| 2 | The abnormal expression of retinoic acid receptor-β,p53 and Ki67 protein,n normal,premalignant and malignant esophageal tissues显示文摘AIM: Esophageal cancer remains a significant healthiproblem worldwide. It is important to investigate alterationsin expression of retinoic acid receptor-β, p53 and Ki67proteins in esophageal carcinogenesis.METHODS: To find biomarkers for early identification ofesophageal cancer, we analyzed the retinoic acid receptor-j3,p53 protein and the proliferation marker Ki67 in surgicalspecimens of normal, mildly, and severely dysplastic andmalignant esophageal tissues by in situ hybridization ofRNA and immunohistochemistry.RESULTS: RAR-β was expressed in 94.3 %(33/35) of normalmucosae, 67.8 %(19/28) of the mild, 58.1% (18/31) of thesevere ieaions and 53.2 % (116/218) of tumor samples. RAR-β mRNA was expressed in 62.7 % (42/67), 55.1% (43/78) and29.2 % (7/24) of well, moderated and poorly differentiatedSSCs. The p53 and Ki67 proteins were 5.9 % (2/34) of thenormal mucosa. P53 and Ki67 stained positively in 10.7 % (3/28) and 21.4 % (6/28) of mild dysplasia, and 51.6 %(16/31)and 58. 1% (18/31) of severely dysplasia respectively.Samples from esophageal cancer showed no higher levers ofp53 and Ki67 expression than seen in severely dysplasticlesions. | 6 | Os isotopic compositions of a cobalt-rich ferromanganese crust profile in Central Pacific显示文摘187Os/188Os ratios in all sub-layers of the Co-rich ferromanganese crust MP5D17 from Line Seamount in Central Pacific have been systematically determined. The 187Os/188Os values in this crust are highly variable, ranging from 0.4262 to 1.0155. From the bottom to the top, the os-mium isotopic ratios of the crust display a regular variation trend, which correlates well with its changes in structure and texture in the vertical profile. In comparison with the evolu-tion curve of Os isotopic composition in seawater over the past 80 Ma, a similar evolution tendency can be found with respect to the Os isotopic composition of the crust in this study. | 7 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. The results reveal that, with increasing cooling rate, both the spacings of primary and secondary dendrite arms decrease, and the dendrite morphologies transit from coarse to superfine dendrite. The secondary dendrite arms trend to be refined and be well developed, and the tertiary dendrite will occur. The predictions of the Kurz/Fisher model and the Hunt/Lu model accord basically with the experimental data for primary dendrite arm spacing. The regression equation of the primary dendrite arm spacings λ1 and the cooling rate Vc is λ1=0.013Vc-0.32. | 12 | Features of colorectal cancer in China stratified by anatomic sites:A hospital-based study conducted in university-affiliated hospitals from 2014 to 2018显示文摘Objective:The clinical and biological characteristics of colorectal cancer have been found to differ depending on the anatomic site of the cancer.However,for Chinese patients,there is limited information on the proportion of cases at each site and the related features.In this study,we explored the location,distribution and other features of colorectal cancers at each anatomic site in Chinese patients.Methods:We conducted a hospital-based study using hospitalization summary reports from 10 Peking University-affiliated hospitals from 2014 to 2018;the reports covered a total of 2,097,347 hospitalizations.Incident cases were chosen as the study population,and their epidemiological features were further analyzed.Results:A total of 20,739 colorectal cancer patients were identified.Rectum was the most common location(48.3%)of the cancer,whereas the proportions of patients with distal and proximal colon cancer were 24.5%and18.6%,respectively.Patients with rectal cancer were predominantly male and were the youngest for all anatomical sites(each P<0.001).The highest proportion of emergency admissions,the longest hospital stays and the highest hospitalization costs were found in patients with proximal colon cancer(each P<0.001).The proximal colon cancer subgroup included the highest proportions of patients with medical histories of cholecystectomy,cholecystolithiasis and/or gallbladder polyps and appendectomy(P=0.009,P<0.001 and P<0.001,respectively).The