| 1 | Allitridi induces apoptosis by affecting Bcl-2 expression and caspase-3 activity in human gastric cancer cells显示文摘AIM: To investigate the mechanism of allitridi-induced apoptosis in human gastric cancer cell line BGC823. METHODS: Growth inhibition by allitridi was analyzed using cell growth curve and MTT assay. Apoptotic cells were detected using staining with Hoechst 33342, and confirmed by flow cytometric analysis and DNA fragmen- tation analysis. The protein expression affected by allitridi was determined using Western blot. The activity of caspase-3 was measured using a fluorescence assay. RESULTS: Allitridi induced apoptosis, and then inhibited cells proliferation in human gastric cancer cell line BGC823. The protein level of Bcl-2 was decreased dramatically, while Bax and p53 were not significantly affected by allitridi. The expression and activity of caspase-3 started to increase after allitridi treatment for 72 h. | 3 | Simultaneous removal of ammonia and N-nitrosamine precursors from high ammonia water by zeolite and powdered activated carbon显示文摘When adding sufficient chlorine to achieve breakpoint chlorination to source water containing high concentration of ammonia during drinking water treatment, high concentrations of disinfection by-products(DBPs) may form. If N-nitrosamine precursors are present, highly toxic N-nitrosamines, primarily N-nitrosodimethylamine(NDMA), may also form. Removing their precursors before disinfection should be a more effective way to minimize these DBPs formation. In this study, zeolites and activated carbon were examined for ammonia and N-nitrosamine precursor removal when incorporated into drinking water treatment processes.The test results indicate that Mordenite zeolite can remove ammonia and five of seven N-nitrosamine precursors efficiently by single step adsorption test. The practical applicability was evaluated by simulation of typical drinking water treatment processes using six-gang stirring system. The Mordenite zeolite was applied at the steps of lime softening, alum coagulation, and alum coagulation with powdered activated carbon(PAC) sorption. While the lime softening process resulted in poor zeolite performance, alum coagulation did not impact ammonia and N-nitrosamine precursor removal. During alum coagulation, more than67% ammonia and 70%–100% N-nitrosamine precursors were removed by Mordenite zeolite(except 3-(dimethylaminomethyl)indole(DMAI) and 4-dimethylaminoantipyrine(DMAP)). PAC effectively removed DMAI and DMAP when added during alum coagulation. | 4 | Recent advances in electrogenerated chemiluminescence biosensing methods for pharmaceuticals显示文摘Electrogenerated chemiluminescence(electrochemiluminescence, ECL) generates species at electrode surfaces, which undergoes electron-transfer reactions and forms excited states to emit light. It has become a very powerful analytical technique and has been widely used in such as clinical testing, biowarfare agent detection, and pharmaceutical analysis. This review focuses on the current trends of molecular recognition-based biosensing methods for pharmaceutical analysis since 2010. It introduces a background of ECL and presents the recent ECL developments in ECL immunoassay(ECLIA), immunosensors, enzyme-based biosensors, aptamer-based biosensors, and molecularly imprinted polymers(MIP)-based sensors. | 5 | Fine-scale genetic structure of Eremosparton songoricum and implication for conservation显示文摘Eremosparton songoricum (Litv.) Vass. is a clonal shrub that reproduces both asexually by under-ground rhizomes and sexually by seeds. It is now a rare species with a narrow distribution in fragmented habitat patches in the Gurbantunggut Desert of Xinjiang, China. The objective of this study was to determine the spatial pattern or structure of genetic variation within population. The within-populations genetic structure of E. songoricum in a plot (10 m×10 m) was analyzed using inter simple sequence repeats (ISSR) markers. Correlograms of Moran's I showed significant positive value was about 7 m, but changed into a negative correlation with the increase of distance, indicating strong genetic structure. The natural character of clonal reproduction, restricted seed and pollen dispersal were the main factors of influencing the spatial pattern. The knowledge of clonal structures within populations was crucial for understanding evolutionary processes and ecological adaptation. | 7 | Effect of water deficiency on relationships between metabolism,physiology,biomass,and yield of upland cotton(Gossypium hirsutum L.)