| 2 | Genetics of hepatocellular carcinoma显示文摘The completely assembled human genome has made it possible for modern medicine to step into an era rich in genetic information and high-throughput genomic analysis. These novel and readily available genetic resources and analytical tools may be the key to unravel the molecular basis of hepatocellular carcinoma (HCC). Moreover, since an efficient treatment for this disease is lacking, further understanding of the genetic background of HCC will be crucial in order to develop new therapies aimed at selected targets. We report on the current status and recent developments in HCC genetics. Special emphasis is given to the genetics and regulation of major signalling pathways involved in HCC such as p53, Wnt- signalling, TGFβ, Ras, and Rb pathways. Furthermore, we describe the influence of chromosomal aberrations as well as of DNA methylation. | 3 | 天津蓟县铁岭组新剖面钾质斑脱岩锆石U-Pb测年及Hf同位素研究显示文摘天津蓟县西北的大喻山铁岭组剖面钾质斑脱岩夹层锆石U-Pb年龄(1439±14Ma)是约束'蓟县剖面'年代格架的重要锚点,但该剖面露头已遭到严重破坏。最近,笔者在蓟县东北、目前已禁止开采的大五尖采石场发现了铁岭组新剖面,并在其二段近底部再次确认多层钾质斑脱岩,测得其中两层LA-ICPMS锆石U-Pb年龄(1445±12Ma以及1442±10Ma);采自原大喻山剖面该组二段近底部斑脱岩夹层的样品,也获得其LA-ICPMS锆石U-Pb年龄(1439±11Ma)。以上三个年龄值在误差范围内一致,与之前在原大喻山剖面及河北平泉刘家沟剖面该组二段近底部斑脱岩夹层所获的SHRIMP锆石U-Pb年龄(1439±14Ma及1437±21 Ma)也完全可比。与此同时,本文对以上3个样品锆石还开展了锆石原位Hf同位素研究。其结果显示,176Hf/177Hf值变化范围为0. 281703~0. 281880,εHf(t)=-6. 7^-0. 2,两阶段模式年龄为2200~2603Ma,高斯分布峰值为~2360Ma。这表明,铁岭组钾质斑脱岩的物源主要来自于新太古代-古元古代早期地壳物质的部分熔融。本研究为蓟县剖面乃至燕山地区铁岭组高精度年代约束及地层格架厘定等,提供了更多年代学佐证,进一步夯实了整个蓟县剖面作为华北克拉通中-新元古界标准剖面的科学性和代表性;同时,对华北中元古界钾质斑脱岩夹层的锆石首次开展Lu-Hf同位素研究,为探讨这一特殊类型沉积岩的源岩及相关研究提供了新思路;新发现剖面也为未来继续围绕铁岭组的各项研究,提供了一处露头良好、年代框架扎实可靠、可长期观测的新平台。 | 郭文琳 苏文博 张健 李惠民 周红英 李怀坤 Frank R ETTENSOHN Warren DHUFF | 2019 | 岩石学报2019,35,8: | 16 |
| 4 | Pharmacogenomics of EGFR-targeted therapies in non-small cell lung cancer:EGFR and beyond显示文摘Commonly observed aberrations in epidermal growth factor receptor(EGFR) signaling have led to the development of EGFR-targeted therapies for various cancers,including non-small cell lung cancer(NSCLC).EGFR mutations and overexpression have further been shown to modulate sensitivity to these EGFR-targeted therapies in NSCLC and several other types of cancers.However,it is clear that mutations and/or genetic variations in EGFR alone cannot explain all of the variability in the responses of patients with NSCLC to EGFR-targeted therapies.For instance,in addition to EGFR genotype,genetic variations in other members of the signaling pathway downstream of EGFR or variations in parallel receptor tyrosine kinase(RTK) pathways are now recognized to have a significant impact on the efficacy of certain EGFR-targeted therapies.In this review,we highlight the mutations and genetic variations in such genes downstream of EGFR and in parallel RTK pathways.Specifically,the directional effects of these pharmacogenetic factors are discussed with a focus on two commonly prescribed EGFR inhibitors:cetuximab and erlotinib.The results of this comprehensive review can be used to optimize the treatment of NSCLC with EGFR inhibitors.Furthermore,they may provide the rationale for the design of subsequent combination therapies that involve the inhibition of EGFR. | Christopher Delaney Samuel Frank R Stephanie Huang | 2015 | Chinese Journal of Cancer2015,34,4: | 6 |

| 6 | Role of ^(18)F-FDG PET/CT in pre and post treatment evaluation in head and neck carcinoma显示文摘Head and neck cancer(HNC) ranks as the 6th most common cancer worldwide, with the vast majority being head and neck squamous cell carcinoma(HNSCC). The majority of patients present with complicated locally advanced disease(typically stage Ⅲ and Ⅳ) requiring multidisciplinary treatment plans with combinations of surgery, radiation therapy and chemotherapy. Tumor staging is critical to decide therapeutic planning. Multiple challenges include accurate tumor localization with precise delineation of tumor volume, cervical lymph node staging, detection of distant metastasis as well as ruling out synchronous second primary tumors. Somepatients present with cervical lymph node metastasis without obvious primary tumors on clinical examination or conventional cross sectional imaging. Treatment planning includes surgery, radiation, chemotherapy or combinations that could significantly alter the anatomy and physiology of this complex head and neck region, making assessment of treatment response and detection of residual/ recurrent tumor very difficult by clinical evaluation and computed tomography(CT) or magnetic resonance imaging(MRI). 