1Targeting ferroptosis suppresses osteocyte glucolipotoxicity and alleviates diabetic osteoporosis显示文摘Diabetic osteoporosis(DOP) is the leading complication continuously threatening the bone health of patients with diabetes. A key pathogenic factor in DOP is loss of osteocyte viability. However, the mechanism of osteocyte death remains unclear. Here, we identified ferroptosis, which is iron-dependent programmed cell death, as a critical mechanism of osteocyte death in murine models of DOP. The diabetic microenvironment significantly enhanced osteocyte ferroptosis in vitro, as shown by the substantial lipid peroxidation, iron overload, and aberrant activation of the ferroptosis pathway. RNA sequencing showed that heme oxygenase-1(HO-1) expression was notably upregulated in ferroptotic osteocytes. Further findings revealed that HO-1 was essential for osteocyte ferroptosis in DOP and that its promoter activity was controlled by the interaction between the upstream NRF2 and c-JUN transcription factors. Targeting ferroptosis or HO-1 efficiently rescued osteocyte death in DOP by disrupting the vicious cycle between lipid peroxidation and HO-1 activation, eventually ameliorating trabecular deterioration. 2Effects of flexibility and strength training on peak hamstring musculotendinous strains during sprinting显示文摘Background:Hamstring injury is one of the most common injuries in sports involving sprinting.Hamstring flexibility and strength are often considered to be modifiable risk factors in hamstring injury.Understanding the effects of hamstring flexibility or strength training on the biomechanics of the hamstring muscles during sprinting could assist in improving prevention strategies and rehabilitation related to these injuries.The purpose of this study was to determine the effects of altering hamstring flexibility or strength on peak hamstring musculotendinous strain during sprinting.Methods:A total of 20 male college students(aged 18-24 years)participated and were randomly assigned to either a flexibility intervention group or a strength intervention group.Each participant executed exercise training 3 times a week for 8 weeks.Flexibility,sprinting,and isokinetic strength testing were performed before and after the 2 interventions.Paired t tests were performed to determine hamstring flexibility or strength intervention effects on optimal hamstring musculotendinous lengths and peak hamstring musculotendinous strains during sprinting.Results:Participants in the flexibility intervention group significantly increased the optimal musculotendinous lengths of the semimembranosus and biceps long head(p<0.026)and decreased peak musculotendinous strains in all 3 bi-articulate hamstring muscles(p<0.004).Participants in the strength-intervention group significantly increased the optimal musculotendinous lengths of all 3 hamstring muscles(p<0.041)and significantly decreased their peak musculotendinous strain during sprinting(p<0.017).Conclusion:Increasing hamstring flexibility or strength through exercise training may assist in reducing the risk of hamstring injury during sprinting for recreational male athletes.Xianglin Wan Shangxiao Li Thomas M.Best Hui Liu Hanjun Li Bing Yu 2021Journal of Sport and Health Science2021,10,2:6

5Patterns of biomass,carbon,and nitrogen storage distribution dynamics after the invasion of pine forests by Bursaphelenchus xylophilus (Nematoda: Aphelenchoididae) in the three Gorges Reservoir Region显示文摘Masson pine stands infected by Pine wilt disease(PWD) in the Three Gorges Reservoir Region of central China were surveyed to quantify the immediate responses and subsequent trajectories of biomass,carbon(C),and nitrogen(N) in stand-level major ecosystem compartments.The biomasses of above-and belowground tree components,as well as of the understory,forest floor,and mineral soil(0–40 cm),were determined within each stand.C and N storage were also estimated for each ecosystem compartment.Overstory biomass decreased steadily with the extent of PWD infection.Understory biomass ranged from1.97 to 4.16 Mg ha^(-1),and the observed value for forest floor biomass was 12.89–22.59 Mg ha^(-1).The highest mean C and N concentrations were found in the stem bark and needles of Masson pine,respectively,while the lowest were found in the semi-to fully decomposed layer of the forest floor and stem wood of Masson pine,respectively.The C and N storage of aboveground trees,tree roots,and the aboveground ecosystem decreased with the extent ofPWD infection.However,the C and N contents of the understory,forest floor,and total mineral soil initially declined after PWD infection before recovering over the following several years.Those result concluded that the biomass,C,and N storage of different forest ecosystem compartments have experienced certain variations following the PWD epidemic.This is vital to understand the shifts in stand-level C and N allocation in PWD-damaged forest stands,as well as for predicting the responses of regional and global C and N cycling.Ruihe Gao Youqing Luo Zhuang Wang Hanjun Yu Juan Shi 2018Journal of Forestry Research2018,29,2:1

