1Clinical characteristics of 24 asymptomatic infections with COVID-19 screened among close contacts in Nanjing,China显示文摘Previous studies have showed clinical characteristics of patients with the 2019 novel coronavirus disease(COVID-19)and the evidence of person-to-person transmission.Limited data are available for asymptomatic infections.This study aims to present the clinical characteristics of 24 cases with asymptomatic infection screened from close contacts and to show the transmission potential of asymptomatic COVID-19 virus carriers.Epidemiological investigations were conducted among all close contacts of COVID-19 patients(or suspected patients)in Nanjing,Jiangsu Province,China,from Jan 28 to Feb 9,2020,both in clinic and in community.Asymptomatic carriers were laboratory-confirmed positive for the COVID-19 virus by testing the nucleic acid of the pharyngeal swab samples.Their clinical records,laboratory assessments,and chest CT scans were reviewed.As a result,none of the 24 asymptomatic cases presented any obvious symptoms while nucleic acid screening.Five cases(20.8%)developed symptoms(fever,cough,fatigue,etc.)during hospitalization.Twelve(50.0%)cases showed typical CT images of ground-glass chest and 5(20.8%)presented stripe shadowing in the lungs.The remaining 7(29.2%)cases showed normal CT image and had no symptoms during hospitalization.These 7 cases were younger(median age:14.0 years;P=0.012)than the rest.None of the 24 cases developed severe COVID-19 pneumonia or died.The median communicable period,defined as the interval from the first day of positive nucleic acid tests to the first day of continuous negative tests,was 9.5 days(up to 21 days among the 24 asymptomatic cases).Through epidemiological investigation,we observed a typical asymptomatic transmission to the cohabiting family members,which even caused severe COVID-19 pneumonia.Overall,the asymptomatic carriers identified from close contacts were prone to be mildly ill during hospitalization.However,the communicable period could be up to three weeks and the communicated patients could develop severe illness.These results highlighted the importance of close contact tracing and longitudinally surveillance via virus nucleic acid tests.Further isolation recommendation and continuous nucleic acid tests may also be recommended to the patients discharged.Zhiliang Hu Ci Song Chuanjun Xu Guangfu Jin Yaling Chen Xin Xu Hongxia Ma Wei Chen Yuan Lin Yishan Zheng Jianming Wang Zhibin Hu Yongxiang Yi Hongbing Shen 2020Science China(Life Sciences)2020,63,5:77
2Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 3Resequencing of 145 Landmark Cultivars Reveals Asymmetric Sub-genome Selection and Strong Founder Genotype Effects on Wheat Breeding in China显示文摘Controlled pedigrees and the multi-decade timescale of national crop plant breeding programs offer a unique experimental context for examining how selection affects plant genomes.More than 3000 wheat cultivars have been registered,released,and documented since 1949 in China.In this study,a set of 145 elite cultivars selected from historical points of wheat breeding in China were re-sequenced.A total of 43.75 Tb of sequence data were generated with an average read depth of 17.94x for each cultivar,and more than 60.92 million SNPs and 2.54 million InDels were captured,based on the Chinese Spring RefSeq genome v1.0.Seventy years of breeder-driven selection led to dramatic changes in grain yield and related phenotypes,with distinct genomic regions and phenotypes tar-geted by different breeders across the decades.There are very clear instances illustrating how introduced Italian and otherforeign germplasm was integrated into Chinese wheat programs and reshaped the genomic landscape of local modern cultivars.Importantly,the resequencing data also highlighted significant asymmetric breeding selec-tion among the three sub-genomes:this was evident in both the collinear blocks for homeologous chromosomes and among sets of three homeologous genes.Accumulation of more newly assembled genes in newer cultivars implied the potential value of these genes in breeding.Conserved and extended sharing of linkage disequilibrium(LD)blocks was highlighted among pedigree-related cultivars,in which fewer haplotype differences were detected.Fixation or replacement of haplotypes from founder genotypes after generations of breeding was related to their breeding value.Based on the haplotype frequency changes in LD blocks of pedigree-related cultivars,we propose a strategy for evaluating the breeding value of any given line on the basis of the accumulation(pyramiding)of bene-ficial haplotypes.Collectively,our study demonstrates the influence of 'founder genotypes' on the output of breeding efforts over many decades and also suggests that founder genotype perspectives are in fact more dy-namic when applied in the context of modern genomics-informed breeding.Chenyang Hao Chengzhi Jiao Jian Hou Tian Li Hongxia Liu Yuquan Wang Jun Zheng Hong Liu Zhihong Bi Fengfeng Xu Jing Zhao Lin Ma Yamei Wang Uzma Majeed Xu Liu Rudi Appels Marco Maccaferri Roberto Tuberosa Hongfeng Lu Xueyong Zhang 2020Molecular Plant2020,13,12:9

