1大鼠在体肠灌流模型研究淫羊藿不同黄酮苷的吸收代谢显示文摘目的:研究淫羊藿5个黄酮苷淫羊藿苷、朝藿定A、朝藿定B、朝藿定C、宝藿苷Ⅰ在大鼠肠道的在体吸收情况。方法:应用大鼠在体肠灌流模型,用高效液相法测定肠灌流液中淫羊藿各黄酮苷的含量,计算各黄酮苷的肠渗透系数。结果:5个黄酮苷在大鼠各肠段的表观渗透系数按十二指肠、空肠、回肠、结肠顺序依次分别为淫羊藿苷:6.365,5.349,1.004,0.601;朝藿定A分别为4.120,3.397,0.401,0.257;朝藿定B分别为:3.645,3.135,0.990,0.434;朝藿定C分别为:3.114,2.882,0.688,0.470;宝藿苷Ⅰ分别为2.324,2.398,1.529,1.045。淫羊藿苷在大鼠肠道会被水解产生2个代谢产物,朝藿定A,B,C各自会被水解产生1个代谢产物,宝藿苷Ⅰ无代谢产物。结论:5个黄酮成分在大鼠肠段不同部位吸收存在差异,各化合物的回肠和结肠肠段表观渗透系数均显著低于十二指肠和空肠肠段,淫羊藿苷,朝藿定A,B,C的肠道代谢产物与其结构与肠黏膜上的酶相关,并且在十二指肠、空肠代谢较多;在回肠、结肠代谢较少。陈彦 贾晓斌 谭晓斌 范晨怡 HuMing 2009中国中药杂志2009,34,22:19
2Developmental Lead Exposure Alters the Distribution of Protein Kinase C Activity in the Rat Hippocampus显示文摘Chronic low-level lead (Pb) exposure in children is known to cause a deficit in learning and memory. In vitro studies have demonstrated that Pb altered protein kinase C (PKC) activityt Especially, hippocampal PKC has been correlated with performance in several learning tasks. The effects of Pb exposure on hippocampal PKC were investigated during development at various postnatal ages: postnatal day (PN) 7, 14, 28, and 56. Two-tenth % Pb acetate was administered to pregnant and lactating dams and then administered to weanling rats in drinking water. PKC activity was measured in both membrane and cytosolic fractions from the hippocampi of the controls and Pb-exposed animals. Pb-induced increase in PKC activity in the cytosolic fraction was obsereved in the PN56 rats. In contrast, PKC activity was decreased by Pb at PN7 in the membrane fraction. Furthermore, a significant decrease in the ratio of membrane to cytosolic PKC activity which is representative of PKC distribution was observed in the PN28 and PN56 Pb-exposed rats relative to the same-age controls. This study indicates that chronic Pb exposure during development influences hippocampal PKC activity and distribution. 3Experimental Study of Capillary Effect in Porous Silicon Using Micro-Raman Spectroscopy and X-Ray Diffraction显示文摘We investigate the capillary effect and the residual stress evolution in the wetting, drying and rewetting stages of porous silicon using x-ray diffraction and micro-Raman spectroscopy. A reversible capillary effect and an irreversible oxidation effect are the driving forces for the residual stress evolution. The lattice expansion of the porous-silicon layer is observed to decrease slightly by x-ray diffraction and the tensile residual stress increases rapidly by micro-Raman spectroscopy, with the change of about 82 MPa for the oxidation effect and the change of 2.78 GPa (enough for cracking) for the capillary effect. Therefore, the capillary effect plays a major role in the residual stress evolution in the stages. A simple microscopic liquid-bridge model is introduced to explain the capillary effect and its reversibility. 4选择性粒细胞吸附装置治疗重症急性肾损伤的研究显示文摘目的初步观察选择性粒细胞吸附装置(selective cytopheretic device,SCD)联合连续性静静脉血液滤过(continuous veno-venous hemofiltration,CVVH)治疗重症急性肾损伤(acute kidney injury,AKI)患者的疗效,并观察不良事件的发生情况。方法本研究重症AKI的定义为临床诊断为缺血性或肾毒性急性肾小管坏死,同时至少有1个肾外器官衰竭或存在脓毒血症。入选者除给予标准的重症监护治疗外,还同时接受SCD联合CVVH治疗。历史对照组为改善急性肾脏疾病照护计划(the program to improve care in acute renal disease,PICARD)研究中年龄和序贯器官衰竭评分(sequential organ failure assessment,SOFA)评分匹配的患者。主要终点事件为院内全因死亡。其他观察指标包括尿量变化及不良反应情况。使用Cox回归模型校正混杂因素,分析SCD联合CVVH治疗模式的疗效是否优于常规CVVH治疗。结果共入选9例重症AKI患者。SCD联合CVVH治疗组院内全因死亡率为22.2%,显著低于历史对照组(77.8%)(x2=5.247,P=0.027)。在校正年龄、SOFA评分、平均尿量变化等混杂因素后,Cox回归模型显示,SCD联合CVVH的疗效优于常规CVVH治疗(T=-2.596,P=0.0222)。治疗7d后,SCD治疗组平均尿量从基线值约500ml/d升高至2000ml/d以上。研究过程中,患者仅有数例轻度不良反应,无严重不良事件发生。结论 SCD可通过灭活激活的白细胞而调控机体免疫反应,最终降低重症AKI患者的死亡率。SCD安全性良好。周莹 谢琼虹 刘骏峰 许钟烨 匡鼎伟 游怀舟 赖凌云 顾勇 郝传明 林善锬 丁峰 H.David Humes 2011中国血液净化2011,10,11:3
