1Multispecies probiotic protects gut barrier function in experimental models显示文摘AIM:To investigate the effect of the probiotic combination Lactibiane Tolerance?(LT)on epithelial barrier function in vitro and in vivo.METHODS:The effect of the multispecies probiotic LT was assessed on several models of epithelial barrier function both in vitro(in basal and inflammatory conditions)and in vivo[visceral hypersensitivity induced by chronic stress or by colonic perfusion of a fecal supernatant(FSN)from patients with irritable bowel syndrome(IBS)].In vitro,we measured the permeability of confluent T84 cell monolayers incubated with or without LT by evaluating the paracellular flux of macromolecules,in basal conditions and after stimulation with lipopolysaccharide(LPS)or with conditioned medium of colonic biopsies from IBS patients(IBS-CM).In vivo,male C57/Bl6 mice received orally NaCl or LT for 15 d and were submitted to water avoidance stress(WAS)before evaluating visceral sensitivity by measuring the myoelectrical activity of the abdominal muscle and the paracellular permeability with 51Cr-EDTA.Permeability and sensitivity were also measured after colonic instillation of FSN.Tight-junctions were assessed by immunoblotting and TLR-4 expression was evaluated by immunohistochemistry RESULTS:Incubation of T84 cell monolayers with LT in basal conditions had no significant effect on permeability(P>0.05 vs culture medium).By contrast,addition of LT bacterial bodies(LT)completely prevented the LPS-induced increase in paracellular permeability(P<0.01 vs LPS 10 ng/mL(LPS 10);P<0.01 vs LPS 100ng/mL(LPS 100),P>0.05 vs culture medium).The effect was dose dependent as addition of 109 LT bacterial bodies induced a stronger decrease in absorbance than 106 LT(109 LT+LPS 10:-20.1%±13.4,P<0.01vs LPS 10;106 LT+LPS 10:-11.6%±6.2,P<0.01 vs LPS 10;109 LT+LPS 100:-14.4%±5.5,P<0.01 vs LPS 100;106 LT+LPS 100:-11.6%±7.3,P<0.05 vs LPS 100).Moreover,the increase in paracellular permeability induced by culturing T84 cells with conditioned medium of colonic biopsies from IBS patients(IBS-CM)was completely inhibited in the presence of 109 LT(P<0.01 vs IBS-CM).LT also significantly prevented the epithelial disruption induced by intracolonic infusion of fecal supernatant from IBS patients(P<0.01 vs IBS FSN)or water avoidance stress P<0.01 vs WAS)in C57/Bl6 mice and increased the expression of occludin in vitro and in vivo,as assessed by immnunoblotting.The WAS-induced effect on visceral sensitivity was prevented by LT treatment since values obtained for all steps of colorectal distension were significantly(P<0.01)different from the WAS group.Finally,LT downregulated the response mediated through TLR-4 in vitro(decrease in tumor necrosis factorαsecretion in response to LPS:-65.8%for 109 LT and-52.5%for 106LT,P<0.01 vs LPS)and in vivo(inhibition of WAS induced an increase in TLR-4 expression in the LT treated mice colon,P<0.01 vs WAS).CONCLUSION:The probiotic LT mix prevented the disruption to the epithelial barrier induced by LPS,stress or colonic soluble factors from IBS patients and prevented visceral hypersensitivity.Mylene Nébot-Vivinus Cherryl Harkat Hanene Bzioueche Christel Cartier Raffaella Plichon-Dainese Lara Moussa Helene Eutamene Dorsa Pishvaie Sophie Holowacz Christian Seyrig Thierry Piche Vassilia Theodorou 2014World Journal of Gastroenterology2014,20,22:12
