1粪菌移植应用于慢传输型便秘的临床疗效及疗效影响因素:一项前瞻性研究的长期随访结果显示文摘背景和目的肠道菌群参与结肠动力的调控,菌群失调是便秘是病因之一。粪菌移植被证实可以恢复肠道平衡。本研究旨在评估粪菌移植治疗慢传输型便秘(STC)的临床疗效和预后影响因素。方法:52例STC患者接受标准的粪菌移植入组研究,对其进行至少6个月的随访。记录入组患者的排便习惯、结肠传输时间、便秘相关的症状(PAC-SYM评分)、生活质量(PAC-QOL评分)、治疗满意度和不良反应。主要疗效指标是平均每周完全自主排便≥3次的患者比例。结果:治疗后34周、912周、2124周,每周完全自主排便≥3次患者的比例分别为50.0%、38.5%和32.7%,均较治疗前显著提高(均P<0.01)。其他排便情况、结肠传输时间、便秘相关症状和生活质量方面,治疗后亦有显著改善;但这些短期改善在治疗后12周和24周逐渐减小。治疗前排便不尽是唯一的疗效影响因素。治疗和随访期间未发现任何治疗相关的不良反应。结论:本研究显示粪菌移植治疗STC安全有效,但远期疗效有所降低。治疗前排便不尽感较少甚至没有的患者接受粪菌移植的治疗效果更佳。Chao Ding Wenting Fan Lili Gu Hongliang Tian Xiaolong Ge Jianfeng Gong Yongzhan Nie Ning Li 2018Gastroenterology Report2018,6,2:7

3Efficacy and Economic Evaluation of Nonbiological Artificial Liver Therapy in Acute-on-chronic Hepatitis B Liver Failure显示文摘Background and Aims:Nonbiological artificial liver(NBAL)is frequently used as a first-line treatment for hepatitis B virus-associated acute-on-chronic liver failure(HBV-ACLF).This study aimed to compare the therapeutic efficacy and cost-effectiveness ratio(CER)of comprehensive medical treatment,plasma exchange(PE),and double plasma molecular adsorption system(DPMAS)plus half-dose PE(DPMAS+PE)in patients with HBV-ACLF.Methods:A total of 186 patients with HBV-ACLF randomly received comprehensive medical treatment,PE,or DPMAS+PE and were prospectively evaluated.Patients were divided into four subgroups based on the pretreatment prothrombin activity(PTA):Group I(PTA>40%),group II(PTA 30–40%),group III(PTA 20–30%),and group IV(PTA<20%).The main outcome measures were 28 day effectiveness;90 day liver transplantation-free survival;change of biochemical parameters;and CER.Results:DPMAS+PE treatment was associated with significantly higher 28 day effectiveness and 90 day liver transplantation-free survival compared with PE treatment in patients with group I liver failure.Clearance of serum total bilirubin(TBIL),AST,and creatinine(Cr)were significantly higher in the DPMAS+PE group than in the PE group.For subjects with group I liver failure,DPMAS+PE treatment had advantages of lower CER values and better cost-effectiveness.Conclusions:Compared with comprehensive medical treatment and PE alone,DPMAS with halfdose sequential PE treatment more effectively improved TBIL,AST,and Cr in HBV-ACLF patients,improved 28 day effectiveness and 90 day survival rates in patients with group I liver failure,and was more cost effective.DPMAS+PE is a viable NBAL approach for treatment of HBV-ACLF.Cichun Wu Wenting Peng Da Cheng Huimin Gu Fei Liu Shifang Peng Lei Fu 2023Journal of Clinical and Translational Hepatology2023,11,2:5
4Effects of stability margin and thrust specific fuel consumption constrains on multi-disciplinary optimization for blended-wing-body design显示文摘Blended-Wing-Body(BWB) configuration, as an innovative transport concept, has become a worldwide research focus in the field of civil transports development. Relative to the conventional Tube-And-Wing(TAW) configuration, the BWB shows integrated benefits and serves as a most promising candidate for future ‘‘green aviation'. The objective of the present work is to figure out the effects of the stability margin and Thrust Specific Fuel Consumption(TSFC) on the BWB design in the framework of Multi-Disciplinary Optimization(MDO). A physically-based platform was promoted to study the effect static stability margin and engine technology level. Low-order physically based models are applied to the evaluation of the weight and the aerodynamic performance. The modules and methods are illustrated in detail, and the validation of the methods shows feasibility and confidence for the conceptual design of BWB aircrafts. In order to find out the relation between planform changes and the selection of stability and engine technology level, two sets of optimizations are conducted separately. The study proves that these two factors have dominant effects towards the optimized BWB designs in both aerodynamic shapes, weight distribution, which needs to be considered during the MDO design process. A balance diagram analysis is applied to find out a reasonable static stability margin range. 