6Inhibition of 12-lipoxygenase reduces proliferation and induces apoptosis of hepatocellular carcinoma cells in vitro and in vivo显示文摘BACKGROUND:12-lipoxygenase(12-LOX) has been reported to be an important gene in cancer cell proliferation and survival,and tumor metastasis.However,its role in hepatocellular carcinoma(HCC) cells remains unknown.METHODS:Expression of 12-LOX was assessed in a diethylnitrosamine-induced rat HCC model,and in SMMC-7721,HepG2 and L-02 cells using immunohistochemical staining and reverse transcriptase-polymerase chain reaction(RT-PCR).GST-π and Ki-67 were determined in vivo by immunohistochemical staining.Apoptosis was evaluated by TUNEL assay.Cell viability and apoptosis were determined by MTT assay and flow cytometry,respectively.Apoptosis-related proteins in SMMC-7721 and HepG2 cells were detected by Western blotting.RESULTS:Immunohistochemical staining and RT-PCR showed that 12-LOX was over-expressed in rat HCC and two HCC cell lines,while the expression was inhibited by baicalein,a specific inhibitor of 12-LOX.Baicalein inhibited cell proliferation and induced apoptosis in rat HCC and both cell lines in a dose-and time-dependent manner.Our in vivo study demonstrated that baicalein also reduced neoplastic nodules.Mechanistically,baicalein reduced Bcl-2 protein expression coupled with a slight increase of the expression of Bax and activation of caspase-3.Furthermore,baicalein inhibited the activation of ERK-1/2(phosphorylated).Interestingly,the effects of baicalein were reversed by 12(S)-HETE,a metabolite of 12-LOX.CONCLUSIONS:Inhibition of 12-LOX leads to reduced numbers of HCC cells,partially caused by increased apoptosis.12-LOX may be a potential molecular target for HCC prevention and treatment.Xi-Ming Xu,Guang-Jin Yuan,Jun-Jian Deng,Hong-Ting Guo,Miao Xiang,Fang Yang,Wei Ge and Shi-You Chen Cancer Center, Renmin Hospital of Wuhan University, Wuhan 430060, China Cancer Center, the 82nd Hospital of the Chinese PLA, Huai’an 223001, China Department of Physiology, Medical College of Wuhan University, Wuhan 430071, China Department of Physiology & Pharmacology, University of Georgia, Athens, GA 30602, USA 2012Hepatobiliary & Pancreatic Diseases International2012,11,2:20

8颈椎前路减压术合并椎动脉损伤显示文摘目的 探讨颈椎前路减压术中椎动脉损伤的危险因素及其处理方法。方法 1980年 1月~ 1998年 12月,对 720例患者行颈椎前路减压术,减压范围包括 1~ 3个椎间隙并行椎体次全切除。术中采用高速磨钻切除骨及椎间盘组织。 630例患者同时行颈椎前路 Caspar钢板内固定术。 3例行椎间减压时向侧方切除过多,导致椎动脉损伤,采用明胶海绵加游离肌片局部填塞止血。结果 720例中椎动脉损伤发生率为 0.4%, 3例均压迫止血成功,无一例出现脑缺血等神经系统症状。结论 颈椎前路减压时不应跨越钩椎关节,局部明胶海绵加游离肌片填塞是有效的椎动脉止血方法。任先军 Caspar W Fuchs GA 2000中华骨科杂志2000,20,8:16
9Change in subfoveal choroidal thickness after argon laser panretinal photocoagulation显示文摘AIM:To evaluate changes in subfoveal choroidal thickness(SFCT) and macular thickness as measured by enhanced depth imaging spectral-domain optical coherence tomography(EDI-OCT) after argon laser panretinal photocoagulation(PRP) in patients with severe diabetic retinopathy.·METHODS:This prospective,comparative case series included 21 patients(28 eyes) with severe diabetic retinopathy.All patients underwent three sessions of PRP.The SFCT and macular thickness were measured using EDI-OCT at baseline and one week after completion of 3 sessions of PRP.·RESULTS:SFCT before PRP was(318.1±96.5)μm and increased to(349.9 ±108.3)μm(P =0.001) after PRP.Macular thickness significantly increased at one week after PRP(from 273.1 ±23.9μm at baseline vs 295.8 ±25.3μm at one week;P <0.001).No significant relationship between the changes in macular thickness and SFCT was observed(r =-0.13,P =0.52).·CONCLUSION:PRP induced increases in both SFCT and macular thickness.Changes in SFCT did not correlate with changes in macular thickness.Ga Eun Cho Hee Yun Cho Yun Taek Kim 2013International Journal of Ophthalmology(English edition)2013,6,4:14

