1Friction characteristics of confined inflatable structures显示文摘The availability of high‐strength fabrics and progress in the development of large‐scale inflatable technology made possible the creation of temporary and quickly deployable structures for protection of underground infrastructure.Inflatable structures are relatively lightweight and portable,and can maintain the required rigidity while in operation.These benefits have prompted the development of inflatable structures for use in confined spaces,such as tunnels and large‐diameter pipes to act as barriers for containing flooding with minimal infrastructure modification.This work presents experimental results obtained from the evaluation of frictional characteristics of the fabric material that constitute the structural membrane of confined inflatable structures developed for protection of underground transportation tunnels and other large conduits.Friction tests at coupon level and slippage tests in a reduced‐scale inflatable structure were performed in order to evaluate the frictional characteristics of Vectran webbings.Tests at coupon level were performed to determine the friction coefficient for different surface types and conditions.Tests with the reduced‐scale inflatable structure contributed to the understanding of the friction characteristics at system level when subjected to different pressurization or depressurization sequences designed to induce slippage.Test results indicate that friction coefficient values at coupon level are about 29 percent higher than values derived from reduced‐scale tests.Eduardo M.SOSA Gregory J.THOMPSON Ever J.BARBERO Subhadeep GHOSH Kevin L.PEIL 2014Friction2014,2,4:5
2Mild closed head traumatic brain injury-induced changes in monoamine neurotransmitters in the trigeminal subnuclei of a rat model:mechanisms underlying orofacial allodynias and headache显示文摘Our recent findings have demonstrated that rodent models of closed head traumatic brain injury exhibit comprehensive evidence of progressive and enduring orofacial allodynias, a hypersensitive pain response induced by non-painful stimulation. These allodynias, tested using thermal hyperalgesia, correlated with changes in several known pain signaling receptors and molecules along the trigeminal pain pathway, especially in the trigeminal nucleus caudalis. This study focused to extend our previous work to investigate the changes in monoamine neurotransmitter immunoreactivity changes in spinal trigeminal nucleus oralis, pars interpolaris and nucleus tractus solitaries following mild to moderate closed head traumatic brain injury, which are related to tactile allodynia, touch-pressure sensitivity, and visceral pain. Our results exhibited significant alterations in the excitatory monoamine, serotonin, in spinal trigeminal nucleus oralis and pars interpolaris which usually modulate tactile and mechanical sensitivity in addition to the thermal sensitivity. Moreover, we also detected a robust alteration in the expression of serotonin, and inhibitory molecule norepinephrine in the nucleus tractus solitaries, which might indicate the possibility of an alteration in visceral pain, and existence of other morbidities related to solitary nucleus dysfunction in this rodent model of mild to moderate closed head traumatic brain injury. 17Reduction of epinephrine in the lumbar spinal cord following repetitive blast-induced traumatic brain injury in rats显示文摘Traumatic brain inju ry-induced unfavorable outcomes in human patients have independently been associated with dysregulated levels of monoamines,especially epinephrine,although few preclinical studies have examined the epinephrine level in the central nervous system after traumatic brain injury.Epinephrine has been shown to regulate the activities of spinal motoneurons as well as increase the heart rate,blood pressure,and blood flow to the hindlimb muscles.Therefore,the purpose of the present study was to determine the impact of repeated blast-induced traumatic brain injury on the epinephrine levels in seve ral function-s pecific central nervous system regions in rats.Following three repeated blast injuries at 3-day intervals,the hippocampus,motor cortex,locus coeruleus,vestibular nuclei,and lumbar spinal cord were harvested at post-injury day eight and processed for epinephrine assays using a high-sensitive electrochemical detector cou pled with high-performance liquid chromatography.Our results showed that the epinephrine levels were significantly decreased in the lumbar spinal cord tissues of blast-induced traumatic brain injury animals compared to the levels detected in age-and sex-matched sham controls.In other function-specific central nervous system regions,although the epinephrine levels were slightly altered following blast-induced tra u matic brain injury,they were not statistically significant.These results suggest that blast injury-induced significant downregulation of epinephrine in the lumbar spinal cord could negatively impact the motor and cardiovascular function.This is the first repo rt to show altered epinephrine levels in the spinal cord following repetitive mild blast-induced traumatic brain injury.Shigeharu Tsuda Mustafa Golam Jiamei Hou Kevin K.W.Wang Floyd J.Thompson Prodip Bose 2024Neural Regeneration Research2024,19,7:0
