1DJ-1 is involved in the multidrug resistance of SGC7901 gastric cancer cells through PTEN/PI3K/Akt/Nrf2 pathway显示文摘Gastric cancer is a common malignancy worldwide.The occurrence of multidrug resistance(MDR)is the major obstacle for effective gastric cancer chemotherapy.In this study,the in-depth molec-ular mechanism of the DJ-1-induced MDR in SGC7901 gastric cancer cells was investigated.The results showed that DJ-1 expression level was higher in MDR variant SGC7901NCR cells than that in its parental SGC7901 cells.Moreover,DJ-1 overexpression conferred the MDR phenotype to SGC7901 cells,while DJ-1 knockdown in SGC7901NCR cells induced re-sensitization to adriamycin,vincristine,cisplatin,and 5-fluorouracil.These results suggested that DJ-1 mediated the develop-ment of MDR in SGC7901 gastric cancer cells.Importantly,further data revealed that the activation of PI3k/Akt and Nrf2 signaling pathway were required for the DJ-1-induced MDR phenotype in SGC7901 gastric cancer cells.Meanwhile,we found that Pl3k/Akt pathway was activated probably through DJ-1 directly binding to and negatively regulating PTEN,consequently resulting in Nrf2 phosphorylation and activation,and thereby inducing Nrf2-dependent P-glycoprotein(P-gp)and Bcl-2 expressions in the DJ-1-mediated MDR of SGC7901 gastric cancer cells.Overall,these results revealed that activating PTEN/PI3K/Akt/Nrf2 pathway and subsequently upregulating P-gp and Bcl-2 expression could be a critical mechanism by which DJ-1 mediates the development of MDR in SGC7901 gastric cancer cells.The new findings may be helpful for understanding the mechanisms of MDR in gastric cancer cells,prompting its further investigation as a molecular target to overcome MDR.Lejia Qiu Zhaoxia Ma Xiaoran Li Yizhang Deng Guangling Duan Le Zhao Xingwang Xu Lin Xiao Haoyue Liu Zhengming Zhu Heping Chen 2020Acta Biochimica et Biophysica Sinica2020,52,11:5
2Distribution and speciation of mercury in the Hongfeng Reservoir,Guizhou Province, China显示文摘The water, pore water, sediment, and fish samples were collected from the Hongfeng Reservoir in No- vember 2003 and February 2004 in accordance with trace metal protocols. The average concentrations of total mer- cury (THg), dissolved mercury (DHg), reactive mercury, dissolved gaseous mercury, total methylmercury, and dis- solved methylmercury in the water columns were 8.00, 5.70, 0.63, 0.05, 0.16, and 0.07 ng/L, respectively. THg and DHg in the water columns, THg in pore water and THg in lake sediments of the Hongfeng Reservoir showed the level of mercury in the Hongfeng Reservoir was higher than in other natural waters in the world due to the loading of a lot of waste water with relatively high concentrations of mercury, whereas methylmercury concentrations in fish (wet weight) varied from 1.73–51.00 ng/g, much lower than in most remote lakes and reservoirs reported in northern Europe and North America. Methylmercury distributions in pore water and sediments showed methylation oc- curred mainly in the upper several centimeters of sediment cores in the Hongfeng Reservoir. The concentrations of dissolved organic carbon, total suspended particles, total Hg, and methylmercury were higher at Houwu than those at Daba in November 2003. 3Evaluation of the potential risks of heavy metal contamination in rice paddy soils around an abandoned Hg mine area in Southwest China显示文摘To assess the potential ecological and health risks of trace elements(Hg,Cd,As,Mn,Sb,Pb,Cu,Ni,Cr,and Zn),a total of 138 soil samples from rice paddies were collected during the rice harvest season in the Wanshan mining area,Guizhou Province,Southwest China.Factors of the pollution load index(PLI),geo-accumulation index(I-Geo),enrichment factor(EF),and risk index(RI)were determined.High concentrations of Hg,Sb,As,Zn,Cd,Cu,and Mn were observed in the soils.The PLI,I-Geo,and EF results all showed high levels of contamination by Hg and Sb and moderate levels of contamination by As,Pb,Zn,Cu,Cd,and Mn.There was no significant contamination from Ni and Cr.The RI was very high,with Hg as the dominant pollutant,as expected,indicating that the historical large-scale Hg mining,as well as artisanal mining,has had a significant impact on the Wanshan area.Moreover,coal combustion,manganese factories,and the use of agrochemicals by the local population could also have an impact on the soil through the introduction of heavy metal loads.To address the current state of contamination,pollutant remediation and the regulation control of the anthropogenic activities in Wanshan are urgently needed.Bayou Adlane Zhidong Xu Xiaohang Xu Longchao Liang Jialiang Han Guangle Qiu 2020Acta Geochimica2020,39,1:5
