1Construction of a cucumber genetic linkage map with SRAP markers and location of the genes for lateral branch traits显示文摘Using SRAP (sequence-related amplified polymorphism) markers a genetic linkage map of cucumber was constructed with a population consisting of 138 F2 individuals derived from a cross of the two cucumber lines, S06 and S52. In the survey of parental polymorphisms with 182 primer combinations, 64 polymorphism-revealing primer pairs were screened out, which generated totally 108 polymorphic bands with an average of 1.7 bands per primer pair and at most 6 bands from one primer pair. The constructed molecular linkage map included 92 loci, distributed in seven linkage groups and spanning 1164.2 cM in length with an average genetic distance of 12.6 cM between two neighboring loci. Based on this linkage map, the quantitative trait loci (QTL) for the lateral branch number (lbn) and the lateral branch average length (lbl) in cucumber were identified by QTLMapper1.6. A major QTL lbn1 located between ME11SA4B and ME5EM5 in LG2 could explain 10.63% of the total variation with its positively effecting allele from S06. 3Micro-trichome as a class I homeodomain-leucine zipper gene regulates multicellular trichome development in Cucumis sativus显示文摘Plant trichomes serve as a highly suitable model for investigating cell differentiation at the single-cell level.The regulatory genes involved in unicellular trichome development in Arabidopsis thaliana have been intensively studied,but genes regulating multicellular trichome development in plants remain unclear.Here,we characterized Cucumis sativus(cucumber) trichomes as representative multicellular and unbranched structures,and identified Micro-trichome(Mict),using map-based cloning in an F_2 segregating population of7,936 individuals generated from a spontaneous mict mutant.In mict plants,trichomes in both leaves and fruits,are small,poorly developed,and denser than in the wild type.Sequence analysis revealed that a 2,649-bp genomic deletion,spanning the first and second exons,occurred in a plant-specific class I homeodomain-leucine zipper gene.Tissue-specific expression analysis indicated that Mict is strongly expressed in the trichome cells.Transcriptome profiling identified potential targets of Mict including putative homologs of genes known in other systems to regulate trichome development,meristem determinacy,and hormone responsiveness.Phylogenic analysis charted the relationships among putative homologs in angiosperms.Our paper represents initial steps toward understanding the development of multicellular trichomes.Jun-Long Zhao Jun-Song Pan Yuan Guan Wei-Wei Zhang Bei-Bei Bie Yun-Li Wang Huan-Le He Hong-Li Lian Run Cai 2015Journal of Integrative Plant Biology2015,57,11:9
4On the measurement of slip length for liquid flow over super-hydrophobic surface显示文摘To design a surface with large slip or larger drag reduction is a pop issue in the fields of liquid transporting and body swimming. In this context, it is a crucial problem to measure the slip length for these surfaces. Here we propose a novel method by using rheometer for this objective. This method is implemented by designing the distribution of the super-hydrophobic area on the sample. Using this method, a slip length of 40 μm for 70 wt% glycerin flow over a super-hydrophobic surface with stripe structure (the period, width and height of ridges are 150 μm, 40 μm and 65 μm, respectively) is measured. The result shows that the slip length measured using this method is in good agreement with former results measured by other methods. 5Construction of a BAC library from cucumber (Cucumis sativus L.) and identification of linkage group specific clones显示文摘A bacterial artificial chromosome (BAC) library consisting of 19,200 clones with an average insert size of 105 kb has been constructed from a cucumber (Cucumis sativus L.) inbred line S94,derived from a cultivar in North China. The entire library was equivalent to approximately 5 haploid cucumber genomes. To facilitate chromosome engineering and anchor the cucumber genetic linkage map to its chromosomes,15 sequence-characterized ampli?ed regions (SCAR) and seven simple sequence repeats (SSR) markers from each link-age group of cucumber were used to screen an ordered array of pooled BAC DNA with polymerase chain reaction (PCR). Fifteen mark-ers gave at least two positive clones. As a result,22 BAC clones representing 7 linkage groups of cucumber were identified,which further validated the genome coverage and utility of the library. 6Prospective study of scoring system in selective intraoperative cholangiography during laparoscopic cholecystectomy显示文摘AIM: To evaluate of scoring system in predicting choledocholithiasis in selective intraoperative cholangiography (IOC) during laparoscopic cholecystectomy (LC).METHODS: The scoring system of predicting choledocholithiasis was developed during the retrospective study in 264 cases, and was tested in 184 to evaluate its predictive value in choledocholithiasis.RESULTS: The scoring system was developed in a retrospective study of 264 cases, the statistical analyses showed the predictive factors included sex, transaminase levels, alkaline phosphatase level, bilirubin level, and common bile duct diameter on ultrasonography. The scoring system was used in 184 cases prospectively, of which, 3 of 162 (1.9%) cases scoring <3 had choledocholithiasis, 17 of 22 (77.3 %) cases scores≥3 had choledocholithiasis. 