12The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 13Interferon-c and interleukin-4 detected in serum and saliva from patients with oral lichen planus显示文摘Our previous salivary study had demonstrated an apparent T helper 2(Th2)-predominance in saliva of oral lichen planus(OLP) patients and suggested a potential of salivary interleukin-4(IL-4) as a biomarker for monitoring disease severity. To further determine the consistency of Th1/Th2 bias of OLP, this study investigated the expression profile of interferon-c(IFN-c) and IL-4 in serum and the relationship of the serum levels of these cytokines with their saliva partners. Sixty ethnic Chinese patients with OLP(40 of the erythematous/ulcerative form and 20 of the reticular form) were recruited for this study, with 40 age–sex-matched healthy volunteers as control group. IFN-c and IL-4 levels in serum and paired saliva samples were screened by enzyme-linked immunosorbent assay. OLP patient showed a low-level IFN-c but high-level IL-4 expression profile in both serum and saliva, with a lower IFN-c/IL-4 ratio. Serum IL-4 level in the erythematous/ulcerative group was significantly higher than that in the reticular group. Serum levels of IFN-c and IL-4 were significantly and positively correlated with their saliva partners. 14Magnetic nanoparticles based cancer therapy: current status and applications显示文摘Nanotechnology holds a promising potential for developing biomedical nanoplatforms in cancer therapy. The magnetic nanoparticles, which integrate uniquely appealing features of magnetic manipulation, nanoscale heat generator, localized magnetic field and enzyme-mimics, prompt the development and application of magnetic nanoparticles-based cancer medicine. Considerable success has been achieved in improving the magnetic resonance imaging(MRI) sensitivity, and the therapeutic function of the magnetic nanoparticles should be given adequate attention. This work reviews the current status and applications of magnetic nanoparticles based cancer therapy. 15In-plane bistatic backward scattering from seabottom with randomly inhomogeneous sediment and rough interface显示文摘The in-plane bistatic backward scattering caused by seabottom with random inhomogeneities and rough interface is studied. This work combines the ray tube integral method of Ivakin et al. and the complex-ray/steepest descent method of Hines. For a given incident grazing angle, the theoretical expression can easily be used to calculate scattering strength for different backward scattering angles. (This is essential for modeling shallow water reverberation by the normal mode approach.) The model is tested against published scattering measurements. 16Honokiol:a promising small molecular weight natural agent for the growth inhibition of oral squamous cell carcinoma cells显示文摘Honokiol (HNK) is a small organic molecule purified from magnolia species and has demonstrated anticancer activities in a variety of cancer cell lines; however, its effect on oral squamous cell carcinoma (OSCC) cells is unknown. We investigated the antitumor activities of HNK on OSCC cells in vitro for the first time. The inhibitory effects of HNK on the growth and proliferation of OSCC cells were demonstrated via in vitro 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and propidium iodide (PI) assays, and the apoptotic cells were investigated by the observation of morphological changes and detection of DNA fragmentation via PI, TdT-mediated dUTP-biotin nick end labeling (TUNEL), and DNA ladder assays, as well as flow cytometry assay. The results showed that HNK inhibited the growth and proliferation of OSCC cells in vitro in a time and dose-dependent manner. The inhibitory effect was associated with the cell apoptosis induced by HNK, evidenced by the morphological features of apoptotic cells, TUNEL-positive cells and a degradation of chromosomal DNA into small internucleosomal fragments. The study also demonstrated here that the inhibition or apoptosis mediated by 15 μg·mL-1 or 20 μg·mL-1 of HNK were more stronger compared with those of 20 μg·mL-1 5-fluorouracil (5-Fu, the control) applied to OSCC cells, when the ratio of OSCC cell numbers were measured between the treatment of different concentrations of HNK to the 5-Fu treatment for 48 h. 17The Oral Microbiome Bank of China显示文摘The human microbiome project(HMP) promoted further understanding of human oral microbes. However, research on the human oral microbiota has not made as much progress as research on the gut microbiota. Currently, the causal relationship between the oral microbiota and oral diseases remains unclear, and little is known about the link between