显示文摘Drought is a common abiotic stress that considerably limits crop production. The objective of this study is to explore the influence of water deficiency on the yield, physiologic and metabolomic attributes in upland cotton cultivars(Gossypium hirsutum L). Cotton cultivars, 'Ishonch' and 'Tashkent-6' were selected to study the relationships among their physiologic, metabolomic and yield attributes during water deficiency. Deficit irrigation was designed by modifying the traditional watering protocol to reduce water use. Results indicate that cotton cultivars respond differently to water deficit stress. Water deficit significantly influenced plant height, the number of internodes, and sympodial branches in both cultivars. However, yield components such as the number of bolls, boll seed, lint mass, and individual plant yield were significantly reduced only in 'Tashkent-6'. The leaf area decreased and the specific leaf weight increased in 'Ishonch' under deficit irrigation conditions. However, 'Tashkent-6' demonstrated significant water loss compared to 'Ishonch', and both cultivars showed reduced transpiration rates. Untargeted metabolite profiles of leaves showed clear separation in 'Ishonch', but not in 'Tashkent-6' under deficit irrigation compared to full irrigation. The individual metabolites such as proline and galactinol showed strong association with yield under water deficit stress. Moreover, this study indicates that leaf area and transpiration intensity influence yield during water deficiency. In summary, the correlation among morpho-physiologic, metabolic, and yield components significantly varied between the two cultivars under water deficiency. The flowering stage was sensitive to water stress for both cultivars. | 9 | Microstructural characteristics and mechanical behavior of B4Cp/6061Al composites synthesized at different hot-pressing temperatures显示文摘B4Cp/6061Al composites have become important structural and functional materials and can be fabricated by powder metallurgy and subsequent hot rolling. In this work, the effects of the hot-pressing temperature on microstructures and mechanical behaviors of the B4Cp/6061Al composites were investigated. The results showed that compared with the T4 heat treated B4Cp/6061Al composite hot pressed at 560℃, the yield strength and failure strain of the composites hot pressed at 580℃ were increased to 235 MPa and 18.4%, respectively. This was associated with the interface bonding strength between the B4C particles and the matrix. However, the reaction products, identified to be MgAl2O4 phases, were detected in the composites hot pressed at 600℃. The formation of the MgAl2O4 phases resulted in the Mg depletion, thus reducing the yield strength to 203.5 MPa after the T4 heat treatment due to the effect of the solid solution strengthening being weakened. In addition, the variation of hardness and electrical conductivity was mainly related to the Mg content in the matrix. | 10 | Combined detection and subclass characteristics analysis of CTCs and CTECs by SE-iFISH in ovarian cancer显示文摘Objective:Hematogenous metastasis is essential for the progression of ovarian cancer(OC),and circulating tumor cells(CTCs)are part of the metastatic cascade.However,the detection rate of CTC is low due to the use of less sensitive detection methods.Therefore,this study aimed to detect CTCs and circulating tumorigenic endothelial cells(CTECs)in patients with OC using subtraction enrichment and immunostaining and fluorescence in situ hybridization(SE-iFISH).Methods:We enrolled a total of 56 subjects,including 20 OC patients and 36 ovarian benign tumor patients.CTCs and CTECs were captured by subtraction enrichment(SE)and counted and classified according to immunofluorescence staining of tumor markers(TMs)carbohydrate antigen 125(CA125)and human epididymis protein 4(HE4)combined with fluorescence in situ hybridization(iFISH)of chromosome 8(Chr8)aneuploidy.The diagnostic value and subtype characteristics of CTCs and CTECs were investigated.Results:The detection rate of CTCs by SE-iFISH was high.Compared with CA125 and HE4,Chr8 aneuploidy was the major identification feature of CTC.CTC counts in OC were statistically higher than those in benign groups.CTC and CTEC with≥pentaploidy were detected in both groups,illustrating the poor diagnostic value of CTC or CTEC.Distributions of triploid and tetraploid CTC subtypes were significantly different,and combined detection of triploid and tetraploid CTCs showed the best diagnostic value.In contrast,the distribution of CTECs in the OC and benign groups had no statistically significant difference.Small CTCs accounted for over 1/3 of the total CTC count.We also found that small CTCs and CTECs primarily comprised triploid cells,while large CTCs and CTECs mainly comprised pentaploidy and beyond.Conclusions:The application of SE-iFISH offered a more comprehensive understanding of heterogeneous CTCs and CTECs in OC.Analysis of subclass characteristics of the CTCs and CTECs according to Chr8 aneuploidy and cell size may broaden their potential clinical utility and deepen mechanistic studies in OC. | Hongyan Cheng Shang Wang Wenqing Luan Xue Ye Sha Dou Zhijian Tang Honglan Zhu Peter Ping Lin Yi Li Heng Cui Xiaohong Chang | 2021 | Chinese Journal of Cancer Research2021,33,2: | 3 |