18A proton birdcage coil integrated with interchangeable single loops for multi-nuclear MRI/MRS显示文摘Energy metabolism is fundamental for life.It encompasses the utilization of carbohydrates,lipids,and proteins for internal processes,while aberrant energy metabolism is implicated in many diseases.In the present study,using three-dimensional(3D)printing from polycarbonate via fused deposition modeling,we propose a multi-nuclear radiofrequency(RF)coil design with integrated 1H birdcage and interchangeable X-nuclei(^(2)H,^(13)C,^(23)Na,and^(31)P)single-loop coils for magnetic resonance imaging(MRI)/magnetic resonance spectroscopy(MRS).The single-loop coil for each nucleus attaches to an arc bracket that slides unrestrictedly along the birdcage coil inner surface,enabling convenient switching among various nuclei and animal handling.Compared to a commercial 1H birdcage coil,the proposed 1H birdcage coil exhibited superior signal-excitation homogeneity and imaging signal-to-noise ratio(SNR).For X-nuclei study,prominent peaks in spectroscopy for phantom solutions showed excellent SNR,and the static and dynamic peaks of in vivo spectroscopy validated the efficacy of the coil design in structural imaging and energy metabolism detection simultaneously.Yi ZHANG Zhiyan QUAN Feiyang LOU Yujiao FANG Garth J.THOMPSON Gao CHEN Xiaotong ZHANG 2024Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2024,25,2:0
19The measurement and modification of hypoxia in colorectal cancer:overlooked but not forgotten显示文摘Tumour hypoxia is the inevitable consequence of a tumour’s rapid growth and disorganized,inefficient vasculature.The compensatorymechanisms employed by tumours,and indeed the absence of oxygen itself,hinder the ability of all treatmentmodalities.The clinical consequence is poorer overall survival,disease-free survival,and locoregional control.Recognizing this,clinicians have been attenuating the effect of hypoxia,primarily with hypoxicmodification or with hypoxia-activated prodrugs,and notable success has been demonstrated.However,in the case of colorectal cancer(CRC),there is a general paucity of knowledge and evidence surrounding themeasurement andmodification of hypoxia,and this is possibly due to the comparative inaccessibility of such tumours.We specifically review the role of hypoxia in CRC and focus on the current evidence for the existence of hypoxia in CRC,themajority of which originates from indirect positron emission topography imaging with hypoxia selective radiotracers;the evidence correlating CRC hypoxia with poorer oncological outcome,which is largely based on themeasurement of hypoxia inducible factor in correlation with clinical outcome;the evidence of hypoxicmodification in CRC,of which no direct evidence exists,but is reflected in a number of indirectmarkers;the prognostic andmonitoring implications of accurate CRC hypoxia quantification and its potential in the field of precision oncology;and the present and future imaging tools and technologies being developed for themeasurement of CRC hypoxia,including the use of blood-oxygenlevel-dependentmagnetic resonance imaging and diffuse reflectance spectroscopy.Teddy Fletcher Alex J.Thompson Hutan Ashrafian Ara Darzi 2022Gastroenterology Report2022,10,1:0
20Using Pharmacokinetic Modeling and Electronic Health Record Data to Predict Clinical and Safety Outcomes after Methylprednisolone Exposure during Cardiopulmonary Bypass in Neonates显示文摘Background:Infants undergoing cardiac surgery with cardiopulmonary bypass(CPB)frequently receive intraoperative methylprednisolone(MP)to suppress CPB-related inflammation;however,the optimal dosing strategy and efficacy of MP remain unclear.Methods:We retrospectively analyzed all infants under 90 days-old who received intra-operative MP for cardiac surgery with CPB from 2014–2017 at our institution.We combined real-world dosing data from the electronic health record(EHR)and two previously developed population pharmacokinetic/pharmacodynamic models to simulate peak concentration(Cmax)and area under the concentration-time curve for 24 h(AUC24)for MP and the inflammatory cytokines interleukin-6(IL-6)and interleukin-10(IL-10).We evaluated the relationships between post-operative,safety,and other clinical outcomes obtained from the EHR with each predicted exposure using non-parametric tests.Results:A total of 142 infants with median post-natal age 8(interquartile range[IQR]:5,37)days received a total dose of 30(19,49)mg/kg of MP.Twelve(8%)died,37(26%)met the composite post-operative outcome,114(80%)met the composite safety outcome,and 23(16%)had a major complication.Predicted median Cmax and AUC24 IL-6 exposure was significantly higher for infants meeting the composite post-operative outcome and those with major complications.Predicted median Cmax and AUC24 MP exposure was significantly higher for infants requiring insulin.No exposure was associated with death or other safety outcomes.Conclusions:Pro-inflammatory IL-6,but not MP exposure,was associated with post-operative organ dysfunction,suggesting current MP dosing may not adequately suppress IL-6 or increase IL-10 to impact clinical outcomes.Prospective study will be required to define the optimal exposure-efficacy and exposure-safety profiles in these infants.Henry P.Foote Huali Wu Stephen J.Balevic Elizabeth J.Thompson Kevin D.Hill Eric M.Graham Christoph P.Hornik Karan R.Kumar 2023Congenital Heart Disease2023,18,3:0