7Irisin Attenuates Osteoarthritis by Inhibiting Apoptosis of Osteocytes Through Activating Erk Signaling Pathway显示文摘Osteoarthritis(OA)is an inflammatory disease involving the joints that is prevalent in the global aging population.The purpose of this study is to determine whether irisin can attenuate osteoarthritis(OA)progression in anterior cruciate ligament transection(ACLT)mice models and the mechanism of irisin therapy effect on OA by increase the resistance of apoptosis in MLO-Y4 cells induced by mechanical stretch in vitro.Methods For in vivo study,3-month-old male C57BL/6 J mice were randomized to three groups,sham-operated,anterior cruciate ligament transection(ACLT)-operated treated with vehicle,and ACLT-operated treated with irisin by intraperitoneal injection once a week.Cartilage erosion was observed by HE staining.Osteoarthritis Research Society International(OARSI)scores were evaluated according to the safranin O stai-ning.The microstructure of tibia cortical bone,trabecular bone,and subchondral bone was analyzed by micro-CT and the bone histomorphometry has been administrated including mineral apposition rate(MAR).Edu staining and cck-8 were used for the detection of the proliferation of MLO-Y4 cells.For mechanical stress,cells were seeded on the collagen-I coated chamber subjected with a peak biaxial stretch of 20%at 1 Hz for 16 hours to induce apoptosis.Flow cytometry was used for the detection of apoptosis and cell cycle.TUNNEL was used for staining the apoptotic cells and rt-PCR was applied for quantifying the expression of mRNA such as Bax,Bcl-2,SOST,c-myc,Opg.Western blot was utilized to confirm the mechanism of how irisin decrease the osteocyte apoptosis.Results In vivo,irisin can attenuate articular cartilage degeneration.Irisin maintains the proportion of hyaline cartilage and calcified cartilage and keep fewer cartilage erosions in ACLT-operated mice.For immunohistochemical(IHC)staining,irisin reduced the expression of caspase3,Bax and matrix metalloproteinase-13 in both cartilage and subchondral bone.Irisin-treated ACLT group shows higher Trabecular number(Tb.N)and bone volume fraction(BV/TV)compared to the vehicle-treated ACLT group.In vitro, irisin significantly increased the proliferation of MLO-Y4 cells detected by Edu and Ki67 staining,and irisin can protect the cells from both mechanical stretchinduced apoptosis detected by FITC-PI flow cytometry and maintain the cell activity by regulating the expression of Bax,Bcl-2,and c-myc.Transcriptome sequencing shows that irisin significantly activates the MAPK signaling pathway and we confirm the result by western blot:irisin effectively activates the Erk signaling pathway through phosphorylation and has a certain activation effect on p38 signaling pathway,no activation was observed for FAK signaling pathway.Conclusions Irisin can attenuate the progression of OA by decrease the apoptosis of osteocyte,which can improve the microarchitecture of subchondral bone.Erk pathway activation plays an important role in reducing the apoptosis of osteocyte.Zihao He Hanjun Li Feng Zhou Jingke Du Shuhong Zhang Tingting Tang Zhifeng Yu 2019医用生物力学2019,34,A01:0
8Semiparametric Bayesian Inference for Accelerated Failure Time Models with Errors-in-Covariates and Doubly Censored Data显示文摘This paper proposes a Bayesian semiparametric accelerated failure time model for doubly censored data with errors-in-covariates. The authors model the distributions of the unobserved covariates and the regression errors via the Dirichlet processes. Moreover, the authors extend the Bayesian Lasso approach to our semiparametric model for variable selection. The authors develop the Markov chain Monte Carlo strategies for posterior calculation. Simulation studies are conducted to show the performance of the proposed method. 