6Prognostic assessment of apoptotic gene polymorphisms in non-small cell lung cancer in Chinese显示文摘Apoptosis plays a key role in inhibiting tumor growth, progression and resistance to anti-tumor therapy. We hypothesized that genetic variants in apoptotic genes may affect the prognosis of lung cancer. To test this hypothesis, we selected 38 potentially functional single nucleotide polymorphisms (SNPs) from 12 genes (BAX, BCL2, BID, CASP3, CASP6, CASP7, CASP8, CASP9, CASP10, FAS, FASLG and MCL1) involved in apoptosis to assess their prognostic significance in lung cancer in a Chinese case cohort with 568 non-small cell lung cancer (NSCLC) patients. Thirty-five SNPs passing quality control underwent association analyses, 11 of which were shown to be significantly associated with NSCLC survival (P<0.05). After Cox stepwise regression analyses, 3 SNPs were independently associated with the outcome of NSCLC (BID rs8190315: P=0.003; CASP9 rs4645981: P=0.007 and FAS rs1800682: P=0.016). A favorable survival of NSCLC was significantly associated with the genotypes of BID rs8190315 AG/GG (adjusted HR=0.65, 95% CI: 0.49-0.88), CASP9 rs4645981 AA (HR=0.22, 95% CI: 0.07-0.69) and FAS rs1800682 GG (adjusted HR=0.67, 95% CI: 0.46-0.97). Time-dependent receptor operation curve (ROC) analysis revealed that the area under curve (AUC) at year 5 was significantly increased from 0.762 to 0.819 after adding the risk score of these 3 SNPs to the clinical risk score. The remaining 32 SNPs were not significantly associated with NSCLC prognosis after adjustment for these 3 SNPs. 7Effects of Traditional Chinese Medicine herbs for tonifying Qi and kidney, and replenishing spleen on intermittent asthma in children aged 2 to 5 years old显示文摘OBJECTIVE: To evaluate the efficacy and safety of Traditional Chinese Medicine(TCM) herbs for tonifying Qi and kidney, and replenishing spleen on intermittent asthma in children aged 2 and 5 years.METHODS: A randomized, single-blind, placebo controlled trial was conducted. Children with intermittent asthma were enrolled and their baseline conditions were measured using a questionnaire. A total of 60 participants, aged 2 to 5, were randomized into either the treatment group(n = 40) or the placebo group(n = 20). The treatment group was treated with granules of TCM herbs for tonifying Qi and kidney, and replenishing spleen, and the placebo group was given placebo granules for 3 months.The number of asthma attacks was counted and TCM syndrome scores were measured at 1, 2, 3, 6, 9,and 12 months. The airway resistance and levels of eosinophil cationic protein for the two groups were observed before and after 3 months. The results were statistically analyzed.RESULTS: Compared with the placebo group, the number of asthma attacks significantly decreased in the treatment group(P < 0.05). For the treatment group, the TCM syndrome scores decreased after 1 and 2 months; there was also a significant difference in scores between the two groups(P < 0.05).The difference remained after the medicine was stopped for 9 months(P < 0.05). After the 3-month treatment, compared with the placebo group, the airway resistance decreased in the treatment group(P < 0.05). 9Association between Arctic autumn sea ice concentration and early winter precipitation in China显示文摘Associations between autumn Arctic sea ice concentration(SIC) and early winter precipitation in China are studied using singular value decomposition analysis. The results show that a reduced SIC almost everywhere in the Arctic Ocean, except the northern Greenland Sea and Canadian Basin, are accompanied by dry conditions over central China, extending northeast from the Tibetan Plateau toward the Japan Sea, the Bohai Sea and the Yellow Sea, and wet conditions over South China and North China. Atmospheric circulation anomalies associated with SIC variability show two wave-train structures, which are persistent from autumn to winter, leading to the identified relationship between autumn Arctic SIC and early winter precipitation in China. 