7Machine learning for detecting mesial temporal lobe epilepsy by structural and functional neuroimaging显示文摘Mesial temporal lobe epilepsy(mTLE),the most common type of focal epilepsy,is associated with functional and structural brain alterations.Machine learning(ML)techniques have been successfully used in discriminating mTLE from healthy controls.However,either functional or structural neuroimaging data are mostly used separately as input,and the opportunity to combine both has not been exploited yet.We conducted a multimodal ML study based on functional and structural neuroimaging measures.We enrolled 37 patients with left mTLE,37 patients with right mTLE,and 74 healthy controls and trained a support vector ML model to distinguish them by using each measure and the combinations of the measures.For each single measure,we obtained a mean accuracy of 74%and 69%for discriminating left mTLE and right mTLE from controls,respectively,and 64%when all patients were combined.We achieved an accuracy of 78%by integrating functional data and 79%by integrating structural data for left mTLE,and the highest accuracy of 84%was obtained when all functional and structural measures were combined.These findings suggest that combining multimodal measures within a single model is a promising direction for improving the classification of individual patients with mTLE.Baiwan Zhou Dongmei An Fenglai Xiao Running Niu Wenbin Li Wei Li Xin Tong Graham J Kemp Dong Zhou Qiyong Gong Du Lei 2020Frontiers of Medicine2020,14,5:2
8SARS Patients-derived Human Recombinant Antibodies to S and M Proteins Efficiently Neutralize SARS-Coronavirus Infectivity显示文摘Objective To develop a specific SARS virus-targeted antibody preparation for emergent prophylaxis and treatment of SARS virus infection. Methods By using phage display technology, we constructed a naive antibody library from convalescent SARS patient lymphocytes. To obtain the neutralizing antibody to SARS virus surface proteins, the library panning procedure was performed on purified SARS virions and the specific Fab antibody clones were enriched by four rounds of repeated panning procedure and screened by highthroughput selection. The selected Fab antibodies expressed in the periplasma of E. Coli were soluble and further purified and tested for their binding properties and antiviral function to SARS virus. The functional Fab antibodies were converted to full human IgG antibodies with recombinant baculovirus/insect cell systems and their neutralizing activities were further determined. Results After four rounds of the panning, a number of SARS-CoV virus-targeted human recombinant Fab antibodies were isolated from the SARS patient antibody library. Most of these were identified to recognize both natural and recombinant SARS spike (S) proteins, two Fab antibodies were specific for the virus membrane (M) protein, only one bound to SARS-CoV nucleocapsid protein. The SARS-CoV S and M protein-targeted Fab or IgG antibodies showed significant neutralizing activities in cytopathic effect (CPE) inhibition neutralization test, these antibodies were able to completely neutralize the SARS virus and protect the Vero cells from CPE after virus infection. However, the N protein-targeted Fab or IgG antibodies failed to neutralize the virus. In addition, the SARS N protein-targeted human Fab antibody reacted with the denatured N proteins, whereas none of the S and M protein specific neutralizing antibodies did. These results suggested that the S and M protein-specific neutralizing antibodies could recognize conformational epitopes which might be involved in the binding of virions to cellular receptors and the fusion activity of the virus.Conclusion The SARS-CoV spike protein and membrane proteins are able to elicite efficient neutralizing antibodies in SARS patients. 12An injectable self-adaptive polymer as a drug carrier for the treatment of nontraumatic early-stage osteonecrosis of the femoral head显示文摘Core decompression(CD)with the elimination of osteonecrotic bone is the most common strategy for treating early-stage nontraumatic osteonecrosis of the femoral head(ONFH).Adjuvant treatments are widely used in combination with CD as suitable methods of therapy.Existing augmentations have to be fabricated in advance.Here,we report a novel injectable glycerin-modified polycaprolactone(GPCL)that can adapt to the shape of the CD cavity.GPCL shows great flowability at 52.6℃.After solidification,its compressive modulus was 120 kPa at body temperature(37℃).This excellent characteristic enables the polymer to provide mechanical support in vivo.In addition,GPCL acts as a carrier of the therapeutic agent zoledronic acid(ZA),demonstrating sustained release into the CD region.ZA-loaded GPCL was injected into ONFH lesions to treat early-stage nontraumatic cases.Compared to that in the CD group,CD+ZA-loaded GPCL injection preserved bone density and increased the collagen level in the femoral head.At the interface between the GPCL and CD tunnel wall,osteogenesis was significantly promoted.In addition,morphological evaluations revealed that the femoral heads in the CD+ZA-GPCL group exhibited improved pressure resistance.These results suggest a strategy effective to preserve the bone density of the femoral head,thus decreasing the possibility of femoral head collapse.This novel injectable polymer has,therefore,considerable potential in clinical applications.Ning Kong Hang Yang Run Tian Guanzhi Liu Yiyang Li Huanshuai Guan Qilu Wei Xueshan Du Yutian Lei Zhe Li Ruomu Cao Yiwei Zhao Xiaohui Wang Kunzheng Wang Pei Yang 2022Bone Research2022,10,3:1
