| 2 | Study of intracellular anabolism of 5-fluorouracil and incorporation in nucleic acids based on an LC-HRMS method显示文摘5-Fluorouracil(5-FU)is an anticancer drug extensively used for different cancers.Intracellular metabolic activation leads to several nucleoside and nucleotide metabolites essential to exert its cytotoxic activity on multiple cellular targets such as enzymes,DNA and RNA.In this paper,we describe the development of a method based on liquid chromatography coupled with high resolution mass spectrometry suitable for the simultaneous determination of the ten anabolic metabolites(nucleoside,nucleotide and sugar nucleotide)of 5-FU.The chromatographic separationwas optimized on a porous graphitic carbon column allowing the analysis of the metabolites of 5-FU as well as endogenous nucleotides.The detection was performed on an Orbitrap■tandem mass spectrometer.Linearity of the method was verified in intracellular content and in RNA extracts.The limit of detection was equal to 12 pg injected on column for nucleoside metabolites of 5-FU and 150 pg injected on column for mono-and tri-phosphate nucleotide metabolites.Matrix effect was evaluated in cellular contents,DNA and RNA extracts for nucleoside and nucleotides metabolites.The method was successfully applied to i)measure the proportion of each anabolic metabolite of 5-FU in cellular contents,ii)follow the consequence of inhibition of enzymes on the endogenous nucleotide pools,iii)study the incorporation of metabolites of 5-FU into RNA and DNA,and iv)to determine the incorporation rate of 5-FUrd into 18 S and 28 S sub-units of rRNA. | Christelle Machon Frederic Catez Nicole Dalla Venezia Floriane Vanhalle Laetitia Guyot Anne Vincent Maxime Garcia Beatrice Roy Jean-Jacques Diaz Jerome Guitton | 2021 | Journal of Pharmaceutical Analysis2021,11,1: | 1 |

| 7 | Induced structural modifications in ZnS nanowires via physical state of catalyst:Highlights of 15R crystal phase显示文摘Peculiar and unique growth mechanisms involved in semiconductor nanowires(NWs)pave the way to the achievement of new crystallographic phases and remarkable material properties,and hence,studying polytypism in semiconductor NWs arouses a strong interest for the next generation of electronic and photonic applications.In this context,the growth of ZnS nanowires has been investigated,as bulk ZnS compound exhibits numerous unstable polytypes at high temperatures,but their stable occurrence is highly anticipated in a nanowire due to its special quasi-dimensional shape and growth modes.In this work,the idea is to provide a change in the growth mechanism via the physical state of catalyst droplet(liquid or solid)and hence,study the induced structural modifications in ZnS nanowires.The HRTEM images of VLS(via liquid alloyed catalyst)grown ZnS NWs show periodic stacking faults,which is precisely identified as a stacking sequence of cubic or hexagonal individual planes leading to an astonishing 15R crystal polymorph.This crystallographic phase is observed for the first time in nanowires.Contrastingly,NWs grown with VSS(via solid catalyst)show crystal polytypes of zinc blende and wurtzite.We calculate and discuss the role of cohesive energies in the formation of such ZnS polytypes.Further,we present the selection rules for the crystallization of such 15R structure in NWs and discuss the involved VLS and VSS growth mechanisms leading to the formation of different crystal phases. | Sumit Kumar Frederic Fossard Gaelle Amiri Jean-Michel Chauveau Vincent Sallet | 2022 | Nano Research2022,15,1: | 1 |

| 13 | Territorial Accessibility and Decision-Making Structure Related to Debris Flow Impacts on Roads in the French Alps显示文摘The Alps are highly impacted by debris flows that cause major problems for companies and transport networks located in the valley bottoms. One such event occurred in the Rif Blanc catchment and affected the road network in the French Alps, as well as adjacent areas across the Italian border, for several days in June 2012. This article presents two independent approaches to vulnerability assessment. Based on investigations conducted during a survey of local authorities following the event, we compared theoretical risk management and real crisis management in terms of decision making and modes of intervention. Functional vulnerability and territorial consequences were analyzed using a best travel time model of accessibility. We show that a bottom-up approach is practiced in case of actual management planning with a central coordination of general council. Conversely theoretical crisis management shows prefect as the key actor supported by several other state institutions. Our analysis also revealed