10Diversity and sub-functionalization of TaGW8 homoeologs hold potential for genetic yield improvement in wheat显示文摘Grain Weight 8(GW8) in rice is a SQUAMOSA Promoter-Binding Protein-Like(SPL) family transcription factor with multiple biological functions. In this study, three GW8 homoeologs were cloned from homoeologous group 7 chromosomes of wheat. Subcellular localization and trans-activation activity assays suggested that Ta GW8 is a transcriptional activator.Ta GW8 genes were preferentially expressed in young spikes and developing grains. Ectopic expressions of Ta GW8 in Arabidopsis and rice resulted in enhanced vegetative growth,earlier flowering and larger seeds. Ta GW8-7 A was the most highly variable of the three homoeologs with four haplotypes(Hap-1/2/3/4). Ta GW8-7 B had two haplotypes(Hap-L/H).Ta GW8-7 A-Hap-2 was associated with high thousand-grain weight(TGW) and large kernel length and showed higher transcriptional levels and binding activity than the other haplotypes. The high frequency of Ta GW8-7 A-Hap-2 in Chinese wheat populations suggested that it had been extensively selected in breeding. This haplotype showed a high potential for exploitation in global wheat breeding because its frequency was low in exotic germplasm. Ta GW8-7 B-Hap-H produced abundant transcripts and was associated with early heading and maturity, less tiller number and high TGW. This haplotype might be suitable for multiple cropping systems due to short wheat season. 11Au-Pd/mesoporous Fe_2O_3:Highly active photocatalysts for the visible-light-driven degradation of acetone显示文摘Three-dimensionally ordered mesoporous Fe_2O_3(meso-Fe_2O_3) and its supported Au, Pd,and Au-Pd alloy(xA uP dy/meso-Fe_2O_3; x = 0.08–0.72 wt.%; Pd/Au molar ratio(y) = 1.48–1.85)photocatalysts have been prepared via the KIT-6-templating and polyvinyl alcohol-protected reduction routes, respectively. Physical properties of the samples were characterized, and their photocatalytic activities were evaluated for the photocatalytic oxidation of acetone in the presence of a small amount of H_2O_2 under visible-light illumination. It was found that the meso-Fe_2O_3 was rhombohedral in crystal structure. The as-obtained samples displayed a high surface area of 111.0–140.8 m^2/g and a bandgap energy of 1.98–2.12 eV. The Au, Pd and/or Au–Pd alloy nanoparticles(NPs) with a size of 3–4 nm were uniformly dispersed on the surface of the meso-Fe_2O_3 support. The 0.72 wt.% AuP d1.48/meso-Fe_2O_3 sample performed the best in the presence of 0.06 mol/L H_2O_2 aqueous solution, showing a 100% acetone conversion within4 hr of visible-light illumination. It was concluded that the good performance of 0.72 wt.%AuPd_(1.48)/meso-Fe_2O_3 for photocatalytic acetone oxidation was associated with its ordered mesoporous structure, high adsorbed oxygen species concentration, plasmonic resonance effect between AuPd_(1.48) NPs and meso-Fe_2O_3, and effective separation of the photogenerated charge carriers. 12Western Indian Ocean SST signal and anomalous Antarctic sea-ice concentration variation显示文摘Teleconnection between El Nino/La Nina-Southern Oscillation (ENSO) phenomenon and anomalous Antarctic sea-ice variation has been studied extensively.In this study,impacts of sea surface temperature in the Indian Ocean on Antarctic sea-ice change were investigated during Janaury 1979 and October 2009.Based on previous research results,sea areas in the western Indian Ocean (WIO;50°–70°E,10 °–20 °S) are selected for the resreach.All variables showed 1-10 year interannual timescales by Fast Founer Tranaform (FFT) transformation.Results show that i) strong WIO signals emerged in the anomalous changes of Antarctic sea-ice concentration;ii) significant positive correlations occurred around the Antarctic Peninsula,Ross Sea and its northwest peripheral sea region iii) negative correlation occurred in the Indian Ocean section of the Southern Ocean,Amundsen Seas,and the sea area over northern Ross Sea;and iv) the atmospheric anomalies associated with the WIO including wind,meridional heat flux,and surface air temperature over southern high latitudes were the possible factors for the teleconnection.LIU Na ZHANG Zhanhai CHEN Hongxia LIN Lina 2011Acta Oceanologica Sinica2011,30,2:2
13The Flavor and Antioxidant Activity Change Pattern of Shrimp Head Paste During Fermentation显示文摘To study the changes of volatile compounds and antioxidant activity of fast fermented shrimp head paste during the fermentation processing,Penaeus vannamei boone head was used as the raw material,and compound starter was added for fermen-tation.During the 14 days of fermentation,the changes of free amino acids,volatile flavor compounds and main components were determined by SPME-GC-MS and electronic nose.The oxidation-resistance changes of water extract at different fermentation stages were evaluated.The results showed that the total free amino acids increased from original 2320 to 5640 mg(100 mL)^(−1).SPME-GC-MS analysis found that aldehydes,pyrazine compounds increased from 10.80%,1.94%to 24.35%,13.63%respectively during the fermentation process.The HO·scavenging ability of shrimp head paste increased from 52%to 86%.Our results showed that the shrimp paste produced from shrimp head fermentation could obtain good flavor and possess certain antioxidant activity of aquatic fermented condiment.YU Jing LU Kuan SUN Jinyuan XIE Wancui SONG Lin CHE Hongxia 2022Journal of Ocean University of China2022,21,1:2
14Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART.Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu 2021Cell Research2021,31,8:2
15Observed features of temperature, salinity and current in central Chukchi Sea during the summer of 2012显示文摘During the summer of 2012, the fifth CHINARE Arctic Expedition was carried out, and a submersible mooring system was deployed in M5 station located at(69°30.155'N,169°00.654'W) and recovered 50 d later. A set of temperature, salinity and current profile records was acquired. The characteristics of these observations are analyzed in this paper. Some main results are achieved as below.(1) Temperature generally decreases while salinity generally increases with increasing depth. The average values of all records are 2.98°C and 32.21 psu.(2)Salinity and temperature are well negatively correlated, and the correlation coefficient between them is –0.84.However, they did not always vary synchronously. Their co-variation featured different characters during different significant periods.(3) The average velocity for the whole water column is 141 mm/s with directional angle of347.1°. The statistical distribution curve of velocity record number gets narrower with increasing depth. More than85% of the recorded velocities are northward, and the mean magnitudes of dominated northward velocities are100–150 mm/s.(4) Rotary spectrum analysis shows that motions with low frequency take a majority of energy in all layers. The most significant energy peaks for all layers are around 0.012 cph(about 3.5 d period), while the tidal motion in mooring area is nonsignificant.(5) Velocities in all layers feature similar and synchronous temporal variations, except for the slight decrease in magnitude and leftward twist from top to bottom. The directions of velocity correspond well to those of surface wind. In case-control GWAS, typical statistical strategy is traditional logistical regression(LR) based on single-locus analysis. However, such a single-locus analysis leads to the well-known multiplicity problem, with a risk of inflating type I error and reducing power. Dimension reduction-based techniques, such as principal component-based logistic regression(PC-LR), partial least squares-based logistic regression(PLS-LR), have recently gained much attention in the analysis of high dimensional genomic data. However, the perfor?mance of these methods is still not clear, especially in GWAS. We conducted simulations and real data application to compare the type I error and power of PC-LR, PLS-LR and LR applicable to GWAS within a defined single nucleotide polymorphism(SNP) set region. We found that PC-LR and PLS can reasonably control type I error under null hypothesis.On contrast, LR, which is corrected by Bonferroni method, was more conserved in all simulation settings. In particular, we found that PC-LR and PLS-LR had comparable power and they both outperformed LR, especially when the causal SNP was in high linkage disequilibrium with genotyped ones and with a small effective size in simulation. 19Arctic autumn sea ice decline and Asian winter temperature anomaly显示文摘Associations between the autumn Arctic sea ice concentration(SIC) and Asian winter temperature are discussed using the singular value decomposition analysis.Results show that in recent 33 years reduced autumn Arctic sea ice is accompanied by Asian winter temperature decrease except in the Tibetan plateau and the Arctic Ocean and the North Pacific Ocean coast.The autumn SIC reduction excites two geopotential height centers in Eurasia and the north Arctic Ocean,which are persistent from autumn to winter.The negative center is in Barents Sea/Kara Sea.The positive center is located in Mongolia.The anomalous winds are associated with geopotential height centers,providing favorable clod air for the Asian winter temperature decreasing in recent 33 years.This relationship indicates a potential long-term outlook for the Asian winter temperature decrease as the decline of the autumn sea ice in the Arctic Ocean is expected to continue as climate warms.LIU Na LIN Lina WANG Yingjie KONG Bin ZHANG Zhanhai CHEN Hongxia 2016Acta Oceanologica Sinica2016,35,7:2
20Optics and Spectral Investigation in Dy^(3+)-Doped Borate Glasses显示文摘Dy 3+ -doped borate glasses (LBLB) with high effective visible fluorescence emission were synthesized. The absorption spectrum and fluorescence spectrum of this glass were measured and analyzed. By using J-O theory, the oscillator strengths for some absorption transitions were calculated according to the absorption spectra. The intensity parameters Ω_t (t=2, 4, 6) of Dy 3+ were determined by using a least-squares fitting approach, and the values are 4.04×10 -20 , 1.30×10 -20 and 1.82×10 -20 cm, respectively. The root-mean-square deviation δ_ rms was calculated. Under UV light excitation, Dy 3+ -doped borate glasses (LBLB) emit intense yellowish white lights.