2Synthesis of novel adsorbent by intercalation of biopolymer in LDH for the removal of arsenic from synthetic and natural water显示文摘This study focuses on the synthesis of nanocomposites named CCA and CZA that were prepared by the incorporation of cellulose(CL)in the Ca/Al and Zn/Al layered double hydroxide(LDH),respectively.These materials were then used for the uptake of As(Ⅲ)and As(V)from aqueous medium.Characterization of both nanocomposites(CCA and CZA)was done using FTIR and Raman analysis to identify the functional groups,N2 adsorption-desorption isotherms to determine the specific surface area and pore geometry and XPS analysis to obtain the surface atomic composition.Some other characters were investigated using simultaneous TGA and DTA and elemental chemical analysis(CHNS/O).The crystallinity of the prepared nanocomposites was displayed by XRD patterns.Furthermore,the sheet-like structure of the LDHs and the irregularity of surface morphology with porous structure were observed by TEM and SEM microphotographs.Optimization of maximum adsorption capacity was adjusted using different parameters including pH,contact time and adsorbent dosage.The pseudo-second-order model was in good fitting with kinetics results.The adsorption isotherm results showed that CZA exhibits better adsorption capacity for As(Ⅲ)than CCA and the Langmuir isotherm model described the data well for both nanocomposites.Thermodynamic studies illustrated the endothermic nature of CCA and exothermic nature on CZA,as well as the fact that the adsorption process is spontaneous.A real water sample collected from well located in Gabes(Tunisia),has also been treated.The obtained experimental results were confirmed that these sorbents are efficient for the treatment of hazardous toxic species such as.Hanen Bessaies Sidra Iftekhar Bhairavi Doshi Jamel Kheriji Mohamed Chaker Ncibi Varsha Srivastava Mika Sillanpaa Bechir Hamrouni 2020Journal of Environmental Sciences2020,32,5:10
3Characterization and physicochemical aspects of novel cellulose-based layered double hydroxide nanocomposite for removal of antimony and fluoride from aqueous solution显示文摘A series of novel adsorbents composed of cellulose(CL)with Ca/Al layered double hydroxide(CC_(x)A;where x represent the Ca/Al molar ratio)were prepared for the adsorption of antimony(Sb(V))and fluoride(F^(-))ions from aqueous solutions.The CC_(x)A was characterized by Fourier-transform infrared spectroscopy(FTIR),Brunauer–Emmett–Teller(BET),elemental analysis(CHNS/O),thermogravimetric analysis(TGA-DTA),zeta potential,X-ray photoelectron spectroscopy(XPS)and scanning electron microscopy with energy dispersive Xray spectroscopy(SEM-EDX)analysis.The effects of varying parameters such as dose,pH,contact time,temperature and initial concentration on the adsorption process were investigated.According to the obtained results,the adsorption processes were described by a pseudo-second-order kinetic model.Langmuir adsorption isotherm model provided the best fit for the experimental data and was used to describe isotherm constants.The maximum adsorption capacity was found to be 77.2 and 63.1 mg/g for Sb(V)and F^(-),respectively by CC_(3)A(experimental conditions:pH 5.5,time 60 min,dose 15 mg/10 mL,temperature 298 K).The CC_(3)A nanocomposite was able to reduce the Sb(V)and F^(-)ions concentration in synthetic solution to lower than 6μg/L and 1.5 mg/L,respectively,which are maximum contaminant levels of these elements in drinking water according to WHO guidelines.Hanen Bessaies Sidra Iftekhar Muhammad Bilal Asif Jamel Kheriji Chaker Necibi Mika Sillanpää Bechir Hamrouni 2021Journal of Environmental Sciences2021,33,4:4

5Chemical constituents, cytotoxic, antifungal and antimicrobial properties of Centaurea diluta Ait. subsp. algeriensis(Coss. & Dur.) Maire显示文摘Objective:To investigate the chemical composition of a moderately polar extract(CHC1_3 soluble part of the MeOH-H_2O extract) obtained from the aerial parts(leaves and flowers) of Centaurea diluta Ait.subsp.algeriensis(Coss.