| 12 | Yizhining elixation improves sustained attention deficit and impulsiveness of spontaneous hypertensive rats显示文摘BACKGROUND: The core symptoms of attention deficit, impulsiveness, hyperactivity of attention deficit hyperactivity disorders (ADHD) can concurrently present in spontaneous hypertensive rats. Neuropathological, biochemical and pharmacological studies also support it. Spontaneous hypertensive rats are one kind of best ADHD animal models, which has been accepted at present. Yizhining elixation can reinforce kidney and calm liver, strengthen spleen and nourish heart, benefit intelligence and focus attention, but its therapeutic effect on treatment of ADHD needs to be further investigated. OBJECTIVE: To observe the effect of Yizhining elixation on the persistent active attention deficit and impulsiveness of rats with spontaneous hypertension. DESIGN: Observational and controlled analysis. SETTING: Department of Pediatrics, the First Affiliated Hospital of Guangzhou University of Chinese Medicine. MATERIALS: Thirty-six male rats with spontaneous hypertension, of clean grade, were involved in this experiment. The involved rats were provided by Shanghai SLAC Laboratory Animal Co., Ltd., and they did not undergo any experiments; Yizhining elixation was composed by Guiban, Shenglonggu, Yuanzhi, Shichangpu, Yejiaoteng, Shudi, Dangshen, Yunling, Fuxiaomai, Wueizi, etc. Crude drug, provided by Outpatient Dispensary, Affiliated Hospital of Guangzhou University of Chinese Medicine, was prepared into water elixation, which contained crude drug of 1.154 g/mL. METHODS: This experiment was carried out in the Department of Pediatrics, First Affiliated Hospital of Guangzhou University of Chinese Medicine between March and July 2005. The involved rats were randomized into 3 groups: Yizhining high dose group, Yizhining low dose group and blank control group, with 12 rats in each. Changes in sustained attention and impulsiveness of rats were detected before and after administration of Yizhining elixation by '5-choice serial response time task' method. Following above-mentioned training and detection, rats in each group were daily drenched with Yizhining elixation once for 14 days successively. Rats in the Yizhining high-dose and low-dose groups were drenched with 12.6 mL/kg and 6.3 mL/kg Yizhining elixation (diluted to 25 mL/kg) respectively, and those in the blank control group were drenched with 12.5 mL/kg normal saline. On the 14th day after administration, the ratios of correct and immature response of rats were re-detected for 3 days successively by formal detection method for later analysis. The sustained attention of rats was evaluated with ratio of correct response: Ratio of correct response=[times of correct response/(times of correct response + times of incorrect response)]; impulsiveness was evaluated with the ratio of immature response: ratio of immature response=[times of immature response/ (times of correct response+times of incorrect response+times of omission+times of immature response)]. MAIN OUTCOME MEASURES: The effects of different doses of Yizhining elixation on the sustained attention (ratio of correct response) and impulsiveness (ratio of immature response) of spontaneous hypertensive rats. RESULTS: Thirty-six spontaneous hypertensive rats were involved in the final analysis. The ratio of correct response of rats in the Yizhining high dose and low dose groups after administration was (98.9±2.30)% and (96.6±4.00)%, respectively, which was significantly higher than that before administration [(80.4±11.5)%, (83.9±8.90)%,P < 0.01] and that in the blank control group[(83.3±6.30)%,P < 0.01]. The ratio of immature response of rats in the Yizhining high dose and low dose groups after administration was (5.04±1.04)%, (7.23±1.22)%, respectively, which was lower than that before administration [(9.85±2.50)%,(9.40±1.73)%,P < 0.05] and that in the blank control group [(9.42±1.64)%,P < 0.01]. | 13 | Desiccation tolerance in bryophytes: the rehydration proteomes of Bryum argenteum provide insights into the resuscitation mechanism显示文摘Bryum argenteum Hedw. is a desiccation tolerant bryophyte and belongs to one of the most important components of the biological soil crusts(BSCs) found in the deserts of Central Asia. Limited information is available on rehydration-responsive proteins in desiccation tolerant plants. As a complement to our previous research analyzing the rehydration transcriptome, we present a parallel quantitative proteomic effort to study rehydration-responsive proteins. Bryophyte gametophores were desiccated(Dry) and rehydrated for 2 h(R2) and 24 h(R24). Proteins from Dry, R2 and R24 gametophores were labeled by isobaric tags for relative and absolute quantitation(iTRAQ) to determine the relative abundance of rehydration-responsive proteins. A total of 5503 non-redundant protein sequences were identified and 4772(86.7%) protein sequences were annotated using Gene Ontology(GO) terms and Pfam classifications. Upon rehydration 239 proteins were elevated and 461 proteins were reduced as compared to the desiccated