distal colon cancer subgroup included the highest proportions of patients with medical histories of diabetes and hypertension(P<0.001,respectively).Conclusions:The patterns of colorectal cancer observed in this study differ from those reported for Western patients and show a significantly higher proportion of patients with rectal cancer.Different epidemiological features were also found based on anatomic sites.Further studies based on tumor location should be conducted to facilitate more accurate screening and treatment. | Ruize Qu Yanpeng Ma Liyuan Tao Xiaoyuan Bao Xin Zhou Bingyan Wang Fei Li Siyi Lu Lin Tuo Siyan Zhan Zhipeng Zhang Wei Fu | 2021 | Chinese Journal of Cancer Research2021,33,4: | 10 |

| 14 | A 2-kb Mycovirus Converts a Pathogenic Fungus into a Beneficial Endophyte for Brassica Protection and Yield Enhancement显示文摘Mycoviruses are viruses that infect fungi,and hypovirulence-associated mycoviruses have the potential to control fungal diseases.However,it is unclear how mycovirus-mediated hypovirulent strains live and survive in the field,and no mycovirus has been applied for field crop protection.In this study,we found that a previously identified small DNA mycovirus(SsHADV-1)can convert its host,Sclerotinia sclerotiorum,from a typical necrotrophic pathogen to a beneficial endophytic fungus.SsHADV-1 downregulates the expression of key pathogenicity factor genes in S.sclerotiorum during infection.When growing in rapeseed,the SsHADV-1-infected strain DT-8 significantly regulates the expression of rapeseed genes involved in defense,hormone signaling,and circadian rhythm pathways.As a result,plant growth is promoted and disease resistance is enhanced.Field experiments showed that spraying DT-8 at the early flowering stage can reduce the disease severity of rapeseed stem rot by 67.6%and improve yield by 14.9%.Moreover,we discovered that SsHADV-1 could also infect other S.sclerotiorum strains on DT-8-inoculated plants and that DT-8 could be recovered from dead plants.These findings suggest that the mycoviruses may have the ability to shape the origin of endophytism.Our discoveries suggest that mycoviruses may influence the origin of endophytism and may also offer a novel strategy for disease control in which mycovirus-infected strains are used to improve crop health and release mycoviruses into the field. | Hongxiang Zhang Jiatao Xie Yanping Fu Jiasen Cheng Zheng Qu Zhenzhen Zhao Shufen Cheng Tao Chen Bo Li Qianqian Wang Xinqiang Liu Binnian Tian David B.Collinge Daohong Jiang | 2020 | Molecular Plant2020,13,10: | 9 |

| 17 | Six-long non-coding RNA signature predicts recurrence-free survival in hepatocellular carcinoma显示文摘BACKGROUND Recent evidence shows that long non-coding RNAs(lncRNAs) are closely related to hepatogenesis and a few aggressive features of hepatocellular carcinoma(HCC). Increasing studies demonstrate that lncRNAs are potential prognostic factors for HCC. Moreover, several studies reported the combination of lncRNAs for predicting the overall survival(OS) of HCC, but the results varied. Thus,more effort including more accurate statistical approaches is needed for exploring the prognostic value of lncRNAs in HCC.AIM To develop a robust lncRNA signature associated with HCC recurrence to improve prognosis prediction of HCC.METHODS Univariate COX regression analysis was performed to screen the lncRNAs significantly associated with recurrence-free survival(RFS) of HCC in GSE76427 for the least absolute shrinkage and selection operator(LASSO) modelling. The established lncRNA signature was validated and developed in The Cancer Genome Atlas(TCGA) series using Kaplan-Meier curves. The expression values of the identified lncRNAs were compared between the tumor and non-tumor tissues. Pathway enrichment of these lncRNAs was conducted based on the significantly co-expressed genes. A prognostic nomogram combining the lncRNA signature and clinical characteristics was constructed.RESULTS The lncRNA signature consisted of six lncRNAs: MSC-AS1, POLR2 J4, EIF3 J-AS1,SERHL, RMST, and PVT1. This risk model was significantly associated with the RFS of HCC in the TCGA cohort with a hazard ratio(HR) being 1.807(95%CI[confidence interval]: 1.329-2.457) and log-rank P-value being less than 