| 1 | Tumor metastasis and the reciprocal regulation of heparanase gene expression by nuclear factor kappa B in human gastric carcinoma tissue显示文摘AIM: To investigate whether NF-κB is activated in human gastric carcinoma tissues and, if so, to study whether there is any correlation between NF-κB activity and heparanase expression in gastric carcinoma.METHODS: NF-κB activation was assayed by immunohistochemical staining in formalin-fixed, paraffin-embedded specimens from 45 gastric carcinoma patients. Electrophoretic mobility shift assay (EMSA) method was used for nuclear protein from these fresh tissue specimens. Heparanase gene expression was quantified using quantitative RT-PCR.RESULTS: The nuclear translocation of RelA (marker of NF-κB activation) was significantly higher in tumor cells compared to adjacent and normal epithelial cells [(41.3±3.52)% vs (0.38±0.22) %, t= 10.993, P= 0.000<0.05; (41.3±3.52)%vs (0±0.31)%, t = 11.484, P = 0.000<0.05]. NF-κB activation was correlated with tumor invasion-related clinicopathological features such as lymphatic invasion,pathological stage, and depth of invasion (Z = 2.148,P= 0.032<0.05; χ2 = 8.758, P= 0.033<0.05; χ2 = 18.531,P = 0.006<0.05). NF-κB activation was significantly correlated with expression of heparanase gene (r= 0.194,P= 0.046<0.05).CONCLUSION: NF-κB RelA (p65) activation was related with increased heparanase gene expression and correlated with poor clinicopathological characteristics in gastric cancers. | 3 | Cryopreservation of cynomolgus monkey (Macacafascicularis)spermatozoa in a chemically defined extender显示文摘Aim: To establish a method for cynomolgus monkey sperm cryopreservation in a chemically defined extender.Methods'' Semen samples were collected by electro-ejaculation from four sexually mature male cynomolgus monkeys.The spermatozoa were frozen in straws by liquid nitrogen vapor using egg-yolk-free Tes-Tris (mTTE) synthetic extender and glycerol as cryoprotectant. The effects of glycerol concentration (1%, 3%, 5%, 10% and 15% [v/v]) and its equilibration time (10min, 30min, 60min and 90min) on post-thaw spermatozoa were examined by sperm motility and sperm head membrane integrity. Results: The post-thaw motility and head membrane integrity of spermatozoa were significantly higher (P<0.05) for 5% glycerol (42.95±2.55 and 50.39±2.42, respectively) than those of the other groups (1%: 19.19±3.22 and 24.84±3.64; 3%: 34.23±3.43 and 41.37±3.42; 10%:15.68±2.36 and 21.39±3.14; 15%: 7.47±1.44 and 12.90±2.18). The parameters for 30min equilibration (42.95±2.55 and 50.39±2.42) were better (P<0.05) than those of the other groups (10min: 31.33±3.06 and 38.98±3.31; 60min: 32.49±3.86 and 40.01±4.18; 90min: 31.16±3.66 and 38.30±3.78). Five percent glycerol and 30min equilibration yielded the highest post-thaw sperm motility and head membrane integrity. | 4 | Analysis of Synonymous Codon Usage Bias in Chlamydia显示文摘Chlamydiae are obligate intracellular bacterial pathogens that cause ocular and sexually transmitted diseases, and are associated with cardiovascular diseases. The analysis of codon usage may improve our understanding of the evolution and pathogenesis of Chlamydia and allow reengineering of target genes to improve their expression for gene therapy. Here, we analyzed the codon usage of C. muridarum, C. trachomatis (here indicating biovar trachoma and LGV), C. pneumoniae, and C. psittaci using the codon usage database and the CUSP (Create a codon usage table) program of EMBOSS (The European Molecular Biology Open Software Suite). The results show that the four genomes have similar codon usage patterns, with a strong bias towards the codons with A and T at the third codon position. Compared with Homo sapiens, the four chlamydial species show discordant seven or eight preferred codons. The ENC (effective number of codons used in a gene)-plot reveals that the genetic heterogeneity in Chlamydia is constrained by the G+C content, while translational selection and gene length exert relatively weaker influences. Moreover, mutational pressure appears to be the major determinant of the codon usage variation among the chlamydial genes. In addition, we compared the codon preferences of C. trachomatis with those of E. coli, yeast, adenovirus and Homo sapiens. There are 23 codons showing distinct usage differences between C. trachomatis and E. coli, 24 between C. trachomatis and adenovirus, 21 between C. trachomatis and Homo sapiens, but only six codons between C. trachomatis