1A CRISPR/Cas12a-empowered surface plasmon resonance platform for rapid and specific diagnosis of the Omicron variant of SARS-CoV-2显示文摘The outbreak of the COVID-19 pandemic was partially due to the challenge of identifying asymptomatic and presymptomatic carriers of the virus,and thus highlights a strong motivation for diagnostics with high sensitivity that can be rapidly deployed.On the other hand,several concerning SARS-CoV-2 variants,including Omicron,are required to be identified as soon as the samples are identified as‘positive’.Unfortunately,a traditional PCR test does not allow their specific identification.Herein,for the first time,we have developed MOPCS(Methodologies of Photonic CRISPR Sensing),which combines an optical sensing technology-surface plasmon resonance(SPR)with the‘gene scissors’clustered regularly interspaced short palindromic repeat(CRISPR)technique to achieve both high sensitivity and specificity when it comes to measurement of viral variants.MOPCS is a low-cost,CRISPR/Cas12a-systemempowered SPR gene-detecting platform that can analyze viral RNA,without the need for amplification,within 38 min from sample input to results output,and achieve a limit of detection of 15 fM.MOPCS achieves a highly sensitive analysis of SARS-CoV-2,and mutations appear in variants B.1.617.2(Delta),B.1.1.529(Omicron)and BA.1(a subtype of Omicron).This platform was also used to analyze some recently collected patient samples from a local outbreak in China,identified by the Centers for Disease Control and Prevention.This innovative CRISPR-empowered SPR platform will further contribute to the fast,sensitive and accurate detection of target nucleic acid sequences with single-base mutations.Zhi Chen Jingfeng Li Tianzhong Li Taojian Fan Changle Meng Chaozhou Li Jianlong Kang Luxiao Chai Yabin Hao Yuxuan Tang Omar AAl-Hartomy Swelm Wageh Abdullah GAl-Sehemi Zhiguang Luo Jiangtian Yu Yonghong Shao Defa Li Shuai Feng William J.Liu Yaqing He Xiaopeng Ma Zhongjian Xie Han Zhang 2022National Science Review2022,9,8:3
2CRISPR-Cas12a-Empowered Electrochemical Biosensor for Rapid and Ultrasensitive Detection of SARS-CoV-2 Delta Variant显示文摘Coronavirus disease 2019(COVID-19)is a highly contagious disease caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).The gold standard method for the diagnosis of SARS-CoV-2 depends on quantitative reverse transcription-polymerase chain reaction till now,which is time-consuming and requires expensive instrumentation,and the confirmation of variants relies on further sequencing techniques.Herein,we first proposed a robust technique-methodology of electrochemical CRISPR sensing with the advantages of rapid,highly sensitivity and specificity for the detection of SARS-CoV-2 variant.To enhance the sensing capability,gold electrodes are uniformly decorated with electro-deposited gold nanoparticles.Using DNA template identical to SARS-CoV-2 Delta spike gene sequence as model,our biosensor exhibits excellent analytical detection limit(50 fM)and high linearity(R2=0.987)over six orders of magnitude dynamic range from 100 fM to 10 nM without any nucleic-acid-amplification assays.The detection can be completed within 1 h with high stability and specificity which benefits from the CRISPR-Cas system.Furthermore,based on the wireless micro-electrochemical platform,the proposed biosensor reveals promising application ability in point-of-care testing.Chenshuo Wu Zhi Chen Chaozhou Li Yabin Hao Yuxuan Tang Yuxuan Yuan Luxiao Chai Taojian Fan Jiangtian Yu Xiaopeng Ma Omar AAl-Hartomy SWageh Abdullah GAl-Sehemi Zhiguang Luo Yaqing He Jingfeng Li Zhongjian Xie Han Zhang 2022Nano-Micro Letters2022,14,10:2
