1Craniocerebral injury promotes the repair of peripheral nerve injury显示文摘The increase in neurotrophic factors after craniocerebral injury has been shown to promote fracture healing. Moreover, neurotrophic factors play a key role in the regeneration and repair of peripheral nerve. However, whether craniocerebral injury alters the repair of peripheral nerve injuries remains poorly understood. Rat injury models were established by transecting the left sciatic nerve and using a free-fall device to induce craniocerebral injury. Compared with sciatic nerve injury alone after 6–12 weeks, rats with combined sciatic and craniocerebral injuries showed decreased sciatic functional index, increased recovery of gastrocnemius muscle wet weight, recovery of sciatic nerve ganglia and corresponding spinal cord segment neuron morphologies, and increased numbers of horseradish peroxidase-labeled cells. 2Antioxidation Activities of Low-Molecular-Weight Gelatin Hydrolysate Isolated from the Sea Cucumber Stichopus japonicus显示文摘Gelatin extracted from the body wall of the sea cucumber (Stichopus japonicus) was hydrolyzed with flavourzyme.Low-molecular-weight gelatin hydrolysate (LMW-GH) of 700-1700 Da was produced using an ultrafiltration membrane bioreactor system.Chemiluminescence analysis revealed that LMW-GH scavenges high free radicals in a concentration-dependent manner;IC50 value for superoxide and hydroxyl radicals was 442 and 285 μg mL-1,respectively.LMW-GH exhibited excellent inhibitory characteristics against melanin synthesis and tyrosinase activity in B16 cells.Furthermore,LMW-GH notably increased intracellular glutathione (GSH),which in turn suppressed melanogenesis.LMW-GH performs antioxidation activity,holding the potential of being used as a valuable ingredient in function foods,cosmetics and pharmaceuticals or nutriceuticals.WANG Jingfeng WANG Yuming TANG Qingjuan WANG Yi CHANG Yaoguang ZHAO Qin XUE Changhu 2010Journal of Ocean University of China2010,9,1:13
3Test Data Generation for Stateful Network Protocol Fuzzing Using a Rule-Based State Machine显示文摘To improve the efficiency and coverage of stateful network protocol fuzzing, this paper proposes a new method, using a rule-based state machine and a stateful rule tree to guide the generation of fuzz testing data. The method first builds a rule-based state machine model as a formal description of the states of a network protocol.This removes safety paths, to cut down the scale of the state space. Then it uses a stateful rule tree to describe the relationship between states and messages, and then remove useless items from it. According to the message sequence obtained by the analysis of paths using the stateful rule tree and the protocol specification, an abstract data model of test case generation is defined. The fuzz testing data is produced by various generation algorithms through filling data in the fields of the data model. Using the rule-based state machine and the stateful rule tree,the quantity of test data can be reduced. 4Semi-valid Fuzz Testing Case Generation for Stateful Network Protocol显示文摘Network protocols are divided into stateless and stateful.Stateful network protocols have complex communication interactions and state transitions.However,the existing network protocol fuzzing does not support state transitions very well.This paper focuses on this issue and proposes the Semi-valid Fuzzing for the Stateful Network Protocol(SFSNP).The SFSNP analyzes protocol interactions and builds an extended finite state machine with a path marker for the network protocol; then it obtains test sequences of the extended finite state machine,and further performs the mutation operation using the semi-valid algorithm for each state transition in the test sequences; finally,it obtains fuzzing sequences.Moreover,because different test sequences may have the same state transitions,the SFSNP uses the state transition marking algorithm to reduce redundant test cases.By using the stateful rule tree of the protocol,the SFSNP extracts the constraints in the protocol specifications to construct semi-valid fuzz testing cases within the sub-protocol domain,and finally forms fuzzing sequences.Experimental results indicate that the SFSNP is reasonably effective at reducing the quantity of generated test cases and improving the quality of fuzz testing cases.The SFSNP can reduce redundancy and shorten testing time.Rui Ma Shuaimin Ren Ke Ma Changzhen Hu Jingfeng Xue 2017Tsinghua Science and Technology2017,22,5:6