and yeast. Therefore, the yeast system may be more suitable for the expression of chlamydial genes. Finally, we compared the codon preferences of C. trachomatis with those of six eukaryotes, eight prokaryotes and 23 viruses. There is a strong positive correlation between the differences in coding GC content and the variations in codon bias (r=0.905, P<0.001). | 5 | Optimization design of prestressed concrete wind-turbine tower显示文摘Wind energy is a clean and renewable energy for which technology has developed rapidly in recent years.Wind turbines are commonly supported on tubular steel towers.As the turbine size is growing and the towers are rising in height,steel towers are required to be sufficiently strong and stiff,consequently leading to high construction costs.To tackle this problem,a new type of prestressed concrete tower was designed employing a novel tower concept having a regular octagon cross section with interior ribs on each side,which was optimized by comparing the natural frequency and stress difference under the same lateral load in different directions of the tower.The designed tower features a tapered profile that reduces the area subjected to wind;the tapered profile reduces the total weight,applied moment and the capital cost.An optimization method was developed employing ABAQUS software and a genetic algorithm.A target function was defined on the basis of the minimum cost of the concrete and prestressed tendon used,and constraints were applied by accounting for the stress,displacements and natural frequency of the tower.Employing the method,a 100 m prestressed concrete tower system for a 5 MW turbine was optimized and designed under wind and earthquake loads.The paper also reports a systematic design procedure incorporating the finite element method and the optimization method for the prestressed concrete wind-turbine towers. | MA HongWang MENG Ran | 2014 | Science China(Technological Sciences)2014,57,2: | 6 |

| 7 | In situ fabrication of organic electrochemical transistors on a microfluidic chip显示文摘Microfluid chips integrating with organic electrochemical transistors (OECTs) are useful for manufacturing biosensors with high throughput and large-scale analyses. We report here the utilization of alternating current (AC) electrodeposition to fabricate OECTs in situ on a microfiuid chip. With this method, the organic semiconductor (OS) layer with a channel length of 8μm was readily prepared without requiring the postbonding process in the conventional construction of microfluidic chips. Poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate)/graphene quantum dots (PEDOT:PSS/GQDs) composites with different morphologies, such as microfilms, nanodendrites and nanowires were electropolymerized. The mass transfer process of the electropolymerization reaction was evidenced to be diffusion limited. Morphologies, growth directions, and chemical structures of OS layers could be tuned by the amplitude and the frequency of the AC voltage. Transfer and output characteristic curves of OECTs were measured on the microfluidic chip. | 10 | FLUVIAL PROCESSES IN THE LOWER JINGJIANG RIVER:IMPACT OF THE THREE GORGES RESERVOIR IMPOUNDMENT显示文摘Sediment supply to the lower Jingjiang River will be subject to substantial reduction after the impoundment of the Three Gorges Reservoir,which could result in an excess of carrying capacity and serious bank erosions in the downstream alluvial channel,threatening the bank protection works and the safety of the Jingjiang Dyke. This paper presents a summary of research works concerning the fluvial processes in the lower Jingjiang River and the possible impact of the Three Gorges Reservoir impoundment on the variation of its channel pattern. Three different predictions have been put forward by researchers:1) the Jingjiang River will evolve towards a more sinuous,meandering channel pattern,with extensive bank erosion taking place along the river; 2) the river channel will be straightened and broadened because no point bar can be formed due to reduced sediment supply while bank erosion develops in the concave bank,and 3) this river reach will maintain its present channel pattern without significant change,although the sinuosity may be slightly reduced,since:a) the Three Gorges Reservoir mainly intercept sediment particles with sizes larger than 0.025mm,and b) the complex interaction between the Yangtze River and the Dongting Lake helps to reduce the negative effect of channel erosion