| 1 | Spatiotemporal microRNA profile in peripheral nerve regeneration:miR-138 targets vimentin and inhibits Schwann cell migration and proliferation显示文摘While the peripheral nervous system has regenerative ability,restoration of sufficient function remains a challenge.Vimentin has been shown to be localized in axonal growth fronts and associated with nerve regeneration,including myelination,neuroplasticity,kinase signaling in nerve axoplasm,and cell migration;however,the mechanisms regulating its expression within Schwann cell(SC) remain unexplored.The aim of this study was to profile the spatial and temporal expression profile of micro RNA(mi RNA) in a regenerating rat sciatic nerve after transection,and explore the potential role of mi R-138-5 p targeting vimentin in SC proliferation and migration.A rat sciatic nerve transection model,utilizing a polyethylene nerve guide,was used to investigate mi RNA expression at 7,14,30,60,and 90 days during nerve regeneration.Relative levels of mi RNA expression were determined using microarray analysis and subsequently validated with quantitative real-time polymerase chain reaction.In vitro assays were conducted with cultured Schwann cells transfected with mi RNA mimics and assessed for migratory and proliferative potential.The top seven dysregulated mi RNAs reported in this study have been implicated in cell migration elsewhere,and GO and KEGG analyses predicted activities essential to wound healing.Transfection of one of these,mi RNA-138-5 p,into SCs reduced cell migration and proliferation.mi R-138-5 p has been shown to directly target vimentin in cancer cells,and the luciferase assay performed here in rat Schwann cells confirmed it.These results detail a role of mi R-138-5 p in rat peripheral nerve regeneration and expand on reports of it as an important regulator in the peripheral nervous system. | Travis B.Sullivan Litchfield C.Robert Patrick A.Teebagy Shannon E.Morgan Evan W.Beatty Bryan J.Cicuto Peter K.Nowd Kimberly M.Rieger-Christ David J.Bryan | 2018 | Neural Regeneration Research2018,13,7: | 6 |

| 4 | Seashore paspalum (Paspalum vaginatum Sw.) performance under shade in multi-environment trials显示文摘Shade caused by trees or buildings can affect the quality and growth of turfgrasses.Thus,breeding for shade tolerance is an essential component of most turfgrass improvement programs.The objective of this study was to evaluate the performance of seashore paspalum(Paspalum vaginatum Sw.)breeding lines under shade in multi-environment trials.Germplasm sources were 20 lines from the University of Georgia seashore paspalum breeding program and two checks,the cultivars'SeaDwarf'and'SeaStar'.Field trials were conducted from 2016 to 2019 under shade structures designed to reduce ambient sunlight at three locations:Citra(FL),Tifton(GA),and Raleigh(NC).The response variables evaluated were percent living ground cover(%GC),dark green color index(DGCI),canopy height(CH)and turfgrass quality(TQ).Data were analyzed by mixed model approaches using ASReml-R,and the t-statistics were used to group the entries.Significant genetic variances were observed in the single-location-repeated-measures analysis for CH and DGCI at Citra and Tifton,%GC at Citra,and TQ at Raleigh.Spearman correlations of the predicted values for entries between locations ranged from low to moderate,with the exception of between Citra and Tifton(0.76)for CH.There were a few breeding lines with superior performance to the entry average for all traits for which the entry variance was significant.In conclusion,genetic variability was observed for all traits and superior seashore paspalum breeding lines for multiple traits were identified under shade. | Beatriz Tome Gouveia Paul Raymer Brian M.Schwartz Esteban F.Rios Kevin E.Kenworthy J.Bryan Unruh Charles Fontanier Esdras M.Carbajal Jason Kruse Natasha K.Restuccia Susana R.Milla-Lewis | 2021 | Grass Research2021,1,1: | 0 |
