| 3 | Genetic dissection of maize seedling root system architecture traits using an ultra-high density bin-map and a recombinant inbred line population显示文摘Maize(Zea mays) root system architecture(RSA)mediates the key functions of plant anchorage and acquisition of nutrients and water. In this study,a set of 204 recombinant inbred lines(RILs) was derived from the widely adapted Chinese hybrid ZD958(Zheng58 Chang7-2),genotyped by sequencing(GBS) and evaluated as seedlings for 24 RSA related traits divided into primary,seminal and total root classes. Signi ficant differences between the means of the parental phenotypes were detected for 18 traits,and extensive transgressive segregation in the RIL population was observed for all traits. Moderate to strong relationships among the traits were discovered. A total of 62 quantitative trait loci(QTL) were identi fied that individually explained from1.6% to 11.6%(total root dry weight/total seedling shoot dry weight) of the phenotypic variation. Eighteen,24 and 20 QTL were identi fied for primary,seminal and total root classes of traits,respectively. We found hotspots of 5,3,4 and 12 QTL in maize chromosome bins 2.06,3.02-03,9.02-04,and 9.05-06,respectively,implicating the presence of root gene clusters or pleiotropic effects. | 4 | Equations for estimating the above-ground biomass of Larix sibirica in the forest-steppe of Mongolia显示文摘Biomass functions were established to estimate above-ground biomass of Siberian larch (Larix sibirica) in the Altai Mountains of Mongolia. The functions are based on biomass sampling of trees from 18 different sites, which represent the driest locations within the natural range of L. sibirica. The best performing regression model was found for the equations y = (D2 H)/(a+bD) for stem biomass, y = aDb for branch biomass, and y=aDb Hc for needle biomass, where D is the stem diameter at breast height and H is the tree height. The robustness of the biomass functions is assessed by comparison with equations which had been previously published from a plantation in Iceland. There, y=aDb H c was found to be the most significant model for stem and total above-ground biomasses. Applying the equations from Iceland for estimating the above-ground biomass of trees from Mongolia resulted in the underestimation of the biomass in large-diameter trees and the overestimation of the biomass in thin trees. The underestimation of thick-stemmed trees is probably attributable to the higher wood density, which has to be expected under the ultracontinental climate of Mongolia compared to the euoceanic climate of Iceland. | 6 | Orally administered extract from Prunella vulgaris attenuates spontaneous colitis in mdr1a^(-/-) mice显示文摘AIM: To investigate the ability of a Prunella vulgaris(P. vulgaris) ethanolic extract to attenuate spontaneous typhlocolitis in mdr1a-/- mice. METHODS: Vehicle(5% ethanol) or P. vulgaris ethanolic extract(2.4 mg/d) were administered daily by oral gavage to mdr1a-/- or wild type FVBWT mice from 6 wk of age up to 20 wk of age. Clinical signs of disease were noted by monitoring weight loss. Mice experiencingweight loss in excess of 15% were removed from the study. At the time mice were removed from the study, blood and colon tissue were collected for analyses that included histological evaluation of lesions, inflammatory cytokine levels, and myeloperoxidase activity. RESULTS: Administration of P. vulgaris extracts to mdr1a-/- mice delayed onset of colitis and reduced severity of mucosal inflammation when compared to vehicle-treated mdr1a-/- mice. Oral administration of the P. vulgaris extract resulted in reduced(P < 0.05) serum levels of IL-10(4.6 ± 2 vs 19.4 ± 4), CXCL9(1319.0 ± 277 vs 3901.0 ± 858), and TNFα(9.9 ± 3 vs 14.8 ± 1) as well as reduced gene expression by more than two-fold for Ccl2, Ccl20, Cxcl1, Cxcl9, IL-1 α, Mmp10, VCAM-1, ICAM, IL-2, and TNFα in the colonic mucosa of mdr1a-/- mice compared to vehicle-treated mdr1a-/-mice. Histologically, several microscopic parameters were reduced(P < 0.05) in P. vulgaris-treated mdr1a-/-mice, as was myeloperoxidase activity in the colon(2.49 ± 0.16 vs 3.36 ± 0.06, P < 0.05). The numbers of CD4+ T cells(2031.9 ± 412.1 vs 5054.5 ± 809.5) and germinal center B cells(2749.6 ± 473.7 vs 4934.0 ± 645.9) observed in the cecal tonsils of P. vulgaris-treated mdr1a-/- were significantly reduced(P < 0.05) from vehicle-treated mdr1a-/- mice. Vehicle-treated mdr1a-/- mice were found to produce serum antibodies to antigens derived from members of the intestinal microbiota, indicative of severe colitis and a loss of adaptive tolerance to the members of the microbiota. These serum antibodies were greatly reduced or absent in P. vulgaris-treated mdr1a-/- mice. | 8 | Response of tree-ring width to climate warming and selective logging in larch forests of the Mongolian Altai显示文摘Aims The Mongolian Altai is an old settlement area,which is populated by pastoral nomads since 2000–3000 years.Forests in this region(at ca.2300 m a.s.l.)are highly fragmented and border on steppe and alpine grasslands,which are used for mobile livestock husbandry.The climate in Central Asia is warming to levels clearly above the global average,which affects the vegetation.Furthermore,the transition from planned to market economy and the decollectivization of livestock 20 years ago has strongly changed land use practices in Mongolia,especially resulting in an increase in recent logging activities.We were interested in the question how climate warming and selective logging influence the annual stem growth and the stand structure.Methods The impact of climate and land use by the pastoral nomads on the annual stem increment of more than 1800 trees of Siberian larch(Larix sibirica)was analyzed.Different groups of trees with divergent growth trends depending on the social position and stand history were identified by non-metric multidimensional scaling and analysis of similarities.Long-term trends in the annual stem increment were analyzed by establishing separate regional growth curves for trees of different age classes.Important Findings Instrumental climate data substantiate an increase of temperature by 2.1°C since 1940 at constant precipitation.Trees benefit from the increased temperatures.Climate–response analysis revealed that radial stem increment was promoted by the temperature in early summer,but also high precipitation in spring and in the year before tree-ring formation.Forest dynamics is also strongly influenced by anthropogenic activities.In addition to the natural forest dynamics,logging resulted in divergent growth trends within given age classes and habitats(forest interior and forest edge);overall,22 groups of trees with different characteristics in the annual radial stem increment were identified.A tree-ring series-based reconstruction of logging intensity since 1935 suggests that moderate selective logging occurred throughout the study period.However,selective logging was strongly intensified after 1990 as the result of the breakdown of the Communist regime in Mongolia and the transition from centrally planned to market economy.Because tree stump densities showed that the ratio of felled to live trees was 2:1 in the interior or even 0.9:1 at the edges of the forests and most logging occurred during the past 20 years,it must be concluded that the forests of the Mongolian Altai are presently exploited far beyond the level of sustainability.A close correlation of the ratio of felled to live trees with the density of summer camps of pastoral nomads in the vicinity suggests that trees are primarily felled by the local population. | Choimaa Dulamsuren Mookhor Khishigjargal Christoph Leuschner Markus Hauck | 2014 | Journal of Plant Ecology2014,7,1: | 5 |