& Dur.) Maire,a species endemic to Algeria and Morocco on which no reports are available to date.To evaluate in vitro the cytotoxic,antifungal and antimicrobial activities of this extract and the cytotoxic and antimicrobial activities of its isolated secondary metabolites.Methods:The cytotoxic effects of the extract were investigated on 3 human cancer cell lines i.e.the A549 non-small-cell lung carcinoma(NSCLC),the MCF7 breast adenocarcinoma and the U373 glioblastoma using a MTT colorimetric assay.Biological data allowed to guide the fractionation of the extract by separation and purification on silica gel 60(CC and TLC).The isolated compounds which were characterized by spectral analysis,mainly HR-ESIMS,HR-EIMS,UV and NMR experiments(~1H,^(13)C,COSY,ROESY,HSQC and HMBC) and comparison of their spectroscopic data with those reported in the literature,were evaluated for cytotoxic activities on six cancer cell lines(A549,MCF7,U373,Hs683 human glioma,PC3 human prostate and B16-F10 murine melanoma).The direct and indirect antibacterial and antifungal activities were determined using microdilution methods for the raw extract and TLC-bioautography and microdilution methods against standard and clinical strains for the isolated compounds.Results:The raw extract reduced cell viability with IC_(50)s of 27,25 and 21 μg/mL on A549,MCF7 and U373,respectively.Five secondary metabolites:two phenolic compounds(vanillin 1,paridol 3),a lignan[(-)-arctigenin 2]and two flavonoid aglycones(eupatilin 4 and jaceosidin 5),were then isolated from this extract.Moderate cytotoxic effects were observed for(-)-arctigenin 2(IC_(50)s:28 and 33μM on Hs683 and B16-F10,respectively),eupatilin 4(IC_(50)s:33 and 47 μM on B16-F10 and PC3,respectively) and jaceosidin 5(IC_(50)s:32 and 40 μM on PC3 and B16-F10,respectively).Conclusions:All the isolated compounds were described for the first time from this species.Although inactive against 7 tested microorganisms(fungi,bacteria and yeast,human or plant pathogens),the raw extract was able to potentiate the effect of beta-lactam antibiotics on methicillin-resistant Staphylococcus aureus(MRSA),reducing the minimal inhibitory concentrations(MICs) by a factor of 2-32-fold.No synergy was found between the extract and streptomycin.From the five isolated compounds only jaseosidin 5 showed a moderate antimicrobial activity.Hanene Zater Joelle Huet Veronique Fontaine Samir Benayache Caroline Stevigny Pierre Duez Fadila Benayache 2016Asian Pacific Journal of Tropical Medicine2016,9,6:2
6A New Method for Sentiment Analysis Using Contextual Auto-Encoders显示文摘Sentiment analysis, a hot research topic, presents new challenges for understanding users' opinions and judg-ments expressed online. They aim to classify the subjective texts by assigning them a polarity label. In this paper, weintroduce a novel machine learning framework using auto-encoders network to predict the sentiment polarity label at theword level and the sentence level. Inspired by the dimensionality reduction and the feature extraction capabilities of theauto-encoders, we propose a new model for distributed word vector representation "PMI-SA" using as input pointwise-mutual-information "PMI" word vectors. The resulted continuous word vectors are combined to represent a sentence. Anunsupervised sentence embedding method, called Contextual Recursive Auto-Encoders "CoRAE", is also developed forlearning sentence representation. Indeed, CoRAE follows the basic idea of the recursive auto-encoders to deeply composethe vectors of words constituting the sentence, but without relying on any syntactic parse tree. The CoRAE model consistsin combining recursively each word with its context words (neighbors' words: previous and next) by considering the wordorder. A support vector machine classifier with fine-tuning technique is also used to show that our deep compositionalrepresentation model CoRAE improves significantly the accuracy of sentiment analysis task. Experimental results demon-strate that CoRAE remarkably outperforms several competitive baseline methods on two databases, namely, Sanders twittercorpus and Facebook comments corpus.
