1Gene therapy: light is finally in the tunnel显示文摘After two decades of ups and downs,gene therapy has recently achieved a milestone in treating patients with Leber’s congenital amaurosis(LCA).LCA is a group of inherited blinding diseases with retinal degeneration and severe vision loss in early infancy.Mutations in several genes,including RPE65,cause the disease.Using adenoassociated virus as a vector,three independent teams of investigators have recently shown that RPE65 can be delivered to retinal pigment epithelial cells of LCA patients by subretinal injections resulting in clinical benefits without side effects.However,considering the whole field of gene therapy,there are still major obstacles to clinical applications for other diseases.These obstacles include innate and immune barriers to vector delivery,toxicity of vectors and the lack of sustained therapeutic gene expression.Therefore,new strategies are needed to overcome these hurdles for achieving safe and effective gene therapy.In this article,we shall review the major advancements over the past two decades and,using lung gene therapy as an example,discuss the current obstacles and possible solutions to provide a roadmap for future gene therapy research.Huibi Cao Robert S.Molday Jim Hu 2011Protein & Cell2011,2,12:9

3Identification of selenocyst-eine insertion sequence(SECIS) element in eukaryotic selenoproteins by RNA Draw program显示文摘The computer program RNA Draw was used to identify the secondary structures in the 3’untranslated regions (3’UTRs) of the mRNAs from 46 eukaryotic seleno-proteins among 7 species. The program found one or two possible SECIS elements in these selenoproteins. The SECIS element consists of a stem-loop or hairpin structure with three conserved sequences of AUGA-(A)AA-GA. SECIS element was not found by the RNA Draw program in randomly selected non-selenoproteins. The results showed that SECIS element is the unique character of the genes ofeukaryotic selenoproteins. 4Potential of Helper-Dependent Adenoviral Vectors in Modulating Airway Innate Immunity显示文摘Innate immune responses form the first line of defense against foreign insults and recently significant advances have been made in our understanding of the initiation of innate immune response along with its ability to modulate inflammation. In airway diseases such as asthma, COPD and cystic fibrosis, over reacting of the airway innate immune responses leads to cytokine imbalance and airway remodeling or damage. Helper-dependent adenoviral vectors have the potential to deliver genes to modulate airway innate immune responses and have many advantages over its predecessors. 5Lung gene therapydHow to capture illumination from the light already present in the tunnel显示文摘Gene therapy has been considered as the most ideal medical intervention for genetic diseases because it is intended to target the cause of diseases instead of disease symptoms.Availability of techniques for identification of genetic mutations and for in vitro manipulation of genes makes it practical and attractive.After the initial hype in 1990s and later disappointments in clinical trials formore than a decade,light has finally come into the tunnel in recent years,especially in the field of eye gene therapy where it has taken big strides.Clinical trials in gene therapy for retinal degenerative diseases such as Leber’s congenital amaurosis(LCA)and choroideremia demonstrated clear therapeutic efficacies without apparent side effects.Although these successful examples are still rare and sporadic in the field,they provide the proof of concept for harnessing the power of gene therapy to treat genetic diseases and to modernize our medication.In addition,those success stories illuminate the path for the development of gene therapy treating other genetic diseases.Because of the differences in target organs and cells,distinct barriers to gene delivery exist in gene therapy for each genetic disease.It is not feasible for authors to review the current development in the entire field.Thus,in this article,we will focus onwhatwe can learn from the current success in gene therapy for retinal degenerative diseases to speed up the gene therapy development for lung diseases,such as cystic fibrosis.Emily Xia Manjunatha Ankathatti Munegowda Huibi Cao Jim Hu 2014Genes & Diseases2014,1,1:2

