1Gut microbiome alterations and its link to corticosteroid resistance in immune thrombocytopenia显示文摘Quantitative metagenomic studies have linked the gut microbiota to autoimmune disorders.Here,we performed deep shotgun metagenomic sequencing of fecal samples from 99 immune thrombocytopenia(ITP)patients and 52 healthy controls.Dysbiosis in the gut microbiome of ITP was detected phylogenetically and functionally,and classifier based on species markers distinguished individuals with ITP from healthy controls.In particular,the abundance of Ruminococcus gnavus,Bifidobacterium longum and Akkermansia muciniphila was markedly increased in treatment-na?ve ITP patients,and the alterations of microbial species were correlated with clinical indices.Functionally,the secondary bile acid biosynthesis and flagellar assembly were depleted in the gut microbiota of ITP,which may contribute to the onset of ITP by affecting the immune system.Furthermore,we found that corticosteroid treatment affected the gut microbiome of ITP.Compared with corticosteroid-sensitive ITP patients,we identified that the corticosteroid-resistant ITP patients displayed a distinct gut microbiome,which was different from that of the treatment-na?ve ITP patients.Together,we provided support for the critical role of gut microbiota in the development of ITP and established a foundation for further research characterizing gut microbiota in relation to corticosteroid resistance of ITP.Yanan Wang Fengqi Liu Gaochao Zhang Yan Su Xueyan Sun Qi Chen Chencong Wang Haixia Fu Yun He Xiaolu Zhu Xiao Liu Meng Lv Xiangyu Zhao Xiaosu Zhao Yueying Li Qianfei Wang Xiaojun Huang Xiaohui Zhang 2021Science China(Life Sciences)2021,64,5:6
2A survey analysis of heavy metals bio-accumulation in internal organs of sea shell animals affected by the sustainable pollution of antifouling paints used for ships anchored at some domestic maritime spaces显示文摘Some samples of sea shell animals stuck and multiplied on the bottom (beneath the seawater) coated with antifouling paints were collected at some domestic maritime spaces, and the content of heavy metals was detected through Inductively Coupled Plasma-Mass Spectroscopy. Meanwhile, comparison with sea shell animals was made on market for edible use. It shows that the content of heavy metals in internal organs of these marine animals is very high due to the large amount of copper and zinc con- tained in the antifouling paints, and this also does severely harm to sea environment and ecology. 3Broad area quantum cascade lasers operating in pulsed mode above 100℃ at λ ~ 4.7 μm显示文摘We demonstrate a broad area(400 μm) high power quantum cascade laser(QCL). A total peak power of 62 W operating at room temperature is achieved at λ~ 4.7 μm. The temperature dependence of the peak power characteristic is given in the experiment, and also the temperature of the active zone is simulated by a finite-elementmethod(FEM). 9MIIP inhibits clear cell renal cell carcinoma proliferation and angiogenesis via negative modulation of the HIF-2α-CYR61 axis显示文摘Objective:In various cancers,migration and invasion inhibitory protein(MIIP)is expressed at low level and is involved in cancer pathogenesis.Herein,we sought to explore the function of MIIP in clear cell renal cell carcinoma(ccRCC).Methods:CCK-8,colony formation,cell cycle,and endothelial cell tube formation assays were performed to evaluate the roles of MIIP in ccRCC proliferation and angiogenesis.To explore the underlyi ng mechanism,we con ducted RNA-sequencing,GSEA,qRT-PCR,Western blot,ELISA,cell transfection,coimmunoprecipitation,and ubiquitination assays in ccRCC cell lines.Furthermore,xenograft tumor growth in nude mice,and Ki-67 and CD31 staining in xenograft tissues were examined.Finally,the association of MIIP expression with clinical pathology and the expression status of HIF-2a and cysteine-rich 61(CYR61)were further analyzed in human RCC tissues through Western blot and immunohistochemistry.Results:Both in vitro and in vivo functional experiments indicated that forced expression of MIIP inhibited ccRCC proliferation and angiogenesis,whereas silencing MIIP either in normal HK-2 cells or in ccRCC cells had the opposite effect(P<0.05).Mechanistically,CYR61 was identified as a gene significantly downregulated by MIIP overexpression,and was required for the suppressive role of MIIP in ccRCC.MIIP was found to promote HSP90 acetylation and thus impair its chaperone function toward HIF-2a.Consequently,RACK1 binds HIF-2a and causes its ubiquitination and proteasomal degradation,thus decreasing the transcription of its target,CYR61.Finally,analyses of clinical samples demonstrated that MIIP is significantly downregulated in cancer vs.normal tissues in RCC cases,and its expression is negatively associated with histological grade,metastasis,the prognosis of patients with RCC,and the expression of HIF-2a and CYR61(P<0.05).Conclusions:MIIP is a novel tumor suppressor in ccRCC via negative regulation of HIF-2a-CYR61 axis.Fengqi Yan Qinhao Wang Mingyuan Xia Yi Ru Wei Hu Guang Yan Xin Xiong Mei Zhang Jiancai Wang Qi Li Bo Zhang He Wang Wei Lin Guojun Wu Xia Li 2022Cancer Biology & Medicine2022,19,6:0

11Tunable optical topological transitions of plasmon polaritons in WTe_(2) van der Waals films显示文摘Naturally existing in-plane hyperbolic polaritons and the associated optical topological transitions,which avoid the nano-structuring to achieve hyperbolicity,can outperform their counterparts in artificial metasurfaces.Such plasmon polaritons are rare,but experimentally revealed recently in WTe_(2)van der Waals thin films.Different from phonon polaritons,hyperbolic plasmon polaritons originate from the interplay of free carrier Drude response and interband transitions,which promise good intrinsic tunability.However,tunable in-plane hyperbolic plasmon polariton and its optical topological transition of the isofrequency contours to the elliptic topology in a natural material have not been realized.Here we demonstrate the tuning of the optical topological transition through Mo doping and temperature.The optical topological transition energy is tuned over a wide range,with frequencies ranging from 429 cm^(−1)(23.3 microns)for pure WTe_(2)to 270 cm^(−1)(37.0 microns)at the 50%Mo-doping level at 10 K.Moreover,the temperature-induced blueshift of the optical topological transition energy is also revealed,enabling active and reversible tuning.Surprisingly,the localized surface plasmon resonance in skew ribbons shows unusual polarization dependence,accurately manifesting its topology,which renders a reliable means to track the topology with far-field techniques.Our results open an avenue for reconfigurable photonic devices capable of plasmon polariton steering,such as canaling,focusing,and routing,and pave the way for low-symmetry plasmonic nanophotonics based on anisotropic natural materials.Yuangang Xie Chong Wang Fucong Fei Yuqi Li Qiaoxia Xing Shenyang Huang Yuchen Lei Jiasheng Zhang Lei Mu Yaomin Dai Fengqi Song Hugen Yan 2023Light(Science & Applications)2023,12,9:0
