1重度肥胖对肺功能及运动试验中呼吸模式的影响显示文摘目的:研究重度肥胖对肺功能及运动试验中呼吸模式的影响。方法:42名女性受试者,正常体重组20名,重度肥胖组22名,进行肺功能及功率车运动试验测定。结果:肥胖组补呼气量、功能残气量、残气量及肺总量均比对照组显著降低(P<0.05)。肥胖组静息状态、无氧阈状态和极量运动状态的氧耗量均比对照组显著增高(P<0.05),而公斤氧耗量均比对照组显著降低(P<0.01)。无氧阈状态和极量运动状态时,肥胖组的分钟通气量、呼吸频率、生理死腔与潮气量的比值以及氧的通气当量均高于对照组,而潮气量均小子对照组,差异有显著性(P〈0.05)。结论:重度肥胖可使肺容量减少、运动中的组织供氧能力及胸壁顺应性、肺顺应性下降,呈现浅快呼吸模式。李靖 李时悦 Feners RJ Buffington CK Cowan GSM 2000新医学2000,31,9:13
2Grade classification of neuroepithelial tumors using high-resolution magic-angle spinning proton nuclear magnetic resonance spectroscopy and pattern recognition显示文摘Clinical data have shown that survival rates vary considerably among brain tumor patients,according to the type and grade of the tumor.Metabolite profiles of intact tumor tissues measured with high-resolution magic-angle spinning proton nuclear magnetic resonance spectroscopy (HRMAS 1H NMRS) can provide important information on tumor biology and metabolism.These metabolic fingerprints can then be used for tumor classification and grading,with great potential value for tumor diagnosis.We studied the metabolic characteristics of 30 neuroepithelial tumor biopsies,including two astrocytomas (grade I),12 astrocytomas (grade II),eight anaplastic astrocytomas (grade III),three glioblastomas (grade IV) and five medulloblastomas (grade IV) from 30 patients using HRMAS 1H NMRS.The results were correlated with pathological features using multivariate data analysis,including principal component analysis (PCA).There were significant differences in the levels of N-acetyl-aspartate (NAA),creatine,myo-inositol,glycine and lactate between tumors of different grades (P<0.05).There were also significant differences in the ratios of NAA/creatine,lactate/creatine,myo-inositol/creatine,glycine/creatine,scyllo-inositol/creatine and alanine/creatine (P<0.05).A soft independent modeling of class analogy model produced a predictive accuracy of 87% for high-grade (grade III-IV) brain tumors with a sensitivity of 87% and a specificity of 93%.HRMAS 1H NMR spectroscopy in conjunction with pattern recognition thus provides a potentially useful tool for the rapid and accurate classification of human brain tumor grades.CHEN WenXue LOU HaiYan ZHANG HongPing NIE Xiu LAN WenXian YANG YongXia XIANG Yun QI JianPin LEI Hao TANG HuiRu CHEN FenEr DENG Feng 2011Science China(Life Sciences)2011,54,7:5
3Development of fluorine-substituted NH_(2)-biphenyl-diarylpyrimidines as highly potent non-nucleoside reverse transcriptase inhibitors:Boosting the safety and metabolic stability显示文摘Our recent studies for nonnucleoside reverse transcriptase inhibitors identified a highly potent compound JK-4b against WT HIV-1(EC_(50)=1.0 nmol/L),but the poor metabolic stability in human liver microsomes(t_(1/2)=14.6 min)and insufficient selectivity(SI=2059)with high cytotoxicity(CC_(50)=2.08μmol/L)remained major issues associated with JK-4b.The present efforts were devoted to the introduction of fluorine into the biphenyl ring of JK-4b,leading to the discovery of a novel series of fluorine-substituted NH_(2)-biphenyl-diarylpyrimidines with noticeable inhibitory activity toward WT HIV-1 strain(EC_(50)=1.8–349 nmol/L).The best compound 5t in this collection(EC_(50)=1.8 nmol/L,CC_(50)=117μmol/L)was 32-fold in selectivity(SI=66,443)compared to JK-4b and showed remarkable potency toward clinically multiple mutant strains,such as L100I,K103N,E138K,and Y181C.The metabolic stability of 5t was also significantly improved(t_(1/2)=74.52 min),approximately 5-fold higher than JK-4b in human liver microsomes(t_(1/2)=14.6 min).Also,5t possessed good stability in both human and monkey plasma.No significant in vitro inhibition effect toward CYP enzyme and hERG was observed.The single-dose acute toxicity test did not induce mice death or obvious pathological damage.These findings pave the way for further development of 5t as a drug candidate.Xin Jin Shuai Wang Limin Zhao Wenjuan Huang Yinxiang Zhang Christophe Pannecouque Erik De Clercq Ge Meng Huri Piao Fener Chen 2023Acta Pharmaceutica Sinica B2023,13,3:1

14Revisiting the Bjerrum's correction factor:Use of the liquidity index for assessing the effect of soil plasticity on undrained shear strength显示文摘The undrained shear strength(s_u) of fine-grained soils that can be measured in situ and in laboratory is one of the key geotechnical parameters.The unconfined compression test(UCT) is widely used in laboratory to measure this parameter due to its simplicity;however,it is severely affected by sample disturbance.The vane shear test(VST) technique that is less sensitive to sample disturbance involves a correction factor against the soil plasticity,commonly known as the Bjerrum's correction factor,μ.This study aims to reevaluate the Bjerrum's correction factor in consideration of a different approach and a relatively new method of testing.Atterberg limits test,miniature VST,and reverse extrusion test(RET)were conducted on 120 remolded samples.The effect of soil plasticity on undrained shear strength was examined using the liquidity index instead of Bjerrum's correction factor.In comparison with the result obatined using the Bjerrum's correction factor,the lindrained shear strength was better represented when s_u values were correlated with the liquidity index.The results were validated by the RET,which was proven to take into account soil plasticity with a reliable degree of accuracy.This study also shows that the RET has strong promise as a new tool for testing undrained shear strength of fine-grained soils.Kamil Kayabali Ozgur Akturk Mustafa Fener Orhan Dikmen Furkan Hamza Harputlugil 2015Journal of Rock Mechanics and Geotechnical Engineering2015,7,6:1