18F-2-fluoro-2-deoxyD-glucose positron emission tomography/CT(18F-FDG PET/CT) has been widely used to assess HNC for more than a decade with high diagnostic accuracy especially in detection of initial distant metastasis and evaluation of treatment response. There are some limitations that are unique to PET/CT including artifacts, lower soft tissue contrast and resolution as compared to MRI, false positivity in post-treatment phase due to inflammation and granulation tissues, etc. The aim of this article is to review the roles of PET/CT in both pre and post treatment management of HNSCC including its limitations that radiologists must know. In contrast to other pancreatic tumors,the solid-pseudopapillary tumor has a favorable prognosis.The 60-year-old female patient we report on here was treated by left pancreatic resection combined with splenectomy for a non-metastasizing tumor of the pancreas. A solid-pseudopapillary tumor was found on histology. The patient had no signs of metastases at present.Since a microscopically invasive tumor growth is assumed,oncologically curative resection should be preferred vs the less radical enucleation. | 9 | Differential response of tomato genotypes to Xanthomonas-specific pathogen-associated molecular patterns and correlation with bacterial spot (Xanthomonas perforans) resistance显示文摘Plants depend on innate immune responses to retard the initial spread of pathogens entering through stomata,hydathodes or injuries.These responses are triggered by conserved patterns in pathogen-encoded molecules known as pathogen-associated molecular patterns(PAMPs).Production of reactive oxygen species(ROS)is one of the first responses,and the resulting‘oxidative burst’is considered to be a first line of defense.In this study,we conducted association analyses between ROS production and bacterial spot(BS;Xanthomonas spp.)resistance in 63 genotypes of tomato(Solanum lycopersicum L.).A luminol-based assay was performed on leaf tissues that had been treated with a flagellin 22(flg22),flagellin 28 and a Xanthomonas-specific flg22(flg22-Xac)peptide,to measure PAMP-induced ROS production in each genotype.These genotypes were also assessed for BS disease response by inoculation with Xanthomonas perforans,race T4.Although there was no consistent relationship between peptides used and host response to the BS,there was a significant negative correlation(r=−0.25,P<0.05)between foliar disease severity and ROS production,when flg22-Xac was used.This response could potentially be used to identify the Xanthomonas-specific PRR allele in tomato,and eventually PAMP-triggered immunity loci could be mapped in a segregating population.This has potential significance in tomato improvement. | Krishna Bhattarai Frank J Louws John D Williamson Dilip R Panthee | 2016 | Horticulture Research2016,3,1: | 3 |

| 14 | Cholangiocarcinoma, gone without the Wnt?显示文摘Cholangiocarcinoma(CCA) is a relatively rare malignancy of the intra- or extra-hepatic bile ducts that is classified according to its anatomical localization as intrahepatic, perihilar or distal. Overall, CCA has a dismal prognosis due to typical presentation at an advanced irresectable stage, lack of effective non-surgical treatments, and a high rate of disease recurrence. CCA frequently arises on a background of chronic liver inflammation and cholestasis. Chronic inflammation is accompanied by enhanced cell turnover with generation of additional inflammatory stimuli, and a microenvironment rich in pro-inflammatory mediators and proliferative factors that enable accumulation of mutations, transformation and expansion of mutated cells. A recent study by Boulter et al implicates the Wnt signaling cascade in cholangiocarcinogenesis. Wnt ligands Wnt7 B and Wnt10 A were found to be highly overexpressed in human CCA tissue. Wnt7 B protein was present throughout the tumor stroma, and often co-localized with a subset of CD68+ macrophages. To address in a direct manner whether Wnt signaling is engaged in development of CCA, Boulter et al explored the Wnt signaling pathway in an experimental model that recapitulates the multi-stage progression of human CCA.Wnt ligands found to be elevated in human CCA were also upregulated during the course of CCA development following thioacetamide treatment. Wnt10 a increased during the(pre-cancerous) regenerative phase, while Wnt7 b induction paralleled tumor growth. Along with upregulation of target genes, the findings demonstrate that the canonical Wnt pathway is progressively activated during cholangio-carcinogenesis. Macrophage depletion,eliminating a major source of Wnt7 b, prevented activation of the canonical Wnt cascade, and resulted in reduced number and volume of tumors in this model. Moreover,specific inhibitors of the canonical Wnt pathway(ICG-001 and C-59) caused reduction of tumor area and number,in xenograft and thioacetamide models of CCA. The aggregated findings show that experimental, and presumably human CCA, is a Wnt-driven tumor.