5Ex vivo 3D osteocyte network construction with primary murine bone cells显示文摘Osteocytes reside as three-dimensionally(3D) networked cells in the lacunocanalicular structure of bones and regulate bone and mineral homeostasis. Despite of their important regulatory roles, in vitro studies of osteocytes have been challenging because:(1) current cell lines do not sufficiently represent the phenotypic features of mature osteocytes and(2) primary cells rapidly differentiate to osteoblasts upon isolation. In this study, we used a 3D perfusion culture approach to:(1) construct the 3D cellular network of primary murine osteocytes by biomimetic assembly with microbeads and(2) reproduce ex vivo the phenotype of primary murine osteocytes, for the first time to our best knowledge. In order to enable 3D construction with a sufficient number of viable cells, we used a proliferated osteoblastic population of healthy cells outgrown from digested bone chips. The diameter of microbeads was controlled to:(1) distribute and entrap cells within the interstitial spaces between the microbeads and(2) maintain average cell-to-cell distance to be about 19 mm. The entrapped cells formed a 3D cellular network by extending and connecting their processes through openings between the microbeads. Also, with increasing culture time, the entrapped cells exhibited the characteristic gene expressions(SOST and FGF23) and nonproliferative behavior of mature osteocytes. In contrast, 2D-cultured cells continued their osteoblastic differentiation and proliferation. 6The effect of tensile and fluid shear stress on the in vitro degradation of magnesium alloy for stent applications显示文摘Magnesium alloys have gained great attention as biodegradable materials for stent applications.Cardiovascular stents are continuously exposed to different types of mechanical loadings simultaneously during service,including tensile,compressive and fluid shear stress.In this study,the in vitro degradation of WE43 wires was investigated under combined effect of tensile loading and fluid shear stress and compared with that experienced an individual loading condition.For the individual mechanical loading treatment,the degradation of magnesium wires was more severely affected by tensile loading than fluid shear stress.Under tensile loading,magnesium wires showed faster increment of corrosion rates,loss of mechanical properties and localized corrosion morphology with the increasing tensile loadings.With the combined stress,smaller variation of the corrosion rates as well as the slower strength degeneration was shown with increasing stress levels,in comparison with the individual treatment of tensile loading.This study could help to understand the effect of complex stress condition on the corrosion of magnesium for the optimization of biodegradable magnesium stents.Xue-Nan Gu Yun Lu Fan Wang Wenting Lin Ping Li Yubo Fan 2018Bioactive Materials2018,3,4:2

8S100 protein expression during induced Schwann cell-like cell differentiation of rat bone marrow mesenchymal cells in vitro显示文摘BACKGROUND: S100 protein can promote axonal growth. Therefore, transplantation of induced bone marrow-derived mesenchymal stem cells (MSCs) that can secrete S100 may provide a beneficial microenvironment for neural regeneration. OBJECTIVE: To explore the changes in S100 expression during rat MSCs differentiation into Schwann cells in vitro. DESIGN, TIME AND SETTING: This cytology experiment was performed at the Jiangsu Key Laboratory of Neuroregeneration, Nantong University in China, from January 2006 to May 2007. MATERIALS: The rabbit anti-S100 polyclonal antibody was purchased from Dako, Denmark; the mouse anti-rat S100 monoclonal antibody was purchased from Sigma, USA. METHODS: MSCs were cultured from adult Sprague-Dawley rat femur and