4The influence of atmospheric Hg on Hg contaminations in rice and paddy soil in the Xunyang Hg mining district,China显示文摘To date, the Xunyang mercury(Hg) mining district is the only ongoing large-scale Hg mining district in China. To understand the influence of Hg contamination mode from the Hg mining and smelting activities, 27 sampling sites in the Xunyang Hg mining district were chosen in this study. Total gaseous mercury(TGM) in ambient air was measured using a Lumex-RA915 automatic Hg analyzer in2011. Rice samples and soil samples from rhizosphere were collected systematically and simultaneously. Total Hg(THg) and methylmercury(Me Hg) concentrations in rice grain and soil samples and Hg speciation with modified sequential selective extractions were measured. The local environment was seriously polluted with Hg. The TGM(302 ± 376 ng·m-3, ranging from 24 to 2220 ng·m^(-3)) in the local ambient air, THg(28 ± 30 mg·kg^(-1), ranging from 0.31 to 121 mg·kg^(-1)) and Me Hg(2.3 ± 1.9 lg·kg^(-1),ranging from 0.24 to 8.9 lg·kg^(-1)) in soil samples were at the sample level with Hg contaminated area. The THg concentration(26 ± 16 lg·kg^(-1)ranging from 4.5 to 71 lg·kg^(-1)) in most of the rice grain samples clearly exceeds the threshold level(20 lg·kg^(-1)) in the Chinese national guidelines for cereals(NY 861-2004). The inorganic mercury(IHg)(9.1 ± 5.6 lg·kg^(-1), ranging from 1.2 to 24 lg·kg^(-1)) and Me Hg(14 ± 9.8 lg·kg^(-1), ranging from 2.1 to 59 lg·kg^(-1))concentration in rice grain samples were at the same level with Hg contaminated area. The main species of Hg in paddy soils reveal strong complex Hg and residue Hg. 5Chemical characterization and sources of PM2.5 at 12-h resolution in Guiyang, China显示文摘The increasing emission of primary and gaseous precursors of secondarily formed atmospheric particulate matter due to continuing industrial development and urbanization are leading to an increased public awareness of environmental issues and human health risks in China.As part of a pilot study, 12-h integrated fine fraction particulate matter(PM_(2.5) filter samples were collected to chemically characterize and investigate the sources of ambient particulate matter in Guiyang City, Guizhou Province, southwestern China. Results showed that the 12-h integrated PM_(2.5) concentrations exhibited a daytime average of 51 ± 22 lg m^(-3)(mean ± standard deviation) with a range of 17–128 lg m^(-3) and a nighttime average of55 ± 32 lg m~(^(-3)) with a range of 4–186 lg m^(-3). The 24-h integrated PM_(2.5) concentrations varied from 15 to157 lg m~(^(-3)), with a mean value of 53 ± 25 lg m^(-3), which exceeded the 24-h PM_(2.5) standard of 35 lg m^(-3) set by USEPA, but was below the standard of 75 lg m^(-3), set by China Ministry of Environmental Protection. Energy-dispersive X-ray fluorescence spectrometry(XRF) was applied to determine PM_(2.5) chemical element concentrations. The order of concentrations of heavy metals in PM_(2.5) were iron(Fe) [ zinc(Zn) [ manganese(Mn) [ lead(Pb) [ arsenic(As) [ chromium(Cr). 6Comparison of in vitro digestion methods for determining bioaccessibility of Hg in rice of China显示文摘Rice consumption can be a major pathway for mercury(Hg) exposure, which can cause health hazards to Chinese residents. To accurately assess the potential health risks, the bioaccessibility of Hg in rice samples collected at background and Hg-contamination sites was examined using four in vitro methods. The results showed that < 50% of the total mercury(THg) was bioaccessible in the studied rice samples. The THg bioaccessibility in the rice samples collected at the Hg-contaminated site was higher than that observed at the background area. The bioaccessibility of arsenic(As) and cadmium(Cd) was also evaluated for comparison of the four in vitro methods used in this study. The Physiologically Based Extraction Test was found to be the most accurate method based on the consistency of the results compared to those reported in previous studies. The estimated daily intakes of THg via rice consumption using the bioaccessibility data were found to meet the recommendation value set by the JECFA and the WHO in both sites. 9Seasonal distribution of total mercury and methylmercury in sediments of the Wujiangdu Reservoir,Guizhou,China显示文摘The concentrations of total mercury and methylmercury in sediments were determined at the dam of the Wujiangdu Reservoir in different seasons. Total mercury (HgT) levels in the whole sediment profile were 254.2±47.0 ng/g in winter, 254.2±31.6 ng/g in spring, and 256.7±60.8 ng/g in summer, without significant variations in different seasons or at different depths. In contrast, the methylmercury (MeHg) compounds were most abundant at the sediment-water interface and decreased progressively with depth. MeHg contents of the sediments during different seasons are highly dependent on microbial activity, and seem to be higher when Hg (II)-methylating microorganisms are active. Thus, MeHg levels tend to rise in the loci where nutrient supplies and biological productivity are favorable.
