Commissioning progress of the FAST显示文摘The Five-hundred-meter Aperture Spherical radio Telescope(FAST) was completed with its main structure installed on September 25, 2016, after which it entered the commissioning phase. This paper aims to introduce the commissioning progress of the FAST over the past two years. To improve its operational reliability and ensure effective observation time, FAST has been equipped with a real-time information system for the active reflector system and hierarchical commissioning scheme for the feed support system, which ultimately achieves safe operation of the two systems. For meeting the high-performance indices, a highprecision measurement system was set up based on the effective control methods that were implemented for the active reflector system and feed support system. Since the commissioning of the FAST, a low-frequency ultra-wideband receiver and 19-beam1.05-1.45 GHz receiver have been mainly used. Telescope efficiency, pointing accuracy, and system noise temperature were completely tested and ultimately achieved the acceptance indices of the telescope. The FAST has been in the process of national acceptance preparations and has begun to search for pulsars. In the future, it will still strive to improve its capabilities and expand its application prospects. | Peng Jiang YouLing Yue HengQian Gan Rui Yao Hui Li GaoFeng Pan JingHai Sun DongJun Yu HongFei Liu NingYu Tang Lei Qian JiGuang Lu Jun Yan Bo Peng ShuXin Zhang QiMing Wang Qi Li Di Li | 2019 | Science China(Physics,Mechanics & Astronomy)2019,62,5: | 11
Resveratrol induces AMPK and mTOR signaling inhibition-mediated autophagy and apoptosis in multiple myeloma cells显示文摘Resveratrol,a natural compound extracted from the skins of grapes, berries,or other fruits, has been shown to have anti-tumor effects against multiple myeloma(MM)via promoting apoptosis and inhibiting cell viability. In addition to apoptosis, autophagy also plays a significant role in anti-tumor effects. However, whether autophagy is involved in anti-MM activity of resveratrol remains unclear. In this study, human MM cell lines U266,RPMI-8226,and NCI-H929 were treated with resveratrol.Cell Counting Kit-8 assay and colony formation assay were used to measure cell viability.Western blot analysis was used to detect apoptosis-and autophagy-associated proteins.3-Methyladenine(3-MA)was applied to inhibit autophagy. Results showed that resveratrol inhibited cell viability and colony formation via promoting apoptosis and autophagy in MM cell lines U266,RPMI-8226,and NCI-H929.Resveratrol promoted apoptosis-related proteins,Caspase-3 activating poly-ADP-ribose polymerase and Caspase-3 cleavage, and decreased the protein level of Survivin in a dose-dependent manner.Additionally, resveratrol upregulated the levels of LC3 and Beclin1 in a dose-dependent way, indicating that autophagy might be implicated in anti-MM effect of resveratrol.Furthermore,3-MA relieved the cytotoxicity of resveratrol by blocking the autophagic flux.Resveratrol increased the phosphorylation of adenosine monophosphate(AMP)-activated protein kinase and decreased the phosphorylation of mammalian target of rapamycin(mTOR)and its downstream substrates p70S6K and 4EBP1 in a dose-dependent manner, leading to autophagy. Therefore, our results suggest that resveratrol exerts anti-MM effects through apoptosis and autophagy, which can be used as a new therapeutic strategy for MM in clinic. | Ruye Ma Dandan Yu Yu Peng Hongfei Yi Yingcong Wang Taofang Cheng Bingqing Shi Guang Yang Weiming Lai Xiaosong Wu Ye Lu Jumei Shi | 2021 | Acta Biochimica et Biophysica Sinica2021,53,6: | 7
A newly discovered role ofmetabolic enzyme PCK1 as a protein kinase to promote cancer lipogenesis显示文摘Lipid metabolism,in particular fatty acid and cholesterol synthesis,is essential to convert nutrients into metabolic intermediates for membrane biosynthesis, energy storage and the generation of signaling molecules.Tumor cells maintain high level of lipid metabolism for rapid cell proliferation[1-4].Transcription of genes required for fatty acid and cholesterol synthesis and cholesterol uptake is controlled by membrane-bound transcription factor sterol regulatory element-binding proteins(SREBPs),including SREBP-1a,SREBP-1c/ADD1 and SREBP-2 isoforms[5].The function of SREBPs is mainly regulated by an escort protein(the SREBP cleavage-activating protein[SCAP])and endoplasmic reticulum(ER)anchor proteins(insulininduced genes[Insigs]),during the feedback loop of cholesterol synthesis[6-8].Under sterol-depleted conditions,SCAP and SREBPs complex is captured by COPIImediated vesicles and transported from the ER to the Golgi apparatus[7,9],where the SREBPs are proteolytically processed by Site-1 protease(S1P)and Site-2 protease(S2P)to yield active amino-terminal fragments that enter the nucleus for gene transcription[10,11].Under high intracellular sterol conditions, abundant cholesterol in the ER membrane binds to SCAP,induces its conformational change, and enables it to bind to Insigs. | Hongfei Jiang Lei Zhu Daqian Xu Zhimin Lu | 2020 | Cancer Communications2020,40,9: | 6
