1De novo characterization of the root transcriptome of a traditional Chinese medicinal plant Polygonum cuspidatum显示文摘Various active components have been extracted from the root of Polygonum cuspidatum.However,the genetic basis for their activity is virtually unknown.In this study,25600002 short reads (2.3 Gb) of P.cuspidatum root transcriptome were obtained via Illumina HiSeq 2000 sequencing.A total of 86418 unigenes were assembled de novo and annotated.Twelve,18,60 and 54 unigenes were respectively mapped to the mevalonic acid (MVA),methyl-D-erythritol 4-phosphate (MEP),shikimate and resveratrol biosynthesis pathways,suggesting that they are involved in the biosynthesis of pharmaceutically important anthraquinone and resveratrol.Eighteen potential UDP-glycosyltransferase unigenes were identified as the candidates most likely to be involved in the biosynthesis of glycosides of secondary metabolites.Identification of relevant genes could be important in eventually increasing the yields of the medicinally useful constituents of the P.cuspidatum root.From the previously published transcriptome data of 19 non-model plant taxa,1127 shared orthologs were identified and characterized.This information will be very useful for future functional,phylogenetic and evolutionary studies of these plants.HAO DaCheng MA Pei MU Jun CHEN ShiLin XIAO PeiGen PENG Yong HUO Li XU LiJia SUN Chao 2012Science China(Life Sciences)2012,55,5:36
2Chemical and biological research of Clematis medicinal resources显示文摘Clematis is a botanical source for various pharmaceutically active components,which has long been used in conventional medicine since the beginning of Chinese civilization.Increasing interest in Clematis medicinal resources has led to additional discoveries of triterpenoid saponins,flavonoids,coumarins,alkaloids and many other compounds in various Clematis species,and to investigations on their chemotaxonomy,molecular phylogeny and pharmacology.In continuation with our studies on Clematis chemistry and biology,we review the chemistry,chemotaxonomy,molecular biology and phylogeny of Clematis and their relevance to drug efficacy and drug development.Various databases and technology have been used in literature search in order to characterize the global scientific effort.It is essential to study more species for both the sustainable utilization of Clematis medicinal resources and finding novel compounds with potential clinical utility.Systems biology and omics technologies will play an increasingly important role in future medicinal research involving bioactive compounds of Clematis.HAO DaCheng GU XiaoJie XIAO PeiGen PENG Yong 2013Chinese Science Bulletin2013,58,10:13
3miR-137 inhibits melanoma cell proliferation through downregulation of GLO1显示文摘Late-stage melanoma is refractory to current therapies. MicroRNAs (miRNAs) can modulate many physiological and pathological processes of melanoma. Studies have demonstrated that miR-137 acts as a tumor suppressor by inhibiting the proliferation of melanoma cells through targeting multiple mRNAs. The glyoxalase system member glyoxalase 1 (GLO1) is the principal scavenging enzyme of methylglyoxal (MG), a toxic byproduct of glycolysis. Using 35S in vivo/vitro labelling analysis for dynamic proteomics (SiLAD), we found that miR-137 downregulated the expression of GLO1 in melanoma cells.Bioinformatics analysis predicted that GLO1 is a direct target of miR-137. This was validated by dual luciferase reporter assay.Quantitative RT-PCR (qRT-PCR) and western blot analysis indicated that miR-137 could decrease endogenous GLO1 expression.Furthermore, siRNA targeting of GLO1 mimicked inhibition of melanoma cell proliferation caused by miR-137 overexpression.Re-expression of GLO1 was able to restore miR-137-mediated suppression of melanoma cell proliferation. 4Research progress in the phytochemistry and biology of Ilex pharmaceutical resources显示文摘Ilex is a botanical source for various health-promoting and pharmaceutically active compounds that have been used in traditional Chinese medicine and food for thousands of years.Increasing interest in Ilex pharmaceutical and food resources has led to additional discoveries of terpenoids,saponins,polyphenols(especially flavonoids),glycosides,and many other compounds in various Ilex species,and to investigation of their chemotaxonomy,molecular phylogeny and pharmacol-ogy.In continuation of our studies on Ilex pharmacology and phylogeny,we review the phytochemistry,chemotaxonomy,molecular biology and phylogeny of Ilex species and their relevance to health-promotion and therapeutic efficacy.The similarity and dissimilarity between Ilex paraguariensis,the source plant of mate tea,and the source plants of large-leaved K udingcha(e.g.,Ilex kudingcha and Ilex latifolia)are discussed.It is essential to utilize emerging technologies in non-Camellia tea studies to promote the sustainable utilization of Ilex resources and the identification and development of novel compounds with potential health and clinical utility.Systems biology and '-omics'technologies will play an increasingly important role in pharmaceutical and food research on the bioactive compounds of Ilex species.Dacheng Hao Xiaojie Gua Peigen Xiao Zhanguo Liang Lijia Xu Yong Peng 2013Acta Pharmaceutica Sinica B2013,3,1:6