9A Randomized Controlled Trial of a Biodegradable Polymer,Microcrystalline Sirolimus-Eluting Stent(MiStent)versus Another Biodegradable Polymer Sirolimus-Eluting Stent(TIVOLI):The DESSOLVE-C Trial显示文摘Objective::Data comparing the outcomes of MiStent(Micell Technologies,Durham,North Carolina,USA)microcrystalline biodegradable polymer(BP)drug-eluting stent(DES)and those of another post-marketing BP-DES,TIVOLI(EssenTech,Beijing,China)are rare.This study sought to compare the angiographic efficacy and clinical outcomes of the microcrystalline BP sirolimus-eluting stent(SES)system MiStent and those of TIVOLI BP-SES.Methods::The DESSOLVE-C trial was a prospective,single-blinded,multicenter,randomized trial(NCT02448524),which randomly assigned patients with de novo coronary lesions to receive MiStent or TIVOLI BP-SES by a 1:1 ratio.The primary endpoint was a non-inferiority comparison of in-stent late lumen loss(LLL)by quantitative coronary angiography at 9 months.The secondary endpoint was device-related clinical cardiovascular composite events(target lesion failure(TLF),composite of cardiac death,target vessel myocardial infarction(MI),and clinically driven target lesion revascularization)and 1-year outcomes.Results::A total of 428 patients(216 patients in the MiStent group and 212 patients in the TIVOLI group)were enrolled and included in an intention-to-treat analysis.MiStent was not only non-inferior but superior to TIVOLI for in-stent LLL at 9 months((0.23±0.37)mm vs.(0.34±0.48)mm,P for non-inferiority<0.001,P for superiority=0.02).Although without significant difference,the rate of TLF in MiStent was quantitatively lower than that in TIVOLI(3.70%vs.6.60%;P=0.17).Conclusion::Compared with TIVOLI BP-SES,the MiStent system was superior in in-stent LLL at 9 months and had a comparable clinical benefit at 1 year in de novo coronary lesions.Bin Wang Sicong Ma Zhiyong Wang Li Zhang Hanjun Pei Yang Zheng Yuejin Yang Zheng Zhang Xinqun Hu Ziwen Ren Feng Zhang Changqian Wang Renqiang Yang Zhiming Yang Yuexi Wang Guosheng Fu Yu Cao Zuyi Yuan Kai Xu Xin Zhao Bo Xu Miaohan Qiu Quanmin Jing 2023Cardiology Discovery2023,3,1:0
10Achieving 20% photovoltaic efficiency by manganese doped methylammonium lead halide perovskites显示文摘We report a transition metal such as manganese doped methylammonium lead halide perovskite(MA(Pb:Mn)I_(3)) solar cell with an power conversion efficiency(PCE) over 20%. The rational design and fabrication of MA(Pb:Mn)I3 lead to the enhancements of all the photovoltaic parameters. To incorporate Mn can effectively eliminate the trap-assist and bi-molecular recombination. The photo-absorption ability at shorter wavelengths(i.e., less than 500 nm) and charge carrier lifetime can be elaborated. More importantly, the existence of the Mn^(2+)-I~--Mn^(3+)motif contributes for the double exchange effect, giving rise to the charge/spin transport. By a combination of linearly and circularly polarized photo-excitations, we have explicitly determined the role of intrinsic spin–orbit coupling(SOC) in MA(Pb:Mn)I_(3). More dark states are expected to be available for the photocurrent generation. 11Thermoelectric properties of tungsten ceramics prepared from nanopowder precursors显示文摘Tungsten trioxide ceramics were prepared from nm-sized powder precursors. Thermoelectric properties of samples were investigated. The results demonstrated that ceramics prepared in this manner have improved electrical conductivity and Seebeck coefficient values. 12Studies on the Compositional Properties of Main Miben Series Pumpkin Varieties in Southern China显示文摘Pumpkin is among the most popular agricultural products in Southern China. To determine the important analytical properties(dry matter, total soluble sugar,dissolvable polysaccharides, mineral content, electricity conductivity, soluble solid content, fruit weight, fat, protein, fiber, pectin, β-carotene and edible rate) of pumpkin in Southern China, this study investigated the physical and chemical properties and texture of raw and steamed pumpkins of 13 Miben series varieties. Miben pumpkins were frozen at-40 ℃ for 24 h, or at-4 ℃ for 24 h, or were steamed for 15 min in boiling water to investigate the effect of different treatments on the textural change, and to reveal the texture and sensory properties of the different varieties. A total of 13 pumpkin varieties of Miben series CP1578, CP2116, XM2-2,Baimi II, Guangmi I, Guangzhouluxia, Jintong, Laojinhanzaoshu, Quanguanjixiang,Newzuhe III, V pumpkin, Zaoshutianmi and Daguomiben were involved in this study.The results showed that the Miben series pumpkins had significant difference in physical properties. The editable rate of Miben series pumpkins was higher than77%. Chemical properties varied among different varieties. Textures of pumpkin flesh after different processes were different.