3EB病毒膜潜伏蛋白LMP-1筛选鼻咽癌的初步结果(英文)显示文摘Objective: Early diagnosis of nasopharyngeal carcinoma (NPC) is an important method to improve the survival rate.However,the sensitivity and specificity of the screening protocols which was widely used in clinic now are considered to be unsatisfactory.Epstein-Barr virus (EBV)-encoded latent membrane protein-1 (LMP-1) is one of the proteins that have been suggested to be a classic oncogene with transformation properties.The current study set out to discuss the clinical significance of LMP-1 on the screening of NPC.Methods: Three hundred patients who visited our institution (Department of Radiation Oncology,Fujian Provincial Cancer Hospital,Fuzhou,China) with ENT symptoms between 2007 and 2008 were involved in this study,and all of them were agreed to be involved in this investigation.Not only did they undergo nasopharyngeal swab to obtain cells for the LMP-1 polymerase chain reaction (PCR) analysis,but also nasopharyngeal biopsy were taken to identify the diagnosis.Results: An amount of DNA that was sufficient for PCR was extracted from 243 (81%) swab samples,the positive rate of LMP-1 of those with non-nasopharyngeal carcinoma was 3.85% (4/108),which was much lower than those with nasopharyngeal carcinoma (P < 0.05).By detecting LMP-1 in nasopharyngeal swabs,NPC was diagnosed with a sensitivity of 88.15% (119 of 135 patients),specificity of 96.30% (104 of 108 patients),a positive predictive value of 95.2% (119 of 123 patients),a negative predictive value of 86.67% (104 of 120 patients),accuracy of 91.77%,and Youden index of 84.45%.Conclusion: The nasopharyngeal swab coupled with PCR-based EBV LMP-1 detection have high sensitivity and specificity,and also good repeatability,it could serve as part of the screening program for high-risk populations.Shaojun Lin Qiaojuan Guo Jin Lin Jingfeng Zong Lu Han Jianji Pan 2011The Chinese-German Journal of Clinical Oncology2011,10,1:2
4Improved strength and ductility of AZ31B Mg alloy sheets processed by accumulated extrusion bonding with artificial cooling显示文摘In this study,accumulated extrusion bonding(AEB)process with application of artificial water cooling was successfully performed to fabricate fined-grains AZ31B Mg alloy sheets at 150℃,200℃ and 250℃.The resultant microstructure and mechanical properties are systematically investigated.It reveals that the processing temperature has an important effect on the microstructural evolution during extrusion.During AEB process at 150℃ and 200℃,{10-12}tensile twinning was activated at early stage of extrusion,and subsequently continuous dynamic recrystallization(CDRX)occurred and dominated the further deformation.However,for the sample extruded at 250℃,hardly any twins can be observed,and new fined dynamic recrystallized grains were found along grain boundaries.Artificial cooling was utilized to reduce the rate of grain growth out of the extrusion die,resulting the grains significantly refined from 11μm to 2.5μm.Local high dislocation density region was also observed in the microstructure of sample processed at 150℃ in artificial cooling condition,and the degree decreased with the processing temperature increase.The results summarized from tensile tests indicated that due to the grain refinement the strength and ductility was significantly enhanced(YS of 186 MPa vs.145 MPa,UTS of 391 MPa vs.336 MPa and FE of 31.5%vs.24.5%compared with the as-received sample).Subsequently,annealing treatment at different temperatures was applied to eliminate the high dislocation density.The sample annealed at 200℃ exhibited the best comprehensive mechanical property with YS of 179 MPa,UTS of 390 MPa and FE of 33.0%.As the annealing temperature increasing,the dislocation density was reduced by static recrystallization(SRX)and grain growth,leading to a decreased strength and ductility.Tingzhuang Han Jingfeng Zou Guangsheng Huang Lifeng Ma Chaojie Che Weitao Jia Lifei Wang Fusheng Pan 2021Journal of Magnesium and Alloys2021,9,5:2
