| 1 | Activation of SIRT1 ameliorates LPS-induced lung injury in mice via decreasing endothelial tight junction permeability显示文摘The integrity of the endothelial barrier is a determinant of the prognosis of lipopolysaccharide (LPS)-induced acute lung injury (ALI). In this study, we investigated whether and how Sirtuin 1 (SIRT1) maintained the vascular integrity during ALI. An experimental model of ALI was established in mice through intratracheal administration of LPS (10 mg/kg). LPS stimulation significantly increased the pulmonary permeability and decreased the expression of SIRT1 and tight junction proteins (TJs), including occludin, claudin-5, tight junction protein 1 and tight junction protein 2. Morphological studies showed that LPS induced obvious lung injury with inammatory cell infiltration in the interstitial and alveolar space, hemorrhage, edema, and the thickened alveolar wall compared to the control mice. Intratracheal administration of the selective SIRT1 activator SRT1720 (6.25 mg/kg) significantly attenuated LPS- induced lung injury, lung hyper-permeability and increased TJs expression, whereas intratracheal administration of the selective SIRT1 inhibitor EX527 (6.25 mg/kg) aggravated LPS-induced ALI. Similar protective effects of SIRT1 on pulmonary cellular permeability were observed in primary human pulmonary microvascular endothelial cells treated with LPS (2 mg/mL) in vitro. We further demonstrated that the RhoA/ROCK signaling pathway was activated in SIRT1 regulation of tight junction permeability. The RhoA/ROCK inhibitor Y-27632 (10 μM) increased the expression of TJs and reversed LPS- or EX527-induced hyper-permeability. In conclusion, SIRT1 ameliorates LPS-induced lung injury via decreasing endothelial tight junction permeability, possibly via RhoA/ ROCK signaling pathway. | 2 | Histological Subtypes of Lung Cancer in Chinese Males from 2000 to 2012显示文摘Objective To characterize the histological and epidemiological features of male lung cancer patients in China. Methods The demographic and histological information about male lung cancer patients identified from 2000-01-01 to 2012-12-31,was collected from the Cancer Hospital of the Chinese Academy of Medical Sciences. Relative frequencies(RF) were estimated for major histological subtypes and compared according to the years of diagnosis and birth. Results The RF of adenocarcinoma(ADC) increased from 21.96% to 43.36% and the RF of squamous cell carcinoma(SCC) decreased from 39.11% to 32.23% from 2000 to 2012 in the 15 427 male lung cancer patients included in this study(Z=17.909,P<0.0001; Z=-6.117,P<0.0001). The RF of ADC increased from 28.72% in 2000-2004,36.88% in 2005-2008 to 48.61% in 2009-2012 in patients born after 1960. The age-adjusted RF of ADC in 2007-2012 increased consistently in all the investigated areas. Conclusion The increased RF of ADC in male lung cancer patients highlights the need for further investigation of the etiologic factors of these tumors. | 3 | Vehicular volatile organic compounds losses due to refueling and diurnal process in China: 2010–2050显示文摘Volatile organic compounds(VOCs) are crucial to control air pollution in major Chinese cities since VOCs are the dominant factor influencing ambient ozone level, and also an important precursor of secondary organic aerosols. Vehicular evaporative emissions have become a major and growing source of VOC emissions in China. This study consists of lab tests, technology evaluation, emissions modeling, policy projections and cost-benefit analysis to draw a roadmap for China for controlling vehicular evaporative emissions. The analysis suggests that evaporative VOC emissions from China's light-duty gasoline vehicles were approximately 185,000 ton in 2010 and would peak at 1,200,000 ton in 2040 without control. The current control strategy implemented in China, as shown in business as usual(BAU) scenario, will barely reduce the long-term growth in emissions. Even if Stage II gasoline station vapor control policies were extended national wide(BAU + extended Stage II), there would still be over 400,000 ton fuel loss in 2050. In contrast, the implementation of on-board refueling vapor recovery(ORVR) on new cars could reduce 97.5% of evaporative VOCs by 2050(BAU + ORVR/BAU + delayed ORVR). According to the results, a combined Stage II and ORVR program is a comprehensive solution that provides both short-term and long-term benefits. | 6 | Identification of a Newly Isolated Getah Virus in the China-Laos Border,China显示文摘In this study, we isolated a virus strain(YN12031) from specimens of Armigeres subalbatus collected in the China-Laos border. BHK-21 cells infected with YN12031 exhibited an evident cytopathic effect(CPE) 32 h post-infection. The virus particles were spherical, 70 nm in diameter, and enveloped; they also featured surface fibers. Molecular genetic analysis revealed that YN12031 was closely related to alpha viruses such as Chikungunya virus and Sindbis virus, and located in the same clade as MM2021, the prototype of Getahvirus(GETV) isolated in Malaysia in 1955. Phylogenetic analysis of the E2 and capsid genes further revealed that YN12031 was located in the same clade as the Russian