Cancer metabolism and tumor microenvironment:fostering each other?显示文摘The changes associated with malignancy are not only in cancer cells but also in environment in which cancer cells live.Metabolic reprogramming supports tumor cells’high demand of biogenesis for their rapid proliferation, and helps tumor cells to survive under certain genetic or environmental stresses.Emerging evidence suggests that metabolic alteration is ultimately and tightly associated with genetic changes,in particular the dysregulation of key oncogenic and tumor suppressive signaling pathways.Cancer cells activate HIF signaling even in the presence of oxygen and in the absence of growth factor stimulation. This cancer metabolic phenotype, described firstly by German physiologist Otto Warburg, ensures enhanced glycolytic metabolism for the biosynthesis of macromolecules. The conception of metabolite signaling,i.e.,metabolites are regulators of cell signaling, provides novel insights into how reactive oxygen species(ROS)and other metabolites deregulation may regulate redox homeostasis, epigenetics, and proliferation of cancer cells. Moreover, the unveiling of noncanonical functions of metabolic enzymes, such as the moonlighting functions of phosphoglycerate kinase 1(PGK1),reassures the importance of metabolism in cancer development. The metabolic, microRNAs,and ncRNAs alterations in cancer cells can be sorted and delivered either to intercellular matrix or to cancer adjacent cells to shape cancer microenvironment via media such as exosome.Among them, cancer microenvironmental cells are immune cells which exert profound effects on cancer cells.Understanding of all these processes is a prerequisite for the development of a more effective strategy to contain cancers. | Yiyuan Yuan Huimin Li Wang Pu Leilei Chen Dong Guo Hongfei Jiang Bo He Siyuan Qin Kui Wang Na Li Jingwei Feng Jing Wen Shipeng Cheng Yaguang Zhang Weiwei Yang Dan Ye Zhimin Lu Canhua Huang Jun Mei Hua-Feng Zhang Ping Gao Peng Jiang Shicheng Su Bing Sun Shi-Min Zhao | 2022 | Science China(Life Sciences)2022,65,2: | 5
Synthesis of MoX2 (X =Se or S) monolayers with high-concentration 1T'phase on 4H/fcc-Au nanorods for hydrogen evolution显示文摘Controlled synthesis of transition metal dichalcogenide (TMD) monolayers with unusual crystal phases has attracted increasing attention due to their promising applications in electrocatalysis. However, the facile and large-scale preparation of TMD monolayers with high-concentration unusual crystal phase still remains a challenge.Herein,we report the synthesis of MoX2 (X =Se or S) monolayers with high-concentration semimetallic 1T'phase by using the 4H/face-centered cubic (fcc)-Au nanorod as template to form the 4H/fcc-Au@MoX2 nanocomposite. The concentrations of 1T'phase in the prepared MoSe2 and MoS2 monolayers are up to 86% and 81%,respectively.As a proof-of-concept application, the obtained Au@MoS2 nanocomposite is used for the electrocatalytic hydrogen evolution reaction (HER) in acid medium, exhibiting excellent performance with a low overpotential of 178 mV at the current density of 10 mNcm^2,a small Tafel slope of 43.3 mV/dec,and excellent HER stability. This work paves a way for direct synthesis of TMD monolayers with high-concentration of unusual crystal phase for the electrocatalytic application. | Zhengqing Liu Xiao Zhang Yue Gong Qipeng Lu Zhicheng Zhang Hongfei Cheng Qinglang Ma Junze Chen Meiting Zhao Bo Chen Ye Chen Xue-Jun Wu Pengfei Yin Lin Gu Yaping Du Hua Zhang | 2019 | Nano Research2019,12,6: | 3
Antidiabetic effect of an engineered bacterium Lactobacillus plantarum-pMG36e-GLP-1 in monkey model显示文摘Glucagon-like peptide-1(GLP-1)reduces postprandial hyperglycaemia, but its short half-life inhibits clinical application. The aim of the current study was to evaluate the treatment efforts of an engineered strain,Lactobacillus plantarum-pMG36e-GLP-1(L.plantarum-pMG36e-GLP-1),that continuously expresses GLP-1 in spontaneous type 2 diabetes mellitus(T2DM)monkeys.After 7 weeks of oral supplementation with L.plantarum-pMG36e-GLP-1,the fasting blood glucose(FPG)of monkeys was significantly(p<0.05)reduced to a normal level and only a small amount of weight was lost. The results of metagenomic sequencing showed that L.plantarum-pMG36e-GLP-1 caused a substantial(p<0.05)reduction in the intestinal pathogen Prevotella and marked enhancement of butyrate-producing Alistipes genera.According to the functional analysis using Kyoto Encyclopaedia of Genes and Genomes(KEGG)pathways,19 metabolism-related pathways were significantly enriched in T2DM monkeys after treatment with L.plantarum-pMG36e-GLP-1.LC-MS faecal metabolomics analysis found 41 significant differential metabolites(11 higher and 30 lower)in monkeys after treatment pathways linked to the metabolism of cofactors and vitamins were the most relevant. The present study suggests that L.plantarum-pMG36e-GLP-1 had an impact on the gut microbial composition and faecal metabolomic profile in spontaneous T2DM monkeys and may be a novel candidate for diabetes treatment. | Jie Luo Hongfei Zhang Jiachen Lu ChaoLin Ma Tingtao Chen | 2021 | Synthetic and Systems Biotechnology2021,6,4: | 3