tibia. Cell proliferation was determined by the MTT method and CD markers, and cell cycle was meas-ured by flow cytometry. MSCs were induced to differentiate into SC cells. SC cells were stained for S100 protein, glial fibrillary acidic protein, and low-affinity nerve growth factor receptor. S100 protein and mRNA levels were evaluated by flow cytometry, Western blot, and reverse transcription-polymerase chain reaction. MAIN OUTCOME MEASURES: S100 protein and mRNA expression. RESULTS: MSCs exhibited high amplification potential over eight passages. Prior to induction, the majority of MSCs were at the G0/G1 phase of the cell cycle. After induction, MSCs displayed morphology changes similar to Schwann cells. Moreover, induction increased S100 mRNA levels. Immunofluorescence showed that MSCs expressed S100 protein, glial fibrillary acidic protein, and low-affinity nerve growth factor receptor at 7 days of induction. Induction also increased S100 protein levels compared with untreated MSCs. 113-39 Distribution Characteristics of Soil Nutrients and Salt Content of Northwest Arid Region in Our Country--with the Industrial Park in Gansu as an Example显示文摘According to the poor nutrient and soil salinization of Wuwei industrial park in Gansu in Northwest arid region,we collected a large number of samples in the region and measured the soil salinity and nutrient content in 020cm. By the statistical analysis and comparison the values of the water content, pH, salinity, organic matter andavailable nitrogen phosphorus and potassium (NPK) in Table 1, our results indicated that: (1) In the sample region,besides the low water content, the degree of soil salinization is predominant and the majority of soil belong to themoderate or severe saline-alkali land. (2) Due to the low nutrient content, the soil is relatively poor. There areobvious differences between the soil organic matter and the available NPK[1]. In short, the nitrogen contents ofthe soil in the region are seriously rare. 13The Halo Concentration and Mass Relation Traced by Satellite Galaxies显示文摘We study the relation between halo concentration and mass(c-M relation) using galaxy catalogs of the Seventh and Eighth Data Releases of the Sloan Digital Sky Survey(SDSS DR7 and DR8). Assuming that the satellite galaxies follow the distribution of dark matter, we derive the halo concentration by fitting the satellite radial profile with a Nararro Frank and White(NFW) format. The derived c-M relation covers a wide halo mass range from10^(11.6)to 10^(14.1)M_(⊙). We confirm the anti-correlation between the halo mass and concentration as predicted in cosmological simulations. Our results are in good agreement with those derived using galaxy dynamics and gravitational lensing for halos of 10^(11.6)– 10^(12.9)M_(⊙), while they are slightly lower for halos of 10^(12.9)– 10^(14.1)M_(⊙).This is because blue satellite galaxies are less concentrated, especially in the inner regions. 14Nacelle-airframe integration design method for blended-wing-body transport with podded engines显示文摘Strong shock waves and flow separation often occur during the integration of nacelle and airframe for blended-wing-bodies with podded engines. To address this problem, this paper presents an integration method with numerical simulations. The philosophy of channeling flow and avoiding the throat effect on the nacelle and airframe is established based on the analysis of flow interference in the initial configuration. A parametric integration design method is proposed from twodimensional plane to three-dimensional space with control mechanisms and selection principles of the key parameters determined by their influences. Results show that strong shock waves and flow separation can be successfully eliminated under the influence of both the reshaped channel and decelerated inflow below the nacelle. Supersonic regions around the nacelle are effectively reduced, concentrating mainly on the lip position.