5IGHG1 promotes motility likely through epithelial-mesenchymal transition in ovarian cancer显示文摘Objective: Ovarian cancer(OC) is one of the leading causes of death for female cancer patients. COC166-9 is an OC-specific monoclonal antibody and we have identified immunoglobulin γ-1 heavy chain constant region(IGHG1) as its antigen. We explore the function of IGHG1 in proliferation, apoptosis and motility of OC cells further in this research.Methods: IGHG1 expression in OC specimens was detected through immunohistochemistry. Real-time quantitative polymerase chain reaction(RT-q PCR) or western blotting assay was used to test IGHG1 expression in OC cells. Viability of OC cells was tested by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay. Flow cytometry or western blotting assay was used to detect cell cycle and apoptosis. Cellular motility was analyzed by using transwell assay and the markers of epithelial-mesenchymal transition(EMT) were tested through immunoblots.Results: Although it exerts negligible effect on the viability and apoptosis of OC cells, IGHG1 could promote migration and invasion of malignant cells in vitro. Mechanistically, IGHG1 increases the expression of N-cadherin and Vimentin while decreases E-cadherin expression. Additionally, IGHG1 expression in OC specimens is higher relative to the paired normal counterparts. Further analysis demonstrates that the increased IGHG1 expression correlates positively with the lymph node metastasis of OC.Conclusions: IGHG1 promotes the motility of OC cells likely through executing the EMT program. 6Sericin can reduce hippocampal neuronal apoptosis by activating the Akt signal transduction pathway in a rat model of diabetes mellitus显示文摘In the present study, a rat model of type 2 diabetes mellitus was established by continuous peritoneal injection of streptozotocin. Following intragastric perfusion of sericin for 35 days, blood glucose levels significantly reduced, neuronal apoptosis in the hippocampal CA1 region decreased, hippocampal phosphorylated Akt and nuclear factor kappa B expression were enhanced, but Bcl-xL/Bcl-2 associated death promoter expression decreased. 7Effects of sericin on heme oxygenase-1 expression in the hippocampus and cerebral cortex of type 2 diabetes mellitus rats显示文摘Previous studies have demonstrated that sericin effectively reduces blood glucose,and protects islet cells,as well as the gonads and kidneys.However,whether sericin improves diabetes mellitus-induced structural and functional problems in the central nervous system remains poorly understood.Rat models of type 2 diabetes mellitus were established by intraperitoneal injection of streptozotocin.The present study observed histological changes in the hippocampus and cerebral cortex,as well as heme oxygenase-1 expression,and explored sericin effects on the central nervous system in diabetic rats.Pathological damage to neural cells in the rat hippocampus and cerebral cortex was relieved following intragastric administration of sericin at a dose of 2.4 g/kg for 35 consecutive days.Heme oxygenase-1 protein and mRNA expressions were decreased in the hippocampus and cerebral cortex of diabetes mellitus rats after sericin treatment.The results suggest that sericin plays a protective effect on the nervous system by decreasing the high expression of heme oxygenase-1 following diabetes mellitus.Zhihong Chen Yaqiang He Wenliang Fu Jingfeng Xue 2011Neural Regeneration Research2011,6,6:4
8Hpyerglycemic Effect of a Mixture of Sea Cucumber and Cordyceps Sinensis in StreptozotocinInduced Diabetic Rat显示文摘Sea cucumber and cordyceps sinensis are used as both food and traditional medicines in Asia. This study was carried out in order to investigate the hpyerglycemic effect of a mixture of sea cucumber(Apostichopus japonicas) and cordyceps sinensis(Cordyceps militaris)(SCC) in diabetic rat and explore the mechanism underlining such an effect. The diabetic model rat was induced with intraperitoneal injection of streptozotocin(STZ). The diabetic model rats were randomly divided into control group(0.9% NaCl), low dose group(300 mg SCC·(kg body weight)-1) and high dose group(1200 mg SCC(kg body weight)-1). Sodium chloride and SCC were