| 2 | Application of a microseismic monitoring system in deep mining显示文摘A microseismic monitoring system was used in the Donggua Shan underground copper mine, and its application was intro- duced. The spacial distribution of the seismic event was monitored effectively during mining with this system. The distribution of the seismic intensity in different time periods and in the different mining districts was obtained via the clustering analysis of the monitored results, and the different intensity concentration districts of seismicity were compartmentalized. The various characteristics and wave- forms of different vibrations in the underground mine were revealed with the help of the micro-seismic monitoring system. | 3 | Recent Changes in Precipitation Extremes in the Heihe River Basin,Northwest China显示文摘Changes in rainfall extremes pose a serious and additional threat to water resources planning and management,natural and artificial oasis stability,and sustainable development in the fragile ecosystems of arid inland river basins. In this study,the trend and temporal variation of extreme precipitation are analyzed using daily precipitation datasets at 11 stations over the arid inland Heihe River basin in Northwest China from 1960 to 2011. Eight indices of extreme precipitation are studied.The results show statistically significant and large-magnitude increasing and decreasing trends for most indices,primarily in the Qilian Mountains and eastern Hexi Corridor. More frequent and intense rainfall extremes have occurred in the southern part of the desert area than in the northern portion. In general,the temporal variation in precipitation extremes has changed throughout the basin. Wet day precipitation and heavy precipitation days show statistically significant linear increasing trends and step changes in the Qilian Mountains and Hexi Corridor. Consecutive dry days have decreased obviously in the region in most years after approximately the late 1980 s,but meanwhile very long dry spells have increased,especially in the Hexi Corridor. The probability density function indicates that very long wet spells have increased in the Qilian Mountains. The East Asian summer monsoon index and western Pacific subtropical high intensity index possess strong and significant negative and positive correlations with rainfall extremes,respectively. | 4 | Influenza A virus use of BinCARD1 to facilitate the binding of viral NP to importinα7 is counteracted by TBK1-p62 axis-mediated autophagy显示文摘As a major component of the viral ribonucleoprotein(vRNP)complex in influenza A virus(IAV),nucleoprotein(NP)interacts with isoforms of importinαfamily members,leading to the import of itself and vRNP complex into the nucleus,a process pivotal in the replication cycle of IAV.In this study,we found that BinCARD1,an isoform of Bcl10-interacting protein with CARD(BinCARD),was leveraged by IAV for efficient viral replication.BinCARD1 promoted the nuclear import of the vRNP complex and newly synthesized NP and thus enhanced vRNP complex activity.Moreover,we found that BinCARD1 interacted with NP to promote NP binding to importinα7,an adaptor in the host nuclear import pathway.However,we also found that BinCARD1 promoted RIG-I-mediated innate immune signaling by mediating Lys63-linked polyubiquitination of TRAF3,and that TBK1 appeared to degrade BinCARD1.We showed that BinCARD1 was polyubiquitinated at residue K103 through a Lys63 linkage,which was recognized by the TBK1-p62 axis for autophagic degradation.Overall,our data demonstrate that IAV leverages BinCARD1 as an important host factor that promotes viral replication,and two mechanisms in the host defense system are triggered—innate immune signaling and autophagic degradation—to mitigate the promoting effect of BinCARD1 on the life cycle of IAV. | Xuyuan Wang Li Jiang Guangwen Wang Wenjun Shi Yuzhen Hu Bo Wang Xianying Zeng Guobin Tian Guohua Deng Jianzhong Shi Liling Liu Chengjun Li Hualan Chen | 2022 | Cellular & Molecular Immunology2022,19,10: | 3 |

However, accumulated evidences demonstrate that active function and trafficking of immune cells, including regulatory T cells, conventional T cells, B cells, dendritic cells, natural killer T (NKT) cells, neutrophils, myeloid-derived suppressor cells and mesenchymal stem cells, are observed in the bone marrow. Furthermore, bone marrow is a predetermined metastatic location for multiple human tumors. In this review, we discuss the immune network in the bone marrow. | 6 | TRIM28 protects CARM1 from proteasome-mediated degradation to prevent colorectal cancer metastasis显示文摘TRIM28 (Tripartite motif-containing protein 28), a member of TRIM family, is aberrantly expressed and reportedly has different functions in many types of human cancer. However, the biological roles of TRIM28 and related mechanism in colorectal cancer (CRC) remain unclear. Here, we showed that TRIM28 was downregulated in colorectal cancer compared with normal mucosa, especially at advanced stages, and acted as an independent prognostic factor of favorable outcome. Functional studies demonstrated that TRIM28 restrained CRC migration and invasion in vitro and in vivo. Mechanistically, we reported that CARM1 (co-activator-associated arginine methyltransferase1) was a critical player downstream of TRIM28. TRIM28 interacted with CARM1, and protected CARM1 from proteasome-mediated degradation through physical protein-protein interaction to suppress CRC