through certain self-adjusting mechanism in fluvial processes. Discrepancy between these predictions shows that further research efforts are needed to understand the impact of Three Gorges Reservoir operation on the downstream fluvial processes. | 12 | Pollen Morphology of Parnassia L. (Parnassiaceae) and Its Systematic Implications显示文摘The pollen morphology of 28 species of Parnassia L. was investigated with light microscopy and scanning electron microscopy (SEM). The shape of pollen grains in this genus varies from subspheroidal to prolate in equatorial view and is three-lobed circular in the polar view. Pollen grains are usually radially symmetrical, isopolar, tricolporate or syntricolporate, with reticulate sculpture. The pollen characteristics among species are fairly similar to each other. Morphological information regarding the pollen grains shows that Parnassia is a natural genus. Based on exine ornamentation observed under SEM, three types of pollen grains were recognized: (i) type Ⅰ, with foveolate-reticulate sculpture; (ii) type Ⅱ, with a finely reticulate sculpture; and (iii) type Ⅲ, with a coarsely reticulate sculpture. Most sections of this genus have one type of sculpture of pollen morphology, but Sect. Nectarotrilobos has three types of sculpture and Sect.Saxifragastrum has two types of sculpture. | 13 | Low temperature impact toughness and fracture mechanism of cast QT400-18L ductile iron with diferent Ni additions显示文摘Different contents of Ni(0.3wt.%to 1.2wt.%)were added to the QT400-18L ductile iron to investigate the effect of Ni addition on the impact toughness of cast ductile irons at low temperatures.The impact toughnesses of the samples at room and low temperatures were tested.The microstructures and fractographs were observed.Results show that with the increase of Ni addition there is a general trend of refinement of the ferrite matrix while the nodule density shows no obvious change.When the Ni content is 0.7wt.%,the matrix structure is the refined ferrite with a very small fraction(about 2%)of pearlite near the eutectic cell boundaries.When the Ni content is further increased,the fraction of pearlite increases significantly and reaches more than 5%when 1.2wt.%Ni is added.The impact toughness at room temperature increases as the content of Ni increases from 0.3 wt.%to 0.7 wt.%,but decreases as the Ni content further increases to 1.2wt.%due to the increase of pearlite fraction.The maximum value of the impact work is 18.5 J at room temperature with 0.7wt.%Ni addition.The average value of the impact work is still more than 13 J even at-30℃.In addition,the fracture mechanism changes from ductile manner to brittleness as the testing temperature decreases from 20℃to-60℃. | Zhang Xinning Qu Yingdong Yang Hongwang Li Rongde | 2013 | China Foundry2013,10,5: | 2 |

However, it remains a challenge to efficiently induce ES cells to differentiate into neurons. Here, we show that murine ES cells can efficiently differentiate into neurons when cultured in glial cell- conditioned medium (GCM) under attaching conditions without the formation of embryoid bodies. In comparison with murine embryonic fibroblast-conditioned medium, we found that GCM has a positive effect on limiting the generation of non-neuronal cells, such as astrocytes. | 17 | IFN-stimulated P2Y13 protects mice from viral infection by suppressing the cAMP/EPACl signaling pathway显示文摘Among the most important sensors of extracellular danger signals, purinergic receptors have been demonstrated to play crucial roles in host defense against infection. However, the function of P2 receptors in viral infection has been little explored. Here we demonstrated that P2Y13 and its ligand ADP play an important role in protecting hosts from viral infections. First, we demonstrate that P2Y13, as a typical interferon-stimulated gene, is induced together with extracellular ADP during viral infection. Most importantly, extracellular ADP restricts the replication of different kinds of viruses, including vesicular stomatitis virus, Newcastle disease virus, herpes simplex virus 1, and murine leukemia virus. This kind of protection is dependent on P2Y13 but not P2Y1 or P2Y12, which are also considered as receptors for ADP. Furthermore, cyclic adenosine monophosphate and EPAC1 are downregu-lated by extracellular ADP through the P2Y13-coupled Gi alpha subunit. Accordingly, inhibition or deletion of EPAC1 significantly eliminates ADP/P2Y13-mediated antiviral activities.
