| 1 | Expression of inducible nitric oxide synthase and cyclooxygenase-2 in pancreatic adenocarcinoma:Correlation with microvessel density显示文摘AIM: Cydooxygenases (COX) are key enzymes for conversion of arachidonic acid to prostaglandins. Nitric oxide synthase (NOS) is the enzyme responsible for formation of nitric oxide.Both have constitutive and inducible isoforms. The inducible isoforms (iNOS and COX-2) are of great interest as regulators of tumor angiogenesis, tumorigenesis and inflammatory processes. This study was to clarify their role in pancreatic adenocarcinomas.METHODS: We investigated the immunohistochemical iNOS and COX-2 expression in 40 pancreatic ductal adenocarcinomas of different grade and stage. The results were compared with microvessel density and clinicopathological data.RESULTS: Twenty-one (52.5%) of the cases showed iNOS expression, 15 (37.5%) of the cases were positive for COX-2.The immunoreaction was heterogeneously distributed within the tumors. Staining intensity was different between the tumors. No correlation between iNOS and COX-2 expression was seen. There was no relationship with microvessel density.However, iNOS positive tumors developed more often distant metastases and the more malignant tumors showed a higher COX-2 expression. There was no correlation with other clinicopathological data.CONCLUSION: Approximately half of the cases expressed iNOS and COX-2. These two enzymes do not seem to be the key step in angiogenesis or carcinogenesis of pancreatic adenocarcinomas. | 2 | 传统DNA疫苗载体与Semliki森林病毒复制子对HIV-1 Pr55^(gag)表达与体液免疫原性的比较性研究显示文摘为探索Semliki森林病毒 (SFV)衍生的复制型DNA载体可否用于HIV疫苗的候选载体 ,对该载体与传统DNA疫苗载体对HIV - 1Pr5 5 gag的表达与体液免疫原性进行了系统比较研究。将野生型 (wtgag)及密码子改造(syngag)的HIV - 1ⅢBgag 基因分别克隆于SFVDNA载体及传统DNA疫苗载体 [pCDNA3 1(+) ],对其Pr5 5 gag细胞内表达水平、Pr5 5 gag病毒样颗粒释放、以及在BALB/c鼠的体液免疫原性进行了比较。在 2 93T、H12 99、C2C12和BHK细胞系中 ,SFV -wtgag可以Rev非依赖方式有效表达Pr5 5 gag ,而 pC -wtgag转染的细胞不能有效表达Pr5 5 gag,从而不能诱导小鼠产生免疫反应。虽然SFV质粒的细胞转化效率明显低于pCDNA载体 ,SFV -wtgag和SFV -syngag在细胞内Pr5 5 gag的表达量与 pC -syngag相似 ,而Pr5 5 gag病毒样颗粒的释放明显低于 pC -syngag。在肌内注射免疫的小鼠中 ,低剂量 (0 1和 1 0 μg)的SFV及 pCDNAgag表达质粒均未诱导出GAG特异性免疫反应。在高剂量 (10 ,30 ,10 0 μg)免疫组中 ,与SFVgag表达质粒相比 ,pC -syngag可诱导出较高水平的TH1型GAG特异性抗体。SFV -syngag较SFV -wtgag可诱导出高水平的体液免疫反应。结果提示 ,SFV衍生的复制子单独使用不能在小鼠诱导出优于传统DNA疫苗载体的HIV - 1GAG特异性? | 张健慧 Jens Wild Kurt Bieler Marcus Graf Ludwig Deml Hans Wolf PeterLiljestrm Ralf Wagner 邵一鸣 | 2002 | 病毒学报2002，18，1： | 13 |
| 3 | Neurovascular complications due to the Hippocrates method for reducing anterior shoulder dislocations显示文摘In spite of the fact that the Hippocrates method hardly has been evaluated in a scientific manner and numerous associated iatrogenic complications have been reported, this method remains to be one of the most common techniques for reducing anterior shoulder dislocations. We report the case of a 69-year-old farmer under coumarin anticoagulant therapy who sustained acute first time anterior dislocation of his dominant right shoulder. By using the Hippocrates method with the patient under general anaesthesia, the brachial vein was injured and an increasing hematoma subsequently caused brachial plexus paresis by pressure. After surgery for decompression and vascular suturing, symptoms declined rapidly, but brachial plexus paresis still was not fully reversible after 3 mo of follow-up. The hazardousness of using the Hippocrates method can be explained by traction on the outstretched arm with force of the operator's body weight, direct trauma to the axillary region by the physician's heel, and the topographic relations of neurovascular structures and the dislocated humeral head. As there is a variety of alternative reduction techniques which have been evalu-ated scientifically and proofed to be safe, we strongly caution against the use of the Hippocrates method as a first line technique for reducing anterior shoulder dislocations, especially in elder patients with fragile vessels or under anticoagulant therapy, and recommend the scapular manipulation technique or the Milch technique, for example, as a first choice. | Markus Regauer Hans Polzer Wolf Mutschler | 2014 | World Journal of Orthopedics2014，5，1： | 7 |
| 4 | 云南瑞丽人免疫缺陷病毒感染者gp120基因C2-V3区的序列测定和亚型分析显示文摘经套式聚合酶链反应(nested-PCR)对17份1995年初采集于云南瑞丽市人免疫缺陷病毒1型(HIV-1)阳性静脉吸毒者外周血单核细胞(PBMCs)的核酸样品进行扩增,从17份样品中获得了HIV-1膜蛋白(env)基因的核酸片段,并对其C2-V3及邻区450个核苷酸序列进行了测定和分析。结果表明,17份样品中存在B和C两种亚型的HIV-1毒株序列,其亚型内的基因离散率分别为58%和22%,与A-E参考亚型及部分B和C亚型代表株序列相比较,属B亚型的12个毒株与包括泰国、缅甸及云南瑞丽代表株yn289在内的B亚型毒株序列十分接近,基因离散率在44%~49%的范围内;属C亚型的5个毒株则与主要代表印度C亚型毒株的共享序列ccon及瑞丽C亚型代表毒株yn272十分相似,其基因离散率均为19%。以上数据进一步确认我们的结论,即HIV-1在瑞丽的流行以B亚型毒株为主、C亚型的传人和流行时间较短。对B亚型毒株V3环序列的分析还发现,位于V3环顶端的四肽序中GPGQ占50%,GPGR则仅占25%,且编码其精氨酸(R)的密码子均为CGA而不是AGA。此结果与我们根据早期瑞丽HIV-1毒株序列研究结果得出的推测? | 管永军 陈钧 邵一鸣 赵全壁 曾毅 张家鹏 段一娟 Josef Kostler Hans Wolf | 1997 | 中华实验和临床病毒学杂志1997，11，1： | 5 |