5The template-assisted zinc ion incorporation in SAPO-34 and the enhanced ethylene selectivity in MTO reaction显示文摘The SAPO-34 catalyst was fine-tuned with zinc cations through a straightforward template-assisted ion incorporation (TH) process, without the necessary template pre-removal and the preparation of NH4- SAPO-34 intermediate, which is more facile, efficient and cost-effective than the conventional ion exchange process. The template-assisted zinc cations incorporated SAPO-34 catalysts were characterized by XRD, XRF, N2 adsorption-desorption, XPS, SEM, EDX,NMR, respectively. 14CRISPR-powered optothermal nanotweezers:Diverse bio-nanoparticle manipulation and single nucleotide identification显示文摘Optothermal nanotweezers have emerged as an innovative optical manipulation technique in the past decade,which revolutionized classical optical manipulation by efficiently capturing a broader range of nanoparticles.However,the optothermal temperature field was merely employed for in-situ manipulation of nanoparticles,its potential for identifying bio-nanoparticles remains largely untapped.Hence,based on the synergistic effect of optothermal manipulation and CRIPSR-based bio-detection,we developed CRISPR-powered optothermal nanotweezers(CRONT).Specifically,by harnessing diffusiophoresis and thermo-osmotic flows near the substrate upon optothermal excitation,we successfully trapped and enriched DNA functionalized gold nanoparticles,CRISPR-associated proteins,as well as DNA strands.Remarkably,we built an optothermal scheme for enhancing CRISPR-based single-nucleotide polymorphism(SNP)detection at single molecule level,while also introducing a novel CRISPR methodology for observing nucleotide cleavage.Therefore,this innovative approach has endowed optical tweezers with DNA identification ability in aqueous solution which was unattainable before.With its high specificity and feasibility for in-situ bio-nanoparticle manipulation and identification,CRONT will become a universal tool in point-of-care diagnosis,biophotonics,and bio-nanotechnology.Jiajie Chen Zhi Chen Changle Meng Jianxing Zhou Yuhang Peng Xiaoqi Dai Jingfeng Li Yili Zhong Xiaolin Chen Wu Yuan Ho-Pui Ho Bruce Zhi Gao Junle Qu Xueji Zhang Han Zhang Yonghong Shao 2023Light(Science & Applications)2023,12,12:0
15Enhancing the aromatic selectivity of cyclohexane aromatization by CO_(2)coupling显示文摘Improving the aromatic selectivity in the alkane aromatization process is of great importance for its practical utilization but challenge to make because the high H/C ratio of alkanes would lead to a serious hydrogen transfer process and a large amount of light alkanes.Herein,CO_(2)is introduced into the cyclohexane conversion process on the HZSM-5 zeolite,which can improve the aromatic selectivity.By optimizing the reaction conditions,an improved aromatic(benzene,toluene,xylene,and C9+)selectivity of 48.2%can be obtained at the conditions of 2.7 MPa(CO_(2)),450℃,and 1.7 h^(−1),which is better than that without CO_(2)(aromatic selectivity=43.2%).In situ transmission Fourier transform infrared spectroscopy spectra illustrate that many oxygenated chemical intermediates(e.g.,carboxylic acid,anhydride,unsaturated aldehydes/ketones or ketene)would be formed during the cyclohexane conversion process in the presence of CO_(2).13C isotope labeling experimental results demonstrate that CO_(2)can enter into the aromatics through the formation of oxygenated chemical intermediates and thereby improve the aromatic selectivity.This study may open a green,economic,and promising way to improve the aromatic selectivity for alkane aromatization process.Xiangxiang Ren Zhong-Pan Hu Jingfeng Han Yingxu Wei Zhongmin Liu 2023Frontiers of Chemical Science and Engineering2023,17,11:0
16Coffee Flower Identification Using Binarization Algorithm Based on Convolutional Neural Network for Digital Images显示文摘Crop-type identification is one of the most significant applications of agricultural remote sensing,and it is important for yield estimation prediction and field management.At present,crop identification using datasets from unmanned aerial vehicle(UAV)and satellite platforms have achieved state-of-the-art performances.However,accurate monitoring of small plants,such as the coffee flower,cannot be achieved using datasets from these platforms.With the development of time-lapse image acquisition technology based on ground-based remote sensing,a large number of small-scale plantation datasets with high spatial-temporal resolution are being generated,which can provide great opportunities for small target monitoring of a specific region.The main contribution of this paper is to combine the binarization algorithm based on OTSU