| 1 | Evaluation of biodegradable electric conductive tube-guides and mesenchymal stem cells显示文摘AIM: To study the therapeutic effect of three tubeguides with electrical conductivity associated to mesenchymal stem cells(MSCs) on neuro-muscular regeneration after neurotmesis.METHODS: Rats with 10-mm gap nerve injury were tested using polyvinyl alcohol(PVA), PVA-carbon nanotubes(CNTs) and MSCs, and PVA-polypyrrole(PPy). The regenerated nerves and tibialis anterior muscles were processed for stereological studies after 20 wk. The functional recovery was assessed serially for gait biomechanical analysis, by extensor postural thrust, sciatic functional index and static sciatic functionalindex(SSI), and by withdrawal reflex latency(WRL). In vitro studies included cytocompatibility, flow cytometry, reverse transcriptase polymerase chain reaction and karyotype analysis of the MSCs. Histopathology of lung, liver, kidneys, and regional lymph nodes ensured the biomaterials biocompatibility. RESULTS: SSI remained negative throughout and independently from treatment. Differences between treted groups in the severity of changes in WRL existed, showing a faster regeneration for PVA-CNTs-MSCs(P < 0.05). At toe-off, less acute ankle joint angles were seen for PVA-CNTs-MSCs group(P = 0.051) suggesting improved ankle muscles function during the push off phase of the gait cycle. In PVA-PPy and PVA-CNTs groups, there was a 25% and 42% increase of average fiber area and a 13% and 21% increase of the 'minimal Feret's diameter' respectively. Stereological analysis disclosed a significantly(P < 0.05) increased myelin thickness(M), ratio myelin thickness/axon diameter(M/d) and ratio axon diameter/fiber diameter(d/D; g-ratio) in PVA-CNT-MSCs group(P < 0.05). CONCLUSION: Results revealed that treatment with MSCs and PVA-CNTs tube-guides induced better nerve fiber regeneration. Functional and kinematics analysis revealed positive synergistic effects brought by MSCs and PVA-CNTs. | 8 | Genoprotection and metabolic benefits of marine macroalgae-Insights into the concept of functional foods through direct and indirect consumption显示文摘The consumption of marine macroalgae has been linked with health benefits,albeit some actions,including genoprotection,remain underexplored.Hence,we evaluated the genoprotective potential of a mixture of Ulva rigida,Fucus vesiculosus and Gracilaria gracilis,through direct consumption or via indirect intake using fish(Sparus aurata)as a vector of algal phytocompounds.Mice(Mus musculus)were subjected to a 1-month dietary supplementation with 5%of macroalgae mix or 10%of fillet of fish previously fed with an algae-supplemented aquafeed(for comparison purposes,10%supplementation with fillet of fish standardly fed was also considered).Thereafter,mice were challenged by the genotoxicant methyl methanesulfonate(MMS),and the genetic damage was evaluated through the micronuclei test.Complementarily,a wider evaluation was carried out encompassing the assessment of energy metabolism,haematological and histological parameters,as well as growth performance.Macroalgae unequivocally protected against MMS-induced genotoxicity.The activity profile of hepatic lactate and isocitrate dehydrogenases promoted by the supplemented diets reflected an improved energy balance.Genoprotection properties were not transferred via fish fed with the algae-supplemented aquafeed,though the transference of phytocomponents with beneficial properties(viz.energy balance improvement)should not be disregarded.Overall,these findings highlighted the genoprotection afforded by marine macroalgae,likely promoted by desmutagenic factors,reinforcing their definition as functional food.The possibility of using fish as an indirect source of algae-borne phytocomponents proved to be plausible and worthy of further investigations. | Ana Marques João Ferreira Ana Cerqueda-Pacheco Vitória Pereira Helena Abreu Rui Pereira Maria João Pires Fernanda Seixas Paula Oliveira Isabel Gaivão Mário Pacheco | 2022 | Food Bioscience2022,47,3: | 0 |
| 9 | Species richness and density evaluation for plants with aggregated distributions:fixed vs.variable area methods显示文摘Aims Knowledge of species richness is of prime importance to both basic and applied aspects of ecological studies.However,quantifying plant species richness in the tropics is potentially time-consuming because of high species diversity.Plant species richness estimates are also frequently biased,because many rare species are not detected.to address these problems,the use of a variable area method has been proposed as an alternative to fixed area methods,but its appli-cability to plants with aggregated distributions has been questioned based on simulation studies.We use empirical data to compare the efficiency and accuracy of a variable area method and a fixed area method for estimating species richness,density and basal area for plants with aggregated distributions,using palms as a model taxon.Methods Adult palms were sampled in twenty 10×30 m transects in an Atlantic Forest in Rio de Janeiro state,Brazil.All individuals were considered in the fixed area method,while in the variable area method only the first six adults in each transect were sampled;in this case,transect length was defined according to the distance of the sixth adult from the beginning of the transect.When fewer than six individuals were observed in a given transect,transect length was extended up to 50 m to search for additional individuals.the efficiency of both methods was compared based on species rarefaction curves,using the chao 1 statistic(for abundance data).For each species we calculated mean density per transect and basal area,according to each sampling method.Sampling effort in terms of the number of individual plants and the area necessary to characterize maximum species richness in each sampling method,as well as mean time taken to sample a single transect,were compared as measures of efficiency.Important findings An accurate estimate of species richness was achieved using both methods,but in the variable area method,a quarter of the number of individuals and half the area was sufficient to characterize maximum species richness.Density and basal area did not differ between meth-ods for any of the species studied.In the fixed area method sampling effort was 90 min per transect,whereas in the variable area method it was 30 min per transect.the variable area method,with its faster assessment of palm species richness,should facilitate greater spatial representativeness by making it easier to sample a larger number of plots at different spatial scales.We thus find sufficient evidence to recommend the variable area method for rapid and robust evaluations of species richness for palms with aggregate distributions,as well as for other plants with similar spatial patterns,in tropical forests. | Rita de Cássia Quitete Portela Alexandra dos Santos Pires Maria Isabel Guedes Braz Eduardo Arcoverde de Mattos | 2017 | Journal of Plant Ecology2017,10,5: | 0 |