the oral microbiota and human systemic diseases. To further understand the contribution of the oral microbiota in oral diseases and systemic diseases, a Human Oral Microbiome Database(HOMD) was established in the US. The HOMD includes 619 taxa in 13 phyla, and most of the microorganisms are from American populations. Due to individual differences in the microbiome, the HOMD does not reflect the Chinese oral microbial status. Herein, we established a new oral microbiome database—the Oral Microbiome Bank of China(OMBC,http://gffzz1f92d53390b34408hwcpo0699cpn66b0x.ffgz.tsg.suse.edu.cn/ombc). Currently, the OMBC includes information on 289 bacterial strains and 720 clinical samples from the Chinese population, along with lab and clinical information. 18Analysis of combustion instability via constant volume combustion in a LOX/RP-1 bipropellant liquid rocket engine显示文摘Turbulent two-phase reacting flow in the chamber of LOX/RP-1 bipropellant liquid rocket engine is numerically investigated in this paper. The predicted pressure and mean axial velocity are qualitatively consistent with the experimental measurements. The self-excited pressure oscillations are obtained without any disturbance introduced through the initial and boundary conditions. It is found that amount of abrupt pressure peaks appear frequently and stochastically in the head regions of the chamber, which are the important sources to drive and strengthen combustion instability. Such abrupt pressures are induced by local constant volume combustion, because local combustible gas mixtures with high temperature are formed and burnt out suddenly due to some fuel droplets reaching their critical state in a rich oxygen surrounding. A third Damkhler number is defined as the ratio of the characteristic time of a chemical reaction to the characteristic time of a pressure wave expansion to measure the relative intensity of acoustic propagation and combustion process in thrusters. The analysis of the third Damkhler number distributions in the whole thrust chamber shows that local constant volume combustion happens in the head regions, while constant pressure combustion presents in the downstream regions. 19再发性低血糖致幼鼠脑损伤的实验研究显示文摘目的探讨再发性低血糖幼鼠脑Caspase-3表达及神经元变性的动态变化。方法将48只15日龄C57BL/6野生型小鼠,分为正常对照组(n=8)及低血糖组(n=40),再分为低血糖后12h,24h,48h,72h组,每组10只)。低血糖组给予短效胰岛素腹腔注射3次,每次剂量为5U/kg。采用免疫组化方法观察小鼠脑内caspase-3表达的动态变化;FJB染色观察小鼠脑内神经元变性的时程及分布情况。结果再发性低血糖后12h幼鼠脑内caspase-3表达即增多,24h达高峰,至72h仍高于正常对照组;开始阳性染色主要位于细胞质,以后逐渐向细胞核转移,caspase-3表达最强的部位是海马齿状回,皮质次之,海马CA1区仅见少量caspase-3阳性细胞。再发性低血糖后12h,脑内即可见FJB阳性细胞,24h及48hFJB阳性细胞逐渐增多,至72h阳性细胞最多,染色最强;从分布上看,纹状体内FJB阳性细胞数最多(尤以枕部皮质与纹状体交界处为著),皮质次之,海马回仅见少量FJB阳性细胞。结论再发性低血糖可致幼鼠脑损伤,神经元凋亡与坏死并存,易损部位为海马齿状回、枕部皮质与纹状体交界处,以选择性神经元死亡为主。辛颖 WERTHER GA RUSSO VC 2006中国现代医学杂志2006,16,5:6
20Effects of salinity on methane gas hydrate system显示文摘Using an approximately analytical formation, we extend the steady state model of the pure methane hydrate system to include the salinity based on the dynamic model of the methane hydrate system. The top and bottom boundaries of the methane hydrate stability zone (MHSZ) and the actual methane hy-drate zone (MHZ), and the top of free gas occurrence are determined by using numerical methods and the new steady state model developed in this paper. Numerical results show that the MHZ thickness becomes thinner with increasing the salinity, and the stability is lowered and the base of the MHSZ is shifted toward the seafloor in the presence of salts. As a result, the thickness of actual hydrate occur-rence becomes thinner compared with that of the pure water case. On the other hand, since lower solubility reduces the amount of gas needed to form methane hydrate, the existence of salts in sea-water can actually promote methane gas hydrate formation in the hydrate stability zone. Numerical modeling also demonstrates that for the salt-water case the presence of methane within the field of methane hydrate stability is not sufficient to ensure the occurrence of gas hydrate, which can only form when the methane concentration dissolved in solution with salts exceeds the local methane solubility in salt water and if the methane flux exceeds a critical value corresponding to the rate of diffusive methane transport.