isolate LEIV/16275/Mag. Analysis of the homology of nucleotides and amino acids in the coding area and E2 gene demonstrated that the YN12031 isolated from the China-Laos border(tropical region) was related closest to the LEIV/16275/Mag isolate obtained in Russia(North frigid zone area) among other isolates studied. These results suggest that GETV can adapt to different geographical environments to propagate and evolve. | 7 | TaqMan Real-time RT-PCR Assay for Detecting and Differentiating Japanese Encephalitis Virus显示文摘Objective To detect Japanese encephalitis virus(JEV) rapidly and distinguish its genotypes, a TaqMan-based reverse transcriptase quantitative polymerase chain reaction(RT-PCR) detection system was developed.Methods By aligning the full-length sequences of JEV(G1-G5), six sets of highly specific TaqMan real-time RT-PCR primers and probes were designed based on the highly conserved NS1, NS2, and M genes of JEV, which included one set for non-specific JEV detection and five sets for the detection of specific JEV genotypes. Twenty batches of mosquito samples were used to evaluate our quantitative PCR assay.Results With the specific assay, no other flavivirus were detected. The lower limits of detection of the system were 1 pfu/mL for JEV titers and 100 RNA copies/μL. The coefficients of variation of this real-time RT-PCR were all < 2.8%. | 10 | Regulating Effects of Novel CpG Chitosan-nanoparticles on Immune Responses of Mice to Porcine Paratyphoid Vaccines显示文摘Objective To study the regulating effects of a novel CpG oligodeoxynuleotide and the synergistic effect of chitosan-nanoparticles (CNP) with CpG on immune responses of mice, which were used to develop a novel immunoadjuvant to boost immune response to conventional vaccines. Methods A novel CpG ODN containing 11 CpG motifs was synthesized and its bioactivities to stimulate the proliferation of lymphocytes of pig in vitro were detected. Then it was entrapped with CNP prepared in our laboratory by the method of ionic cross linkage, and immunized Kunming mice were co-inoculated with paratyphoid vaccine. The peripheral blood was collected weekly from the tail vein of inoculated mice to detect the contents of IgG, IgA, IgM, and specific antibody against salmonella as well as the levels of interleukin-2 (IL2), IL-4, and IL-6 by SABC-ELISA assay. The numbers of leucocytes, monocytes, granuloytes, and lymphocytes were calculated separately using the routine method. The experimental mice were orally challenged with virulent salmonella 35 days after inoculation. Results This CpG ODN could remarkably provoke the proliferation of lymphocytes of pig in vitro in contrast with the control (P<0.05). Compared with those of the control, immunoglobulins, including IgG, IgA, IgM, and specific antibodies to paratyphoid vaccine, increased significantly in sera from the CpG or CpG-CNP-vaccinated mice (P<0.05). IL-2, IL-4, and IL-6 increased remarkably in sera from immunized mice (P<0.05). The leucocytes, monocytes, granuloytes, and lymphocytes of the mice immunized with CpG or CpG-CNP were also increased in number (P<0.05). After the challenge, these immunity values were elevated in the mice vaccinated with CpG or CpG-CNP. The immunized mice all survived, while the control mice fell ill with evident lesions with diffuse hemorrhage in stomach, small intestine, and peritoneum. | 11 | Molecular Characterization of Full-length Genome of Japanese Encephalitis Virus Genotype V Isolated from Tibet, China显示文摘Objective To determine the molecular characterization of full-length genome of Japanese encephalitis virus(JEV) genotype V. Methods The full-length nucleotide sequences of JEV strains isolated from different locations and sources were used in sequence and phylogenetic analysis. Results The full-length genome of genotypes V JEV, XZ0934, and Muar strain were composed of 10 983 and 10 988 nucleotides respectively and shared a lower level of identity with JEV genotypes I-IV, ranging from 78.4%(G I, KV1899) to 79.7%(G III, JaGAr01), for the nucleotide sequences, and from 90.0%(G I, KV1899) to 91.8%(G III, JaGAr01) for the amino acid sequences. The open reading frame(ORF) of JEV genotype V spanned nucleotides 96 to 10 397 and encoded 3 433 amino acids. Interestingly, a comparison with JEV genotype I-IV revealed that 3 nucleotides(encoded with a serine residue) were inserted in the NS4A gene of JEV genotype V, and the insertion of nucleotides was also found in downstream of the ORF stop codon in 3'-untranslated region. Moreover, numerous amino acid mutations were observed in 3 functional domains of the E gene of JEV genotype V. Conclusion The molecular characterization of JEV genotype V is significantly different from that of the known genotypes I-IV. Especially with regard to pain experience, the IC has been supposed to participate in both sensory-discriminative and affective-motivational aspects of pain. In this review, we discuss the latest data proposing that subregions of the IC are involved in isolated pain networks: the posterior sensory circuit and the anterior emotional network. Due to abundant connections with other brain areas, the IC is likely to serve as an interface where