that a debris flow event with a local impact on the road network has territorial consequences at a regional scale. This study contributes to the discussion about how to minimize the vulnerability of alpine transport networks prone to debris flows. | 19 | Livestock induces strong spatial heterogeneity of soil CO2, N2O and CH4 emissions within a semi-arid sylvo-pastoral landscape in West Africa显示文摘Greenhouse gas(GHG) emissions from the surface soils and surface water receiving animal excreta may be important components of the GHG balance of terrestrial ecosystems, but the associated processes are poorly documented in tropical environments, especially in tropical arid and semi-arid areas. A typical sylvo-pastoral landscape in the semi-arid zone of Senegal, West Africa, was investigated in this study. The study area(706 km2 of managed pastoral land) was a circular zone with a radius of 15 km centered on a borehole used to water livestock. The landscape supports a stocking rate ranging from 0.11 to 0.39 tropical livestock units per hectare depending on the seasonal movements of the livestock. Six landscape units were investigated(land in the vicinity of the borehole, natural ponds, natural rangelands, forest plantations, settlements, and enclosed plots). Carbon dioxide(CO_2), nitrous oxide(N_2O) and methane(CH_4) fluxes were measured with static chambers set up at 13 sites covering the six landscape units, and the 13 sites are assumed to be representative of the spatial heterogeneity of the emissions. A total of 216 fluxes were measured during the one-year study period(May 2014 to April 2015). At the landscape level, soils and surface water emitted an average 19.8 t C-CO_2 eq/(hm2·a)(CO_2: 82%, N_2O: 15%, and CH_4: 3%), but detailed results revealed notable spatial heterogeneity of GHG emissions. CO_2 fluxes ranged from 1148.2(±91.6) mg/(m2·d) in rangelands to 97,980.2(±14,861.7) mg/(m2·d) in surface water in the vicinity of the borehole. N_2O fluxes ranged from 0.6(±0.1) mg/(m2·d) in forest plantations to 22.6(±10.8) mg/(m2·d) in the vicinity of the borehole. CH_4 fluxes ranged from –3.2(±0.3) mg/(m2·d) in forest plantations to 8788.5(±2295.9) mg/(m2·d) from surface water in the vicinity of the borehole. This study identified GHG emission 'hot spots' in the landscape. | 20 | Exploring the effects of dietary inulin in rainbow trout fed a high‑starch,100%plant‑based diet显示文摘Background High dietary carbohydrates can spare protein in rainbow trout(Oncorhynchus mykiss)but may affect growth and health.Inulin,a prebiotic,could have nutritional and metabolic effects,along with anti-inflammatory properties in teleosts,improving growth and welfare.We tested this hypothesis in rainbow trout by feeding them a 100%plant-based diet,which is a viable alternative to fishmeal and fish oil in aquaculture feeds.In a two-factor design,we examined the impact of inulin(2%)as well as the variation in the carbohydrates(CHO)/plant protein ratio on rainbow trout.We assessed the influence of these factors on zootechnical parameters,plasma metabolites,gut microbiota,production of short-chain fatty acids and lactic acid,as well as the expression of free-fatty acid receptor genes in the mid-intestine,intermediary liver metabolism,and immune markers in a 12-week feeding trial.Results The use of 2%inulin did not significantly change the fish intestinal microbiota,but interestingly,the high CHO/protein ratio group showed a change in intestinal microbiota and in particular the beta diversity,with 21 bacterial genera affected,including Ralstonia,Bacillus,and 11 lactic-acid producing bacteria.There were higher levels of butyric,and valeric acid in groups fed with high CHO/protein diet but not with inulin.The high CHO/protein group showed a decrease in the expression of pro-inflammatory cytokines(il1b,il8,and tnfa)in liver and a lower expression of the genes coding for tight-junction proteins in mid-intestine(tjp1a and tjp3).However,the 2%inulin did not modify the expression of plasma immune markers.Finally,inulin induced a negative effect on rainbow trout growth performance irrespective of the dietary carbohydrates.Conclusions With a 100%plant-based diet,inclusion of high levels of carbohydrates could be a promising way for fish nutrition in aquaculture through a protein sparing effect whereas the supplementation of 2%inulin does not appear to improve the use of CHO when combined with a 100%plant-based diet. | Raphaël Defaix Jep Lokesh Laura Frohn Mickael Le Bechec Thierry Pigot Vincent Veron Anne Surget Sandra Biasutti Frederic Terrier Sandrine Skiba-Cassy Jerome Roy Stephane Panserat Karine Ricaud | 2024 | Journal of Animal Science and Biotechnology2024,15,2: | 0 |