0.001. The best candidates of the six-lncRNA signature were younger male patients with HBV infection in relatively early tumor-stage and better physical condition but with higher preoperative alpha-fetoprotein. All the lncRNAs were significantly upregulated in tumor samples compared to non-tumor samples(P < 0.05). | 18 | New Re-Os Isotopic Constrains on the Formation of the Metalliferous Deposits of the Lower Cambrian Niutitang Formation显示文摘The Terreneuvian Epoch(541–521 Ma)is also an important period for metallogenesis in South China, as was represented by the widespread occurrences of Ni-Mo polymetallic layers on the antecedent shallow platform margin and the V-enriched black shales in deeper slope-basin settings. In this study, we have measured Re-Os isochron ages of Ni-Mo polymetallic layers(Songlin, Niuchang, Sancha, Chuanpengwan), V-rich black shales(Bahuang), and non-metalliferous black shales(Shuidong) in the basal Niutitang Formation in Guizhou and Hunan province, South China. The Ni-Mo polymetallic layers and V-enriched black shales have similar Re-Os isochron ages, suggesting concurrent deposition of these two types of metalliferous ores. This suggestion is consistent with the traditional stratigraphic correlation by using the nodular phosphorite bed directly underlying these metalliferous layers as a stratigraphic marker. Furthermore, the metalliferous ores and non-metalliferous black shales have similar initial ^(187)Os/^(188) Os ratios of 0.8–0.9, arguing for a dominant seawater origin with minor contribution of hydrothermal activity. Furthermore, Re-Os isotopic data also imply that Ni-Mo and V ore might have derived from the same source. We suggest that the spatial distribution of metalliferous ores can be explained by the development of non-sulfidic anoxic-suboxic wedge(NSASW) in the slope-basin and sulfidic wedge in the previous platform margin. Upwelling of deep water first transects the mildly reduced, organic rich NSASW, in which V(V) is reduced to V(IV), and is preferentially removed from seawater by organometallic complex formation. As a result, V-rich black shale deposits in the slope-basin of Yangtze Platform. Based on constructive techniques, we give an accurate enumeration formula for n-variable balanced RSBFs when n is a power of a prime. Furthermore, an original and efficient method to enumerate all n-variable (n prime) 1st order correlationimmune functions is presented. The exact number of 1st order correlation immune RSBFs with 11 variables is 6925047156550478825225250374129764511077684773805520800 and the number of 13 variables has 189 digits. | 20 | CAS9 is a genome mutator by directly disrupting DNA-PK dependent DNA repair pathway显示文摘With its high efficiency for site-specific genome editing and easy manipulation,the clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR associated protein 9(CAS9)system has become the most widely used gene editing technology in biomedical research.In addition,significant progress has been made for the clinical development of CRISPR/CAS9 based gene therapies of human diseases,several of which are entering clinical trials.Here we report that CAS9 protein can function as a genome mutator independent of any exogenous guide RNA(gRNA)in human cells,promoting genomic DNA double-stranded break(DSB)damage and genomic instability.CAS9 interacts with the KU86 subunit of the DNA-dependent protein kinase(DNA-PK)complex and disrupts the interaction between KU86 and its kinase subunit,leading to defective DNA-PK-dependent repair of DNA DSB damage via non-homologous end-joining(NHEJ)pathway.XCAS9 is a CAS9 variant with potentially higher fidelity and broader compatibility,and dCAS9 is a CAS9 variant without nuclease activity.We show that XCAS9 and dCAS9 also interact with KU86 and disrupt DNA DSB repair.Considering the critical roles of DNA-PK in maintaining genomic stability and the pleiotropic impact of DNA DSB damage responses on cellular proliferation and survival,our findings caution the interpretation of data involving CRISPR/CAS9-based gene editing and raise serious safety concerns of CRISPR/CAS9 system in clinical application. | Shuxiang Xu Jinchul Kim Qingshuang Tang Qu Chen Jingfeng Liu Yang Xu Xuemei Fu | 2020 | Protein & Cell2020,11,5: | 8 |