cross-modal shaping of pain occurs. In chronic pain,however, this mode of emotional awareness and the modulation of pain are disrupted. | 16 | Long-term inorganic plus organic fertilization increases yield and yield stability of winter wheat显示文摘An understanding of wheat yield and yield stability response to fertilization is important for sustainable wheat production. A 36-year long-term fertilization experiment was employed to evaluate the yield and yield stability of winter wheat. Five fertilization regimes were compared,including(1) CK, no fertilizer;(2) NPK, inorganic fertilizer only;(3) O, organic fertilizer only;(4)NPKO, 50% of NPK plus 50% of O, and(5) HNPKO, 80% of NPK plus 80% of O. The greatest yield increase was recorded in HNPKO, followed by NPKO, with O producing the lowest mean yield increase. Over the 36 years, the rate of wheat yield increase in fertilized plots ranged from95.31 kg ha-1 year-1 in the HNPKO to 138.65 kg ha-1 year-1 in the O. Yield stability analysis using the additive main effects and multiplicative interactions(AMMI) method assigned 62.3%, 26.3%,and 11.4% of sums of squares to fertilization effect, environmental effect, and fertilization ×environment interaction effect, respectively. The combination of inorganic and organic fertilization(NPKO and HNPKO) appeared to produce more stable yields than O or NPK, with lower coefficients of variation and AMMI stability value. However, wheat grown with O seemed to be the most susceptible to climate change and the least productive among the fertilized plots.Significant correlations of grain yield with soil properties and with mean air temperature were observed. To establish successful in fections, plant bacterial, oomycete, fun gal, and viral pathogens have evolved at least three major strategies to disrupt SA-mediated defense? The first strategy is to reduce SA accumulation directly by converting SA into its inactive derivatives. The second strategy is to interrupt SA biosynthesis by targeting the ICS1 pathway. In the third major strategy, plant pathogens deploy different mechanisms to interfere with SA downstream signaling. | 18 | A Reverse-transcription Recombinase-aided Amplification Assay for the Rapid Detection of the Far-Eastern Subtype of Tick-borne Encephalitis Virus显示文摘Objective Tick-borne encephalitis virus(TBEV) is an emerging pathogen in Europe and North Asia that causes tick-borne encephalitis(TBE). A simple, rapid method for detecting TBEV RNA is needed to control this disease. Methods A reverse-transcription recombinase-aided amplification(RT-RAA) assay was developed. This assay can be completed in one closed tube at 39℃ within 30 minutes. The sensitivity and specificity of RT-RAA were validated using non-infectious synthetic RNA representing a fragment of the NS5 region of the wild-type(WT) TBEV genome and the Senzhang strain. Additionally, 10 batches of tick samples were used to evaluate the performance of the RT-RAA assay. Results The analytical limit of detection of the assay was 20 copies per reaction of the TBEV synthetic transcript and 3 plaque-forming units(pfu) per reaction of TBEV titers. With the specific assay, no signal due to other arboviruses was observed. Of the 10 batches of tick samples obtained from the Changbai Mountains of China, three were TBEV-positive, which was consistent with the results of the quantitative real-time PCR assay. | 19 | Reduced β2-GPI is associated with increased platelet aggregation and activation in patients with prolonged isolated thrombocytopenia after allo-HSCT显示文摘We aimed to measure platelet function and its relationship with β2-GPI in prolonged isolated thrombocytopenia(PT) after allogeneic hematopoietic stem cell transplantation(allo-HSCT).Fifty-six patients with PT and 60 allo-HSCT recipients without PT(non-PT controls) were enrolled.Platelet aggregation and activation,β2-GPI and anti-β2-GPI antibody levels,vWF antigen,and vWF activity were analyzed.The effect of β2-GPI on platelet aggregation was also measured ex vivo.Results showed that ADP-induced platelet aggregation significantly increased(39%±7.5% vs.23%±8.5%,P=0.032),and the platelet expression of both CD62 p(33.6%±11.6% vs.8.5%±3.5%,P<0.001) and PAC-1(42.4%±7.6% vs.6.8%±2.2%,P<0.001) was significantly higher in patients with PT than in those without PT.Significantly lower β2-GPI levels(164.2±12 μg m L^-1 vs.234.2±16 μg mL^-1,P<0.001),higher anti-β2-GPI IgG levels(1.78±0.46 U mL^-1 vs.0.94±0.39 U mL^-1,P<0.001),and increased vWF activity(133.06%±30.50% vs.102.17%±25.90%,P<0.001) were observed in patients with PT than in those without PT.Both ADPinduced platelet aggregation(n=116,r^2=-0.5042,P<0.001) and vWF activity(n=116,r^2=-0.2872,P<0.001) were negatively correlated with β2-GPI levels.In summary,our data suggested that platelet aggregation and activation were significantly higher in patients with PT than in those without PT,which might be associated with reduced β2-GPI levels.The reduced β2-GPI levels might be due to the existence of anti-β2-GPI IgG. | Haixia Fu Jingzhong Zhao Lanping Xu Kaiyan Liu Yu Wang Huan Chen Wei Han Jingzhi Wang Fengrong Wang Xiaojun Huang Xiaohui Zhang | 2019 | Science China(Life Sciences)2019,62,7: | 4 |
