| 2 | 基因枪法获得优质HMW亚基基因表达的转基因小麦显示文摘为探讨利用基因工程技术进行小麦品质改良的可行性,用基因枪法对湖北省3个小麦品种鄂恩1号、鄂麦11号和鄂麦12号分别进行优质高分子量麦谷蛋白亚基(HMW-GS)基因1Ax1和1Dx5+1Dy10的转导。试验结果表明,经基因枪轰击的幼胚外植体的再生频率和转化频率明显比幼穗高;基因型间愈伤组织再生能力和转化频率的差异,直接影响着小麦转化的成功率;3个品种的幼胚发育时期与其再生频率均呈显著负相关(r=-0.93或-0.95);在可控环境条件下,花后12~14d的供试植株幼胚的转化频率最高(鄂恩1号、鄂麦11号和鄂麦12号分别为4.5%、2.9%和2%)。研究表明,鄂恩1号、鄂麦11号适龄期的幼胚试材,用等摩尔比的优势亚基基因1Dx5和1Dy10与选择标记基因以2:1摩尔比混合的沉淀物包裹金粉,经600~900psi氦气压轰击后,在含0.5mg·L-12,4-D的MS培养基中诱导愈伤、含0.1mg·L-12,4-D和5mg·L-1玉米素的R培养基中分化、3mg·L-1L-PPT选择压下继代分化筛选,可获得稳定可育的T0代转基因植株以及胚乳特异性表达的T1代转基因种子。 | 施农农 何光源 李克秀 Jones H.D. | 3 | A Review of Botulism in China显示文摘The epidemiological characteristics and etiology of botulisin in China, as well as the distribution of different types of Clustridium bolulinum in China; are described.Through 1989, 15 provinces and autonomous regions reported the occurrence of botulism.There were 2861 cases involved in 745 outbreaks.Among the cases 421 died, with a case fatality of 14.7%.The main epidemiological characteristics of botulism in China are: (i) the major foods causing botulism are homemade fermented bean products which accounted for 62.6% of the cases; (ii) the incubation period is longer (3 h-54 days) than that described in the western literature (mostly 2-7 days); (iii) the peak occurrence is from February to May; (iv) the progression of symptoms and signs is slower than that of western cases.All types of C.bolulinum.with the exception of type G, have been found in China.The distribution of various types of C.botnlinum is significantly different between southern and northern China; this is related to the latitude and is correlated with the prevalence of this disease.Most of the botulism outbreaks occurred above 30° north latitude in northern China and outbreaks rarely occurred below 30° north latitude.Nationwide surveys showed that the average detection rate of C bolulinum spores in soil and foods in the northern parts of China was 14.8%.while it was only 2.5% in the south.C.bolulinum types A, B, E.and F, which are involved in human botulism, were frequently found in the North, while types C and D, which are involved only in animal intoxication, were found more frequently in the south. 1990 Academic Press.Inc. | GAO Q.Y. | 4 | Effects of temperature and thermally-induced microstructure change on hydraulic conductivity of Boom Clay显示文摘Boom Clay is one of the potential host rocks for deep geological disposal of high-level radioactive nuclear waste in Belgium. In order to investigate the mechanism of hydraulic conductivity variation under complex thermo-mechanical coupling conditions and to better understand the thermo-hydromechanical(THM) coupling behaviour of Boom Clay, a series of permeability tests using temperaturecontrolled triaxial cell has been carried out on the Boom Clay samples taken from Belgian underground research laboratory(URL) HADES. Due to its sedimentary nature, Boom Clay presents acrossanisotropy with respect to its sub-horizontal bedding plane. Direct measurements of the vertical(Kv)and horizontal(Kh)hydraulic conductivities show that the hydraulic conductivity at 80℃ is about 2.4 times larger than that at room temperature(23℃), and the hydraulic conductivity variation with temperature is basically reversible during heatingecooling cycle. The anisotropic property of Boom Clay is studied by scanning electron microscope(SEM) tests, which highlight the transversely isotropic characteristics of intact Boom Clay. It is shown that the sub-horizontal bedding feature accounts for the horizontal permeability higher than the vertical one. The measured increment in hydraulic conductivity with temperature is lower than the calculated one when merely considering the changes in water kinematic viscosity and density with temperature. The nuclear magnetic resonance(NMR) tests have also been carried out to investigate the impact of microstructure variation on the THM properties of clay. The results show that heating under unconstrained boundary condition will produce larger size of pores and weaken the microstructure. The discrepancy between the hydraulic conductivity experimentally measured and predicted(considering water viscosity and density changes with temperature) can be attributed to the microstructural weakening effect on the thermal volume change behaviour of Boom Clay. Based on the experimental results, a hydraulic conductivity evolution model is proposed and then implemented in ABAQUS. Three-dimensional(3D) numerical simulation of the admissible thermal loading for argillaceous storage(ATLAS) Ⅲ in situ heating test has been conducted subsequently, and the numerical results are in good agreement with field measurements. | W.Z.