5Thermally stable photoluminescence and long persistent luminescence of Ca3Ga4O9:Tb^3+/Zn^2+显示文摘A green long persistent luminescence(LPL) phosphor Ca_3 Ga_4 O_9:Tb^(3+)/Zn^(2+) was prepared. Ca_3 Ga_4 O_9 matrix exhibits blue self-activated LPL due to the creation of intrinsic traps. When Tb3+ is doped, the photoluminescence(PL) and LPL colors change from blue to green with their intensities significantly enhanced. The doping of Zn^(2+) evidently improves the PL and LPL performances of the Ca_3Ga_4O_9 matrix and Ca_3Ga_4O_9:Tb^(3+). The thermoluminescence(TL) spectra show that a successive trap distribution is formed by multiple intrinsic traps with different depths in the Ca_3 Ga_4 O_9 matrix, and the incorporation of Tb^(3+) and Zn^(2+) effectively increases the densities of these intrinsic traps. 6NiSe_(2)/Ni(OH)_(2) Heterojunction Composite through Epitaxial-like Strategy as High-Rate Battery-Type Electrode Material显示文摘Constructing heterojunction is a promising way to improve the charge transfer efficiency and can thus promote the electrochemical properties.Herein,a facile and effective epitaxial-like growth strategy is applied to NiSe2 nano-octahe-dra to fabricate the NiSe2-(100)/Ni(OH)2-(110)heterojunction.The heterojunction composite and Ni(OH)2(performing high electrochemical activity)is ideal high-rate battery-type supercapacitor electrode.The NiSe2/Ni(OH)2 electrode exhibits a high specific capacity of 909 C g^-1 at 1 A g^-1 and 597 C g^-1 at 20 A g^-1.The assembled asymmetric supercapacitor composed of the NiSe2/Ni(OH)2 cathode and p-phenylenediamine-functional reduced graphene oxide anode achieves an ultrahigh specific capacity of 303 C g^-1 at 1 A g^-1 and a superior energy density of 76.1 Wh kg^-1 at 906 W kg^-1,as well as an outstanding cycling stability of 82%retention for 8000 cycles at 10 A g^-1.To the best of our knowledge,this is the first example of NiSe2/Ni(OH)2 heterojunction exhibiting such remarkable supercapacitor performance.This work not only provides a promising candidate for next-generation energy storage device but also offers a possible universal strategy to fabricate metal selenides/metal hydroxides heterojunctions.Hao Mei Zhaodi Huang Ben Xu Zhenyu Xiao Yingjie Mei Haobing Zhang Shiyu Zhang Dacheng Li Wenpei Kang Dao Feng Sun 2020Nano-Micro Letters2020,12,5:3
7Research progress of Chinese herbal medicine compounds and their bioactivities:Fruitful 2020显示文摘Traditional Chinese medicines(TCMs)have continued to be a treasure trove.The study of chemodiversity and versatility of bioactivities has always been an important content of pharmacophylogeny.There is amazing progress in the discovery and research of natural components with novel structures and significant bioactivities in 2020.In this paper we review 271 valuable natural products,including terpenoids,steroids,flavonoids,phenylpropanoids,phenolics,nitrogen containing compounds and essential oil,etc.,isolated and identified from TCMs published in journals of Chinese Traditional and Herbal Drugs(Zhong Cao Yao)and Chinese Herbal Medicines(CHMs),and focus on their structures,source organisms,and relevant bioactivities,paying special attention to structural characteristics of novel compounds and newly revealed pharmacological properties of known compounds.It is worth noting that natural products with antitumor activity still constitute the primary object of research.Among the reported compounds,two new triterpenoids,i.e.,ursolic acid 3-O-β-cis-caffeate and mollugoside E,display remarkable cytotoxicity against PC-9 and HL-60 cell lines,respectively.Three known phenolic compounds,i.e.,pyoluteorin,4-hydroxy-3-methoxy cinnamaldehyde and 3,7-dimethoxy-5-hydroxy-1,4-phenanthrenequinone,exhibit significant cytotoxicity against multiple cell lines.Numerous studies on the free radical scavenging activity of reported compounds are currently underway.In vitro,three known phenolic compounds,i.e.,3,4-Odicaffeoylquinic acid methyl ester,3,4,5-O-tricaffeoylquinic acid methyl ester and arbutin,had more considerable antioxidant activities than vitamin C.The anti-inflammatory,anti-diabetic,hypolipidemic,neuroprotective and antimicrobial activities of isolated compounds are also encouraging.The structural characteristics and bioactivities of TCM compounds highlighted here reflect the enormous progress of CHM research in 2020 and will play a positive role in the future drug discovery and development.According to pharmacophylogeny,the phylogenetic distribution of compounds with different natures and flavors can be explored,with view to better mining TCM resources.Xiaojie Gu Dacheng Hao Peigen Xiao 2022Chinese Herbal Medicines2022,14,2:2

