| 4 | Pre-operative radiochemotherapy of locally advanced rectal cancer显示文摘AIM: To evaluate results of pre-operative radiochemotherapy followed by surgery for 15 patients with locally advanced un-resectable rectal cancer.METHODS: 15 patients with advanced non-resectable rectal cancer were treated with pre-operative irriadiation of 40-46 Gy plus concomitant chemotherapy (5-FU+LV and 5′-DFuR) (RCS group). For comparison, 27 similar patients,treated by preoperative radiotherapy (40-50 Gy) plus surgery were served as control (RS group).RESULTS: No radiochemotherapy or radiotherapy was interrupted and then was delayed because of toxicities in both groups. The radical resectability rate was 73.3% in the RCS group and 37.0% (P=0.024) in RS group. Sphincter preservation rates were 26.6% and 3.7% respectively (P=0.028). Sphincter preservation rates of lower rectal cancer were 27.3 % and 0.0 % respectively (P=0.014). Response rates of RCS and RS groups were 46.7 % and 18.5 %(P=0.053). The tumor downstage rates were 8 (53.3%)and 9 (33.3%) in these groups (P=0.206). The 3-year overall survival rates were 66.7 % and 55.6% (P=0.485), and the disease free survival rates were 40.1% and 33.2%(P=0.663). The 3-year local recurrent rates were 26.7%and 48.1% (P=0.174). No obvious late effects were found in either groups.CONCLUSION: High resectability is possible following preoperative radiochemotherapy and can have more sphincters preserved. It is important to improve the quality of the patients′ life even without increasing the survival or local control rates. | 9 | Establishment of a Sandwich ELISA Method for Detection of Vascular Endothelial Growth Factor in Serum Samples of Hepatocellular Carcinoma Patients显示文摘Objective To establish a sandwich ELISA method for detecting vascular endothelial growth factor (VEGF) in sera of population and the patients with hepatocellular carcinoma (HCC). Methods Full length and two truncated human VEGF cDNA sequences were amplified from a commercial plasmid pBLAST49-hVEGF by PCR and inserted into the prokaryotic-expression plasmid pET-32a or pGEX-2T. Various VEGF proteins were expressed and purified from E. coli in His-Trx or GST fusion forms. The specific VEGF antibodies were elicited in experimental rabbits and mice by immunization of the full length VEGF fusion protein His-Trx-VEGF1-165. After purification of antibodies with chromatograph of Protein G, a sandwich ELISA technique was established. Serum VEGF levels were evaluated in 229 adults and 291 HCC patients. Results SDS-PAGE displayed that the molecular weights of the expressed full length (His-Trx-VEGF1-165), N-terminal (His-Trx-VEGF1-100) and C-terminal (GST-VEGF100-165) human VEGF fusion proteins were about 38KD, 31KD, and 33KD, respectively. Western blots confirmed that the prepared antisera were able to recognize both prokaryoticly and eukaryoticly expressed recombinant VEGF proteins. Assays of serially diluted His-Trx-VEGF1-100 by the established sandwich ELISA method showed that the linear range of the standard curve was 0.625-320 ng/mL, with the squared correlation coefficient R2=0.991. Screening of a serum panel containing 291 serum samples of HCC patients and 229 health adults revealed that the average VEGF level in HCC patients was higher than that in healthy controls, with a statically significant difference. | 12 | Meiotic nuclear divisions 1(MND1)fuels cell cycle progression by activating a KLF6/E2F1 positive feedback loop in lung adenocarcinoma显示文摘Background:Considering the increase in the proportion of lung adenocarcinoma(LUAD)cases among all lung cancers and its considerable contribution to cancer-related deaths worldwide,we sought to identify novel oncogenes to provide potential targets and facilitate a better understanding of the malignant progression of LUAD.Methods:The results from the screening of transcriptome and survival analyses according to the integrated Gene Expression Omnibus(GEO)datasets and The Cancer Genome Atlas(TCGA)data were combined,and a promising risk biomarker called meiotic nuclear divisions 1(MND1)was selectively acquired.Cell viability assays and subcutaneous xenograftmodelswere used to validate the oncogenic role ofMND1 in LUADcell proliferation and tumor growth.Aseries of assays,including mass spectrometry,co-immunoprecipitation(Co-IP),and chromatin immunoprecipitation(ChIP),were performed to explore the underlying mechanism.Results:MND1 up-regulation was identified to be an independent risk factor for overall survival in LUAD patients evaluated by both tissue microarray staining and third party data analysis.In vivo and in vitro assays showed that MND1 promoted LUAD cell proliferation by regulating cell cycle.The results of the Co-IP,ChIP and dual-luciferase reporter assays validated that MND1 competitively bound to tumor suppressor Kruppel-like factor 6(KLF6),and thereby protecting E2F transcription factor 1(E2F1)from KLF6-induced transcriptional repression.Luciferase reporter and ChIP assays found that E2F1 activated MND1 transcription by binding to its promoter in a feedback manner.Conclusions:MND1,KLF6,and E2F1 form a positive feedback loop to regulate cell cycle and confer DDP resistance in LUAD.MND1 is crucial for malignant progression and may be a potential therapeutic target in LUAD patients. | Quanli Zhang Run Shi Yongkang Bai Lijuan Meng Jingwen Hu Hongyu Zhu Tongyan Liu Xiaomeng De Siwei Wang Jie Wang Lin Xu Guoren Zhou Rong Yin | 2021 | Cancer Communications2021,41,6: | 2 |