protein sample. Differentially up-regulated proteins were classified into a number of categories including reactive oxygen species scavenging enzymes, detoxifying enzymes, Late Embryogenesis Abundant(LEA) proteins, heat shock proteins, proteasome components and proteases, and photosynthesis and translation related proteins. | 14 | Thermal and elemental analysis of the combustion chamber deposits in a large-scale two-stroke marine diesel engine显示文摘It is of significance to understand the chemical content of carbon deposits in the large-scale two-stroke(LSTS) marine diesel engine because of adverse effect on the engine performance, oil consumption and emissions. In this work, two different combustion chamber deposits in an LSTS marine diesel engine were studied using thermogravimetry analysis(TGA), elemental analysis(EA) and synchrotron X-ray fluorescence(SXRF). One was on the piston top and the other on the piston land, termed PTCD and PLCD, respectively. For the PTCD sample, the 97% residue in the TGA and 1.4% carbon content in the EA indicated the main compositions of PTCD were metal salts or oxides and ashes, significantly different from the previous findings of the highest carbon content in deposits from the small four stroke engines. The different chemical content between PTCD and PLCD implied higher thermal load in the LSTS marine diesel engine led to a nearly complete thermal decomposition of PTCD. The higher calcium content in PTCD and PLCD indicated the additives of cylinder oil should be the main source of metal content of PTCD and PLCD. Calcium distribution in the SXRF results was indicative of the potential layered structure in PTCD and PLCD. | 18 | Cytokine nanosponges suppressing overactive macrophages and dampening systematic cytokine storm for the treatment of hemophagocytic lymphohistiocytosis显示文摘Hemophagocytic lymphohistiocytosis(HLH)is a highly fatal condition with the positive feedback loop between continued immune cell activation and cytokine storm as the core mechanism to mediate multiple organ dysfunction.Inspired by macrophage membranes harbor the receptors with special high affinity for proin-flammation cytokines,lipopolysaccharide(LPS)-stimulated macrophage membrane-coated nanoparticles(LMNP)were developed to show strong sponge ability to both IFN-γand IL-6 and suppressed overactivation of macrophages by inhibiting JAK/STAT signaling pathway both in vitro and in vivo.Besides,LMNP also efficiently alleviated HLH-related symptoms including cytopenia,hepatosplenomegaly and hepatorenal dysfunction and save the life of mouse models.Furthermore,its sponge effect also worked well for five human HLH samples in vitro.Altogether,it’s firstly demonstrated that biocompatible LMNP could dampen HLH with high potential for clinical transformation,which also provided alternative insights for the treatment of other cytokine storm-mediated pathologic conditions such as COVID-19 infection and cytokine releasing syndrome during CAR-T therapy. | Honglan Wang Huiwen Liu Jia Li Chunying Liu Hui Chen Junying Li Chunyan Sun Tao Guo Zhiqing Pang Bo Zhang Yu Hu | 2023 | Bioactive Materials2023,,3: | 1 |
| 19 | Effects of the dispersed droplet sizes on the critical behavior of pseudobinary microemulsion显示文摘The critical behavior of pseudobinary microemulsion systems{water/sodium di(2-ethylhexyl)sulfo- succinate(AOT)/n-decane}with various droplet sizes was studied by measurements of refractive index. | 20 | Dietary fat intake and liver cancer risk: A prospective cohort study in Chinese women显示文摘Objective:This study aimed to determine whether dietary fat intake increased liver cancer risk in Chinese women from a prospective population-based cohort.Methods:A total of 72,704 Chinese women were followed up from the time of baseline recruitment(1996–2000)to the end of 2016.Dietary fat intake was calculated using a validated food frequency questionnaire.The Cox regression model was used to assess the hazard ratio(HR)and 95%confidence intervals(CI)for dietary fat intake and liver cancer risk.Results:We identified 252 incident liver cancer cases out of 1,267,845 person-years during the overall follow-up time.Null associations,neither in quartiles nor per standard deviation(SD)increment,were detected between liver cancer risk and dietary total fat,fat subtypes and subtype ratios,and food sources.The HR(95%CI)of the 1-SD increment was 1.03(0.90–1.17)for total fat,1.06(0.93–1.20)for saturated fat,1.06(0.93–1.21)for monounsaturated fat,and 1.00(0.89–1.13)for polyunsaturated fat.Similar null associations were observed in stratification analyses according to body mass index and menopausal status.Conclusions:In our prospective cohort study,no significant association was observed in Chinese women between dietary fat and liver cancer risk,and in stratification and sensitivity analyses. | Xiaowei Ji Jing Wang Zhuoying Li Qiuming Shen Jiayi Tuo Jinghao Bi Yuting Tan Honglan Li Yongbing Xiang | 2022 | Cancer Biology & Medicine2022,19,3: | 1 |