and the convolutional neural network(CNN)model to improve coffee flower identification accuracy using the time-lapse images(i.e.,digital images).A certain number of positive and negative samples are selected from the original digital images for the network model training.Then,the pretrained network model is initialized using the VGGNet and trained using the constructed training datasets.Based on the well-trained CNN model,the coffee flower is initially extracted,and its boundary information can be further optimized by using the extracted coffee flower result of the binarization algorithm.Based on the digital images with different depression angles and illumination conditions,the performance of the proposed method is investigated by comparison of the performances of support vector machine(SVM)and CNN model.Hence,the experimental results show that the proposed method has the ability to improve coffee flower classification accuracy.The results of the image with a 52.5°angle of depression under soft lighting conditions are the highest,and the corresponding Dice(F1)and intersection over union(IoU)have reached 0.80 and 0.67,respectively.Pengliang Wei Ting Jiang Huaiyue Peng Hongwei Jin Han Sun Dengfeng Chai Jingfeng Huang 2020Plant Phenomics2020,2,1:0
17MalFSM:Feature Subset Selection Method for Malware Family Classification显示文摘Malware detection has been a hot spot in cyberspace security and academic research.We investigate the correlation between the opcode features of malicious samples and perform feature extraction,selection and fusion by filtering redundant features,thus alleviating the dimensional disaster problem and achieving efficient identification of malware families for proper classification.Malware authors use obfuscation technology to generate a large number of malware variants,which imposes a heavy analysis burden on security researchers and consumes a lot of resources in both time and space.To this end,we propose the MalFSM framework.Through the feature selection method,we reduce the 735 opcode features contained in the Kaggle dataset to 16,and then fuse on metadata features(count of file lines and file size)for a total of 18 features,and find that the machine learning classification is efficient and high accuracy.We analyzed the correlation between the opcode features of malicious samples and interpreted the selected features.Our comprehensive experiments show that the highest classification accuracy of MalFSM can reach up to 98.6% and the classification time is only 7.76 s on the Kaggle malware dataset of Microsoft.KONG Zixiao XUE Jingfeng WANG Yong ZHANG Qian HAN Weijie ZHU Yufen 2023Chinese Journal of Electronics2023,32,1:0
18Mechanism of Lingbao Huxin Dan(灵宝护心丹)in the treatment of bradyarrhythmia complicated with coronary heart disease:a network pharmacology analysis显示文摘OBJECTIVE:To investigate the mechanism of action of the Lingbao Huxin Dan(灵宝护心丹)in treating bradycardia arrhythmia with coronary heart disease(BACHD)by network pharmacology.METHODS:The active ingredients of the Lingbao Huxin Dan were screened on the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform and Bioinformatics tools designed for the analysis of molecular mechanisms of Chinese medicine platform;target prediction was conducted with the SwissTargetPrediction database,and Cytoscape 3.8 was used to construct a drug ingredient-target network.The Genecards,Online Mendelian Inheritance in Man,and DrugB ank databases were searched for disease targets.Venn plots were used to display the common targets of BA-CHD and active ingredients.The STRING platform was used to construct a protein-protein interaction network.The Metascape data platform was used for Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis to construct a signaling pathway network of the active ingredients of the Lingbao Huxin Dan.RESULTS:There were 121 active ingredients,899 related targets,39 targets important in BA-CHD and 14 targets which intersected between the active ingredients and BA-CHD.There were 27 core therapeutic ingredients,153 biological processes,18 cell ingredients and 20 molecular