7Quantitative trait locus analysis of lateral branch-related traits in cucumber(Cucumis sativus L.)using recombinant inbred lines显示文摘A group of 224 recombinant inbred lines (RILs) was derived from a narrow cross between 2 cucumber (Cucumis sativus L.) lines, namely, S94 (Northern China type with weak lateral branch growth potential and early lateral branch sprouting time) and S06 (Northern European type with strong lateral branch growth potential and late lateral branch sprouting time). These lines were then used for investigating lateral branch-related traits. A total of 36 quantitative trait loci (QTLs) were detected for the following 4 lateral branch-related traits: lateral branch average length (LBAL), lateral branch total length (LBTL), lateral branch number (LBN), and first lateral branch node (FLBN). Further, each QTL explained 3.1% (lbtl2.1, spring) to 32.3% (lbn2.3, spring) of the observed phenotypic variance. Eleven QTLs (lbal1.1, lbtl1.1, lbn1.2, flbn1.2, etc.) for different traits were found to be clustered on the e23m18d-ME23EM6c section (7.4 cM) of linkage group (LG) 1; further, 15 QTLs (lbal2.1, lbtl2.1, lbn2.1, flbn2.1, etc.) were found to be clustered on the S94A1-ME4SA4a section (13.9 cM) of LG2. Twenty-one QTLs explained more than 10% of the phenotypic variance. Moreover, lbtl1.3 (autumn, 26.2%, logarithm of odds (LOD) = 17.4; spring, 26.9%, LOD = 17.9) had stable position and contribution in both seasons. 8Relationship Between Coleoptile Length and Drought Resistance and Their QTL Mapping in Rice显示文摘By using a set of recombinant inbred line(RIL)population involving in 195 lines derived from a cross of Zhenshan 97B (lowland variety)and IRAT109(upland variety),the correlation analysis between coleoptile length(CL)and drought resistance index (DRI)and their QTL identification were conducted.There existed a significantly positive relationship between CL and DRI with the correlation coefficient of 0.2206**under water stress conditions.Under normal and water stress conditions,a total of eleven and four QTLs for CL and DRI,respectively,were detected on chromosomes 1,2,4,5,6,7,9,11 and 12 by using a linkage map including 213 SSR markers,which explained 4.84%to 22.65%of phenotypic variance.Chromosomes 1 and 9 possessing the QTLs for DRI harbored simultaneously QTLs for CL,and qCL9 shared the same chromosome location with qDRI9(RM160-RM215).Comparing the QTLs related to drought resistance in other studies,QTLs for CL and DRI were located in the same or adjacent marker interval as those related to root traits,such as number,dry weight,depth,and length of root.Moreover,sixteen and three pairs of epistatic loci for CL and DRI were found,which accounted for 56.17%and 11.93%of the total variation in CL and DRI,respectively.HU Song-ping YANG Hua ZOU Gui-hua LIU Hong-yan LIU Guo-lan MEI Han-wei CAI Run LI Ming-shou LUO Li-jun 2007Rice science2007,14,1:4
9Rapid Determination of Silicon Content in Rice显示文摘A method for rapid determination of silicon content in rice was introduced. The reliability of this method was verified byusing a recombinant inbred line (RIL) population of rice cross Zhenshan 97B / Milyang 46. Two hundred and forty-nine RILs weretransplanted in two replications. Simple correlation coefficients on the silicon content in the hull, flag leaf and stem in rice betweenduplicate samples of 498 rice materials were 0.97954, 0.97026 and 0.98848, respectively. Ten representative samples wereselected for measurement using the high-temperature alkaline fusion method. 10Binding between Prion Protein and Aβ Oligomers Contributes to the Pathogenesis of Alzheimer’s Disease显示文摘A plethora of evidence suggests that protein misfolding and aggregation are underlying mechanisms of various neurodegenerative diseases,such as prion diseases and Alzheimer's disease(AD).Like prion diseases,AD has been considered as an infectious disease in the past decades as it shows strain specificity and transmission potential.Although it remains elusive how protein aggregation leads to AD,it is becoming clear that cellular prion protein(PrP^c)plays an important role in AD pathogenesis.Here,we briefly reviewed AD pathogenesis and focused on recent progresses how PrP^c contributed to AD development.In addition,we proposed a potential mechanism to explain why infectious agents,such as viruses,conduce AD pathogenesis.Microbe infections cause AD deposition and upregulation of PrP^c,which lead to high affinity binding between AD oligomers and PrP^c.The interaction between PrP^c and AP oligomers in turn activates the Fyn signaling cascade,resulting in neuron death in the central nervous system(CNS).Thus,silencing PrP^c expression may turn out be an effective treatment for PrP^c dependent AD.Chang Kong Hao Xie Zhenxing Gao Ming Shao Huan Li Run Shi Lili Cai Shanshan Gao Taolei Sun Chaoyang Li 2019Virologica Sinica2019,34,5:4

