-mutant glioblastomas(cohort B).The median progression-free survival and median overall survival in cohort A were 9.33 and 13.76 months for unmethylated cases,and 18.37 and 41.61 months for methylated cases,and in cohort B were 6.97 and 9.10 months for unmethylated cases,and 23.40 and 26.40 months for methylated cases.In addition,we confirmed that the MGMT promoter methylation was significantly(P=0.0001)correlated with longer OS in IDH-mutant patients with GBM,independently of age,gender distribution,tumor type(primary or recurrent/secondary),and the extent of resection.Conclusions:MGMT promoter methylation has predictive value in IDH-mutant glioblastoma,but its cutoff value should be higher than that for IDH-wildtype glioblastoma. | Ruichao Chai Guanzhang Li Yuqing Liu Kenan Zhang Zheng Zhao Fan Wu Yuzhou Chang Bo Pang Jingjun Li Yangfang Li Tao Jiang Yongzhi Wang | 2021 | Cancer Biology & Medicine2021,18,1: | 2 |
| 18 | Effects of High Temperature Oxidation on Mechanical Properties of Ti_3AlC_2显示文摘The oxidation tests of Ti_3AlC_2 were conducted at 1100 and 1200?C in air for 48 and 360 h, respectively,and the effects of high temperature oxidation on the flexural strength and hardness of Ti_3AlC_2 were investigated. The microstructure, grain size and phase compositions of Ti_3AlC_2 substrate didn't change after oxidation, hence the oxide removed Ti_3AlC_2 substrate maintained its initial flexural strength and hardness. However, the flexural strength of oxide retained Ti_3AlC_2 decreased by about 5%. Acoustic emission monitoring indicated that during the process of three-point bending test, the formed Al_2O_3 scale on Ti_3AlC_2 surface fractured firstly in a cleavage manner, then the substrate/oxide interface cracked,and finally the Ti_3AlC_2 substrate fractured. The mechanical degradation was caused by the preferential formation of cracks in brittle Al_2O_3 scale as well as at defective and lacunose grain boundaries of the substrate where stress concentration generated. The mechanical degradation was insensitive to oxidation temperature and time in the present conditions. | 19 | Low threshold random lasing in dye-doped and strongly disordered chiral liquid crystals显示文摘Random lasing was experimentally investigated in pyrromethene 597-doped strongly disordered chiral liquid crystals (CLCs) composed of the nematic liquid crystal SLC1717 and the chiral agent CB15. The concentration of the chiral agent tuned the bandgap, and disordered CLC microdomains were achieved by fast quenching of the mixture from the isotropic to the cholesteric phase. Random lasing and band edge lasing were observed synchronously, and their behavior changed with the spectral location of the bandgap. The emission band for band edge lasing shifted with the change of the bandgap, while the emission band for random lasing remained practically constant. | 20 | Wavelength dependence of light propagation in a water suspension of anisotropic scatterers显示文摘We experimentally study the wavelength dependence of light propagation in a water suspension of lithium niobate microcrystalline particles.First,the ballistic transmission in the visible range of the suspension is measured.The nonlinear relationship is observed between the transport mean free path and the wavelength of the incident light.Secondly,we measure the coherent backscattering (CBS) of the sample at different wavelengths.The full width at half maximum of the CBS cone at 532 nm is about 1.24 times as large as that at 671 nm.The results indicate that the light with a long wavelength propagates further than the short wavelength light and the localization state of the short one is stronger.Finally,we investigate the light-controllable CBS experiments in the disordered materials of anisotropic scatterers,which show that the configuration of pump light with the longer wavelength and the probe light with the shorter wavelength performs better. | SHI Fan ZHANG XinZheng LI Jun WANG PiDong XU Yan YU XuanYi XU JingJun | 2011 | Science China(Physics,Mechanics & Astronomy)2011,54,11: | 2 |