13Study of micro-trichome (mict) reveals novel connections between transcriptional regulation of multicellular trichome development and specific metabolism in cucumber显示文摘Trichomes that cover the epidermis of aerial plant organs play multiple roles in plant protection.Compared with a unicellular trichome in model plants,the development mechanism of the multicellular trichome is largely unclear.Notably,variations in trichome development are often accompanied by defects in the biosynthesis of cuticle and secondary metabolites;however,major questions about the interactions between developmental differences in trichomes and defects in metabolic pathways remain unanswered.Here,we characterized the glabrous mutant mict/csgl1/cstbh via combined metabolomic and transcriptomic analyses to extend our limited knowledge regarding multicellular trichome development and metabolism in cucumber.Mict was found to be explicitly expressed within trichome cells.Transcriptomic analysis indicated that genes involved in flavonoid and cuticle metabolism are significantly downregulated in mict mutants.Further metabolomic analysis confirmed that flavonoids,lipids,and cuticle compositions are dramatically altered in mict mutants.Additional studies revealed that Mict regulates flavonoid,lipid,and cuticle biosynthesis by likely directly binding to downstream functional genes,such as CsTT4,CsFLS1,CsCER26,and CsMYB36.These findings suggest that specific metabolic pathways(e.g.,flavonoids and cuticle components)are co-regulated by Mict and provide insights into transcriptional regulation mechanisms of multicellular trichome development and its specific metabolism in cucumber.Jian Pan Leyu Zhang Guanqun Chen Haifan Wen Yue Chen Hui Du Junlong Zhao Huanle He Hongli Lian Huiming Chen Jianxin Shi Run Cai Gang Wang Junsong Pan 2021Horticulture Research2021,8,1:1

15Efficient Transposition of the Retrotransposon Tnt1 in Cucumber(Cucumis sativus L.)显示文摘Tnt1 is an active retrotransposon originally identified in tobacco(Nicotiana tabacum L.)(Grandbastien et al.,1989),but its transposition activity could be activated through tissue culture in other plant species.The insertions are stable and inheritable in the progeny,which has made it a valuable and versatile tool for developing insertional mutagenesis libraries in several plant species.Here,we explored its utility for mutagenesis in cucumber(Cucumis sativus L.).T_3 Tnt1 transgenic cucumber plants were subjected to tissue culture to regenerate self-pollinated progeny.With PCR and analyses and Southern hybridization,we found regenerated plants maintained the original Tnt1 insertion and created new insertions suggesting characteristic re-transposition activity of Tnt1 during this process.Using genome walking,some flanking sequences of Tnt1 insertions were recovered in regenerated plants.The results demonstrated that Tnt1 could be stably inherited and re-transposable during tissue culture in cucumber and that it is feasible to use for developing an insertional mutagenesis library for cucumber.ZHANG Qi DU Hui LV Duo XIAO Tingting PAN Jian HE Huanle WANG Gang CAI Run WENG Yiqun PAN Junsong 2018Horticultural Plant Journal2018,4,3:0
16Adsorption of emerging sodium p-perfluorous nonenoxybenzene sulfonate(OBS) onto soils: Kinetics, isotherms and mechanisms显示文摘The widespread use of sodium p-perfluorous nonenoxybenzene sulfonate(OBS), a typical alternative to perfluorooctane sulfonate, has resulted in potential threats to the environment, but the adsorption behavior of OBS in soils has not yet been reported. In this study, the adsorption behaviors of OBS on five soils with different physicochemical properties were investigated. The rate of OBS adsorption was fast, and most of the OBS uptake was completed within 12 h. The good model fit of OBS adsorption to the pseudo-second-order and Elovich models indicated the occurrence of chemical adsorption. The adsorption isotherms of OBS on the soils were better described by the Freundlich model than by the Langmuir model, suggesting that the OBS adsorption sites on the soils were heterogeneous. This is possibly associated with various adsorption mechanisms including hydrophobic, π-π, hydrogen bonding, and electrostatic interactions,further confirmed by the good model fit to the D-R isotherm. Adsorption of OBS occurred on the soils, and the adsorption process was spontaneous and endothermic. In addition, the soils were more suitable for OBS adsorption at lower pH values due to the stronger electrostatic adsorption. The OBS adsorption on the soils decreased with the increase of soil depth from 0 to 30 cm. Moreover, the presence of organic matter and ammonia nitrogen in the soils was favorable for OBS adsorption, and these parameters decreased with increasing soil depth, making OBS adsorption less prominent in the deeper soil. 18Sparse flight spotlight mode 3-D imaging of spaceborne SAR based on sparse spectrum and principal component analysis显示文摘The spaceborne synthetic aperture radar(SAR)sparse flight 3-D imaging technology through multiple observations of the cross-track direction is designed to form the cross-track equivalent aperture,and achieve the third dimensionality recognition.In this paper,combined with the actual triple star orbits,a sparse flight spaceborne SAR 3-D imaging method based on the sparse spectrum of interferometry and the principal component analysis(PCA)is presented.Firstly,interferometric processing is utilized to reach an effective sparse representation of radar images in the frequency domain.Secondly,as a method with simple principle and fast calculation,the PCA is introduced to extract the main features of the image spectrum according to its principal characteristics.Finally,the 3-D image can be obtained by inverse transformation of the reconstructed spectrum by the PCA.The simulation results of 4.84 km equivalent cross-track aperture and corresponding 1.78 m cross-track resolution verify the effective suppression of this method on high-frequency sidelobe noise introduced by sparse flight with a sparsity of 49%and random noise introduced by the receiver.Meanwhile,due to the influence of orbit distribution of the actual triple star orbits,the simulation results of the sparse flight with the 7-bit Barker code orbits are given as a comparison and reference to illuminate the significance of orbit distribution for this reconstruction results.This method has prospects for sparse flight 3-D imaging in high latitude areas for its short revisit period.ZHOU Kai LI Daojing CUI Anjing HAN Dong TIAN He YU Haifeng DU Jianbo LIU Lei ZHU Yu ZHANG Running 2021Journal of Systems Engineering and Electronics2021,32,5:0