12Factors influencing the results of treatment of bile duct injuries during laparoscopic cholecystectomy显示文摘BACKGROUND: The short-term results of repair of lapa- roscopic bile duct injuries have been well discussed, but the long-term results have been rarely reported. This study was undertaken to evaluate the factors influencing the outcome of repair of bile duct injuries caused by laparoscopic chole- cystectomy. METHODS: The outcomes of repair of bile duct injuries caused by laparoscopic cholecystectomy in 31 patients were reviewed retrospectively, and the effects of injury recogni- tion, cholangiography, repair modality and techniques on the long-term results were analyzed. RESULTS: Bile duct injuries were repaired successfully in 19 (95%) of 20 patients with injuries who had been recog- nized intraoperatively, and in 10 (90%) of 11 patients with injuries who had been recognized postoperatively. Repair was successful in 29 (93% ) of the 31 patients after complete cholangiography. Closure of partial division, laceration, or small perforation of the bile duct with or without T tube drainage was satisfactory in the 23 patients. End to end re- pair over T tube was successful in 2 transection patients, who were detected intraoperatively. Roux-en-Y hepatico- jejunostomy was used successfully to repair transection, ex- cision or stricture of the bile duct in 4 of 5 patients (80% ). 14Prion Protein Protects Cancer Cells against Endoplasmic Reticulum Stress Induced Apoptosis显示文摘Unfolded protein response(UPR) is an adaptive reaction for cells to reduce endoplasmic reticulum(ER) stress. In many types of cancers, such as lung cancer and pancreatic cancer, cancer cells may harness ER stress to facilitate their survival and growth. Prion protein(PrP) is a glycosylated cell surface protein that has been shown to be up-regulated in many cancer cells. Since PrP is a protein prone to misfolding, ER stress can result in under-glycosylated PrP, which in turn may activate ER stress. To assess whether ER stress leads to the production of under-glycosylated PrP and whether underglycosylated PrP may contribute to ER stress thus leading to cancer cell apoptosis, we treated different cancer cells with brefeldin A(BFA), thapsigargin(Thps), and tunicamycin(TM). We found that although BFA, Thps, and TM treatment activated UPR, only ATF4 was consistently activated by these reagents, but not other branches of ER stress. However, the canonical PERK-eIF2α-ATF4 did not account for the observed activation of ATF4 in lung cancer cells. In addition, BFA,but neither Thps nor TM, significantly stimulated the expression of cytosolic PrP. Finally, we found that the levels of PrP contributed to anti-apoptosis activity of BFA-induced cancer cell death. 16Kinetic Characteristics of Neutralizing Antibody Responses Vary among Patients with COVID-19显示文摘Objective The coronavirus disease 2019(COVID-19) pandemic continues to present a major challenge to public health. Vaccine development requires an understanding of the kinetics of neutralizing antibody(NAb) responses to severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).Methods In total, 605 serum samples from 125 COVID-19 patients(from January 1 to March 14, 2020)varying in age, sex, severity of symptoms, and presence of underlying diseases were collected, and antibody titers were measured using a micro-neutralization assay with wild-type SARS-CoV-2.Results NAbs were detectable approximately 10 days post-onset(dpo) of symptoms and peaked at approximately 20 dpo. The NAb levels were slightly higher in young males and severe cases, while no significant difference was observed for the other classifications. In follow-up cases, the NAb titer had increased or stabilized in 18 cases, whereas it had decreased in 26 cases, and in one case NAbs were undetectable at the end of our observation. Although a decreasing trend in NAb titer was observed in many cases, the NAb level was generally still protective.Conclusion We demonstrated that NAb levels vary among all categories of COVID-19 patients. 18Simulation of Silty Clay Compressibility Parameters Based on Improved BP Neural Network Using Bayesian Regularization显示文摘Soil compressibility parameters are important indicators in the geotechnical field and are affected by various factors such as natural conditions and human interference.When the sample size is too large,conventional methods require massive human and financial resources.In order to reasonably simulate the compressibility parameters of the sample,this paper firstly adopts the correlation analysis to select seven influencing factors.Each of the factors has a high correlation with compressibility parameters.Meanwhile,the proportion of the weights of the seven factors in the Bayesian neural network is analyzed based on Garson theory.Secondly,an output model of the compressibility parameters of BR-BP silty clay is established based on Bayesian regularized BP neural network.Finally,the model is used to simulate the measured compressibility parameters.The output results are compared with the measured values and the output results of the traditional LM-BP neural network.The results show that the model is more stable and has stronger nonlinear fitting ability.The output of the model is basically consistent with the actual value.Compared with the traditional LMBP neural network model,its data sensitivity is enhanced,and the accuracy of the output result is significantly improved,the average value of the relative error of the compression coefficient is reduced from 15.54%to 6.15%,and the average value of the relative error of the compression modulus is reduced from 6.07%to 4.62%.The results provide a new technical method for obtaining the compressibility parameters of silty clay in this area,showing good theoretical significance and practical value.CAI Run PENG Tao WANG Qian HE Fanmin ZHAO Duoying 2020Earthquake Research in China2020,34,3:1