metastasis. Further, TRIM28 suppressed the migration and invasion of CRC cells through inhibiting WNT/β-catenin signaling in a CARM1-dependent manner, but independent of CARM1′s methyltransferase activity. The protein expression of CARM1 was positively correlated with TRIM28 in CRC tissues. Patients with high levels of TRIM28 and CARM1 had improved prognosis, whereas patients with low TRIM28 and CARM1 expression had the poor outcomes. Thus, our study reveals an inhibitory role of TRIM28 in CRC metastasis, which was achieved through a TRIM28-CARM1-WNT/β-catenin axis. | 11 | Spatial Heterogeneity of Marine Environment Changes:A Case Study in Laizhou Bay and Its Adjacent Waters,China显示文摘Temporal and spatial heterogeneity identifications of marine environment and its changes have great significances in marine spatial planning and offshore pollution control.In this study,considering the integration of marine ecological environment and sea-land interaction,we built a spatialized evaluation model named Marine Environment Change Spatial Assessment(MECSA)to quantify the marine environment status and changes.In MECSA,we applied the geospatial model and the pressure-state-response(PSR)model for processing and integrating evaluation indicators.A case study in the Laizhou Bay showed that the marine environment quality was generally in a declining state from 2009 to 2015.In 2015,the Marine Environment Index(MEI)had decreased by 0.1 compared with 2009,although the two years all reached a‘Good'level.The spatial MEI layers of the two years showed a same distribution:the southwestern part was in poorer status,with a fan ring shape gradually getting better to the northeast.The Marine Organisms Response Index(MORI)contributed the most to the MEI.Therefore,future marine environmental assessment and spatial planning should focus on the identification the marine environment with its changes from the perspective of spatial heterogeneity and systemicity.Based on single indicators and comprehensive evaluation results,we can propose spatially targeted policies and recommendations scientifically. | WANG Youxiao YU Ge HU Guobin LIU Dahai | 2021 | Journal of Ocean University of China2021,20,1: | 0 |

| 14 | M6PR interacts with the HA2 subunit of influenza A virus to facilitate the fusion of viral and endosomal membranes显示文摘Influenza A virus(IAV) commandeers numerous host cellular factors for successful replication. However, very few host factors have been revealed to be involved in the fusion of viral envelope and late endosomal membranes. In this study, we identified cation-dependent mannose-6-phosphate receptor(M6PR) as a crucial host factor for the replication of IAV. We found that siRNA knockdown of M6PR expression significantly reduced the growth titers of different subtypes of IAV, and that the inhibitory effect of M6PR siRNA treatment on IAV growth was overcome by the complement of exogenously expressed M6PR. When A549 cells were treated with siRNA targeting M6PR,the nuclear accumulation of viral nucleoprotein(NP) was dramatically inhibited at early timepoints post-infection, indicating that M6PR engages in the early stage of the IAV replication cycle. By investigating the role of M6PR in the individual entry and post-entry steps of IAV replication, we found that the downregulation of M6PR expression had no effect on attachment, internalization, early endosome trafficking,or late endosome acidification. However, we found that M6PR expression was critical for the fusion of viral envelope and late endosomal membranes. Of note, M6PR interacted with the hemagglutinin(HA) protein of IAV, and further studies showed that the lumenal domain of M6PR and the ectodomain of HA2 mediated the interaction and directly promoted the fusion of the viral and late endosomal membranes,thereby facilitating IAV replication. | 20 | Presence of a long nuclear-localization signal sequence in homeodomain transcription factor Nkx 1.2显示文摘Homeodomains,a 60-amino acid sequence encoded by 180 nucleotides,are highly conserved DNA-binding motifs that are present in a variety of transcription factors in species ranging from yeast to humans.The NKX proteins belong to the homeodomain(HD)-containing transcription factor family.They play vital roles in the regulation of morphogenesis.NKX1-2 is one member of the NKX subfamily.At present,information about its nuclear localization signal(NLS)sequence is limited.We studied the NLS sequence of zebrafish Nkx1.2 by introducing sequence changes such as deletion,mutation,and truncation,and identified an NLS motif(QNRRTKWKKQ)that is localized at the C-terminus of the homeodomain.Moreover,the deletion of two amino acid residues(RR)in this NLS motif prevents Nkx1.2 from entering the nucleus,indicating that the two amino acids are essential for Nkx1.2 nuclear localization.However,the NLS motif alone is unable to target cytoplasmic protein glutathione S-transferase(GST)to the nucleus.An intact homeodomain is necessary for mediating the complete nuclear transport of cytoplasmic protein.Unlike most nuclear import proteins with short NLS sequences,a long NLS is present in zebrafish Nkx1.2.We also demonstrated that the sequences of homeodomain of NKX1.2 are well conserved among different species.This study is informative to verify the function of the NKX1.2 protein. | Xinyi LI Lihui CHEN Xinyuan WANG Chen SUN Guangdong JI Guobin HU Zhenhui LIU | 2024 | Journal of Oceanology and Limnology2024,42,2: | 0 |