15Prediction of sites under adaptive evolution in flavin-containing monooxygenases: Selection pattern revisited显示文摘Flavin-containing monooxygenase (FMO), like cytochrome P450 (CYP), is a monooxygenase that uses the reducing equivalents of NADPH to reduce one atom of molecular oxygen to water, while the other atom is used to oxidize the substrate. Recently, it was shown that some CYP isoforms have been subject to positive selection. However, it is unknown whether the highly conserved phase I detoxification enzyme, FMO, has undergone similar positive Darwinian selection. We used maximum-likelihood models of codon substitution, evolutionary fingerprinting, and cross species comparison to investigate the occurrence of adaptive evolution in FMO sequences. We used recent genomic data from a range of species, including vertebrates and invertebrates. We present the evidence for the occurrence of adaptive evolution in mammalian FMO 3, 4, 5, and fugu FMOs but not in mammalian FMO 1, FMO 2, frog FMOs, other fish FMOs and invertebrate FMOs. The sites under adaptive evolution were significantly associated with the insertion domain in mammalian FMO 5. We identified specific amino acid sites in FMOs 3–5 that are likely targets for selection based on the patterns of parallel amino acid change. The most likely role of adaptive evolution is the repair of mutations that permitted optimal NADP+ binding and improved catalytic efficiency. The occurrence of positive selection during the evolution of phase I detoxification enzymes such as FMOs 3–5 and fugu FMO suggests the occurrence of both high selection pressure acting on species within their unique habitats and significant changes in intensity and direction (forms of xenobiotics and drugs) resulting from changes in microhabitat and food.HAO DaCheng XIAO PeiGen 2011Chinese Science Bulletin2011,56,12:1

19Study on Vegetation Characteristics & Soil Properties of Secondary Grassland in Farmland Areas Abandoned for Different Years in Minqin Oasis显示文摘[Objectives]This study was conducted to investigate the vegetation community characteristics and soil properties of secondary grassland in abandoned farmland areas of different years in Minqin Oasis. [Methods] By the method of space-for-time substitution, the changes of vegetation community characteristics(species composition, structural characteristics and important values) and soil microbial and enzyme activity were determined, and the correlation between various soil factors was analyzed. [Results](1) There were 39 species belonging to 32 genera of 15 families in the research area, including 8 shrub plants, 15 perennial herbs and 16 annual herbs.(2) Soil microbial biomass carbon, nitrogen and phosphorus contents were higher in the upper layer than in the lower layer, and the phenomenon of “surface aggregation” was obvious. The differences were different with the change of abandoned years.(3) Soil catalase content was the highest among different plots, and soil phosphatase, sucrase and urease all showed a trend of fluctuating decline with the increase in abandoned years. Soil enzyme changes were different in different soil layers. 20Deepening insights into food and medicine continuum within the context of pharmacophylogeny显示文摘The concept of food and medicine continuum(FMC)is traced back to Shennong’s Classic of Materia Medica(Shénnóng Bencǎo Jing in Chinese)of the 1st century(Heinrich et al.,2022),which recorded 120 herbal medicines of the‘top grade’with nourishing properties;they can be consumed for extended periods of time without obvious side effects,indicating their functional food and medicinal benefits(Xiao,2023).In recent decades,the health-promoting actions of numerous edible herbal medicines and phytometabolites therein have been scrutinized.For instance,many FMC plants,such as Panax ginseng C.A.Mey.,Camellia sinensis(Linn.)O.Ktze.,Ganoderma lucidum(Leyss.ex Fr.)Karst.,Lycium barbarum L.,etc.,display effects of enhancing immunity(Pan et al.,2023;Yao et al.,2023).The benefits of dietary interventions and phytometabolites on improving cognitive functions in elderly population have been supported by accumulating evidence(Heinrich et al.,2022).Therefore,food and medicine dual-use products are important in the prevention and intervention of chronic diseases and senile illnesses.Dacheng Hao Changxiao Liu 2023Chinese Herbal Medicines2023,15,1:0