| 13 | Warhead power assessment based on double hierarchy hesitant fuzzy linguistic term sets theory and gained and lost dominance score method显示文摘Warhead power assessment of the anti-ship missile plays a vital role in determining the optimal design of missile, thus having important strategic research significance. However, in the assessment process, expert’s judgement will directly affect the assessment accuracy. In addition,there are many criteria involved in the missile design alternatives. Some criteria with poor performance may be compensated by other criteria with excellent performance, and then it is impossible to find the truly optimal alternative. Aimed at solving these problems, this paper proposes a synthetical assessment process based on fuzzy hesitant linguistic term set and the Gained and Lost Dominance Score(GLDS) method. In order to improve the assessment accuracy of experts and solve the problem that experts generate different opinions, combined with the advantages of fuzzy hesitant sets and linguistic term sets, the double hierarchy hesitant fuzzy linguistic term sets are used in this paper to improve the accuracy of expert’s judgement. In order to effectively combine expert’s experience with the data of criteria, the evidence theory and entropy weight method are used to transfer the expert’s judgement to the weight. In order to avoid selecting defective alternative of missile design, the GLDS is used to fuse expert information and criteria information. Sensitivity analysis shows that the assessment process has sensitivity to some extent. However, when the fluctuation of expert’s assessment makes the fluctuation of θ in the range of-5% to 5%, the impact on the results is not quite conspicuous. | 14 | Kinetic Characteristics of Neutralizing Antibody Responses Vary among Patients with COVID-19显示文摘Objective The coronavirus disease 2019(COVID-19) pandemic continues to present a major challenge to public health. Vaccine development requires an understanding of the kinetics of neutralizing antibody(NAb) responses to severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).Methods In total, 605 serum samples from 125 COVID-19 patients(from January 1 to March 14, 2020)varying in age, sex, severity of symptoms, and presence of underlying diseases were collected, and antibody titers were measured using a micro-neutralization assay with wild-type SARS-CoV-2.Results NAbs were detectable approximately 10 days post-onset(dpo) of symptoms and peaked at approximately 20 dpo. The NAb levels were slightly higher in young males and severe cases, while no significant difference was observed for the other classifications. In follow-up cases, the NAb titer had increased or stabilized in 18 cases, whereas it had decreased in 26 cases, and in one case NAbs were undetectable at the end of our observation. Although a decreasing trend in NAb titer was observed in many cases, the NAb level was generally still protective.Conclusion We demonstrated that NAb levels vary among all categories of COVID-19 patients. | 16 | Accumulation of freshwater in the permanent ice zone of the Canada Basin during summer 2008显示文摘A combination of δ^(18)O and salinity data was employed to explore the freshwater balance in the Canada Basin in summer 2008.The Arctic river water and Pacific river water were quantitatively distinguished by using different saline end-members.The fractions of total river water,including the Arctic and Pacific river water,were high in the upper 50 m and decreased with depth as well as increasing latitude.In contrast,the fraction of Pacific river water increased gradually with depth but decreased toward north.The inventory of total river water in the Canada Basin was higher than other arctic seas,indicating that Canada Basin was a main storage region for river water in the Arctic Ocean.The fraction of Arctic river water was higher than Pacific river water in the upper 50 m while the opposite was true below 50 m.As a result,the inventories of Pacific river water were higher than those of Arctic river water,demonstrating that the Pacific inflow through the Bering Strait is the main source of freshwater in the Canada Basin.Both the river water and sea-ice melted water in the permanent ice zone were more abundant than those in the region with sea-ice just melted.The fractions of total river water,Arctic river water,Pacific river water increased northward to the north of 82°N,indicating an additional source of river water in the permanent ice zone of the northern Canada Basin.A possible reason for the extra river water in the permanent ice zone is the lateral advection of shelf waters by the Trans-Polar Drift.The penetration depth of sea-ice melted waters was less than 30 m in the southern Canada Basin,while it extended to 125 m in the northern Canada Basin.The inventory of seaice melted water suggested that sea-ice melted waters were also accumulated in the permanent ice zone,attributing to the trap of earlier melted waters in the permanent ice zone via the Beaufort Gyre. | TONG Jinlu CHEN Min1 YANG Weifeng ZHANG Run PAN Hong ZHENG Minfang QIU Yusheng HU Wangjiang ZENG Jian | 2017 | Acta Oceanologica Sinica2017,36,8: | 1 |