9Porcine methionine sulfoxide reductase B3:molecular cloning,tissue-specific expression profiles,and polymorphisms associated with ear size in Sus scrofa显示文摘Background: In Sus scrofa, methionine sulfoxide reductase B3(MSRB3) is a crucial candidate gene for ear size, and an important conformational trait of pig breeds. However, challenges in MSRB3 c DNA amplification have prevented further identification of MSRB3 allelic variants influencing pig ear size.Results: We cloned a full-length c DNA sequence of porcine MSRB3 by rapid-amplification of c DNA ends. The3,765-bp gene contained a 5'-untranslated region(UTR)(190 bp), a coding region(552 bp), and a 3'-UTR(3,016 bp) and shared 84 %, 84 %, 87 %, 86 %, and 70 % sequence identities with human, orangutan, mouse, chicken, and zebrafish,respectively. The gene encoded a 183-amino acid protein, which shared 88 %, 91 %, 89 %, 86 %, and 67 % identities with human, orangutan, mouse, chicken, and zebrafish, respectively. Tissue expression analysis using q RT-PCR revealed that MSRB3 was expressed in the heart, liver, lung, kidney, spleen, ear, muscle, fat, lymph, skeletal, and hypothalamic tissues. Three single nucleotide polymorphisms(SNPs) were identified in MSRB3: c.-735 C > T in the 5' flanking region,c.2571 T > C in the 3'-UTR, and a synonymous mutation of c.484 T > C in the coding region. The SNPs c.-735 C > T and c.2571 T > C were significantly associated with ear size in a Large White × Minzhu F2 population other than in Beijing Black pigs. Subsequently, at SNP c.-735 C > T, the m RNA of MSRB3 was significantly higher expressed in ears of individuals with the TT genotype(Minzhu) than those with CC(Large White).Conclusions: The porcine MSRB3 owned a 3,765-bp full-length c DNA sequence and was detected to express in ear tissue. Two SNPs of this gene were shown to be significantly associated with ear size in a Large White × Minzhu intercross population instead of Beijing Black pig population. What's more, the individuals with higher m RNA expression of MSRB3 have larger ear sizes. 19Genome-wide search for candidate genes determining vertebrae number in pigs显示文摘Longer porcine carcasses may be expected to have more vertebrae. Therefore, vertebrae number in pigs is an economically important trait. To examine the genetic basis of this trait, we genotyped 578 F_2 Large White ?Minzhu pigs using the Porcine SNP60K Bead Chip. A genome-wide association study(GWAS) identified 36 significant single nucleotide polymorphisms(SNPs) on the chromosomes SSC1(294.28–300.32 Mb) and SSC7(102.22–109.39 Mb). A 6.04-Mb region that contained all 13 significant SNPs on SSC1 also contained the gene NR6A1, previously reported to influence the number of vertebrae in pigs. However, the reported putative casual mutation of NR6A1 c.748 C > T showed no genome-wide significant association with the trait, suggesting it was not a causal mutation in our population. The remaining 23 significant SNPs on SSC7 were concentrated in a 7.17-Mb region, which was within a quantitative trait locus interval for number of vertebrae. TMED10 was the closest gene to the most significant SNP and might be a candidate.Haplotype sharing and block analysis refined the QTL to an interval of about 3 Mb containing 29 candidate genes.Of these 29 genes, the previously reported possible casual mutation of VRTN g.19034 A > C was not found to be a causal mutation in our population. Exploration of these genes via additional genetic and functional studies in mammals revealed that TGFβ3 could be a good candidate on SSC7. 20Modification of Fe_3O_4 Magnetic Nanoparticles by L-dopa or Dopamine as an Enzyme Support显示文摘Fe_3O_4 magnetic nanoparticles were prepared by co-precipitation of Fe2+ and Fe3+ in an ammonia solution,and its size was about 36 nm measured by an atomic force microscope.Fe3O4 magnetic nanoparticles were modified by L-dopa or dopamine using sonication method.The analysis of FTIR clearly indicated the formation of Fe-O-C bond.Direct immobilization of trypsin(EC:3.4.21.4)on Fe3O4 magnetic nanoparticles with L-dopa and dopamine spacer was investigated using glutaraldehyde as a coupling agent.No significant changes in the size and magnetic property of the three kinds of magnetic nanoparticles linked with or without trypsin were observed.The existence of the spacer molecule on magnetic nanoparticles could greatly improve the activity and the storage stability of bound trypsin through increasing the flexibility of enzyme and changing the microenvironment on nanoparticles surface compared to the naked magnetic nanoparticles.PENG Hong ZHANG Xiao HUANG Kaixun XU Huibi 2008Journal of Wuhan University of Technology(Materials Science)2008,23,4:1