Effect of heat treatment on the microstructure of a Ni–Fe based superalloy for advanced ultra-supercritical power plant applications显示文摘The effect of heat treatment on the microstructure and microhardness of a Ni–Fe based superalloy for700 °C advanced ultra-supercritical coalfi red power plants was investigated. Results showed that the main phases in the alloy were γ, γ′, MC and M_23C_6, and no harmful phase was observed in the alloy.M_23C_6-type carbides discretely distributed nearby grain boundaries as the alloy was aged at above840 °C. The microhardness decreased with increasing aging temperature. The coarsening of γ′ led to the increment of microhardness at 780 °C and 810 °C for a short aging time, and a signi fi cant decrease in microhardness after aging at 840 °C. The aging temperature had more signi fi cant role on the microstructure than holding time. Therefore, to obtain optimum strengthening effect for this alloy, the aging temperature should not exceed 810 °C. | Xinbao Zhao Yingying Dang Hongfei Yin Jintao Lu Yong Yuan Zhen Yang Jingbo Yan Yuefeng Gu | 2016 | Progress in Natural Science:Materials International2016,26,2: | 2
Highly efficient perovskite solar cells based on symmetric hole transport material constructed with indaceno[1,2-b:5,6-b’]dithiophene core building block显示文摘Two novel hole transport materials (HTMs) with indaceno[1,2-b:5,6-b’]dithiophene (IDT) as core building blocks, termed IDT1 and IDT2,were designed and synthesized. The side alkyl chains were introduced to regulate and control the morphology and stacking behavior of HTMs,and the peripheral triarylamine arms were introduced to adjust the energy levels and to facilitate efficient hole transport.Applied in mesoporous structured perovskite solar cells (PSCs),HTM IDT1 achieved higher power conversion efficiency (PCE,19.55%) and better stability than Spiro-OMeTAD (19.25%) and IDT2 (15.77%) based PSC.These results suggest the potential of IDTl as a promising HTM for PSCs. | Cheng Wu Cheng Chen Li Tao Xingdong Ding Mengmeng Zheng Hongping Li Gongqiang Li Hongfei Lu Ming Cheng | 2020 | Journal of Energy Chemistry2020,29,4: | 2
Gut,metabolism and nutritional Support for COVID-19:Experiences from China显示文摘There is little research that focuses on the relationship between the gut, metabolism, nutritional support and COVID-19.As a group of Chinese physicians, nutritionists and scientists working on the frontline treating COVID-19 patients,we aim to integrate our experiences and the current clinical evidence to address this pressing issue in this article.Based on our clinical observations and available evidence,we recommend the following practice.Firstly, the Nutritional Risk Screening 2002 tool should be used routinely and periodically;for patients with a score≥3,oral nutritional supplements should be given immediately.Secondly, for patients receiving the antiviral agents lopinavir/ritonavir,gastrointestinal side effects should be monitored for and timely intervention provided.Thirdly, for feeding, the enteral route should be the first choice. In patients undergoing mechanical ventilation, establishing a jejunal route as early as possible can guarantee the feeding target being achieved if gastric dilatation occurs.Fourthly,we suggest a permissive underfeeding strategy for severe/critical patients admitted to the intensive care unit during the first week of admission, with the energy target no more than 20 kcal/kg/day(for those on mechanical ventilation, this target may be lowered to 10–15 kcal/kg/day)and the protein target around 1.0–1.2 g/kg/day.If the inflammatory condition is significantly alleviated, the energy target may be gradually increased to 25–30 kcal/kg/day and the protein target to 1.2–1.5 g/kg/day.Fifthly,supplemental parenteral nutrition should be used with caution.Lastly,omega-3 fatty acids may be used as immunoregulators, intravenous administration of omega-3 fatty emulsion(10 g/day)at an early stage may help to reduce the inflammatory reaction. | Ting Yuanand Wei Chen Hua Jiang Jian-Cheng Zhang Jun Zeng Lu Wang Yu Wang Charles Damien Lu Lei Deng Hongfei Deng Kai Wang Ming-Wei Sun Ping Zhou Ting Yuan Wei Chen | 2020 | Burns & Trauma2020,8,1: | 2