intragastrically administered once a day for 35 d. Changes in fasting serum glucose and serum insulin content, oral glucose tolerance and liver and muscle glycogen content were routinely evaluated. Pancreas tissue and β-cells of islets were observed under both optical and transmission electronic microscope, respectively. The abundance of glucose metabolism-relating genes in gastrocnemius and epididymal adipose tissue was determined with either reverse transcription PCR(RT-PCR) or western blotting. Results showed that SCC significantly decreased fasting serum glucose content, improved glucose tolerance and increased serum insulin and glycogen content; repaired STZ-injured β-cells of diabetic rat, and increased the expression of phosphatidylinositol 3 kinase(PI(3)K), protein kinase B(PKB) and glucose transporter 4(Glut4) encoding protein in both gastrocnemius and adipose tissue, and Glut4 encoding gene in peripheral tissue. 12Study on the effects of the different polar group of EPA-enriched phospholipids on the proliferation and apoptosis in 95D cells显示文摘EPA-enriched phosphatidylcholine(EPA-PC)and EPA-enriched phosphatidylethanolamine(EPA-PE)are newly identified marine phospholipids.The polar group of phospholipids is known to influence EPA-phospholipid activity.However,the differences in anti-tumor effects between EPA-PC and EPA-PE have not been reported.In this study,we evaluated the effects of two forms of EPA on the proliferation and apoptosis in the lung-cancer cell line 95D as well as possible molecular mechanisms.Our results showed that EPA-PC effectively inhibited proliferative activity and promoted apoptosis of 95D cells in a dose-dependent manner,while EPA-PE had no effect on cell proliferation,although it slightly promoted apoptosis.Western blot results showed that EPA-PC and EPA-PE upregulated the expression of PPARy,RXRoc,and PTEN,and downregulated the PI3K/AKT signaling pathway.Furthermore,EPA-PC and EPA-PE induced the expression of the pro-apoptotic gene,Bax,and reduced the expression of the anti-apoptotic gene,Bcl-xl.Additionally,EPA-PC and EPA-PE promoted the release of cytochrome c and activated the apoptotic enzyme-cleaved caspase-3.These data suggest that the anti-tumor effect of EPA-phospholipids may be exerted via a PPARy-related mechanism.EPA-PC was more efficacious as compared to EPA-PE,which might be due to the different polar groups of phospholipids.Yao Guo Qin Zhao Yingying Tian Yuanyuan Liu Ziyi Yan Changhu Xue Jingfeng Wang 2021Marine Life Science & Technology2021,3,4:1

17Altered acetylcholinesterase levels in the spinal cord anterior horn and dorsal root ganglion following sciatic nerve ischemia显示文摘BACKGROUND: Peripheral nerve ischemia has been shown to result in ischemic fiber degenera-tion and axoplasmic transport disturbance. However, the effect on acetylcholinesterase (AChE) ex- pression in relevant cells following sciatic nerve ischemia remains unclear. OBJECTIVE: To observe AChE concentration changes following peripheral nerve ischemia. DESIGN, TIME AND SETTING: The present comparative observation, neuroanatomical experiment was performed at the Central Laboratory Animal of Chengde Medical College between 2006 and 2007. MATERIALS: A total of 20 healthy, adult, Wistar rats were randomized into two groups (n = 10): 8-day ischemia and 14-day ischemia. METHODS: Ischemia injury was induced in the unilateral sciatic nerve (experimental side) through ligation of the common iliac artery. The contralateral side received no intervention, and served as the control side. Rats in the 8-day ischemia and 14-day ischemia groups were allowed to survive for 8 and 14 days, respectively. MAIN OUTCOME MEASURES: The L5 lumbar spinal cord and the L5 dorsal root ganglion were removed from both sides and sectioned utilizing a Leica vibrating slicer. AChE cellular expression was detected using Karnovsky-Root, and the number of AChE-positive cells and average gray value were analyzed using a MiVnt image analysis system. RESULTS: In the 8-day ischemia group, AChE-positive cell numbers were significantly less in the dorsal root ganglion and spinal cord anterior horn of the experimental side, but the average gray value was significantly greater, compared with the control side (P < 0.05). These changes were more significant in the 14-day ischemia group than in the 8-day ischemia group (P < 0.01).