functions obtained by GO enrichment analysis.The KEGG pathway analysis yielded 19 signaling pathways.CONCLUSION:RBA-CHD may treat BA-CHD by regulating adrenergic receptor beta-1,alpha 1-αadrenergic receptor,calcium voltage-gated channel subunit alpha1 C,alpha-1β-adrenergic receptor,nitric oxide synthase 2,beta-2 adrenergic receptor,voltagedependent calcium channel subunit alpha-2/delta-1,angiotensin-converting enzyme,Raf-1 proto-oncogene serine/threonine-protein kinase,and other targets,potentially by affecting adrenergic receptor binding and calcium channel opening,to regulate the activity of cardiomyocytes.HAN Huagang LI Ziqiang OUYANG Jingfeng WANG Tianquan DONG Lingyan CAO Junling 2023Journal of Traditional Chinese Medicine2023,43,5:0
19Epidemiological characteristics and clinical manifestations of pediatric patients with COVID-19 in China: A multicenter retrospective study显示文摘Importance:The Coronavirus disease 2019(COVID-19)global pandemic poses a considerable challenge for pediatricians.Objective:This study aimed to identify the epidemiological characteristics and clinical features of pediatric patients with COVID-19 in China.Methods:This multicenter retrospective study included pediatric patients from 46 hospitals in China,covering 12 provinces and two municipalities.Epidemiological,demographic,clinical,laboratory,treatment,and outcome data were analyzed.Results:In total,211 pediatric patients with COVID-19 were included in this study.The median age was 7.0 years(range:22 days to 18 years).Approximately 16.3%of the patients exhibited asymptomatic infections,23.0%had upper respiratory tract infections,and 60.7%had pneumonia,including two with severe pneumonia and one with critical illness.Approximately 78.7%of the pediatric patients occurred in familial clusters.The most three common symptoms or signs at onset in children with COVID-19 were fever(54.5%),cough(49.3%),and pharyngeal congestion(20.8%).Only 17.6%of the patients presented with decreased lymphocyte count,whereas 13.6%had increased lymphocyte count.Among the patients with pneumonia who exhibited abnormal chest computed tomography findings,18.2%(23/127)of the patients had no other symptoms.Generally,the chest radiographs showed abnormalities that affected both lungs(49.6%);ground-glass opacity(47.2%)was the most common manifestation.The cure and improvement rates were 86.7%(183/211)and 13.3%(28/211),respectively.Only one patient with an underlying condition received invasive mechanical ventilation;none of the patients died.Interpretation:Similar to adults,children of all age groups are susceptible to COVID-19.Fortunately,most pediatric patients have mild symptoms or remain asymptomatic,despite the high incidence of pneumonia.Decreased proportions of white blood cells and lymphocytes are less frequent in children than in adults.Yan Bai Liwei Gao Xianfeng Wang Lili Zhong Jingfeng Li Shenggang Ding Yuejie Zheng Jun Liu Yuxia Cui Lina Wang Mingfeng Han Jun Liu Xinping Jiang Min Jiang Junhua Li Ning Chen Yunxiao Shang Hourong Zhou Yi Xu Gen Lu Xing Chen Jizhi Xu Qihong Fan Yu Tang Jiang Wu Cen Li Xiaoxiang Yang Chunxi Chen Yonghong Yang Gary Wing-Kin Wong Adong Shen Tianyou Wang Runming Jin Baoping Xu Kunling Shen 2021Pediatric Investigation2021,5,3:0
20Direction Finding with Cyclostationarity Analysis Against Frequency Interference显示文摘In this paper, a novel anti-interference direction finding(DF)method for amplitude comparison method based on cyclostationarity is proposed. With the periodic properties of the communication signals, the desired signal’s amplitude value can be effectively obtained even though there is an interference signal whose frequency spectrum overlaps with the desired signal in the environment,and the corresponding incident angle can be estimated accurately with the amplitude comparison method. The influence of interference signal on the amplitude comparison method is discussed and the proposed method’s theoretical feasibility is also analyzed. Compared with the conventional method,simulations are provided to demonstrate the anti-interference capability of the proposed method.
